개 난자의 체외성숙에 미치는 호르몬과 Na-Pyruvate의 영향 (Effects of Hormone and Na-Pyruvate on the In Vitro Maturation of Canine Oocytes)

  • 본 연구는 호르몬과 Na-pyruvate의 첨가가 개 미성숙난자의 체외성숙에 미치는 영향을 검토하였다. 개의 발정주기와 관계없이 암캐의 난소에서 회수한 난자를 NCSU-37 배양액에서 72시간 체외 배양하였다. 호르몬의 영향을 검토하기 위하여 배양액 중에 다양한 호르몬을 $24{\sim}72$시간 첨가하여 성숙율을 검사하였으며, Na-pyruvate 첨가 농도의 영향을 검토하였다. $E_2$ 단독 첨가구에서는 제2감수분열 중기(MII) 단계까지 성숙이 관찰되었으나(5.7%), 타 처리구에서는 MII기까지의 성숙이 이루어지지 않았다(P<0.05), $E_2$$6{\sim}24$시간만 처리하였을 때는 MII기로 성숙되는 난자가 없었으나 72시간 처리하였을 때는 6.0%로 유의적으로 높게 나타났다(P<0.05). Na-pyruvate의 농도를 0.25 mM 첨가한 구보다 2.5 mM 첨가한 구에서 제1감수분열 중기(MI) 단계까지의 성숙율이 증가하는 경향을 보였으나, MII 단계까지의 성숙율은 차이가 없었다. 본 실험의 결과는 개 난자의 체외성숙시 $E_2$가 첨가된 배양액에서 72시간 배양시에 성숙율을 증진시킬 수 있으나, Na-pyruvate의 농도는 개 체외배양에 있어 큰 영향을 미치지 않는 것을 나타낸다.

Loss of Function in GIGANTEA Gene is Involved in Brassinosteroid Signaling

  • Brassinosteroids (BRs) are plant steroid hormones that play essential roles in growth and development. Mutations in BR-signaling pathways cause defective in growth and development like dwarfism, male sterility, abnormal vascular development and photomorphogenesis. Transition from vegetative to reproductive growth is a critical phase change in the development of a flowering plant. In a screen of activation-tagged Arabidopsis, we identified a mutant named abz126 that displayed longer hypocotyls when grown in the dark on MS media containing brassinazole (Brz), an inhibitor of BRs biosynthesis. We have cloned the mutant locus using adapter ligation PCR walking and identified that a single T-DNA had been integrated into the ninth exon of the GIGANTEA (GI) gene, involved in controling flowering time. This insertion resulted in loss-of-function of the GI gene and caused the following phenotypes: long petioles, tall plant height, many rosette leaves and late flowering. RT-PCR assays on abz126 mutant showed that the T-DNA insertion in GIGANTEA led to the loss of mRNA expression of the GI gene. In the hormone dose response assay, abz126 mutant showed: 1) an insensitivity to paclobutrazole (PAC), 2) an altered response with 6-benzylaminopurine (BAP) and 3) insensitive to Brassinolide (BL). Based on these results, we propose that the late flowering and tall phenotypes displayed by the abz126 mutant are caused by a loss-of-function of the GI gene associated with brassinosteroid hormone signaling.

Differential Expression Analysis of Candidate Genes Related with Growth according to Dietary Supplementation of Curcuma longa in Chickens

  • This experiment was conducted to investigate the genetic effects of candidate genes on the growth of spleen and liver tissues using dietary Curcuma longa (C. longa) supplementation. Expression analyses of candidate genes regarding animal growth was performed in order to determine the factors affecting the growth related to immune components of Curucumin, Turmerone, and Zingiberene as the bile secretion Paratolyl methyl carbinol (PTMC). The animals were divided into four groups of five chicks supplied with experimental diets of C. longa at 0.25, 0.5 and 1% and controls. The 19 growth-related genes were known to cell maturation, differentiation significant expression patterns in this analysis. Expression of growth response-related genes in chicks supplemented with 1% of C. longa showed better growth performance than chicks with 0.25 and 0.5% in spleen (p<0.05). The IGF1, MSTN, POU1F1, ADCYAP1 gene were known to central roles in mediating gonadotropin function, regulating steroidogenesis and promoting oocyte growth and maturation. Sex steroids, androgen and estrogen can affect sex differentiation and also can affect muscle development. On the other hand, GHSR and FABP3 gene showed significant expression patterns in this analysis. The results would be used as basic information for the variation of growth-related genes expression on the cell growth, sex cell growth, and sex hormones according to dietary supplementation with C. longa in chickens.

