Antioxidative Activity of Ethanol Extract from Korean Medicinal Plants (국내산 약용식물 추출물의 항산화 효과 검색과 용매 분획물의 비교)

  • To evaluate the antioxidative activity of the ethanol extracts from 116 domestic medicinal plants, each extract was added to palm oil and lard in 1,000 ppm level, and the induction time was measured by Rancimat method. Almost all plant extracts tested showed more or less antioxidative activity. The extracts which had strong antioxidative activity to palm oil and lard were Caesalpinia sappan L., Paeonia japonica Miyabe et Takeda, Dendrobium moniliforme (L.) Sw and Crcumae longa L. These extracts were fractionated with chloroform, ethyl acetate, butanol and water. Ethyl acetate layer of Caesalpinia sappan L. and Paeonia japonica Miyabe et Takeda showed marked antioxidative activity, and chloroform layer of Dendrobium moniliforme (L.) Sw and Crcumae longa L. ethanol extract had stronger antioxidative activity than all the other layers.

  • Antioxidative activity of the extract from Caesalpinia sappan L. by various solvent was compared with several commercial antioxidants, using the Rancimat method. AI (antioxidative index; induction period of oil containing extract/induction period of control oil) of all extracts were higher than commercial antioxidants, such as BHA, ${\delta}-tocopherol$ and ascorbic acid. The ethanol extract was fractionated by liquid liquid extraction. Ethyl acetate fraction showed higher AI than the whole crude extract. When comparing POV and TBA value of palm oil and lard containing different level of each fraction, the oxidative stability of ethyl acetate fraction at 200 ppm level on palm oil and lard were similar to that of BHT at 200 ppm level, and better than BHA, ${\delta}-tocopherol$ and control.

  • Antioxidant activities of ethanol and water extracts of flesh, leaves, and peels of green and purple Kohlrabi were measured by rancimat method, total phenolic compound (TPC), DPPH radical scavenging effect, chelating effect, and reducing power analysis. The highest TPC was observed in ethanol extract from green peel and water extract from purple leaf with 12.00 and 11.70 mg/g of dry sample, respectively. The ethanol extracts from purple Kohlrabi leaf and peel exhibited strong DPPH radical scavenging effects with reducing power while water extracts from purple Kohlrabi leaf and peel also showed strong DPPH radical scavenging with chelating effects. Antioxidant index of ethanol and water extracts from green and purple Kohlrabi measured by rancimat was lower than butylated hydroxytoluene. These results suggest that extracts from purple Kohlrabi leaf and peel exhibited higher antioxidant activities than those of green Kohlrabi, and can be potentially used as proper natural antioxidants.

Isolation of Antioxidative Components from the Bark of Rhus verniciflua STOKES Screened from Some Chinese Medicinal Plants (한약재로부터 선발된 옻나무 수피 추출물로부터 항산화 활성물질의 분리)

  • To develop new natural antioxidants, antioxidative activity of ethanol (75%) extracts from 50 edible or medicinal plants were examined on lard and palm oil by Rancimat method ($120^{\circ}C$, 20 L/hr). The extracts from Rhus verviciflua STOKES showed comparatively strong antioxidative activity on test. Of the solvents used for extraction, chloroform extract exhibited the strongest antioxidant activity. AI (antioxidant index: induction period of oil containing extract/induction period of control oil) of chloroform extract was higher than that of commercial antioxidant, such as BHT, BHA and ${\delta}-tocopherol$. Free phenolic acid fraction (200 ppm) of the chloroform extract from 75% EtOH extract of Rhus verniciflua STOKES (RCF) showed stronger activity than that of BHT, BHA, and ${\delta}-tocopherol$ at the same concentration. RCF-11 and RCF-13 fractions separated by silicagel column chromatography from the RCF showed stronger activity than other fractions by the Rancimat method.

The Oxidation Stability and Flavor Acceptability of Oil from Roasted Soybean (볶은 대두로부터 착유한 대두유의 산화 안정성 및 향기 기호도)

  • The effects of oxidation stability and flavor acceptability of soybean oil by roasting and refining treatment were investigated in this study. The peroxide values of crude oil from unroasted soybean(COUS) were more increased than those of crude oil from unroasted soybean(CORS) during storage under fluorescence light at $45^{\circ}C$. Induction time by the Rancimat Method was 44.9 and above 88.7 hours on COUS and CORS respectively. The order of oxidation stability of oil was crude>degummed>alkali-refining>deodorized>bleached oil in terms of peroxide value, while that of oxidation stability of oil was crude>degummed>deodorized>alkali-refining>bleached oil in regard to induction time by the Rancimat Method. The correlation coefficients between induction time and two charateristics(absorbance of browning reaction products and phosphorous contents) were highly significant, while that between induction time and tocopherol contents was not high during refining stages. The scores for the sensory evaluation of flavor on sesame oil and CORS was 9.269 and 8.269 respectively, but it was not significant between two oils.

