Rhodopsin Chromophore Formation and Thermal Stabilities in the Opsin Mutant E134Q/M257Y (옵신 mutant E134Q/M257Y의 로돕신 형성과 열안정성 분석)

  • Rhodopsin, a dim light photoreceptor, has been regarded as one of the model systems for the structural and functional study of G protein-coupled receptors (GPCRs). Constitutively active mutant GPCRs leading to the activation of heterotrimeric GDP/GTP-binding protein signaling in the absence of ligand binding are of interest for the study of the activation mechanism in GPCRs. The present study focused on the opsin mutant E134Q/M257Y, which showed a moderate level of constitutive activity and the formation of two distinct rhodopsin chromophores with absorption maxima of 500 nm and 380 nm, depending on the presence of an inverse agonist, 11-cis-retinal, and an agonist, all-trans-retinal, respectively. Reconstitution of the mutant rhodopsin upon incubation with different ratios of 11-cis-retinal and the all-trans-retinal, as well as upon sequential binding of the two retinals, indicated its preferential binding to 11-cis-retinal. The thermal stability of the 11-cis-retinal-bound form of the E134Q/M257Y mutant was lower than that of the mutants containing a single replacement but higher than that of the all-trans-retinal-bound forms. The mutant also showed a lower stability in its opsin state as compared with that of the wild-type opsin but had little effects on the binding affinity to 11-cis-retinal. Information obtained in this study will be helpful for analyzing the structural changes associated with the activation of rhodopsin and GPCRs.

Reliability Analysis Using Field-Data of 5.56 mm Rifle (야전운용제원을 이용한 5.56 mm 소총 신뢰도 분석)

  • Reliability is an important factor in weapons systems. Low reliability causes the weapons system to fail to function properly, which directly leads to the weakening of combat capability. This paper classifies the structure of the 5.56 mm rifle, which is currently used by the Korean army, into a total of seven assemblies and describes the eight functions necessary for the rifle to operate normally. In addition, the concept of reliability was defined as the MRBF, and the Poisson process model and TTT plot were explained as a reliability analysis theory for the repair function system. Next, the field-data obtained by defining failure as the replacement of parts other than periodic exchange of parts were refined, and the reliability was analyzed by entering the refined field operation specifications into the Minitab program. As a result, the reliability of the rifle was determined to be 251.73. The assembly parts that required improvement was identified as the barrel, lower body, and butt stock assembly, and 10 detailed parts needed to be improved. Finally, the limits of the reliability analysis using the field-data currently available for small caliber firearms were considered.

Autogenous Shrinkage of Cement Paste Considering Disjoining Pressure in Thin Adsorbed Region (흡착 영역 분리 압력을 고려한 시멘트페이스트의 자기 수축)

  • Meniscus, adsorbed layer thickness, capillary pressure and disjoining pressure was deduced in extended meniscus region in cement paste pore by hydrostatic equilibrium. From the results, the relationship between pore size and adsorbed layer thickness could be derived and adsorbed layer thickness represents $0.299{\sim}2.700nm$ according to pore size $1nm{\sim}1{\mu}m$. Especially, disjoining pressure rapidly Increased in less than 10 nm pore size according to adsorbed layer thickness. Therefore, it is interpreted that autogenous shrinkage of cement paste is highly increases in formation of less than 10 nm pore size. Predictions of autogenous shrinkage in cement paste considering driving force for autogenous shrinkage with capillary pressure and disjoining pressure was low in comparison with experiment values between $1{\sim}4$ days and high in later period. These tendency could be thought that pore damage by mercury injection in early age makes shrinkage driving force underestimate and assumption for unsaturated independent pore makes overestimate. These interactions might be needed corrections considering on hydration or pore replacement model.

  • The purpose of this study is to examine the structural elements of HMR in Korea foods and explore the way HMR products using Korean foods can be developed at this time of increased interest. Through an investigation of its importance by attributes and their partial values, hypothetical HMR products using Korean foods were estimated. In order to develop the optimal HMR goods of Korean food, a preference survey was conducted after selecting 9 profiles using conjoint analysis with orthogonal design, and 4 holdout sets were generated and used for cross-validity authorization and reliability of the model. The results of this study showed that customers put cooking levels, menu price, and the location of purchase into importance when selecting HMR products of Korean foods. They preferred to eat the products after sufficiently heating them and buy the products sold online and through home shopping programs, with the price range of 10,000 won and over. It was concluded that more customers can be attracted if a variety of HMR products using Korean foods which can be prepared readily anywhere and at any time are developed.

