Effects of Maturation Time on In-vitro Production of Korean Native Cow Embryos (체외성숙 시간이 한우 수정란 생산에 미치는 영향)

  • The present study was performed to investigate the first polar body(PB) extrusion during in-vitro maturation(IVM) and to examine the effect of different maturation time on the embryo development of Korean Native Cows(KNC) with regard to blastocyst(BL) cell numbers and pregnancy rates. PB extrusion did not take place for the first 12 hours(hr) of IVM, and most of KNC oocytes extruded PB from 14 to 20 hr after the onset of maturation. There was no significant difference in cleavage and 8-cell stage rates among the treatment groups, but BL and BL/8-cell rates were significantly higher(P<0.05) in 18 hr maturation group(31.0$\pm$5.7 and 82.0$\pm$5.1%) than 22 and 24 hr maturation group. The proportion of BL formed on day 7 and 8 was significantly higher(P<0.05) in 18 hr maturation group(85%) than in 24 hr maturation group(55%). There was a significant difference(P<0.05) in inner cell mass, trophectoderm and total cell number between day 7 BL produced by in-vivo and IVM 18 hr and day 8 BL produced by IVM 18 hr and 24 hr. Pregnancy rates are also significantly higher(P<0.05) in in-vivo(56.3%) and IVM 18 hr day 7(50.0%) group than day 8 treatment groups(18 hr: 16.7%, 24 hr: 10.5%). These results suggest that KNC oocytes achieve developmental competency within 20 hr of IVM, and "short" IVM (18 hr) is more effective than "long" IVM(24 hr) in embryo development rates, BL cell numbers and pregnancy rates.

  • The objective of this study was to determine physiological and mechanical factors effecting on the pregnancy rates following artificial insemination in dairy cattle. Estrus for artificial insemination of cows was checked whether the outflow of mucus from the elements was out flow or not. There were no significant difference of gestation rates with mucus release (31.76%) and without mucus release (24.03%). The pregnancy rates were 39.02% in 10~20mm of follicle size group and 27.08% (p<0.05) in 20~30mm diameter of the follicles. There were not different pregnancy rates between twice inseminated dairy cattle and more than 3 times inseminated cattle. The pregnancy rate was 28.57% under automatic milking system. In contrast, under artificial milking system pregnancy rate was 56.85%. Two systems significantly (p<0.05) were different. These results suggest that pregnancy rates were not effected by physiological system, but mechanical condition.

Effects of Hormones and Glucose Levels during the In Vitro Culture in Medium on In Vitro Fertilization and Developmental Rates of Porcine Oocytes (돼지 수정란의 체외수정 및 발생에 미치는 호르몬 및 Glucose 첨가의 영향에 관한 연구)

  • The study was conducted to determine the optimal hormone and glucose levels during the in vitro culture of bovine oocytes matured and fertilized in vitro for blastocyst development. Oocytes matured in TCM 199 + 10% FCS + hormones and glucose were fertilized in vitro in a TALP medium with swim up separated and heparin-treated epididymal cauda spermatozoa. Oocytes were cultured for 2~5 days in synthetic oviduct fluid medium (SOFM) supplemented with 10% FGS and with different hormone and glucose levels, and further cultured 5 days same medium in SOFM. The results are summarized as follows : The in vitro maturation and penetration rates of porcine oocytes cultured in TCM 199 media containing PMSG, hCG, PMSG + hCG, hCG + $\beta$ estradiol, PMSG + $\beta$ estradiol 0 to20 hours after insemination were 88.0% and 81.8%, 82.6% and 68.4%, 80.0% and 75.0%, 80.0% and 65.0%, 77.3% and 64.7%, respectively. The in vitro maturation and penetration rates of porcine oocytes cultured in TCM 199 media containing PMSG, hCG, PMSG + hCG, hCG + $\beta$ estradiol, PMSG + $\beta$ estradiol 20 to 40 hours after insemination were 92.0% and 87.0%, 92.0% and 82.6%, 91.3% and 81.0%, 85.2% and 73.9%, 87.5% and 81.0%, respectively. The cleavage and in vitro developmental rates to blastocyst of porcine oocytes cultured in TCM 199 media containing 0.05 mM, 0.10 mM, 0.30 mM, 0.50 mM, 1.00 mM, and 3.00 mM glucose lelvels 0~3 days after insemination were 31.5~48.1% and 10.0~16.7%, respectively. The cleavage and in vitro developmental rates to blastocyst of porcine oocytes cultured in TCM 199 media containing 0.05 mM, 0.10 mM, 0.30 mM, 0.50 mM, 1.00 mM, and 3.00 mM glucose levels 4~8 days after insemination were 30.0~53.8% and 8.7~19.2%, respectively. The cleavage and in vitro developmental rates to blastocyst were higher in TCM 199 media containing various glucose levels 0~3 days after insemination than 4~8 days.

