구조물 및 기기의 내진성능 평가를 위한 고주파수 지진에 의한 원자력발전소의 지진응답 증폭계수 (Seismic Response Amplification Factors of Nuclear Power Plants for Seismic Performance Evaluation of Structures and Equipment due to High-frequency Earthquakes)

  • Analysis of the 2016 Gyeongju earthquake and the 2017 Pohang earthquake showed the characteristics of a typical high-frequency earthquake with many high-frequency components, short time strong motion duration, and large peak ground acceleration relative to the magnitude of the earthquake. Domestic nuclear power plants were designed and evaluated based on NRC's Regulatory Guide 1.60 design response spectrum, which had a great deal of energy in the low-frequency range. Therefore, nuclear power plants should carry out seismic verification and seismic performance evaluation of systems, structures, and components by reflecting the domestic characteristics of earthquakes. In this study, high-frequency amplification factors that can be used for seismic verification and seismic performance evaluation of nuclear power plant systems, structures, and equipment were analyzed. In order to analyze the high-frequency amplification factor, five sets of seismic time history were generated, which were matched with the uniform hazard response spectrum to reflect the characteristics of domestic earthquake motion. The nuclear power plant was subjected to seismic analysis for the construction of the Korean standard nuclear power plant, OPR1000, which is a reactor building, an auxiliary building assembly, a component cooling water heat exchanger building, and an essential service water building. Based on the results of the seismic analysis, a high-frequency amplification factor was derived upon the calculation of the floor response spectrum of the important locations of nuclear power plants. The high-frequency amplification factor can be effectively used for the seismic verification and seismic performance evaluation of electric equipment which are sensitive to high-frequency earthquakes.

  • In Korea, most nuclear power plants were designed based on the design response spectrum of Regulatory Guide 1.60 of the NRC. However, in the case of earthquakes occurring in the country, the characteristics of seismic motions in Korea and the design response spectrum differed. The seismic motion in Korea had a higher spectral acceleration in the high-frequency range compared to the design response spectrum. The seismic capacity may be reduced when evaluating the seismic performance of the equipment with high-frequency earthquakes compared with what is evaluated by the design response spectrum for the equipment with a high natural frequency. Therefore, EPRI proposed the inelastic energy absorption factor for the equipment anchorage. In this study, the seismic performance of welding anchorage was evaluated by considering domestic seismic characteristics and EPRI's inelastic energy absorption factor. In order to reflect the characteristics of domestic earthquakes, the uniform hazard response spectrum (UHRS) of Uljin was used. Moreover, the seismic performance of the equipment was evaluated with a design response spectrum of R.G.1.60 and a uniform hazard response spectrum (UHRS) as seismic inputs. As a result, it was confirmed that the seismic performance of the weld anchorage could be increased when the inelastic energy absorption factor is used. Also, a comparative analysis was performed on the seismic capacity of the anchorage of equipment by the welding and the extended bolt.

  • 소포체는 세포막의 합성뿐만 아니라 세포막에 존재하거나 세포외로 분비되어져야 할 단백질을 합성하는 세포내 소기관이다. 소포체에서 단백질이 합성되어질 경우 이황화결합이 형성되고 glycosylation 등의 수식이 일어나며, 이와 동시에 folding과 assembly과정을 거쳐 삼차원적 구조로 성숙이 되는데 이 과정은 folding enzyme과 molecular chaperone의 도움을 받아 이루어진다. 소포체 내에 존재하는 molecular chaperone 중 가장 잘 알려진 것으로 BiP이 있다. BiP의 기능은 N-terminus의 ATPase domain에 의해 조절되고 ATPase domain은 이것과 선택적으로 결합하는 조절인자에 의해 ATPase의 활성이 영향을 받는다. BiP의 핵산치환조절인자로서 발견된 BAP은 ATPase domain에 결합된 ADP를 ATP로 치환하는 것으로 기능이 알려져 있다. 이 BAP의 핵산치환기능이 BiP의 샤페론 작용에 어떤 영향을 미치는지를 in vitro에서 항체 heavy chain을 이용하여 알아보았다. BAP은 ATP보다 ADP가 결합되어 있는 BiP과 더 잘 결합을 하며, in vitro에서 BiP과 결합하고 있는 unfolded 단백질을 BAP은 BiP으로부터 해리하였다. 또한 소포체내에 존재하는 Hsp70 homologue chaperone인 BiP과 Grp170에 대한 BAP의 결합특이성을 anti-Grp170과 anti-BAP 항체로 co-immunoprecipitation을 하여 확인한 결과 BAP은 Grp170과 결합을 하지 않았다. 따라서 BAP은 ER내에 존재하는 동일한 family group에 속하는 Grp170과 BiP에 대하여 BiP에만 특이성을 갖는 것으로 나타났다.