Identification of Antioxidative Components from Ethanol Extracts of Dalbergia odorifera T.CHEN (강진향(Dalbergia odorifera T.CHEN) 에탄올 추출물로부터 항산화 활성물질의 구조동정)

  • The chloroform layer from 75% ethanol extract of Dalbergia odorifera T.CHEN showed strong antioxidative activity on lard and palm oil as tested by Rancimat method. Antioxidative active compound isolated and identified by silica gel column chromatography, thin layer chromatography, mass spectrophotometer, $^1H-NMR$ and $^{13}C-NMR$ was identified as mucronulatol (3(R&S)-3,7-Dihydroxy-2',4-dimethoxyisoflavan). Results of Rancimat method revealed the induction period of Mucronulatol increased longer than those of synthetic antioxidant, BHA and BHT, at the same concentration. Mucronulatol combined with ${\delta}-tocopherol(200ppm)$, and with ascorbic acid (200 ppm) and citric acid (200 ppm) on lard and palm oil, respectively, showed strong synergistic effects.

  • The free phenolic acid fraction of the chloroform extract from 75% ethanol extract of Rhus verniciflua STOKES (RCF) showed stronger antioxidative activity than BHT, BHA and ${\delta}-tocopherol$ at the same concentration. RCF components were isolated and identified by silica gel column chromatography, thin layer chromatography, mass spectrometer and $^1H-NMR\;and\;^{13}C-NMR$. The antioxidative activity was confirmed by electron donating activity, Rancimat method and thiobarbituric acid test in liposome system. RCF-11 could be further separated into three fractions. The antioxidative active compounds were purified and identified as gallic acid, butin and butein. The RCF-13 was purified and identified as sulfuretin.

Screening of the Physiological Activity of Solvent Extracts of Paulownia coreana Bark and Antioxidative Effect of the Extracts on an Edible Oil

  • Paulownia coreana is a medicinal, edible and industrial plant with the largest leaf, and is native to Korea. We evaluated the reducing power activities, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities (RSAs), nitrite scavenging activities (NSAs) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation RSAs by solvent extraction of P. coreana bark by using $50^{\circ}C$ hot water and $25^{\circ}C$ methanol. The antioxidative effect of P. coreana bark extract on corn seed oil was evaluated using the Rancimat test. The extraction yields on dry weight basis with 15 folds of hot water and methanol were 23.88% and 5.30%, respectively; further the flavonoid content in the hot water extract was over 2.5 times more than that in the methanol extract. The DPPH RSA of the methanol extract was substantially higher than that of the water extract, whereas the NSA of the water extract was higher than that of the methanol extract at pH 1.2. The ABTS RSAs of the two extracts were almost the same as that of ascorbic acid and 2,6-ditertiarybutyl hydroxytoluene. The two extracts of P. coreana bark in this study were found to slightly improve the oxidation stability of corn seed oil.

Antioxidative and Radical Scavenging Properties of Extracts from Geum japonicum

  • Extracts from Geum japonicum (Bammu in Korean) were analyzed for their antioxidative activities and scavenging effects on free radicals. The ethyl acetate fraction of G. japonicum methanol extract (EFGJ) showed a remarkable scavenging activity on the 1,1-diphenyl-2-picrylhydrazyl radical. EFGJ also showed excellent antioxidative activity on linoleic acid during long-period storage and on rat liver microsome peroxidation system, and good anti-peroxidation effect on lipid in Rancimat system using lard, palm oil, and perilla oil, as compared with BHT and ${\alpha}$-tocopherol. Varying effects of antioxidative activity of the EFGJ on various fatty acids were observed. The prevention of linoleic acid and linolenic acid peroxidation was superior to ${\alpha}$-tocopherol, but inferior to BHT. The methanol extract of G. japonicum did not show mutagenicity as revealed through SOS chromotest. Based on the results, the extracts of G. japonicum may be useful source as natural antioxidants.

Allelopathic Potential and Antioxidant Activity of Leaf Extracts from Several Wild Plant Species

  • Several wild plant species are known to contain biologically active substances that are allelopathic to weed species as well as antioxidant to foods. Plant extracts or residues from leaves of 4 species, Achyranthes japonica (speedwell), Cucumis sativus (Cucumber), Trifolium repens (white clover), and Vicia angustifolia (narrowleaf vetch) were bioassayed against Medicago sativa (alfalfa) or Echinochloa crus-galli (barnyard grass) to determine their allelopathic effects, and used for measurement of antioxidant activities. The aqueous extracts applied on filter paper significantly inhibited root growth of alfalfa. Aqueous extracts or residues from V. angustifolia showed the most inhibitory effect on alfalfa or barnyard grass seedling growth and followed by A. japonica and T. repens. Oxidative stability by Rancimat method, antioxidant activity by TBA (2-thiobarbituric acid) method and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity for the ground samples or methanol extracts were the greatest in V. angustifolia, although were less than those of commonly used antioxidants, BHT (butylated hydroxytoluene) and ascorbic acid. These results suggest that the wild plant species had potent allelopathic and antioxidant activities, and that their activities differed depending on plant species.