The Effect of Various Concentrations of Taurine during In vitro Fertilization on the Development of Bovine Embryos Fertilized with Spermatozoa from Three Different Bulls

  • We investigated the effect of various concentrations of taurine during in vitro fertilization (IVF) on the embryonic development up to the blastocyst stage of bovine oocytes fertilized with three different Japanese Black bulls (Bull A, B and C). In vitro matured oocytes were fertilized with various concentrations of taurine (0, 1, 10, 50 and 100 mM) in the presence of 2.5 or 5.0 mM caffeine plus $25{\mu}g$/ml heparin (CH) for 6 hr or $100{\mu}g$/ml heparin (H) for $24{\pm}2$ h. After IVF, the cleavage rates from the 2 to 16 cell stage determined at 3 days and the development rates up to the blastocyst stage determined at 7-8 days from the onset of IVF were assessed. Although the cleavage rates for the taurine concentration groups were not significantly increased in any of the three bulls in the CH groups, the development rates up to the blastocyst stage of the 50 mM taurine group of Bulls A and B, and of the 1 to 50 mM groups of Bull C were increased (p<0.05) compared to those of the control (0 mM taurine) groups. On the other hand, none of the bulls in the H groups showed any significant increase either in the cleavage rates or blastocyst formation rates in any taurine concentrations groups compared with those of the control groups. These results indicate that the addition of 50 mM taurine to a fertilization medium containing caffeine and heparin may stimulate embryonic development up to the blastocyst stage when fertilized with different bull semen.

Optimum Feeding Rates in Juvenile Olive Flounder Paralichthys olivaceus Fed Practical Expanded Pellet at Low and High Water Temperatures (저수온기 및 고수온기에 있어서 배합사료를 공급한 넙치(Paralichthys olivaceus) 치어의 적정 공급율)

  • Two feeding trials were conducted to investigate the optimum feeding rates in juvenile olive founder Paralichthys olivaceus fed practical expanded pellet(EP) containing 58.1% crude protein, 10.7% crude lipid, and 10.8% ash at low and high water temperatures. In the first experiment, triplicate groups of 15 fish with average weight of 7.7 g were fed at one of seven feeding rates(0, 1, 1.5, 2, 2.25, or 2.5% of body weight/day or to satiation) for 4 weeks at low water temperature. In the second experiment, quadruplicate groups of 20 fish with average weight of 5.5 g were fed at one of eight feeding rates(0, 3, 3.5, 4, 4.25, 4.5, or 4.75% of body weight/day or to satiation) for 4 weeks at high water temperature. Based on growth performance, we estimated that the optimum feeding rates for juvenile olive flounder were 1.97-2.51% and 4.82-6.36% of body weight/day at low and high water temperatures, respectively.