  • TBM (Tunnel Boring Machine) tunnel should be carry out with the adopted machine until the end of excavation because of impossibility of replacement or modification of machine. Observation of the face of the tunnel is difficult, especially in EPB(Earth Pressure Balance) shield TBM, predict changes in the ground condition with analyzing data, collected during the excavation, and it should be reflected in construction. Until recently, subjects of studies on TBM are mainly the determination of machine and the development of advance rate prediction model, according to the characteristics of ground which is the target of excavation. However, study focused on the estimation of ground conditions and the improvement in operational methods using excavation data of TBM equipment, the principal of the excavation, has been done not so much. This study examine the variances in advance rate depending on changes in operating conditions and evaluate the optimal operating conditions of adopt machine, using working data obtained from EPB shield TBM project. The result of this study is suggested as follows. First, cutter head RPM and total thrust force are biggest influences on advance rate, Second, it is recommended for proper advance rate that total thrust force is controlled while optimum cutter head RPM is kept, Third, according to the increasing trend of total thrust force, the changes in ground conditions can be predicted, the appropriate operating conditions can be determined.

  • Across the secondary school, students deal with the algebraic conditions like as identity, inverse, commutative law, associative law and distributive law. The algebraic structures, group, ring and field, are determined by these algebraic conditions. But the conditioning of these algebraic structures are not mentioned at all, as well as the meaning of the algebraic structures. Thus, students is likely to be considered the algebraic conditions as productions from the number sets. In this study, we systematize didactically the meanings of algebraic conditions and algebraic structures, considering connections between the number systems and the solutions of the equation. Didactically systematizing is to construct the model for student's natural mental activity, that is, to construct the stream of experience through which students are considered mathematical concepts as productions from necessities and high probability. For this purpose, we develop the program for the gifted, which its objective is to teach the meanings of the number system and to grasp the algebraic structure conceptually that is guaranteed to solve equations. And we verify the effectiveness of this developed program using didactical experiment. Moreover, the program can be used in ordinary students by replacement the term 'algebraic structure' with the term such as identity, inverse, commutative law, associative law and distributive law, to teach their meaning.

A Case-control Study of the Relationships between Reproductive Factors and Degree of Dysplasia of the Colorectal Adenoma and Cancer (대장 선종 이형성 및 대장암과 임신, 출산, 월경 요인의 관련성에 관한 환자-대조군 연구)

  • Objectives : Evidence for an effect of rerroductive factors on colorectal carcinogenesis is not yet consistent. Little research has been conducted to investigate whether reproductive factors were associated with colorectal adenomas that are the precursors of colorecta1 cancer, We evaluated the relationships between reproductive factors and the degree of dysplasia of the colorectal adenoma and cancer as colorectal adenoma-carcinoma sequence. Methods : For this study, 241 adenoma cases with histopathologically confirmed incident colorectal adenoma, 76 cancer cases with colorectal cancer and 1677 controls were collected from Our Lady of Mercy Hospital, The Catholic University of Korea, during 1994-1999. Before colonoscopy, information on demographic characteristics, reproductive factors, life style habits and dietary intake were obtained by interviewed questionnaire. Adjusted OR and 95% CI were estimated by using polytomous logistic regression model, Potential confounders that were selected based on the goodness of fit Statistics and interaction between risk factors were considered in this adjustment. The Wald statistic was calculated to test the heterogeneity of the odds ratios for each case. Results .: Postmenopausal women with natural menopause were found to be positively associated with the risk of mild dysplasia adenoma (multivariate-adjusted OR : 2.59, 95% CI=1.1-0.2). Parity was found to be negatively associated with the risk of colorectal lancer (age-adjusted OR : 0.40, 95% CI=0.2-0.9), but did not significantly decrease the risk of colorectal cancer (multivariate-adjusted OR : 0.95, 95% CI=0.3-2.9). Me associations were seen between a9e at menarche, breast feeding, induced abortion, oral contraceptive use, menopausal types, menopausal age or hormone replacement therapy (HRT and the degree of dysplasia of the colorectal adenoma and cancer. However, none of these associations differed' significantly between the degree of dysplasia of the colorectal adenoma and cancer. Conclusions : These findings suggest that postmenopausal women with natural menopause may experience increased risk of mild dysplasia adenorna among colorectal adenoma-carcinoma sequence.