The Cellular Localization of GnRH and LHR in Aged Female Mice

  • Gonadotropin releasing hormone (GnRH) centrally plays a role in control of the hypothalamic-pituitary-gonadal axis-related hormone secretions in the reproductive neuroendocrine system. In addition, hormone receptors like luteinizing hormone receptor (LHR) are important element for hormones to take effect in target organ. However, ageing-dependent changes in terms of the distribution of GnRH neurons in the brain and LHR expression in the acyclic ovary have not been fully understood yet. Therefore, we comparatively investigated those ageing-dependent changes using young (1-5 months), middle (11-14 months) and old (21-27 months) aged female mice. Whereas a number of GnRH positive fibers and neurons with monopolar or bipolar morphology were abundantly observed in the brain of the young and middle aged mice, a few GnRH positive neurons with multiple dendrites were observed in the old aged mice. In addition, acyclic ovary without repeated development and degeneration of the follicles was shown in the old aged mice than others. LHR expression was localized in theca cells, granulosa cell, corpora lutea and atretic follicle in the ovaries from young and middle aged mice, in contrast, old aged mice had few positive LHR expression on the follicles due to acyclic ovary. However, the whole protein level of LHR was higher in the ovary of old aged mice than others. These results are expected to be used as an important basis on the relationship between GnRH and LHR in old aged animals as well as in further research for reproduction failure.

Effects of exogenous hormones treatment on spermiation and plasma levels of gonadal steroids in Roughscale sole, Clidoderma asperrimum

  • Roughscale sole (Clidoderma asperrimum) is only wild caught because basic reproductive research on this species is lacking and gamete production in an artificial setting has not been successful. Exogenous hormone treatment has been used to induce gonadal maturation and final spermiation in wild-caught individuals. In this study, the effects of an exogenous hormone on spermiation in roughscale sole was investigated by implanting different concentrations of a salmon gonadotrophin-releasing hormone analog (sGnRH; 0, 25, 50, and 100 ㎍/kg body weight) into male fishes. The control group did not produce sperm after 21 days post-implantation, and the duration of spermiation was shorter compared to the other groups. The spermiation period and milt amount differed among the hormone-treated groups according to the hormone concentration used. Milt volumes in the groups treated with 25 and 100 ㎍/kg sGnRH increased compared to the control group, whereas exogenous hormone treatment had no effect on the movable sperm ratio. The spermatocrit was high at the beginning of spermiation in all groups and then tended to decrease gradually over time except in the experimental group treated with 100 ㎍/kg sGnRH. Plasma levels of testosterone, 11-ketotestosterone, 17α, 20β-dihydroxy-4-pregnen-3-one were not significantly affected by the sGnRH treatments. Our results demonstrate that it is possible to prolong the spermiation period and increase milt volume by treating male roughscale soles with an exogenous hormone. In addition, the artificial hormone treatment did not affect sperm motility.

Heat stress during summer reduced the ovarian aromatase expression of sows in Korea

  • It has been addressed that heat stress due to high atmospheric temperature during summer in Korea induces impaired release of reproductive hormones, followed by occurring abnormal ovarian cyclicity, lower pregnancy ratio, and reduced litter size. Therefore, the present study attempted to compare seasonal change (spring versus summer) of the ovarian aromatase expression, an enzyme for converting testosterone into estrogen. While serum estrogen level in summer group was significantly lower than that of spring group, testosterone was not different between groups. Consistent with estrogen level, the ovarian aromatase expression in summer at follicular phase was significantly lower than the counterpart of spring. The ovarian aromatase expression was positively related with serum estrogen level significantly (r=0.689; P=0.008) and strongly negative correlation was identified (r=-0.533; P=0.078) with atmospheric temperature. The ovarian aromatase expression was not detected in immature ovarian follicles but specifically localized in the granulosa cell layers in both seasons. However, the aromatase intensity in the granulosa cell layers was stronger in spring than summer. Because testosterone level was not different between groups, it could be concluded that the lower level of estrogen during summer might be derived by not lack of substrate but lower expression of ovarian aromatase by heat stress.

