A New Method for Antimicrobial Susceptibility Testing of Vitro-cultured Bacteria by Means of Resonance Light Scattering Technique

  • A new method for antimicrobial susceptibility testing of vitro-cultured bacteria on an ordinary fluorescence spectrometer was developed. The viable bacteria reduced 3-(4,5 dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) to produce insoluble particles that displayed intense resonance scattering light. The assay showed a linear relationship between the number of viable bacteria and the intensity of resonance scattering light. Dead bacteria were unable to reduce MTT. Methicillin-resistant Staphylococcus aureus exposed to flavonoids from Marchantia convoluta showed a flavonoids concentration-dependent inhibition of the ability to reduce MTT. In the assay, less than 12 h was required to attain susceptibility results and fewer bacteria were utilized than in traditional methods. The RLS technique could, in combination with the MTT assay, be a rapid and sensitive measuring method to determine the in vitro activity of new antimicrobials.

  • The spectra of scattering light fromlaser-produced plasma near its fundamental and second harmonic wavelength were observed respectively by means of spectroscopic technique. The experimental results and the generation mechanism of nonlinear effects such as the second garmonics and the brillouin scattering were analysed theoretically. The spectra of reflected laser light became wider than that of incident laser light. And the peak of spectrum of reflected light shifted to red-side from that of incident light. The second harmonic light is generated from the nonlinear interaction of the incident laser light and the electron plasma wave excited in resonance region by the oblique incidence of laser light to the plasma. The Brillouin backscattering from laser-produced plasmas of hydrogen and deuterium has shown an isotope effect in the red-side region of the generated second harmonic light. This isotope shift is explained by the parametric instability at the cutoff (resonance) region using frequency-and phase-matching conditions of the waves.

Advantage of the Intensive Light Scattering by Plasmonic Nanoparticles in Velocimetry

  • Tracers are one of the critical factors for improving the performance of velocimetry. Silver and gold nanoparticles as tracers with localized surface-plasmon resonance are analyzed for their scattering properties. The scattering cross sections, angular distribution of the scattering, and equivalent scattering cross sections from 53° and 1.5° half-angle cones at 532 nm are calculated, with particle sizes in the nanoscale range. The 53° and 1.5° half-angle cones used as examples correspond respectively to the collection cones for microscope objectives in microscopic measurements and camera lenses in macroscopic measurements. We find that there is a transitional size near 35 nm when comparing the equivalent scattering cross sections between silver and gold nanoparticles in water at 532 nm. The equivalent scattering cross section of silver nanoparticles is greater or smaller than that of gold nanoparticles when the particle radius is greater or smaller than 35 nm respectively. When the radius of the plasmonic nanoparticles is smaller than about 44 nm, their equivalent scattering cross sections are at least ten times that of TiO2 nanoparticles. Plasmonic nanoparticles are promising for velocimetry applications.

Velocity measurements in complex flows of non-Newtonian fluids

  • Experimental methods for making quantitative measurements of velocity fields in non-Newtonian fluids are reviewed. Techniques based on light scattering spectroscopy - laser Doppler velocimetry and homodyne light scattering spectroscopy, techniques based on imaging the displacement of markers - including particle image velocimetry and molecular tagging velocimetry, and techniques based on nuclear magnetic resonance imaging are discussed. The special advantages and disadvantages of each method are summarized, and their applications to non-Newtonian flows are briefly reviewed. Example data from each technique are also included.

Effect of Silver Nanoparticles with Indium Tin Oxide Thin Layers on Silicon Solar Cells

  • AThe effect of localized surface plasmon on silicon substrates was studied using silver nanoparticles. The nanoparticles were formed by self-arrangement through the surface energy using rapid thermal annealing (RTA) technique after the thin nanolayer of silver was deposited by thermal evaporation. By the theoretical calculation based on Mie scattering and dielectric function of air, indium tin oxide (ITO), and silver, the strong peak of scattering cross section of silver nanoparticles was found at 358 nm for air, and 460 nm for ITO, respectively. Accordingly, the strong suppression of reflectance under the condition of induced light of $30^{\circ}$ occurred at the specific wavelength which is almost in accordance with peak of scattering cross section. When the external quantum efficiency was measured using silicon solar cells with silver nanoparticles, there was small enhancement peak near the 460 nm wavelength in which the light was resonated between silver nanoparticles and ITO.

