Iso-catalase Profiles of Deinococcus spp.

  • The obligate aerobic Deinococcus are highly resistant against lethal effect of UV-and ionizing-radiation. Only five mesophilic Deinococcus species, i. e. D. radiodurans, D. radiophilus, D. proteolyticus, D. radiopugnans, and D. grandis are known. Since an indispensable role of catalase has been suggested in protecting cells against oxidative stress and UV radiation, Deinococcal catalase activity of each species and electrophoretic profiles of catalases were investigated on gel. Total catalase activity was varied among the species in the aerobically grown culture at stationary phase. The occurrence of multiple forms of catalases with different molecular weights in four species of Deinococcus and of a single catalase in D. radiopugnans suggests that each species shows the unique catalase profiles on gel. Some Deinococcal catalases also exhibit peroxidase activity. Since Deinococcus spp. are less-distinct to each other in their morphology, biochemical and physiological properties, the catalase profiles on PAGE would be useful in identifying the species of Deinococcus.

  • A method to estimate the radiatio power to the surrounding formation due to axial vibration of the pipe in a fluid-filled borehole has been developed, by using the propagation modes of stress wave in an infinitely-long and uniform drilling borehole surrounded by a radially-infinite homogeneous formation. Also, the equivalent damping coefficient for the axial vibration of the pipe has been derived. As an example, results for a real drilling borehole has been presented. The analysis of the elastic motion of the infinite formation which has cylidrical cavity is simplified with the geometric axisymmetry and the low-frequency assumption so that the analytic solution is obtained.

  • Hermansky-Pudlak Syndrome (HPS) 환자에서 조기에 발생되는 폐섬유화의 원인을 알아보고자, 생쥐 HPS 모델인 ep/ep,pe/pe 돌연변이종의 폐 항산화계의 환성과 방사선에 대한 반응을 측정하였다. HPS 폐에서는 대조군에 비해 glutathione이 더 산화되어 있었고, catalase, glutathione S-transferase(GST) 등의 항산화효소의 활성이 저하되어 있었으며, 10 Gy의 방사선을 조사하였을 때, glutathione 양이 감소하였고, 대조군 폐에서 보여지는 방사선에 의한 ${\gamma}$-glutamylcysteine ligase(GCL), glutathione peroxidase(GPx) 활성의 유의성 있는 증가가 관찰되지 않았다. 이 결과로부터 HPS 환자의 폐는 항산화계 활성이 저하되어 있을 뿐 아니라, 산화적 스트레스가 가해 졌을 때 적응 반응이 매우 취약하여 산화적 환경에 노출된 폐의 병증을 유발할 수 있음을 추측할 수 있다.

  • In this study, the boundary element analysis in dynamics for the multilayered semi-infinite plane is developed using the fundamental solution for moving loads. Also the indirect method and superposition method are introduced to consider the multilayered systems and moving loads. At each layer the fundamental solution can be obtained by solving the governing equation which is transformed by the Fourier transform. The governing equation can be solved by three conditions; continuity conditions of displacement and stress, the traction free condition at the surface and the radiation condition at the surface and the radiation condition at the infinite distance. To verify the solution and the developed algorithm, the theoretical solution for the homogeneous layer and commercial FEM program is compared with the results of this study.

  • Polymeric composite insulators have been in use for outdoor insulation. However, our knowledge about their long-term performance in a outdoor environment is still very limited. Especially, these insulator are subjected to the environmental stress such as ultraviolet radiation. Hence, in this paper. the influence of UV radiation on the aging of the shed materials was evaluated. For the aging evaluation, these insulators studied by experiment methods such as contact angle measurement. oxidation induced time. SEM/EDX, FTRI-ATR, tracking test for the different insulator samples. With the increased UV exposure time, samples are shown to exhibit degradation comparable to those of new ones

  • The purpose of this study was to find out harmful effects of concrete, an essential material in modern architecture, on the human body. Based on the results from animal testing, we can consider the effects on the human body. The results of this study were as follows; Experimental Mouses in concrete have low body temperature due to cold radiation and more aggressive due to cold stress. Therefore, Cold radiation, a property of concrete, makes body temperature lower and affect the body's immune function.

