Effects of Oral Rutaecarpine on the Pharmacokinetics of Intravenous Chlorzoxazone in Rats

  • It has been reported that hepatic microsomal cytochrome P450(CYP) 2E1 is responsible for the metabolism of chlorzoxazone(CZX) to 6-hydroxychlorzoxazone. The present study was undertaken to assess the possible interaction of rutaecarpine, an alkaloid originally isolated from the unripe fruit of Evodia rutaecarpa, with CZX. Male Spraque-Dawley rats were administered with 80 mg/kg/day of oral rutaecarpine for three consecutive days. Twenty four hr after the pre-treatment with rutaecarpine, the rats were treated with 20 mg/kg of intravenous CZX. Rat hepatic microsomes isolated from rutaecarpine-treated rats showed greater(50% increase) activity of p-nitrophenol hydroxylase(a marker of CYP2E1) when compared with the control rats. Compared with control rats, the AUC of CZX was significantly smaller(84% decrease) possibly due to significantly faster CL(646% increase) in rats pretreated with rutaecarpine. This could be, at least partially, due to induction of CYP2E1 by rutaecarpine.

  • Hemoptysis is a common clinical symptom responsible for about 10% of the patients who has the pulmonary disease. In Korea, pulmonary tuberculosis is still the most common cause of the hemoptysis. This study is a chlinical report of Korean Traditional Medical Therapy for the patient of hemopysis with old pulmonary tuberculosis. The patient was suffering from hemoptysis, headache, sweating, feeling of uneasiness and fatigue. After administration of herbal medicines (Bokryungbosim-tang, Yukmijihwangwon-gagambang) and acupunture treatment for 34days, these symptoms of the patient were improved. Thus, we concluded that Oriental Medical Treatment can improve the symptoms of the patient who is suffering from hemoptysis.

Control Effects of an Antibiotic Produced by Streptomyces sp. B25 on Tobacco Mosaic Virus and Determination of Its Molecular Structure

  • The culture filtrate of Streptomyces sp. B25, which was identified in this experiment, was tested for the control of tobacco mosaic virus (TMV) with the susceptible tobacco cultivar, NC 82, under the field conditions following the preliminary examination of its characters for TMV control. Control efficacy of the culture filtrate against TMV infection continued over 50% up to 6 days after treatment, and its systemic effect was about 30% of the direct effect. In field conditions control efficacy of the culture filtrate against TMV infection was 95.3 % at 2 weeks after TMV inoculation, and decreased to 58.3 % at 3 weeks after inoculation. Five fold-dilution of the culture filtrate showed about half of the control efficacy by the stock culture filtrate. Analysis of the antibiotic material responsible for the inhibition of TMV infection through nuclear magnetic resonance experiments revealed that the antibiotic is antimycin $A_1$, which is firstly reported as an anti-phytoviral antibiotic in this experiment.

Role of Kupffer Cells in Vasoregulatory Gene Expression During Endotoxemia

  • Although hepatic microcirculatory dysfunction occurs during endotoxemia, the mechanism responsible for this remains unclear. Since Kupffer cells provide signals that regulate hepatic response in inflammation, this study was designed to investigate the role of Kupffer cells in the imbalance in the expression of vasoactive mediators. Endotoxemia was induced by intraperitoneal E. coli endotoxin (LPS, 1 mg/kg body weight). Kupffer cells were inactivated with gadolinium chloride ($GdCl_3$, 7.5 mg/kg body weight, intravenously) 2 days prior to LPS exposure. Liver samples were taken 6 h following LPS exposure for RT-PCR analysis of mRNA for genes of interest: endothelin (ET-1), its receptors $ET_A$ and $ET_B$, inducible nitric oxide synthase (iNOS), heme oxygenase (HO-1), and tumor necrosis factor-$\alpha$ (TNF-$\alpha$). mRNA levels for iNOS and TNF-$\alpha$ were significantly increased 31.8-fold and 26.7-fold in LPS-treated animals, respectively. This increase was markedly attenuated by $GdCl_3$, HO-1 expression significantly increased in LPS-treated animals, with no significant difference between saline and $GdCl_3$ groups. ET-1 was increased by LPS. mRNA levels for $ET_A$ receptor showed no change, whereas $ET_B$ transcripts increased in LPS-treated animals. The increase in $ET_B$ transcripts was potentiated by $GdCl_3$. We conclude that activation of Kupffer cells plays an important role in the imbalanced hepatic vasoregulatory gene expression induced by endotoxin.

Studied on Enzymes Responsible for Polyamine Level Effects of IAA on the activities of lysine decarboxylase and diamine oxidase and polyamine level in seedlings of soybean (Polyamine 농도에 미치는 효소에 관한 연구 대두의 유묘에서 IAA가 Lysine Decarboxylase와 Diamine Oxidase의 활성과 Polyamine함량에 미치는 영향)

  • The effect of IAA on lysine decarboxylase(LDC)activity, diamine oxidase (DAO) activity and the polyamine level in seedlings of soybean were investigated. The seedlings of soybean were treated with IAA (10-8M, 10-6M, 10-4M) and LDC activity, DAO activity and polyamine levels of cotyledon, elongating hypocotyl, elongated hypocotyl and the root were measured after 4, 6, 8, 10 days. Increased concentration of IAA stimulated the LDC activity and increased the level of polyamine in the seedlings, which reducing the DAO activity and polyamine level was increased in the seedlings. The light also stimulated the IAA effect on polyamine level and DAO activity, and polyamine level was higher in the seedlings grown in the light than those in the dark. Cadaverine was the most abundant polyamine in the seedlings of soybean. The levels of diamines, cadaverine, putrescine were higher in the elongated hypocotyl and root than in the elongating hypocotyl, but the levels of spermidine and spermine were relatively higher in the cotyledon than in the other parts. Accumulation of diamines by high concentration of IAA is postulated to be a phenomenon of inhibition in seedlings growth.