Indirect Cost Effects on Life-Cycle-Cost Effective Optimum Design of Steel Box Girder Bridge (강상자형교의 LCC 최적설계에 미치는 간접비용의 영향)

  • This paper presents the effects of indirect costs on Life-Cycle-Cost(LCC) effective optimum design of steel-box girder bridges. The LCC formulations considered in the LCC optimization of the bridges consist of initial cost and expected rehabilitation costs including repair/replacement costs, loss of contents or fatality and injury losses, and indirect costs such as road user costs and indirect socio-economic losses. To demonstrate the LCC-effectiveness for optimum design of the bridges, an actual steel box girder bridge having two continuous spans(2@50m=100m) is considered as a numerical example. And also, in this paper, various sensitivity analyses are performed to investigate the effects of indirect costs caused by traffic conditions such as number of detour route, number of lane on detour route, length of detour route, and traffic volumes on the LCC-effective optimum design. From the numerical investigations, it may be concluded that indirect costs caused by traffic network may sensitively influence on the LCC-effective optimum design of steel-box girder bridges. Therefore, it may be stated that the traffic conditions should be considered as one of the important items in the LCC-effective optimum design of the bridges.

Application of Random Over Sampling Examples(ROSE) for an Effective Bankruptcy Prediction Model (효과적인 기업부도 예측모형을 위한 ROSE 표본추출기법의 적용)

  • If the frequency of a particular class is excessively higher than the frequency of other classes in the classification problem, data imbalance problems occur, which make machine learning distorted. Corporate bankruptcy prediction often suffers from data imbalance problems since the ratio of insolvent companies is generally very low, whereas the ratio of solvent companies is very high. To mitigate these problems, it is required to apply a proper sampling technique. Until now, oversampling techniques which adjust the class distribution of a data set by sampling minor class with replacement have popularly been used. However, they are a risk of overfitting. Under this background, this study proposes ROSE(Random Over Sampling Examples) technique which is proposed by Menardi and Torelli in 2014 for the effective corporate bankruptcy prediction. The ROSE technique creates new learning samples by synthesizing the samples for learning, so it leads to better prediction accuracy of the classifiers while avoiding the risk of overfitting. Specifically, our study proposes to combine the ROSE method with SVM(support vector machine), which is known as the best binary classifier. We applied the proposed method to a real-world bankruptcy prediction case of a Korean major bank, and compared its performance with other sampling techniques. Experimental results showed that ROSE contributed to the improvement of the prediction accuracy of SVM in bankruptcy prediction compared to other techniques, with statistical significance. These results shed a light on the fact that ROSE can be a good alternative for resolving data imbalance problems of the prediction problems in social science area other than bankruptcy prediction.

The Effect of Silk Fibroin/Nano-hydroxyapatite/Corn Starch Composite Porous Scaffold on Bone Regeneration in the Rabbit Calvarial Defect Model (가토 두개골 결손 모델에서 실크단백과 나노하이드록시아파타이트, 옥수수 녹말 복합물을 이용한 골 이식재 개발)

  • Purpose: This study evaluated the capability of bone formation with silk fibroin/nano-hydroxyapatite/corn starch composite scaffold as a bone defect replacement matrix when grafted in a calvarial bone defect of rabbits $in$ $vivo$. Methods: Ten New Zealand white rabbits were used for this study and bilateral round-shaped defects were formed in the parietal bone (diameter: 8.0 mm). The silk fibroin 10% nano-hydroxyapatite/30% corn starch/60% composite scaffold was grafted into the right parietal bone (experimental group). The left side (control group) was grafted with a nano-hydroxyapatite (30%)/corn starch (70%) scaffold. The animals were sacrificed at 4 weeks and 8 weeks. A micro-computerized tomography (${\mu}CT$) of each specimen was taken. Subsequently, the specimens were decalcified and stained with Masson's trichrome for histological and histomorphometric analysis. Results: The average ${\mu}CT$ and histomorphometric measures of bone formation were higher in the control group than in the experimental group at 4 weeks and 8 weeks after surgery though not statistically significant ($P$ >0.05). Conclusion: The rabbit calvarial defect was not successfully repaired by silk fibroin/nano-hydroxyapatite/corn starch composite scaffold and may have been due to an inflammatory reaction caused by silk powder. In the future, the development of composite bone graft material based on various components should be performed with caution.
