Antioxidant Effects and Inhibitory Effect on NO Synthesis by Extracts of Canavalia lineata (해녀콩 (Canavalia lineata (THUNB.) DC.) 추출물의 항산화 효과 및 NO 생성 억제 효과)

  • Scavenging effects of DPPH radical and hydroxyl radical, inhibition of linoleic acid oxidation, inhibition of NO synthesis and iNOS expression were tested with extracts and chromatographic subfractions of Canavalia lineata obtained at Jeju island. Chloroform extract and its subfractions gave moderate effects on scavenging DPPH radical and hydroxyl radical, They also inhibited linoleic acid oxidation, and NO synthesis. Inhibition of NO synthesis resulted from the repression of iNOS gene expression. Ethyl acetate extract and its subfractions showed excellent effects on scavenging DPPH radical and hydroxyl radical, while they were cytotoxic.

Free radical Scavenging Activities and Inhibitory Effects on Xanthine oxidase of Buckwheat (Suwon No. 5) (수원 5호 메밀의 유리라디칼 소거 및 Xanthine oxidase 활성 저해)

  • To examine the free radical scavenging activities from buckwheat, Suwon No. 5 was extracted with various solvents. The seed of Suwon No. 5 had a high contents of carbohydrate, but the vegetable of that had a 31.4% of protein, 28.6% of ash and 25.7% carbohydrate as moisture free basis. Phenols contents of vegetable extracts had higher than those of seed extracts. Acetone extract of vegetable showed the greatest electron donating ability. Butanol and acetone extracts of vegetable showed high inhibitory effect on lipid peroxidation. Acetone extract of vegetable has also excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effects of extracts on xanthine oxidase were measured. Acetone extract had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was 2.2$\mu\textrm{g}$.

Fermented Saccharina japonica with Phellinus linteus Mycelia Enhances its Radical Scavenging Activities (상황버섯 균사체로 발효한 다시마 추출물의 항산화 활성)

  • To develop new functional materials, Saccharina japonica (S. japonica) was cultivated with Phellinus linteus (P. linteus) mycelia by solid culture. S. japonica was fermented with P. linteus mycelia. The various radical scavenging activities of extracts of fermented S. japonica with P. linteus mycelia (FSPM) were investigated and compared to the antioxidant capabilities of unfermented S. japonica and P. linteus mycelia. The antioxidant activities of FSPM extracts were evaluated using ferric reducing antioxidant power (FRAP) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. FSPM showed stronger free radical scavenging abilities than P. linteus mycelia or S. japonica alone. These results indicate that fermentation of S. japonica changes its chemical nature and could provide beneficial antioxidant effects.

Enhanced DPPH Radical Scavenging Activity of Lactobacillus plantarum K-21 Isolated from Kimchi and its Various Antioxidant Effects (김치유래 Lactobacillus plantarum K-21의 DPPH 라디칼 제거활성 증진 및 다양한 항산화 효과)

  • Lactic Acid Bacteria (LAB) are among the representative probiotics that have been used for a long time in fermented food. Although there are many studies on detecting the radical scavenging activity of LAB, few studies have been conducted on the environmental factors that improve scavenging activity. This study investigated the environmental factors affecting the DPPH radical scavenging and various antioxidant activities of Kimchi-derived Lactobacillus plantarum K-21 with antihypertensive and radical scavenging activities. The optimal conditions for scavenging DPPH radicals were glucose 2%, bactopeptone 0.5%, Tween 80 0.05%, L-cysteine 0.05%, and an initial pH 6.5 at 35℃. Under optimal conditions, the DPPH radical scavenging activity was 94.8±2.2%, which was 1.5 times higher than that of the basic medium. In addition, L. plantarum K-21 had other antioxidant activities; ABTS radical scavenging (93.6±1.5%), hydroxyl radical scavenging (8.5±0.9%), metal chelating (65.9±0.5%), NO scavenging (53.1±19%), SOD-like (25.1±1.5%), and reducing power (11.7±1.4%) activities were detected. Therefore, L. plantarum K-21 may act not only as a starter for lactic acid-fermented foods with improved functionality but also as a drug for various diseases caused by oxidative stress.

Antioxidant and Anti-wrinkle Effects of Steamed Three Ginseng Extracts (숙성된 3종 인삼추출물의 항산화 및 주름개선 효과)

  • Steamed ginseng is well known as a tonic medicine for restoring and enhancing human health. Steamed ginseng had more pharmacologically activity than white ginseng. The effects of steamed ginseng on transplantable tumors, proliferation of lymphocyte and rat liver lipid peroxidation were studied. This study was performed to evaluate the antioxidation and antiwrinkle effects of three ginseng extracts. Raw ginseng (RGS) and dried ginseng (DGS) mature like red ginseng in addition to the ready-made red ginseng (GS) purchased in the market, were steamed and extracted by red ginseng extractor. Three extracts of steamed ginseng were investigated to determine effects of superoxide radical scavenging activity, hydroxyl radical scavenging activity, autooxidation inhibition of linoleic acid, collagenase inhibition and collagen synthesis in normal fibroblast. RGS showed not only the highest superoxide radical scavenging activity at a concentration of 100 ug/mL but also the hydroxyl radical scavenging activity higher than vitamin C. Also RGS showed the highest activity in inhibition of autooxidation of linolic acid, collagen synthesis, and collagenase inhibition.