Analyses of subsurface drainage effects of farmland with respect to pipe and envelop material (관재료 및 피복재료별 농경지 암거배수 효과 분석)

  • Analyses of subsurface drainage effects of farmland with respect to pipe and envelop material are made by the laboratory experiments using soil box to give basic information for the subsurface drainage system planning and design. Three different diameter PVC perforated pipes and a mesh pipe are used with envelop materials such as sand, rice bran, and crushed stone. Steady state subsurface drainage flow rate increased as envelop material changed from sand to rice bran and crushed stone. This indicates that as the hydraulic conductivity of the envelop material increases, the drainage flow rate increases. On the other hand, for a given envelop material, the mesh pipe which has the largest openning area shows the largest flow rate while small diameter PVC pipes show small flow rates. This tells that as the openning area and pipe diameter increase, the flow rate increases, too. Therefore, selection of pipe and envelop material should be made in accordance with the design drainage flow rate. Unsteady state subsurface drainage flow rate with respect to time differs for different envelop material. In case the sand was used as an envelop material, the small diameter PVC pipes show larger flow rates than the large diameter PVC pipe and mesh pipe. When the rice bran was used, the mesh pipe shows the largest flow rate, while small diameter pipes show smaller flow rates. In case the crushed stone was used as an envelop material, the large diameter PVC pipe and mesh pipe show larger flow rates, while small diameter pipes show a little bit smaller flow rates. However, the variation of flow rates among different pipes is the smallest when the crushed stone is used. The flow rate curve with respect to the pipe changes little for the crushed stone envelop which has a large hydraulic conductivity, while that changes much for the sand and rice bran envelops. However, it is difficult to draw a consistent relationship between the drainage flow rate and pipe for all the envelop materials. Since the subsurface drainage experiments are made only under the restricted laboratory condition in this study, further study including field experiment is required.

The Effect of Health Care Program for Newly Married Couple in a Community Health Care Center (일 보건소의 신혼부부 건강프로그램의 효과)

  • Purpose: The research purpose was analysis of the effect of a health education program for newly-married executed from 2003 to 2005 as a part of health care service from community health center and providing the fundamental source for health education needed during newly-married. Method: The data collection with structured questionnaires was conducted during October to December in 2006. From the health center 106 brides who participate the program and 130 brides who didn't participate. Health care program for newly married couple were composed health examinations and health educations. Health examinations were CBC, LFT and Ag & Ab prevallence rates of hepatitis B, rubella. Health educations were done two times, the first individual health education was done at enrolled in health center for establishment of desirable couple relationship, family planning, contraception method and management of pre-pregnancy. The second individual health education was done for explanation of blood examination results, prevention of the congenital deformity and the vertical transmission of hepatitis B, management for pregnancy, breast feeding method, introduction to safety delivery method and encouraging self-study using by materials made by health center after two weeks at revisiting health center. SPSS/PC(ver. 12.0) and $X^2$-test, t-test was used to analyze the collected and tabulated data. Socio-demographic characteristics and regional characteristics of residence area of two groups shows no significant difference. Result: As a variables of experimental effect. The natural childbirth rates of participants group was significantly higher than Non-participants group(p=.012). Breast feeding rates for participants group was also statistically significant higher than Non-participants group. Contraception rates showed no significant difference. The rubella vaccination rates (p=.001) and The hepatitis B vaccination rates (p=.012) shows statistical differences. Conclusion: Therefore health care program for newly married couple in a community health care center was effective generally. We supposed that explore participation ways for nonparticipants, expand the program to community health care center in whole country.

An Analysis of Effects of Changes in Foreign Exchange Rates on the Domestic Energy Prices : Diesel, Heavy Oil, and LNG (환율변동이 국내 에너지가격에 미치는 영향 분석 : 경유, 중유, LNG를 중심으로)

  • Oil and LNG products are characterized by the facts that the raw materials are all imported and financing is dependent heavily upon foreign countries. This makes the oil and LNG products sensitive to changes in foreign exchange rates. However, the extent to which they respond to changes in foreign exchange rates, particularly the extent of price changes, vary considerably, due to the differences in the structures of price determination. The purposes of this paper are twofolds. The first one is to analyze the structures of price determination of diesel, heavy oil, and LNG. The second one is to analyze the effects of changes in foreign exchange rates on the prices of and price competitiveness of the fuels in question through the sensitivity analysis. The results of the sensitivity analysis indicate that diesel price is most sensitive and heavy oil price is least sensitive to changes in foreign exchange rates.