  • Legal issues whether augmented reality(AR) game service companies are responsible for damages that their game users cause to the community and residents are occurring these days. These kind of damages affect not only game users but also third parties who do not play the AR game. This paper explores the recent cases and analyze the type of legal issues such as trespass, nuisance and unjust enrichment. To determine the necessity of regulating the AR game company, constitutional balancing test, causation, specificity and persistence of the damage and other various standards should be applied.

Experimental study on geopolymer concrete prepared using high-silica RHA incorporating alccofine

  • This paper describes the experimental investigation carried out to develop geopolymer concrete using rice husk ash (RHA) along with alccofine. The study reports the fresh and hardened properties of the geopolymer concrete (GPC) activated using alkaline solution. GPC were prepared using different RHA content (350, 375 and $400kg/m^3$), the molarity of the NaOH (8, 12 and 16M). The specimens were cured at $27^{\circ}C$ and $90^{\circ}C$. GPC was activated using NaOH, $Na_2SiO_3$, and alccofine. Prepared GPC samples were tested for compressive and splitting tensile strengths after 3, 7 and 28 days. RHA was suitable to produce geopolymer concrete. Results indicate that behavior of GPC prepared with RHA is similar to fly ash based GPC. Workability and strength can be improved by incorporating the alccofine. Further, alccofine and heat curing improve the early age properties of the GPC. Heat curing is responsible for the initial polymerization of GPC which leads to high workability and improved mechanical properties of the GPC. High strength can be achieved by using the high concentration alkaline solution in terms of molarity and at elevated heat curing. Further, RHA based geopolymer concrete has tremendous potential as a substitute for ordinary concrete.

Animal Model of LPRD (Laryngopharyngeal Reflux Disease) (인후두 위산 역류증의 동물 모형의 개발)

  • Background and Objectives: Complications arising from endotracheal intubation are uncommon but, when they do occur, can be significant. Placement of an endotracheal tube frequently results in trauma to the underlying laryngeal and tracheal tissue, although the trauma is usually reversible. Occasionally, these changes can be of a more permanent nature and result in severe impairment of the airway and/or voice. It is proposed that a common factor-gastroesophageal reflux-might be responsible. This study was performed in order to develop the animal model of LPRD using rats and investigated that LPRD could produce significant damage to larynx especially vocal cords. Materials and Methods : The each four rats were used in the experiment and control study. Each was anesthetized and larynx was exposed and injured in the unilateral aritenoid. Injured site was contact with normal saline(control group) and synthetic gastric juice(experimental group). The larynx was examined after 7days in normal environment. Results : All was survived in the control group and two was survived in the experimental group. In the control group, some inflammation cells was found but in the experimental group, granulation was found. Conclusion : We developed animal model of LPRD using rat and thought LPRD may Play an important role in the development of permanent laryngeal injury.

Skin Toxicity of Toluene on Liver Damage-Induced Rats (간손상 유발실험동물에서 Toluene의 피부독성)

  • To evaluate the cutaneous injury in liver damaged rats by toluene application to the skin, toluene(35mg/㎤) was sequentially applied for 5 days to the dorsal skin of liver damaged rats with $CCl_4$ (6 times ever other day:0.1$m\ell$/100 g body weight-50% $CCl_4$in olive oil). The cutaneous ultrastructural changes were unexoectably not observed in liver from $CCl_4$-treated rats although necrotic liver damage appeared under light microscope. In these animals by the application of toluene to rat skin the cutaneous xanthine oxidase activity was significantly increased(p<0.05), but cytochrome P450 content was not different from that of the control or only $CCl_4$-treated rats. On the other hand, the cutaneous superoxide dismutase and catalase activities in liver damaged animals were significantly respectively(p<0.05, p<0.001), decreased by toluene application to the skin compared with control and especially the former enzyme activity was significantty decreased(p<0.01), compared with that of liver damaged rate rat but glutathione peroxidase, glutathione-S-transferase activities were not significantly different from those of the control or liver damaged rats. Futhermore, the reduced gluathione content of skin was also significantly decreased by toluene application to the liver damaged animals. In conclusion, the great deposits of cerrous peroxide and ultramorphological changes in skin tissue of liver damaged animals by toluene application may be responsible for the oxygen free radical.

Spinal Nerve Root Compression by Acute Inflammatory Granuloma after Spine Surgery -A case report- (척추수술 후 급성 염증성 육아조직 형성으로 인한 신경근 압박 -증례 보고-)

  • This report describes a case of spinal nerve root compression due to an acute inflammatory granuloma after lumbar surgery. A 39 year-old man with a history of increasing back pain with a 3-week duration was diagnosed with a herniated intervertebral disc (HIVD). The diagnosis of a HIVD was confirmed by magnetic resonance imaging (MRI) with indications for surgery. A discectomy and a partial laminectomy was performed and the symptoms were alleviated immediately after surgery for a five-day period. However, a slowly progressing pain was subsequently noted along a different dermatome. There was no pain relief despite the patient being given pharmacological treatments, combined with an epidural steroid injection. The follow up MRI images showed severe compression of the nerve roots by a epidural lesion. Another procedure was performed 17 days after the initial operation. The lesion responsible for the compression of the nerve roots was found to be an acute inflammatory granuloma. The pain was relieved after the second procedure and there were no other symptoms or neurological problems. This case is remarkable in that a granuloma formed relatively quickly and grew to such a size that it was able to severely compress the surrounding nerve roots.