Temporal Changes in the Local Expression of Central Hormone-Regulating Factors in Rat Testis

  • Present study aimed to investigate the temporal changes in expression of some reproductive hormones in testis, originally found in hypothalamus and pituitary. Rats were sacrificed on postnatal day 23 (PND23; immature), pubertal (PND53) and PND 81 (young adult). The testicular RNAs were extracted, and semi-quantitative PCRs for gonadotropin-releasing hormone (GnRH), kisspeptin 1 (KiSS1), pituitary adenylate cyclase-activating polypeptide (PACAP), LH subunits and LH receptor were performed. Transcript levels of GnRH and KiSS1 at PND23 were significantly higher than levels of PND53 and PND81 (p<0.001). PACAP mRNA level at PND23 was significantly lower than those of PND53 and PND81 (p<0.001). The mRNA levels of both testis type and pituitary type luteinizing hormone β subunit (tLHβ and pLHβ, respectively) at PND23 were significantly lower than levels of PND53 and PND81 (p<0.001). The mRNA level of glycoprotein hormone common alpha subunit (Cgα) at PND23 was significantly lower than those of PND53 and PND81 (p<0.001). Present study revealed the intratesticular expression of KiSS1 and GnRH showed a very similar trend while the expression of PACAP in the testis showed reversed pattern. The expressions of LHβ subunits (tLHβ and pLHβ) were very low during immature stage then increased significantly during puberty and early adulthood. Our attempt to study the local role(s) of intratesticular factors will be helpful to achieve precise understanding on the testis physiology and pathology.

인공수정 후 외인성 Progesterone과 Estradiol 병용 및 Progesterone 단독 투여 기간이 젖소의 번식 성적에 미치는 영향 (Effect of Duration Treated on Reproductive Performance in Holstein Cows Treated with Progesterone with and without Estradiol)

  • 본 연구는 젖소에 있어서 인공수정 후 외인성 progesterone과 estradiol을 병용 투여하거나 progesterone을 단독 투여하는 경우에 처리기간별 젖소의 번식 성적에 미치는 영향을 조사하기 위하여 대조군, CIDR에 estradiol 캡슐을 부착시킨 progesterone과 estradiol 병용 투여군(PE군) 및 CIDR에서 estradiol 캡슐을 제거한 progesterone 단독 투여군(P군)로 구분하여, 인공수정 후 14일째에 각 투여군에 따른 CIDR를 질내에 삽입하여 그 날로부터 7일째에 제거하였고(P-7), 5일구의 경우, 인공수정 후 16일째에 CIDR를 질내에 삽입하여 그 날로부터 5일째에 제거하였으며(P-5), 3일구의 경우, 인공수정 후 18일째에 CIDR를 질내에 삽입하여 그 날로부터 3일째에 제거하는 경우에(P-3) 첫 수정시 수태율, 인공수정 후 200일 동안 수태까지의 일수 및 수태당 종부 횟수를 조사하여 다음과 같은 결과를 얻었다. 1. P4군 및 P4+E2군에서 처리기간별 첫 수정시 수태율은 대조구의 경우 15.8%였고, P-7구, P-5구 및 P-3구의 경우, 각각 40.0%, 37.5% 및 28.6%였으며, PE-7구, PE-5구 및 PE-3구의 경우, 각각 62.5%, 50.0% 및 0%였다. 2. P4군 및 P4+E2군에서 처리 기간별 인공수정 후 수태까지의 일수는 대조구의 경우 64.9일이었고, P-7구, P-5구 및 P-3구의 경우, 각각 63.0일, 59.1일 및 8.0일이었으며, PE-7구, PE-5구 및 PE-3구의 경우, 각각 0.0일, 18.9일 및 83.3일로 PE-7구가 대조구 및 PE-3구에 비하여 유의적(p<0.05)으로 단축되는 경향을 나타내었다. 3. P4군 및 P4+E2군에서 처리 기간별 수태당 종부 횟수는 대조구의 경우 2.7회였고, P-7구, P-5구 및 P-3구의 경우, 각각 2.3회, 1.9회 및 1.3회였으며, PE-7구, PE-5구 및 PE-3구의 경우, 각각 1.0회, 1.7회 및 2.8회로 PE-7구가 대조구 및 PE-3구에 비하여 유의적(p<0.05)으로 감소는 경향을 나타내었다.

인공수정 후 외인성 Progesterone과 Estradiol 병용 투여 및 Progesterone 단독 투여가 젖소의 번식 성적에 미치는 영향 (Reproductive Performance of Holstein Cattle Treated with Progesterone and Combination of Progesterone and Estradiol)

