Source Identification and Trends in Atmospheric Particulate-bound Mercury at Seoul and Baengnyeong, South Korea (서울과 백령도의 대기 중 입자상 수은의 분포 특성 및 발생원 추정연구)

  • $PM_{2.5}$-bound mercury (PBM) was monitored at weekly intervals for three years (from 2014 to 2016) at an urban (Seoul) and rural site (Baengnyeong) in South Korea. The average PBM concentrations in $PM_{2.5}$ samples over the entire sampling period were $12{\pm}11pg/m^3$ and $36{\pm}34pg/m^3$ for Baengnyeong and Seoul, respectively. Seasonal differences were pronounced, with concentrations being highest in winter due to local meteorological conditions (high gas-particle coefficient due to low temperature and low mixing layer height in winter) as well as seasonal factors, such as coal combustion for heating purposes in China. In Baengnyeong, the significant positive correlation of PBM with $PM_{2.5}$, air pollutants, and heavy metals suggested that coal combustion in China might be the most important source of ambient mercury in Korea. In winter, no correlation of PBM with $PM_{2.5}$, air pollutants, and heavy metals was seen in Seoul. Furthermore, Seoul showed higher $PBM/PM_{2.5}$ and $Pb/PM_{2.5}$ ratios in winter due to the strong atmospheric oxidation-reduction reaction conditions as well as local and regional PBM sources. We conclude that immediate attention must be given to addressing PBM levels in Korea, including considering it as a key component of future air quality monitoring activities and mitigation measures.

A Study on Changes of the Benthic Environment and Microbial Community in Estuarine Polluted Sediments by Mixing Granulated Coal Ash (석탄회 조립물이 혼합된 하구 오염 퇴적물의 환경 및 미생물 구조 변화에 관한 연구)

  • In this study, the benthic environmental and microbial community structure were investigated by mixing granulated coal ash(GCA) and contaminated estuary sediments. Estuary sediments and GCA were mixed in a ratio of 8:2 and allowed to interact for 1 month, then sediment environmental factors were investigated. The pH of the experimental sediment was mixed increased to 11. The concentration of DIP(Dissolved inorganic phosphorus) in the experimental case decreased by 30 % compared to the control case, and this should be due to formation of calcium phosphate through the chemical reaction of DIP and calcium which diluted from GCA. The high abundance of Gammaproteobacteria seen in the experimental sediment compare to the control can af ect the DIP reduction. The DIN(Dissolved inorganic nitrogen) concentration increased over two times in the experimental case than the control, and this should be due to the high pH condition and release of NH4+-N from the GCA. Microorganisms related to nitrogen circulation were not identified in both the control and experimental cases. It was confirmed that the GCA were effective in reducing the DIP concentration in contaminated estuary sediment, and that benthic microbial communities were shown to influenced the phosphorus circulation.

  • In this review, recent foodborne outbreaks caused by viruses in the Republic of Korea (2010-2019) were analyzed. The human norovirus was found to be the major foodborne virus causing an average of 94.9% of the viral outbreaks. Reverse-transcription polymerase chain reaction (PCR) with electrophoresis has been widely used to detect viruses, but several rapid detection methods, including real-time PCR, multiplex PCR, and quantum dot assay, have also been suggested. For norovirus inactivation studies, surrogates such as murine norovirus and feline calicivirus have been widely used to identify the reduction rate owing to the limitations in laboratory cultivation. Conversely, direct cell infection studies have been conducted for other foodborne viruses such as adenovirus, astrovirus, rotavirus, and hepatitis A or E virus. Moreover, virucidal mechanisms using various physical and chemical treatments have been revealed. These recent studies suggest that rapid in situ detection and effective control are valuable for ensuring food safety against viral infections.