Translation Initiation Factor 4E (eIF4E) is Regulated by Cell Death Inhibitor, Diap1

  • Translation initiation factor 4E (eIF4E) is a key regulator of protein synthesis. Abnormal regulation of eIF4E is closely linked to oncogenic transformation. Several regulatory mechanisms affecting eIF4E are discussed, including transcriptional regulation, phosphorylation and binding of an inhibitor protein. However it is not clear how the level of eIF4E protein is regulated under basal conditions. Here we demonstrate that Diap1 (Drosophila Inhibitor of Apoptosis Protein), a cell death inhibitor, binds directly to eIF4E and poly-ubiquitinates it via its E3 ligase activity, promoting its proteasome-dependent degradation. Expression of Diap1 caused a reduction of Cyclin D1 protein level and inhibited the growth stimulation induced by overexpression of eIF4E. Taken together, our results suggest that the level of eIF4E protein is regulated by Diap1, and that IAPs may play a role in cap-dependent translation by regulating the level of eIF4E protein.

  • Transforming growth factor-$\beta$ (TGF-$\beta$) is a highly pleiotropic cytokine playing pivotal roles in immune regulation. TGF-$\beta$ facilitates tumor cell survival and metastasis by targeting multiple cellular components. Focusing on its immunosuppressive functions, TGF-$\beta$ antagonists have been employed for cancer treatment to enhance tumor immunity. TGF-$\beta$ antagonists exert anti-tumor effects through #1 activating effector cells such as NK cells and cytotoxic $CD8^+$ Tcells (CTLs), #2 inhibiting regulatory/suppressor cell populations, #3 making tumor cells visible to immune cells, #4 inhibiting the production of tumor growth factors. This review focuses on the effect of TGF-$\beta$ on T cells, which are differentiated into effector T cells or newly identified tumor-supporting T cells.

Regulatory Effect of Inflammatory Reaction by Anglicae Dahuricae Radix

  • Anglicae dahuricae Radix (ADR), the dried roots of Angelica Dahurica Benth et Hook (Umbelliferae), is a traditional herbal medicine used to treat colds, headache, skin diseases such as acne and possess analgesic, antipyretic and drainage effects. In the present study, the author evaluated the effect of ADR on regulation of inflammatory reaction. ADR reduced the ear-swelling responses derived from compound 48/80 in dose-dependent manner significantly. ADR inhibited the PMA plus A23187-induced productions of IL (Interleukin)-8, IL-1β, granulocyte macrophage-colony stimulating factor (GM-CSF) and tumor necrosis factor (TNF)-α from human mast cells (HMC)-1. In addition, ADR blocked PMA plus A23187-induced ERK1/2 phosphorylation. Ⅰ suggest that ADR regulates inflammatory reaction through inhibition of inflammatory cytokines such as IL-8, IL-1β and GM-CSF.

