유제품에 들어 있는 우유와 산양유를 동정하기 위해 미토콘드리아의 12S rRNA 유전자를 목표로 하는 primer을 이용하는 duplex PCR 분석을 적용하였다. 소와 산양의 특이성 primer을 이용한 duplex PCR 분석은 우유와 산양유 DNA에 대해 각각 233 bp와 326 bp의 특이성 단편을 나타냈다. Duplex PCR 분석이 라벨에 표시된 성분을 확인하기위하여 시중마트에서 구입한 15개 유제품에 적용하였다. Duplex PCR 분석 결과 4개 시유, 3개 요구르트, 1개 전지분유는 표시된 성분과 완전히 일치하였다. 그러나 7개의 조제분유 중 5개만 표시성분과 일치하고 2개 조제분유제품은 산양유와 우유가 각각 오염되어 있는 것으로 나타났다. 제안된 duplex PCR 분석은 산양유에 들어있는 우유를 0.1%까지 측정할 수 있는 민감하고 신속한 방법이다. Duplex PCR 분석은 유제품 속에 들어있는 우유와 산양유를 one-step 방법으로 동시에 탐지할 수 있다.
대장균군 검사용 간이 시험지는 본 실험실에서 국내 최초로 고안, 개발하였으며 이 간이 시험지법은 현장 검사법의 하나로 대장균군이 내는 succinic acid dehydrogenase 때문에 tetrazolium salt가 환원되어 적색 반점을 형성하는 것을 이용한 방법으로서 이 간이 시험지의 제조는 대체로 종래의 표준 평판법과 거의 동일한 조성의 배지와 시약을 사용하여 여지에 흡착시킨 후, 건조시켜($60^\circ$C) 멸균한 것으로 표준 평판법과 어떤 상관관계가 있는가를 검토하였다. 이 간이 시험지의 제조에서는 bile salt No. 3를 deoxycholate로 대체하여 제조 원가를 절감하였고, 또한 일본에서 현재 시판되고 있는 제품과 품질 비교시험을 하여 더 좋은 결과를 얻었으며 종래의 표준 평판법과 비교하였을 때도 오히려 표준 평판법 (24-48시간 배양)보다 빠른 시간(16-20시간 배양)내에 판정할 수 있는 이 점이 있으며, 표준 평판법에서는 없어서는 안될 배지나 배양 접시, pipette등의 자료 및 기구가 일체 필요없고 언제 어디서나 현장에서 직접 시험할 수 있어 매우 간편하며 또한 저렴한 가격으로 제조 할 수 있는 경제성이 높은 이점을 갖고 있다.
Outbreaks of foodborne diseases associated with Vibrio species such as V. parahaemolyticus, V. vulnificus, and V. cholerae frequently occur in countries having a dietary habit of raw seafood consumption. For rapid identification of different Vibrio species involved in foodborne diseases, whole-cell protein pattern analysis for 13 type strains of 12 Vibrio species was performed using SDS-PAGE analysis. Pathogenic Vibrio species such as V. parahaemolyticus, V. vulnificus, V. cholerae, V. alginolyticus, V. fluvialis, and V. mimicus were included in the 12 Vibrio species used in this study. Each of the 12 Vibrio species showed clearly specific band patterns of its own. Two different strains of V. parahaemolyticus showed two different SDS-PAGE whole-cell protein patterns, giving the possibility of categorizing isolated strains in the same V. parahaemolyticus species into two subgroups. The 36 Vibrio isolates collected from sushi restaurants in Busan were all identified as V. parahaemolyticus by comparing their protein patterns with those of Vibrio type strains. The identified isolates were categorized into two different subgroups of V. parahaemolyticus. The whole-cell protein pattern analysis by SDS-PAGE can be used as a specific, rapid, and simple identification method for Vibrio spp. involved in foodborne diseases at the subspecies level.
Ground cherry (Physalis pubescens) is the most prominent species in the Solanaceae family due to its nutritional content, and prospective health advantages. It is grown all over the world, but notably in northern China. In 2019 firstly bacterial leaf spot (BLS) disease was identified on P. pubescens in China that caused by both BLS pathogens Xanthomonas euvesicatoria pv. euvesicatoria resulted in substantial monetary losses. Here, we compared whole genome sequences of X. euvesicatoria to other Xanthomonas species that caused BLS diseases for high similarities and dissimilarities in genomic sequences through average nucleotide identity (ANI) and BLAST comparison. Molecular techniques and phylogenetic trees were adopted to detect X. euvesicatoria on P. pubescens using recQ, hrpB1, and hrpB2 genes for efficient and precise identification. For rapid molecular detection of X. euvesicatoria, loop-mediated isothermal amplification, polymerase chain reaction (PCR), and real-time PCR techniques were used. Whole genome comparison results showed that the genome of X. euvesicatoria was more closely relative to X. perforans than X. vesicatoria, and X. gardneri with 98%, 84%, and 86% ANI, respectively. All infected leaves of P. pubescens found positive amplification, and negative controls did not show amplification. The findings of evolutionary history revealed that isolated strains XeC10RQ, XeH9RQ, XeA10RQ, and XeB10RQ that originated from China were closely relative and highly homologous to the X. euvesicatoria. This research provides information to researchers on genomic variation in BLS pathogens, and further molecular evolution and identification of X. euvesicatoria using the unique target recQ gene through advance molecular approaches.