A Study on the Influence of the Structural Characteristics of Cu/CeO2 Catalyst on the Low-Temperature Oxidation of Carbon Monoxide (Cu/CeO2 촉매의 구조적 특성이 일산화탄소 저온 산화반응에 미치는 영향 연구)

  • This study confirmed the effect of the Cu/CeO2-X catalyst on the CO oxidation activity at low temperature through the catalyst's structure and reaction characteristics. The catalyst was prepared by the wet impregnation method. Cu/CeO2_X catalysts were manufactured by loading Cu (active metal) using CeO2 (support) formed at different calcination temperatures (300-600 ℃). Manufactured Cu/CeO2_X catalysts were evaluated for the low-temperature activity of carbon monoxide. The Cu/CeO2_300 catalyst showed an activity of 90% at 125 ℃, but the activity gradually decreased as the calcination temperature of the CeO2-X and Cu/CeO2_600 catalysts showed an activity of 65% at 125 ℃. Raman, XRD, H2-TPR, and XPS analysis confirmed the physicochemical properties of the catalysts. Based on the XPS analysis, the lower the calcination temperature of the CeO2 was, the higher the unstable Ce3+ species (non-stoichiometric species) ratio became. The increased Ce3+ species formed a solid solution bond between Cu and CeO2-X, and it was confirmed by the change of the CeO2 peak in Raman analysis and the reduction peak of the solid solution structure in H2-TPR analysis. According to the result, the formation of the solid solution bond between Cu and Ce has been enhanced by the redox properties of the catalysts and by CO oxidation activity at low temperatures.

Selective Catalytic Reduction (SCR) of NOx with NH3 on Sb-promoted VWTi Catalysts (Sb 첨가에 따른 VWTi 촉매의 암모니아 선택적 촉매 환원(SCR)을 통한 질소산화물 저감)

  • VWTi, which is used as a commercial catalyst in NH3-SCR, exhibits excellent denitrification performance at 300 to 400 ℃, but there is a problem that efficiency decreases at low temperatures below 300 ℃. Research on catalysts containing promoter to increase low-temperature denitrification efficiency is steadily progressing. However, research on the cause of the improvement in low-temperature denitrification efficiency of the catalyst and the catalyst properties is insufficient. In this study, it was confirmed that by adding Sb to VWTi, denitrification performance was improved by more than 10% in NH3-SCR reaction below 300 ℃. At this time, the space velocity and the size of the catalyst particles were controlled to exclude the influence of external/internal diffusion. In addition, the catalytic properties according to the presence or absence of Sb were investigated by performing BET, TEM/EDS, O2-TPD, H2-TPR and DRIFTs analysis. It was judged that the addition of Sb increased the adsorbed oxygen species on the surface of the catalyst, thereby enhancing the redox properties of the catalyst at low temperature and exhibiting excellent denitrification performance.

Odor Reduction of Pig Wastewater Using Magnesia (in-situ test) (마그네시아를 이용한 돈분 폐수의 악취 저감(현장 시험))

  • In this study, we tried to obtain the optimal conditions to reduce odors generated from pig wastewater using magnesia (MgO) through in-situ test after producing a reactor for removing odors. For this purpose, the filling amount of magnesia, the injection amount of pig wastewater, the aeration method, the aeration amount and the aeration time were considered. The field experiment was conducted at Cheongwoon Livestock Farm, which has a pig wastewater reservoir. As the amount of magnesia added to the weight of wastewater (500 kg) increases, the amount of ammonia (NH3) and hydrogen sulfide (H2S) generated tended to gradually decrease. As a result of the test, ammonia and hydrogen sulfide in the pig wastewater decreased up to 65% and 77%, respectively, for 2 days aeration after 0.8% of magnesia was added to the reaction tank. The initial pH of the pig wastewater in the reactor was 8.2, and the pH was found to be 9.2 when magnesia was added up to 0.8%. In the light of this trend, it can be known that magnesia gradually increases the pH in the pig wastewater and makes it weakly alkaline. As the pH increases, part of the ammonia gas present in the pig wastewater vaporizes into the air and the remaining part is removed by precipitation after chemical bonding with dissolved magnesium ions and phosphate ions. In order to remove the odor of pig wastewater and turn it into compost, most of the existing livestock farms go through a six-month aeration process using microorganisms. In contrast, the current study proved the effect of removing odors from pig wastewater within 2 days through chemical reactions that do not affect microbial activity.

