Backbone assignment of the intrinsically disordered N-terminal region of Bloom syndrome protein

  • Bloom syndrome protein (BLM) is a pivotal RecQ helicase necessary for genetic stability through DNA repair processes. Our investigation focuses on the N-terminal region of BLM, which has been considered as an intrinsically disordered region (IDR). This IDR plays a critical role in DNA metabolism by interacting with other proteins. In this study, we performed triple resonance experiments of BLM220-300 and presented the backbone chemical shifts. The secondary structure prediction based on chemical shifts of the backbone atoms shows the region is disordered. Our data could help further interaction studies between BLM220-300 and its binding partners using NMR.

태양광전원 수용을 위한 MVDC 배전망의 경제성평가 모델링에 관한 연구 (A Study on Economic Evaluation Modeling of MVDC Distribution System for Hosting Capacity of PV System)

  • MVDC 배전기술은 현재 급격하게 도입되고 있는 태양광전원의 접속지연 문제를 해결하기 위한 효과적인 대안으로 평가되고 있지만, DC 배전망용 기기들을 개발해야 하므로 DC 배전망의 구축비용은 경제적인 측면에서의 문제점을 가지고 있다. 따라서, 본 논문에서는 태양광전원의 수용을 위한 MVDC 배전망의 도입 타당성을 평가하기 위하여, 태양광전원 단지를 용량에 따라 규모별로 정의하고, 이를 수용하기 위한 배전망을 건설하는 경우에 대하여 규모별로 수용성 모델을 제시한다. 이 모델은 배전망의 전원공급방식에 따라 AC 및 DC 배전망으로 구분되며, 수용할 태양광전원 단지의 용량에 따라 수백 MW급은 대규모, 수십 MW급은 중규모, 수 MW급은 소규모로 정의된다. 또한, 본 논문에서는 AC 및 DC 배전망의 건설비, 전력변환설비의 교체비, 운용비로 구성된 비용요소와 태양광전원의 발전수익에 따른 전력량 요금 및 REC 요금으로 구성된 편익요소를 고려하여 MVDC 배전망의 경제성평가 모델링을 제시한다. 이를 바탕으로 현재가치 환산법과 원금균등상환 방식을 이용하여 MVDC 배전망의 경제성을 평가한 결과, 태양광전원의 수용 규모에 따라 일정 연계거리 이후에서는 DC 배전망의 구축비용이 기존의 AC 배전망보다 경제적임을 알 수 있어, 본 논문에서 제시한 경제성평가 모델링의 유용성을 확인하였다.

건조 하수슬러지의 석탄 혼소를 위한 연료특성 및 경제성 평가에 관한 연구 (A Study on the Evaluation of Fuel Characteristic and Economic Benefit for Co-combustion of Dried Sewage Sludge with Coal)

  • 본 연구에서는 건조 하수슬러지의 화석연료와의 혼소 가능성 평가를 연소특성, 경제적 이익 2가지 관점에서 평가하였다. 공업분석 결과 A하수처리장의 건조 하수슬러지의 휘발분은 36.48%, B하수처리장은 40.10%로 석탄에 비해 매우 높았으며, 연료비는 0.12, 0.14로 매우 낮아 연료로써의 품질은 낮은 것으로 조사되었다. 특히 연소성 지수가 30,000kcal/kg 이상으로 연료의 기준인 3,000~5,000kcal/kg을 크게 초과하여 건조 하수슬러지의 개질이 필요할 것으로 판단된다. 건조 하수슬러지의 경제성 평가 결과 석탄의 절감에 따른 이익은 연간 73.4억원, REC인증 이익은 56.4억원이 발생하는 것으로 산정되었다. 반면에 회분처리비용 증가에 따른 손실은 2.6억원, 건조 하수슬러지의 혼소에 소요되는 설비의 투자 및 운영비가 4.2억원으로 산정되어 건조 하수슬러지의 혼소에 따른 경제적 이익은 연간 123억원으로 추산되었다. 다만, 건조 하수슬러지의 생산에 소요되는 동력 및 건조 하수슬러지의 혼소로 인한 환경오염 등에 대한 비용은 본 연구에서 고려되지 못하였으며 향후 이에 대한 연구가 추가적으로 필요할 것이다.

