Substrate Specificity of the Yeast Protein Tyrosine Phosphatase, PTP1, Overexpressed from an Escherichia coli Expression System

  • A Saccharomyces cerevisiae Protein Tyrosine Phosphatase, PTP1, was expressed from an Escherichia coli expression system and milligram quantities of active PTP1 were purified chromatographically. The substrate specificity of the recombinant PTP1 was probed using synthetic phosphotyrosine-containing peptides corresponding to the regulatory phosphorylation sites of the yeast MAP kinase homologues $Fus3_{176-186}$, $Kss1_{179-189}$, and $Hog1_{170-180}$. Peptide sequences derived from the MAP kinase homologues were chosen arbitrarily as starting points for sequence variation studies even though they are not likely to be candidates for physiological substrates of PTP1. Phosphotyrosyl-$Hog1_{170-180}$ peptide showed a $K_M$ value of 877 ${\mu}M$ and phosphorylated $Kss1_{179-189}$ and $Fus3_{176-186}$ peptides showed lower $K_M$ values of 74 ${\mu}M$ and 51 ${\mu}M$ each. To study the effect of sequence variations of the peptide, amino acids of the undecapeptide $Hog1_{170-180}$ (DPQMTGpYVSTR) were sequentially substituted by an alanine residue. More extensive variations of each amino acid revealed positional importance of each amino acid residue. Based on these results, we derived a peptide sequence (DADEpYDA) that is recognized by PTP1 with an affinity ($K_M$ is 4 ${\mu}M$) significantly higher than that of the peptides derived from the phosphorylation sites of Fus3, Kss1, and Hog1.

Autonomously Mitochondrial Replicating Sequence of Aspergillus nidulans (Aspergillus nidulans mtDNA의 자가복제절편)

  • We isolated the ANRI fragment from Aspergillus nidulons that could autononlously replicate and enhance transformation efficiency about $10^4$ fold compared lo the integrative vector in Saccha,omgcer cerevisioe. In A. nidulans recombinant plasmid pLJ16-4.5 which carries the 4.5 kb EcoRI fragment of ANRI showed a 170-[old increase of transformation efficiency compared to the integrative vector pLJ16 and could be recovered from iransfonnants as an intact form. Estimated copy number of transforming plasmid pLJ16-4.5 was scored as 2 to 3 copies in transformed A. nidulans. Recoinbinant plasinid pILJ16-4.5 is inilotically unstable; being lost Irom 65% of aswual progeny of transformants on selective medium and 90% on complete medium. Southern analysis of transformant DNA showed that the pILJ16-4.5 is maintained in free form. The sequencing data showed that ANRl fragment was originated from mitochondiral DNA of A. nid~ilans and contained high AT content as much as 74.7%. One ARS consensus sequence (A/T)TTr4T(A/G)TTT(AiT). I I ARS-like sequence (agreement 10 of 11) and ABFl binding core consensus sequence (TCN7ACG). Also six gyrase binding core consensus sequence (YRTGNYNNY: y=C or T, R=A or G, N=A, G, C or T) of $\Phi$X174 and SV40 DNA and one b site (CACTTTACC) combining with gyrase in ColEl are shown. ANRl can be developed as a repl&ng plasinid for lransfoimation system in A. nirlulmis.

  • In order to obtain yeast cells producing a bacteriocin, Subpeptin JM4-A or Subpeptin JM4-B, the 48 bp oligonucleotides corresponding to Subpeptin JM4-A and Subpeptin JM4-B genes including codon for start and stop were chemically synthesized and cloned into pAUR123, an yeast expression vector. Transformed yeast cells exhibited growth inhibition of Bacillus subtilis, Escherichia coli and Pseudonomas aeruginosa. This result indicates that yeast cells producing Subpeptin JM4-A or Subpeptin JM4-B possess bacteriocidal properties against both Gram positive B. subtilis and Gram negative E. coli and P. aeruginosa cells. The recombinant yeast strains constructed in this study can be applied in the food preservative or. animal foodfeed.

  • In order to obtain yeast cells producing a bacteriocin OR-7, the 180 bp polynucleotide corresponding to the OR-7 gene including codons for start and stop was chemically synthesized and cloned into pAUR123, an yeast expression vector. Transformed yeast cells exhibited growth inhibition of Bacillus subtilis, Campylobacter jeuni, Escherichia coli and Pseudomonas aeruginosa. This result indicates that yeast cells producing OR-7 possess bacteriocidal properties against both Gram positive B. subtilis and Gram negative C. jejuni, E. coli and P. aeruginosa cells. The recombinant yeast strain constructed in this study can be applied in the food preservative or animal feed.

Effect of Non-homologous Spacing in Target DNA Sequence on the Frequency of Cloning Based Homologous Recombination (Target DNA 염기서열 내에 존재하는 비상동성 간격이 상동성재조합을 이용한 클로닝 빈도에 미치는 영향)

  • Transformation-Associated Recombination (TAR) cloning technique allows selective isolation of chromosomal regions and genes from complex genomes. The procedure requires knowledge of relatively small genomic sequences that reside adjacent to the chromosomal region of interest. This technique involves homologous recombination during yeast spheroplast transformation between genomic DNA and a TAR vector that has 5' and 3' gene targeting sequences. In this study, we examined the effect of non-homologous spacing sequence in target hooks on homologous recombination using a plasmid model system. The efficiency of homologous recombination between the modified his3-TRP1-his3 fragments and HlS3 gene on plasmid were analyzed by the characterization of $Ura^+$ transformants. The numbers of $Ura^+$ transformant showed same level when seven different modified his3-TRP1-his3 fragments were used. But the percentage of positive recombinants. $Trp^+His^-$, dramatically decreased when used the modified his3-TRP1-his3 fragments contained incorrect spacing of nonhomologous region. As a result, we suggest that incorrect spacing inhibits the homologous recombination between target hook and substrate DNA. Therefore, we should consider the correct spacing in target hook when the target hook are used for cloning of orthologue gene.

Oestrogenic Activity of Parabens In Vitro Estrogen Assays (에틸, 프로필, 이소프로필, 부틸, 이소부틸 파라벤의 In Vitro 검색시험 연구에서의 내분비독성)

  • The use of underarm and body care cosmetics with oestrogenic chemical excipients (particularly the parabens) and the hypothesized association with breast cancer incidence, particularly in women. It is noted that the type of cosmetic product is irrelevant (e.g. antiperspirant/deodorant versus body lotion, moisturizers or sprays versus creams) and attention must focus on issues of actual exposure to chemicals through continued dermal application of body care products and the endocrine/hormonal activity and toxicity of the chemicals in the formulations. To evaluate the estrogenic activities of parabens such as ethylparaben, butylparaben, propylparaben, isobutylparaben and isopropylparaben, we used recombinant yeasts containing the human estrogen receptor [Saccharomyces cerevisiae ER+LYS 8127], human breast cancer MCF-7 cell lines and human estrogen receptor ${\alpha}\;and\;{\beta}$. In E-screen assays, isopropylparaben is the most estrogenic paraben, and in ER competition assay, isobutylparaben is the most estrogenic paraben. We evaluated isopropylparaben was most active in the recombinant yeast assay, followed by propylparaben, ethylparaben, isobutylparaben and butylparaben. Results from this study demonstrate that parabens are observed in human endocrine system. Therefore, we have shown that the parabens is induced the estrogenic activities similar to $17{\beta}$-estradiol and Bisphenol-A.