EID3 Promotes Glioma Cell Proliferation and Survival by Inactivating AMPKα1

  • Objective : EID3 (EP300-interacting inhibitor of differentiation) was identified as a novel member of EID family and plays a pivotal role in colorectal cancer development. However, its role in glioma remained elusive. In current study, we identified EID3 as a novel oncogenic molecule in human glioma and is critical for glioma cell survival, proliferation and invasion. Methods : A total of five patients with glioma were recruited in present study and fresh glioma samples were removed from patients. Four weeks old male non-obese diabetic severe combined immune deficiency (NOD/SCID) mice were used as transplant recipient models. The subcutaneous tumor size was calculated and recorded every week with vernier caliper. EID3 and AMP-activated protein kinase α1 (AMPKα1) expression levels were confirmed by real-time polymerase chain reaction and Western blot assays. Colony formation assays were performed to evaluate cell proliferation. Methyl thiazolyl tetrazolium (MTT) assays were performed for cell viability assessment. Trypan blue staining approach was applied for cell death assessment. Cell Apoptosis DNA ELISA Detection Kit was used for apoptosis assessment. Results : EID3 was preferentially expressed in glioma tissues/cells, while undetectable in astrocytes, neuronal cells, or normal brain tissues. EID3 knocking down significantly hindered glioma cell proliferation and invasion, as well as induced reduction of cell viability, apoptosis and cell death. EID3 knocking down also greatly inhibited tumor growth in SCID mice. Knocking down of AMPKα1 could effectively rescue glioma cells from apoptosis and cell death caused by EID3 absence, indicating that AMPKα1 acted as a key downstream regulator of EID3 and mediated suppression effects caused by EID3 knocking down inhibition. These findings were confirmed in glioma cells generated patient-derived xenograft models. AMPKα1 protein levels were affected by MG132 treatment in glioma, which suggested EID3 might down regulate AMPKα1 through protein degradation. Conclusion : Collectively, our study demonstrated that EID3 promoted glioma cell proliferation and survival by inhibiting AMPKα1 expression. Targeting EID3 might represent a promising strategy for treating glioma.

Curcumin represses lipid accumulation through inhibiting ERK1/2-PPAR-γ signaling pathway and triggering apoptosis in porcine subcutaneous preadipocytes

  • Objective: Excessive lipid accumulation in adipocytes results in prevalence of obesity and metabolic syndrome. Curcumin (CUR), a naturally phenolic active ingredient, has been shown to have lipid-lowering effects. However, its underlying mechanisms have remained largely unknown. Therefore, the study aims to determine the effect of CUR on cellular lipid accumulation in porcine subcutaneous preadipocytes (PSPA) and to clarify novel mechanisms. Methods: The PSPA were cultured and treated with or without CUR. Both cell counting Kit-8 and lactate dehydrogenase release assays were used to examine cytotoxicity. Intracellular lipid contents were measured by oil-red-o staining extraction and triglyceride quantification. Apoptosis was determined by flow cytometry and the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-nick end labelling assay. Adipogenic and apoptosis genes were analyzed by quantitative polymerase chain reaction and Western blot. Results: The CUR dose-dependently reduced the proliferation and lipid accumulation of PSPA. Noncytotoxic doses of CUR (10 to 20 μM) significantly inhibited extracellular signal-regulated kinase 1/2 (ERK1/2) phosphorylation and expression of adipogenic genes peroxisome proliferation-activity receptor-γ (PPAR-γ), CCAAT/enhancer binding protein-α, sterol regulatory element-binding protein-1c, adipocyte protein-2, glucose transporter-4 as well as key lipogenic enzymes fatty acid synthase and acetyl-CoA carboxylase, while ERK1/2 activation significantly reversed CUR-reduced lipid accumulation by increasing PPAR-γ. Furthermore, compared with differentiation induced media treated cells, higher dose of CUR (30 μM) significantly decreased the expression of AKT and B-cell lymphoma-2 (BCL-2), while increased the expression of BCL-2-associated X (BAX) and the BAX/BCL-2 expression ratio, suggesting triggered apoptosis by inactivating AKT and increasing BAX/BCL-2 ratio and Caspase-3 expression. Moreover, AKT activation significantly rescued CUR inhibiting lipid accumulation via repressing apoptosis. Conclusion: These results demonstrate that CUR is capable of suppressing differentiation by inhibiting ERK1/2-PPAR-γ signaling pathway and triggering apoptosis via decreasing AKT and subsequently increasing BAX/BCL-2 ratio and Caspase-3, suggesting that CUR provides an important method for the reduction of porcine body fat, as well as the prevention and treatment of human obesity.