  • 본 연구는 젖소에 있어서 인공수정 후 외인성 progesterone과 estradiol을 병용 투여하거나 progesterone을 단독 투여하는 경우에 젖소의 번식 성적에 미치는 영향을 조사하기 위하여 대조군, CIDR에서 estradiol 캡슐을 부착시킨 progesterone과 estradiol 병용 투여군(P4+E2군) 및 CIDR에서 estradiol 캡슐을 제거한 progesterone 단독 투여군(P4군)로 구분하여, 인공수정 후 14일째에 질내에 삽입한 후 삽입한 날로부터 7일째에 제거하였다. 수태된 개체에 대해서는 수태율을 분석하였고, 수태되지 않은 개체에 대해서는 수정 후 수태까지의 일수, 수태당종부 횟수, 수태가 되지 않은 경우의 차기 발정에 따른 발정강도 및 CIDR 삽입 기간 동안 및 제거한 후의 혈중 progesterone 농도를 조사하여 다음과 같은 결과를 얻었다. 1. P4+E2군 및 P4군에 따른 수태율에 있어서는 대조군의 경우 15.4%였고, P4+E2군의 경우 60.0% 및 P4군 38.9%로 P4+E2군의 수태율이 가장 높은 경향을 나타내었다. 2. 혈청 중 P4 농도 측정을 위하여 처리 후 0일째(발정 주기 14일째), 2일째(발정 주기 15일째), 3일째(발정주기 17일째), 6일째(발정 주기 20일째), 8일째(발정 주기 1일째), 9일째(발정 주기 2일째), 10일째(발정 주기 3일째), 13일째(발정 주기 6일째)에 채혈하여 측정한 결과, 처리 후 8일째(발정주기 1일째), 9일째(발정주기 2일째) 및 13일째 (발정 주기 6일째)에는 P4+E2군이 대조군 비임신군 및 대조군 임신군에 비하여 유의적으로 높은 경향을 나타내었다.(p<0.05).

자궁근종과 성호르몬 대사물과의 연관성 (Correlation between Steroid Hormone Metabolites and Leiomyomas of Uterus)

  • Objective: To elucidate 1) whether there are any differences in the urine concentrations of steroid hormone metabolites between patients with leiomyoma and normal controls 2) the correlation between urinary profiles of steroid hormones and leiomyomas of the uterus according to their type, location, volume, and weight. Materials of Methods : The study population consisted of 37 premenopausal patients with uterine leiomyoma and the control group consisted of 25 premenopausal normal volunteer women without uterine leiomyoma. Confirmation of the existence of uterine leiomyoma was done by ultrasonography and histopathological examination after surgery. The volume of the leiomyoma was estimated by trans-abdominal and/or trans-vaginal ultrasonography. The Leiomyomas were divided into 3 types (subserosal, intramural and submucosal). Seventeen patients had subserosal type of leiomyoma, 10 with the intramural type and 10 with the submucosal type. The locations of the leiomyoma were also divided into 3 groups (fundus, body and isthmus). Seventeen patients showed a fundus location, 10 in body, and 10 in isthmus. We compared urinary profiles of the endogenous steroids between patients with leiomyomas and normal controls, and also investigated the relationship between urinary profiles of the endogenous steroids and leiomyomas according to their type, location, volume and weight by using highly sensitive Gas Chromatography-Mass Spectrometry (GC-MS) system. Results: The mean ages of the patients with leiomyomas and the control group were $43.1{\pm}5.6$ and $40.6{\pm}7.2$ years, the weights were $63.4{\pm}7.3$ and $59.4{\pm}8.1\;kg$, and their heights were $155.4{\pm}4.8$ and $159.3{\pm}4.8\;cm$ respectively. Seventeen patients had subserosal, 10 had intramural, and 10 had submucosal leiomyomas. There were 17 patients with leiomyoma located in fundus, 10 in body and 10 in isthmus. $17{\beta}$-estradiol, 5-AT, 11-keto ET, $11{\beta}$-hydroxy An, $11{\beta}$-hydroxy Et, THS, THA, THE, a-cortolone, a-cortol, $\beta$-cortol, $11{\beta}$-OH Et/$11{\beta}$-OH An and E2/E1 were significantly increased in patients with leiomyoma than in the control group. $17{\beta}$-estradiol was significantly increased in the intramural and the submucosal types than in the subserosal type. There was no significant difference in the concentrations of urinary steroids according to the locations of leiomyomas. There was no significant relationship between the concentration of urinary steroids and the volume of the leiomyomas. $17{\beta}$-estradiol significantly decreased as the weight of uterus increased (r=-0.322, p=0.04). Conclusion: The concentrations of steroid hormone metabolites were generally increased in patients with leiomyoma but were not significantly related to the volume and weight of the leiomyomas. Our study suggests that steroid hormones may be involved in the initiation of leiomyomas but may not be involved in their progression. In addition, the concentrations of steroid hormone metabolites are not related to the leiomyoma type and location.