In Vitro Free Radical Scavenging and Hepatoprotective Activities of Taraxacum mongolicum (포공영의 자유라디칼 소거 및 간세포 보호활성)

  • The methanol (MeOH) extract and ist fractions of Taraxacum mongolicum (Compositae) were examined for their scavenging effects on 1,1-diphenyl-2- phenylhydrazyl (DPPH) and superoxide radicals, and hepatoprotective effects on tacrine-induced cytotoxicity in human hepatoma cell line, Hep G2 cells. Both methylene chloride $(CH_2Cl_2)$ and butanol (n-BuOH) soluble fractions of the MeOH extract showed the free radicals scavenging and hepatoprotective effects. From these results, it is suggested that hepatoprotective effect of these fractions partly relies on their free radical scavenging activity.

Antioxidant and Cell Protection Effects of Ethanolic Extract from Cirsium setidens, Allium hookeri, Aronia melancocarpa (곤드레, 삼채, 아로니아 주정 추출물의 항산화 및 세포 보호 효과)

  • Cirsium setidens, Allium hookeri, Aronia melancocarpa, are popular dietary ingredients for Asian cuisine or beverage. this study attempted to search and report the antioxidant activity of extracts of aronia, gondre and samchae, which are used for food in Korea. As a result, total phenol content and DPPH radical scavenging activity tended to increase, and Gondre extract showed the highest DPPH radical scavenging activity effect. The results of a study on the ABTS radical scavenging activity of extracts of Gondre, Aronia, and Samchae showed a tendency for radical scavenging activity to increase as the concentration increased. Gondre hot water extract showed the highest ABTS radical scavenging activity. In this way, antioxidant activity was shown in the order of Gondre, Aronia, and Samchae, and the concentration showed a high activity effect at high concentrations. In the cell protection effect results, the concentration of Gondre extract was 500 ㎍/mL, the Samchae alcohol extract was used at concentrations of 2.5, 5, 7.5, and 10 ㎍/mL, and the Aronia extract was used at concentrations of 125 ㎍/mL. and showed a cytoprotective effect at 250 ㎍/mL. We hope that these research results will be used as basic data for the development of various functional foods.

Antioxidant activity of Opuntia robusta and its total polyphenol and flavonoid contents

  • The aim of this study was to investigate the antioxidant effects of O. robusta stem extract (ORE) and to determine the total polyphenol and flavonoid contents. Free radical scavenging properties were evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)), and ·OH (hydroxyl radical) scavenging assays. Total polyphenol and flavonoid contents were measured using Folin-Ciocalteu reagent and aluminum chloride colorimetric methods, respectively. Active compounds of ORE were determined using high-performance liquid chromatography (HPLC). The results of the study showed that ORE exhibited DPPH, ABTS, and ·OH radical scavenging activities in a dose-dependent manner. Especially, 1,000 ㎍/mL of ORE showed the strongest radical scavenging properties against DPPH, ABTS, and ·OH. ORE contained total polyphenol content of 57.4 mg GAE/g and total flavonoid content of 5.4 mg QE/g, which may contribute to their antioxidant effects. As a result of HPLC, the contents of active compounds in ORE, dihydrokaempferol (0.65 ㎍/mL), and 3-O-methylquercetin (1.10 ㎍/mL) were confirmed. In conclusion, ORE may be useful as a functional material with antioxidant properties.

Antioxidant Activity of Hawthorn Fruit in vitro

  • The antioxidant activity of hawthorn fruit (Crataegus pinnatifida Bunge var. typica Schneider) extracts was investigated by several in vitro antioxidants properties, including DPPH free radical scavenging activity, total phenolic and flavonoid contents, hydroxyl radical scavenging activity, reducing power activity, iron-chelating capacity and nitrite scavenging activity. Among the extracts in this study, the 70% EtOH extract showed higher antioxidant activity than the others. The $IC_{50}$ value of DPPH free radical scavenging activity was $99.26\;{\mu}g/mL$. Furthermore, the 70% EtOH extract also showed significantly high total phenolic and flavonoids contents and reducing power activity. However, the MeOH extract exhibited stronger effects on hydroxyl radical scavenging activity, iron-chelating capacity and nitrite scavenging activity. All the results implicated that, the hawthorn fruit may has the available potential to be utilize as a potential source of natural antioxidant.

Evaluation of the Biological Activity of Extracts from Star-Anise (Illicium verum)

  • This study was carried out to investigate the biological activities of star anise extracts obtained from four different solvents (water, methanol, ethanol and chloroform) by measuring total polyphenol and flavonoid contents, DPPH radical scavenging, hydroxyl radical scavenging, nitrite scavenging activity and antimicrobial activity. The methanol extract showed the highest extraction yield, followed by ethanol, water and chloroform. The properties of the extracting solvents significantly affected the total polyphenol content. Methanol extracts contained more total polyphenols than any other extracts. The highest DPPH radical scavenging activity was found in methanol extract. The hydroxyl radical and nitrite scavenging activity were highest in methanol and ethanol extracts. In antimicrobial activity, water extract showed stronger activities than methanol and ethanol extract against Micrococcus luteus and Bacillus subtilis, and no inhibitory effects on Gram negative bacteria were found in all extracts at the concentration used.