A Case Study of Resolving Conflict in Energy Infrastructure Siting by the Solar PV Project

  • The growing demand for new energy infrastructure has often been encountered with the difficulties of siting in power plants and electric transmission lines. Siting such large-scale and complex facilities produces so many-sided issues that it is highly necessary to develop an approach to resolving the related problems and conflicts. This paper introduces how the stakeholders have handled the issues and resolved conflicts with residents opposed the construction of 765 kV transmission line. The solar photovoltaic power generation, called "Hee-Mang Sunlight Power project", is used for persuading residents to agree with constructing high-voltage transmission line and sharing benefits. It is considered how the project performance such as generation output and resident's profits is and proposed what the project should be revised and supplement. The project is shown that the intractable spiting in energy systems can be smartly resolved with cost-effective institutional solutions instead of relatively expensive technical ones.

Implications of Exonuclease Activity of Bacteriophage P2 Old Protein for Lambda Exclusion

  • Temperate bacteriophage P2 has a nonessential gene called old(overcoming lysoginization defection). In the presence of old, the growth of the host (Escherichia coli) with recBC- genotype is ingibited, and another bacteriophage, lambda, cannot superinfect. The Old protein has been shown to possess an exonuclease actibity. Three mutant P2s(old 1, old 17, old 49) which did gene was coned into expression vectors to produce hexahistidine-tagged proteins. The proteins were affinity-purified and shown to lose its exonuclease activity on both double-stranded and single-stranded DNA substrates. Thus, it was concluded that the lambda exclusion was related to Old's exonuclease activity.

IP기반 H.264 디코더 설계를 위한 동기화 파이프라인 최적화 (An optimization of synchronous pipeline design for IP-based H.264 decoder design)

  • This paper presents a synchronous pipeline design for IP-based H.264 decoding system. The first optimization for pipelining aims at efficiently resolving the data dependency due to motion compensation/intra prediction feedback data flow in H.264 decoder. The second one would enhance the efficiency of execution per each pipelining stage to explore the optimized latency and stage number. Thus, the 3 stage pipeline of CAVLD&ITQ|MC/IP&Rec.|DF is obtained to yield the best throughput and implementation. In experiments, it is found that the synchronous pipelined H.264 decoding system, based on existing IPs, could deal with Full HD video at 125.34MHz, in real time.

  • This paper presents nonlinear and parallel design for synchronous pipelining in IP-based H.264 decoder implementation. Since H.264 decoder includes the dataflow of feedback loop, the data dependency requires one NOP stage per pipelining latency to drop the throughput into 1/2. Further, it is found that, in execution time, the stage scheduled for MC is more occupied than that for CAVLD/ITQ/DF. The less efficient stage would be improved by nonlinear scheduling, while the fully-utilized stage could be accelerated by parallel scheduling of IP. The optimization yields 3 nonlinear {CAVLD&ITQ}|3 parallel (MC/IP&Rec.)| 3 nonlinear {DF} pipelined architecture for IP-based H.264 decoder. In experiments, the nonlinear and parallel pipelined H.264 decoder, including existing IPs, could deal with full HD video at 41.86MHz, in real time processing.

Integration of Headspace Solid Phase Micro-Extraction with Gas Chromatography for Quantitative Analysis of Formaldehyde

  • A study was carried out to evaluate the solid phase micro-extraction (SPME) for formaldehyde emission analysis of uncoated plywood. In SPME, formaldehyde was on-fiber derivatized through headspace extraction and analyzed by gas chromatography coupled with mass spectrometry (GC/MS). The SPME was compared with desiccators (DC-JAS 233), small-scale chamber (SSC-ASTM D6007) and liquid-liquid extraction (LLE-EPA 556) methods which were performed in accordance with their respective standards. Compared to SSC (RSD 4.3%) and LLE (RSD 5.0%), the SPME method showed better repeatability (RSD 1.8%) and not much difference from DC (RSD 1.4%). The SPME has proven to be highly precise (at 95% confidence level) with better recovery (REC 102%). Validation of the SPME method for formaldehyde quantitative analysis was evidenced. In addition, the SPME by air sampling directly from plywood specimens (SPME-W) correlated best with DC ($r^2$ = 0.983), followed by LLE ($r^2$ = 0.950) and SSC ($r^2$ = 0.935).