현대 사회에서의 디지털 데이터의 빠른 증가로 디지털 포렌식이 핵심적인 역할을 하고 있으며, 파일 유형 식별은 그 중에서 중요한 부분 중 하나이다. 파일 유형을 빠르고 정확하게 식별하기 위해서 인공지능을 사용한 파일 유형 식별 모델 개발 연구가 진행되고 있다. 그러나 기존 연구들은 일부 국내 점유율이 높은 파일을 식별할 수 없어, 국내에서 사용하기에 부족함이 있다. 따라서 본 논문에서는 CNN과 GRU를 활용한 더욱 정확하고 강력한 파일 유형 식별 모델을 제안한다. 기존 방법의 한계를 극복하기 위해 제안한 모델은 FFT-75 데이터셋에서 가장 우수한 성능을 보이며, 국내에서 높은 점유율을 가지는 HWP, ALZ, EGG와 같은 파일 유형도 효과적으로 식별할 수 있다. 제안한 모델과 세 개의 기존 연구 모델(CNN-CO, FiFTy, CNN-LSTM)을 서로 비교하여 모델 성능을 검증하였다. 최종적으로 CNN과 GRU 기반의 파일 유형 식별 및 분류 모델은 512바이트 파일 조각에서 68.2%의 정확도를, 4096바이트 파일 조각에서는 81.4%의 정확도를 달성하였다.
A PCR assay for the rapid detection of Vibrio vulnificus strains was developed using a virulence gene for elastase found in various Vibrio species. The DNA sequences in the elastase gene facilitated the identification of a species-specific probe for pathogenic V. vulnificus strains from both clinical and environmental sources. Using an elastase gene-based PCR reaction, a species-specific 507-bp PCR product was visualized by agarose gel electrophoresis. Three different DNA extraction methods were then compared to improve the simplicity and rapidity of detection. A PCR assay using the conventional DNA extraction or boiling method was able to detect as few as 25 V. vulnificus cells, making the detection limits at least 1-log-scale lower than that for the EDT A-treated DNA extraction method. In particular, the boiling method, which does not require purification of the chromosomal DNA, was very effective in terms of simple and rapid detection. Meanwhile, the detection limit in a mixed bacterial culture that included other bacteria, such as Escherichia coli or Bacillus subtilis, was two V. vulnificus cells, which was 1-log-scale lower than that for the control. Accordingly, when coupled with a new DNA extraction method, the elastase gene-based PCR can provide a rapid, specific, and sensitive method for identifying V. vulnificus in clinical and environmental samples.

Interests in the field of rapid methods and automation in microbiology have been growing steadily on an international scale in recent years. International meetings concerned this problem have been held in elsewhere in the world countries since the past twenty years. But, unfortunately in the field of microbial examination in food hygiene, this problem have not yet been developed so much as in the field of clinical microbiology. Today, I would like to introduce you here present aspects of rapid and automation technologies, those which are manly carrying in milk and meats industries. My illustration will be given recent improved technologies using automatic apparatus and instruments along with process of microbial count procedure. Recent direct microbiological counting system (ChemeScan \ulcorner) as real time ultrasensitive analysis created by Cheminex Ltd., France is now most evolutional instrument to provide direct microbial counts, down to one cell, within 30 minutes. The results from these evaluations how a good correlation between the ChemScan system and the standard plate count method. This system will be successful application for not only in the field of pharmacology but also food microbiology. In addition, current identification of microbes by sophisticated instruments suitable for food microbiology, one of which Biology is manual system (BIOLOG\ulcorner), provides reference-level capability at a modes price. For the manual system, the color reactions in the microplate are read by eye and manually keyed into personal computer. Species identification appears on the computer screen within seconds, along with biotype patterns, a list of closely related species, and other useful statistics. In present this is useful application for microbial ecology and epidemiological survey. RiboPrinter system newly produced by DuPont is now focusing among microbiologists in the world, and is one of the biggest microbial characterization system using a DNA-based approach. The technology analyzer is bacterial culture for its genetic fingerprint or riboprint pattern. Finally Bio-cellTracer system for automatic measurement of fungal growth and Fukitori-Maseter, a Surface Hygiene Monitoring Kit by using swabe procedure in food processing environment are briefly illustrated in this presentation.