Effect of Cytochalasin B Treatment on the Improvement of Survival Rate in Vitrified Pig Oocyte

  • To improve survival rates of vitrified pig oocytes, the treatment of cytoskeletal stabilizer on an appropriate time is one of the possible approaches. However, the exact treatment timing and effect of cytoskeletal stabilizer such as cytochalasin B (CB) is not well known during oocyte vitrification procedures. Thus, the present study was conducted to determine optimal treatment timing of CB during vitrification and warming procedures. In experiment 1, the survival rates of the postwarming pig oocytes were analyzed by fluorescein diacetate (FDA) assays with 4 classifications. In results, post-warming oocytes showed significantly (p<0.05) decreased number of alive oocytes (31.8% vs. 86.4%) compared to fresh control. In detail, the significant difference (p<0.05) was found only in strong fluorescence (18.2% vs. 70.5%) not in intermediate fluorescence groups (13.6% vs. 15.9%). In experiment 2, CB was treated before (CB-Vitri) and after (Vitri-CB) vitrification. In results, group of Vitri-CB showed significantly (p<0.05) higher (91.6%) survival rates compared to group of CB-Vitri (83.7%), significantly (p<0.05) and comparable with group of Vitri Control (88.7%) by morphological inspection. In FDA assay results, group of Vitri-CB showed significantly (p<0.05) higher (44.2%) survival rates compared to groups of CB-Vitri (36.7%) and Vitri Control (35.1%). In conclusion, the increased survival rates of post-warming pig oocyte treated with Vitri-CB method are firstly described here. The main finding of present study is that the CB treatment during recovery could be helpful to refresh the post-warming pig oocyte resulting its improved survival rates.

The Effects of Varying Sampling Flow Rates on the Measurements of Total Nitrate and Sulfate in Dry Acid Deposition

  • One technique for determining dry acid deposition fluxes involves measurement of time - averaged ambient concentrations of dry acid deposition species using filter packs (FP) coupled with estimates of mean deposition velocities for the exposure period. A critical problem associated with filter pack data comparisons between various field sampling networks is the use of diverse sampling flow rates and duration protocols. Field experiments were conducted to evaluate the effects of varying sampling flow rates, from 1.5 to 10 standard liters per minute, on total nitrate and sulfate measurements of specific dry acid deposition species . Collocated FP samplers were used to determine sampling and analysis data reproducibility and representativeness . Ambient air samples were simultaneously collected using groups of filter packs operated at various flow rates over identical 7 day periods. The species measured were sulfur dioxide, particulate sulfate , nitric acid and particulate nitrate. Statistical results (ANOVA; alpha level 5%) showed that neither the low nor high sampling flow rates caused a significant difference in the measurements of total sulfate and adjusted total nitrate (ATN) . However, it was concluded that for high flow rate sampling measurements, total nitrate (TN) could be affected during extended sampling durations because of potential nitric acid overloading and breakthrough. Although the previous workers (Costello, 1990; Quillian, 1990) used much higher sampling flow rates (~ 17 sLpm) than employed here, it was assumed that for a high loading (> 50$\mu\textrm{g}$ HNO$_3$) of nitric acid on the Nylon filters, a significant fraction (~10%) of nitric acid could pass through the Nylon filters and be collected on the carbonate impregnated filters. It was concluded that even at the highest sampling flow rate employed (10 sLpm) at the Cary Forest site, nitric acid breakthrough was less than 10% of the total HNO$_3$ collected. However, for a heavily polluted urban airshed or with longer sampling times , higher filter loadings could result in substantial nitric acid breakthrough and HNO$_3$concentrations would be underestimated.