Bacillus amyloliquefaciens CJW15와 SSD8이 만드는 항균물질들의 특성 (Properties of Antimicrobial Substances Produced by Bacillus amyloliquefaciens CJW15 and Bacillus amyloliquefaciens SSD8)

  • 청국장에서 강력한 항균력을 지닌 2개의 Bacillus 균주들인 CJW15와 SSD8이 분리되었다. 16S rRNA와 recA 유전자들 염기서열 결정에 의해 두가지 균주 모두 B. amyloliquefaciens로 동정되었다. CJW15는 B. cereus ATCC14579, Listeria monocytogenes ATCC19111, Lactococcus lactis ATCC11454 들의 증식을 강력히 억제하며 SSD8은 B. cereus ATCC14579와 Enterococcus faecium ATCC19953 증식을 억제하였다. 두 균주들의 항균력은 $100^{\circ}C$, 15분 처리에도 감소하지 않았고 산성인 pH 3과 알칼리인 pH 12에서도 안정하였다. 트립신, 펩신, 프로테아제 K, 프로테아제 효소처리에 의해 CJW15 항균력은 변화가 없었지만 SSD8 항균력은 절반으로 감소하였다. 두 균주 공히 surfactin, fengycin, iturin, iturinA와 같은 lipopeptide 생합성 유전자들을 지니고 있고 subtilin과 같은 박테리오신 유전자들도 지니고 있다. 또 두 균주들은 혈전용해능을 지니고 있다.

Biological Functions of the COOH-Terminal Amino Acids of the $\alpha$-Subunit of Tethered Equine Chorionic Gonadotropin

  • Glycoprotein hormones have a common $\alpha$-subunit that is involved in the signaling pathway together with G protein, adenylcyclase and cAMP induction; however, it is an unclear how this common structure is related to hormonal action. To determine the biological functions of the COOH-terminal amino acids in the $\alpha$-subunit of these glycoprotein hormones, a tethered-molecule was constructed by fusing the $NH_2$-terminus of the $\alpha$-subunit to the COOH-terminus of the $\beta$-subunit of equine chorionic gonadotropin (eCG). The following deletion mutants were created by PCR; Ile was inserted at position 96 to form ${\Delta}96$, Lys was substituted at position 95 to form ${\Delta}95$, His was inserted at position 93 to form ${\Delta}93$ and Tyr was substituted at position 87 to form ${\Delta}87$. Each mutant was transfected into CHO-K1 cells. Tethered-wt eCG, and ${\Delta}96$, ${\Delta}95$, and ${\Delta}93$ mutants were efficiently secreted into the medium but the ${\Delta}87$ mutant was not secreted. Interestingly, the RT-PCR, real-time PCR, and northern blot analyses confirmed that the RNA was transcribed in the ${\Delta}87$ mutant. However, the ${\Delta}87$ mutant protein was not detected in the medium or the intracellular fraction of the cell lysates. The LH- and FSH-like activities of the recombinant proteins were assayed in terms of cAMP production using rat LH/CG and rat FSH receptors. The metabolic clearance rate (MCR) was determined by injecting rec-eCG (2 IU) into the tail vein. The ${\Delta}95$ and ${\Delta}93$ mutants were completely inactive in both the LH- and FSH-like activity assays. The ${\Delta}96$ mutant showed slight activity in the LH-like activity assay. In comparison to the wild type, the activity of the ${\Delta}96$ mutant in the FSH-like activity assay was the highest among all the mutants. The MCR assay in which rec-eCG was injected showed a peak at 10 min in all the treatment groups, which disappeared 4 h after injection. These results imply a direct interaction between the receptor and the COOH-terminal region of the a-subunit. The data also reveal a significant difference in the mechanism by which the eCG hormone interacts with the rLH and rFSH receptors. The COOH-terminal region of the $\alpha$-subunit is very important for the secretion and functioning of this hormone.