Comparison of Li(I) Precipitation from the Leaching Solution of the Dust from Spent Lithium-ion Batteries Treatment between Sodium Carbonate and Ammonium Carbonate (폐리튬이온전지 처리시 발생한 더스트 침출용액으로부터 Na2CO3와 (NH4)2CO3에 의한 리튬(I) 석출 비교)

  • Smelting reduction of spent lithium-ion batteries results in metallic alloys, slag, and dust containing Li(I). Precipitation of Li2CO3 was performed using the synthetic leachate of the dust. Herein, the effects of the precipitant and addition of non-aqueous solvents on the precipitation of Li(I) were investigated. Na2CO3 was a more effective precipitating agent than (NH4)2CO3 owing to the hydrolysis reaction of dissolved ammonium and carbonate. The addition of acetone or ethanol improved the Li(I) precipitation percentage for both the precipitants. When using (NH4)2CO3, the Li(I) precipitation percentage increased at a solution pH of 12. Under the same conditions, the Li(I) precipitation percentage using Na2CO3 was much higher than that using (NH4)2CO3.

MicroRNA-21 promotes epithelial-mesenchymal transition and migration of human bronchial epithelial cells by targeting poly (ADP-ribose) polymerase-1 and activating PI3K/AKT signaling

  • Epithelial-mesenchymal transition (EMT) is known to be involved in airway remodeling and fibrosis of bronchial asthma. However, the molecular mechanisms leading to EMT have yet to be fully clarified. The current study was designed to reveal the potential mechanism of microRNA-21 (miR-21) and poly (ADP-ribose) polymerase-1 (PARP-1) affecting EMT through the PI3K/AKT signaling pathway. Human bronchial epithelial cells (16HBE cells) were transfected with miR-21 mimics/inhibitors and PARP-1 plasmid/small interfering RNA (siRNA). A dual luciferase reporter assay and biotin-labeled RNA pull-down experiments were conducted to verify the targeting relationship between miR-21 mimics and PARP-1. The migration ability of 16HBE cells was evaluated by Transwell assay. Quantitative real-time polymerase chain reaction and Western blotting experiments were applied to determine the expression of Snail, ZEB1, E-cadherin, N-cadherin, Vimentin, and PARP-1. The effects of the PI3K inhibitor LY294002 on the migration of 16HBE cells and EMT were investigated. Overexpression of miR-21 mimics induced migration and EMT of 16HBE cells, which was significantly inhibited by overexpression of PARP-1. Our findings showed that PARP-1 was a direct target of miR-21, and that miR-21 targeted PARP-1 to promote migration and EMT of 16HBE cells through the PI3K/AKT signaling pathway. Using LY294002 to block PI3K/AKT signaling pathway resulted in a significant reduction in the migration and EMT of 16HBE cells. These results suggest that miR-21 promotes EMT and migration of HBE cells by targeting PARP-1. Additionally, the PI3K/AKT signaling pathway might be involved in this mechanism, which could indicate its usefulness as a therapeutic target for asthma.