Modification of cell wall structural carbohydrate in the hybrid poplar expressing Medicago R2R3-MYB transcription factor MtMYB70

  • The isolation, cloning, and characterization of an R2R3-MYB transcription factor gene (MtMYB70) from the model legume Medicago truncatula is reported. MtMYB70 consists of a 768-bp coding sequence corresponding to 255 amino acids. Sequence alignment revealed that MtMYB70 cDNA contains conserved R2R3-type MYB domains with highly divergent C terminal regions. MtMYB70 was found to have relatively low sequence homology with known R2R3-MYB genes. Phylogenetic analysis placed the R2R3-MYB domain of MtMYB70 closest to PtMYB1, a known activator of lignin biosynthesis. Overexpression of MtMYB70 under the control of the 35S promoter in transgenic poplar did not cause a significant difference in total lignin content relative to the control, but glucan content was significantly increased in transgenic poplar. Therefore, MtMYB70 might have regulatory role in the biosynthesis of cell wall structural carbohydrates.

  • Heat Shock Transcription Factor (HSF), and the promoter heat Shock Element (HSE), are among the most highly conserved transcriptional regulatory elements in nature. HSF mediates the transcriptional response of eukaryotic cells to heat, infection and inflammation, pharmacological agents, and other stresses. While HSF is essential for cell viability in yeast, oogenesis and early development in Drosophila, extended life-span in C. elegans, and extra-embryonic development and stress resistance in mammals, little is known about its full range of biological target genes. We used whole genome analyses to identify virtually all of the direct transcriptional targets of yeast HSF, representing nearly three percent of the genomic loci. The majority of the identified loci are heat-inducibly bound by yeast HSF, and the target genes encode proteins that have a broad range of biological functions including protein folding and degradation, energy generation, protein secretion, maintenance of cell integrity, small molecule transport, cell signaling, and transcription. Approximately 30% of the HSF direct target genes are also induced by the diauxic shift, in which glucose levels begin to be depleted. We demonstrate that phosphorylation of HSF by Snf1 kinase is responsible for expression of a subset of HSF targets upon glucose starvation.

Baicalin이 조골세포의 생성 및 활성에 미치는 영향 (Effects of Baicalin on the Proliferation and Activity of Osteoblastic Cells)

  • Baicalin은 Scutellaria baicalensis에서 분리되는 flavonoid의 일종으로, 다양한 생물학적 활성을 나타내는 물질로 알려져 있다. Baicalin은 항균, 항염증, 진통작용을 나타내며, nuclear factor-kappaB의 활성을 억제한다고 보고되었다. 최근에 다양한 flavonoid 들이 골조직 대사에 관여함이 밝혀졌으며, 본 연구에서는 baicalin이 골조직의 주요세포인 조골세포의 생성 및 활성에 미치는 영향을 관찰하기 위하여, 세포증식율, 세포생존율, 염기성 인산분해효소 활성 및 osteoprotegerin 생성량의 변화를 관찰하였다. 그 결과 baicalin은 조골세포의 세포 증식과 생존율에는 영향을 미치지 못하였으나, 염기성 인산분해효소의 활성과 osteoprotegerin의 생성량을 증가시켰다. 따라서 baicalin은 골조직에서 조절물질로 역할을 할 것으로 예견된다.

X-ray radiation at low doses stimulates differentiation and mineralization of mouse calvarial osteoblasts

  • Radiotherapy is considered to cause detrimental effects on bone tissue eventually increasing bone loss and fracture risk. However, there is a great controversy on the real effects of irradiation itself on osteoblasts, and the mechanisms by which irradiation affects osteoblast differentiation and mineralization are not completely understood. We explored how X-ray radiation influences differentiation and bone-specific gene expression in mouse calvarial osteoblasts. Irradiation at 2 Gy not only increased differentiation and mineralization of the cells, but also upregulated the expression of alkaline phosphatase, type I collagen, osteopontin, and osteocalcin at early stages of differentiation. However, irradiation at higher doses (>2 Gy) did not stimulate osteoblast differentiation, rather it suppressed DNA synthesis by the cells without a toxic effect. Additional experiments suggested that transforming growth factor-beta 1 and runt-transcription factor 2 play important roles in irradiation- stimulated bone differentiation by acting as upstream regulators of bone-specific markers.