Objectives : Paeng-hwal is described as an insect herbal medicine used for digestive diseases in the Dong-ui-bo-gam. The origin of this herbal medicine is limited to several small crabs, such as Helice tridens. These crab species cohabitat in the same environment and share similar morphological characteristics, making it very difficult to distinguish and collect the individual species for use in dietary supplements or herbal medicines. This study was conducted to develop a genetic identification tool for discriminating among these closely related small crab species. Methods : CO1 DNA barcode regions of 15 samples from 6 species of small crabs were analyzed to obtain the individual sequences. To identify the correct species, comparative analyses were carried out using the database of the NCBI GenBank and the NIBR. SCAR primers were designed to develop simple and rapid assay methods using inter-species specific sequences. Optimal SCAR assay conditions were established through gradient PCR, and the limit of detection (LOD) was determined. Results : Six species of small crabs (Helicana tridens, Macrophthalmus abbreviatus, Helicana tientsinensis, Helicana wuana, Chiromantes dehaani, and Hemigrapsus penicillatus), which are distributed as Paeng-hwal, were identified through CO1 sequences analysis. We also developed SCAR markers to distinguish between six small crabs at the species level. Furthermore, we established the optimal PCR assay methods and the LOD of each individual species. Conclusions : The rapid and simple SCAR-PCR assay methods were developed to identify the species and control the quality of herbal medicines for Paeng-hwal based on the genetic analyses of CO1 DNA barcodes.
본 연구에서는 계대배양과 세균 동정 시험에 소요되는 시간을 단축하고, 혈류감염의 새로운 검사 방법을 모색하여 간단하고 신속 정확한 동정 결과를 도출할 목적으로 질량분석기를 이용하여 다음과 같은 결과를 얻었다. 혈액배양에서 한 가지 세균만 배양된 검체는 총 254개였으며, Vitek 2에서 208주(81.8%)의 세균을 동정되었으며, 45주는 동정되지 않았다. 동정된 세균 중 그람양성 세균이 146개(57.5%), 그람음성 세균이 108개(42.5%)이었다. 전체적으로 233개는 종(species) 수준까지 동정 되었으며, 21개는 속(genus)수준까지 동정 되었다. 동정 오류는 Propionibacterium acnes를 Clostridium bifermentans로 동정 되었다. 균종별로는 enterobacteriaceae, glucose non-fermentative bacilli (GNFB), staphylococci의 정확도는 각각 81/83 (97.6%), 12/15 (80.0%), 72/85 (84.7%)로 나타났다. Vitek 2에 의한 표준법과 Vitek MS에 의한 직접법에 의한 동정의 일치율은 81.8%이었으며, 45주는 동정되지 않았다. 동정되지 않은 세균들의 대부분은 그람양성 세균(n=37)이었고, 그람양성 세균은 streptococci (14), coagulase-negative staphylococci (CNS) (11), enterococci (3), Staphylococcus aureus (2), Micrococcus spp. (2), Bacillus spp. (2) 그리고 Actinomyces odontolyticus, Finegoldia mag na, Peptostreptococcus spp.가 각각 1건씩 이었다. 결과 보고 시간은 기존의 검사 방법보다 24~72시간까지 단축되었다. 산소성 배양과 무산소성 배양 사이의 동정률의 차이는 없었으나 무산소성 배양을 사용하면 용해 과정이 필요 없어 검체 준비 시간을 단축할 수 있었다. 이상의 연구 결과로 혈액배양병에서 직접 동정하는 방법은 정확하고 결과 보고 시간이 신속하여 환자의 치료에 매우 유용할 것이라 생각된다. 향후 추가적인 연구에서는 본 연구에서 정확성이 부족했던 사슬알균(streptococci)과 혈장응고효소 음성 포도알균(CNS)을 동정하기 위한 방법을 더욱 개선할 필요가 있을 것으로 사료된다.
시대의 환경적 변화에 따른 소프트웨어 개발의 발달은 빠른 개발과 높은 생산성을 향상시키기 위한 소프트웨어의 재사용 기술의 확산으로 컴포넌트 기반 개발 방법론이 널리 사용되기 시작했다. 이러한 컴포넌트 기반 개발에서 재사용 가능한 독립적인 컴포넌트의 식별은 컴포넌트 기반 시스템 구축을 위하여 가장 중요한 작업이다. 컴포넌트 식별 방법을 제시하고 있는 기존 방법론들에서는 비즈니스 컴포넌트를 식별함에 있어서 개발자의 경험적 토대를 기반으로 독립적인 컴포넌트를 식별하도록 제시하고 있으므로 평이한 개발자에 의한 비즈니스 컴포넌트 식별이 쉽지 않은 문제점을 가지고 있다. 따라서 본 논문에서는 시스템 컴포넌트를 먼저 식별한 후 비즈니스 컴포넌트를 식별하고 비즈니스 컴포넌트를 식별하기 위하여 분석 클래스 간의 메소드 호출 유형과 메소드 호출 방향에 의한 클래스 간의 종속적 특성과 의존의 강도를 부여하여 효율적으로 컴포넌트를 식별할 수 있는 기준과 방법을 제안한다. 또한 사례 연구를 통하여 시스템 컴포넌트를 중심으로 비즈니스 컴포넌트가 효율적으로 식별됨을 검증한다.