Light transmission in nanostructures

  • We investigate transmission of light in nanoscale structures. We present spatial and temporal domain measurements of the dephasing of surface plasmon excitations in metal films with periodic nano-hole arrays. By probing coherent spatial SP propagation lengths of a few f1. $\mu$m and an ultrafast decay of the SP polarization on a 10 fs timescale, we demonstrate that the SP transmission peaks are homogeneously broadened by the SP radiative lifetime. The pronounced wavelength and hole size dependence of the dephasing rate shows that the microscopic origin of the conversion of SP into light is a Rayleigh-like scattering by the periodic hole array. We have experimentally studied the dephasing of surface plasmon excitations in metallic nano-hole arrays. By relating nanoscopic SP propagation, ultrafast light transmission and optical spectra, we demonstrate that the transmission spectra of these plasmonic bandgap structures are homogeneously broadened. The spectral line shape and dephasing time are dominated by Rayleigh scattering of SP into light and can varied over a wide range by controlling the resonance energy and/or hole radius. This opens the way towards designing SP nano-optic devices and spatially and spectrally tailoring light -matter interactions on nanometer length scales.

[Ru(phen)2(dppz)]2+ Assemble on the Surface of the SDS Micelle and Its Application for the Determination of DNA

  • The solution of $[Ru(phen)_2(dppz)]^{2+}$ and SDS has high Resonance Light Scattering (RLS) signals due to $[Ru(phen)_2(dppz)]^{2+}$ assemble on the surface of the SDS micelle. Because of the high affinity ($KB\geq10^6\;L\;mol^{-1}$) between $[Ru(phen)_2(dppz)]^{2+}$ and DNA, the adding of DNA in the solution of $[Ru(phen)_2(dppz)]^{2+}$-SDS makes the dissociation of $[Ru(phen)_2(dppz)]^{2+}$-SDS, and results in decreasing of the RLS signals and increasing of the absorbance. Based on this, a novel method is proposed for DNA assay. Under optimum condition, good linear relationship was obtained within the concentration range of 0.018-1.26 $\mu g\;mL^{-1}$, the linear equation is $I_{RLS}$ = 504.8-348.8 c (c: $\mu g\;mL^{-1}$) and the correlation coefficient (r) is 0.9992. The detect limit for calf thymus DNA is 8.6 ng $mL^{-1}$. The proposed method was successful applied to determine the extracted colibacillus plasmid DNA.

  • Quantitative analysis of optical scattering intensities from a Au nanoparticle with a diameter of 100 nm, which is effected by the localized surface plasmon resonance (LSPR), were numerically carried out by using a dark-field detection scheme on prism basal plane for two different beam incident modes of reflectance (R-mode) and transmittance (T-mode). Two-dimensional finite difference time domain (FDTD) algorithm was adopted, and its applicabilibility was verified by comparing the simulation results with the theoretical ones. Simulation results of the scattered light intensities from a Au nanoparticle revealed that the scattered intensity of the T-mode was much stronger than that of R-mode. Comparison of the calculated results with the theoretical intensity distribution on the prism showed that the scattered intensity is marimized when the evanescent field, which is generated from the interface of prism and air at TIR angle, is coupled with Au nanoparticle.

  • In this study, we synthesize silica-core gold-satellite nanoparticles (SGNPs) for the surface-enhanced Raman scattering (SERS) based sensing applications. They consist of gold satellite nanoparticles (AuNPs) fixed on the silica core nanoparticles, which sizes of AuNPs can be tunned by varying the amount of reactants (growth solution and reducing agent). Their surface plasmon resonance (SPR) properties were characterized by using UV-vis spectroscopy, showing that the growth of AuNPs on silica cores leads to the light absorption in the longer wavelength region. Furthermore, the size increase of AuNPs exhibited the dramatic change in SERS activity due to the formation of hot spots. The optimized SGNPs showing enhancement factor ${\sim}3.8{\times}10^6$ exhibited a detection limit of rhodamine 6G (R6G) as low as $10^{-8}M$. These findings suggest the importance of size control of SGNPs and their SPR properties to develop highly efficient SERS sensors.