Hypoxic Microenvironmental Control of Stress Protein and Erythropoietin Gene Expression

  • The presence of hypoxic cells in solid tumors has long been considered a problem in cancer treatment such as in radiation therapy or treatment with some anticancer drugs. It has been suggested that hypoxic cells are involved in the development of a more aggressive phenotype and contribute to metastasis. In this study, as an attempt to understand how tumor cells adapt to hypoxic stress, we investigated the regulation of the hypoxia-induced expression of proteins that control essential processes of tumor cell survival and angiogenesis. We first examined whether hypoxia induces stress protein gene expression of murine solid tumor RIF cells. We also examined hypoxia-induced changes in angiogenic gene expression in these cells. Finally, we investigated the association of the elevated levels of stress proteins with the regulation of hypoxia-induced angiogenic gene expression. Results demonstrated that hypoxia induced the expression of the erythropoietin (EPO) gene and at least two major members of stress proteins, heat shock protein 70 (HSP70) and 25 (HSP25) in RIF tumor cells. Evidence that the expression of EPO gene was greatly potentiated in TR cells suggested that the elevated levels of HSPs may play an important role in the regulation of the hypoxia-induced EPO gene expression. One of the RIF variant cell lines, TR, displays elevated levels of HSPs constitutively. Taken together, our results suggest that a hypoxic tumor microenvironment may promote the survival and malignant progression of the tumor cells by temporarily increasing the level of stress proteins and expressing angiogenic genes. We suspect that stress proteins may be associated with the increase of the angiogenic potential of tumor cells under hypoxia.

Differential Growth Response and Gene Expression in Relation to Capsidiol Biosynthesis of Red Pepper Plant and Cultured Cells by γ-Ray and UV Stress (방사선과 자외선에 대한 고추 식물체 및 배양세포의 생장반응과 Capsidiol 생합성 유전자의 발현 차이)

  • Differential responses of red pepper plant and cultured cells to enhanced ${\gamma}$-ray($^{60}$ Co) and ultraviolet(UV) stress were investigated. In seed treatment, 1 Gy of ${\gamma}$-ray increased seedling dry weight up to 19.1%, but 50 Gy treatment markedly ingibited seed germination and subsequent growth of seedling. UV treatment to seed did not change the germination ability of seeds and the growth of seedlings regardless of duration of UV treatment until 24 hrs. In case of UV treatment to seedlings, plant injury was seriously progressed even after the seedlings were returned to no UV condition, and eventually all the leaves showed chlorosis by the stress. However, progress of plant injury by ${\gamma}$-ray stress slower than that caused by UV stress, and even at the high dose of ${\gamma}$-ray 50 Gy, did not caused the cholrosis of stressed plant leaf. Amount of electrolytes leakage from plant leaf by UV treatment for 24hrs was increased up to 28.8 folds in comparison with untreated control, whereas that of 50 Gy of ${\gamma}$-ray was increased only 1.2 folds. UV stress induced the production of capsidiol, antimicrobial phytoalexin, by activation of gene expression involved in capsidiol biosynthesis, such as sesquiterpene cyclase and cyclase and cytochrome P450 hydroxylase in the leaf and cultured cell, but ${\gamma}$-ray stress induced neither the production of capsidiol nor expression of the genes.

Linkage Between Brown Planthopper Resistance Gene and Salt Tolerance in Rice (벼멸구 저항성 유전자와 내염성과의 연관)

  • Using two japonica rice cultivars (Ilpumbyeo and Sanghaehyanghyella), which are distinguishable by the brown planthopper (BHP) resistance maker (R208), a relationship between the BPH resistance gene (Os-Bil) and salt-tolerance was investigated. To do this, changes in the expression level of Os-Bil by the salt stress were quantified by the real-time PCR in the two cultivars, and compared with those in other two indica rice cultivars (Pokkali and IR29). In Ilpumbyeo, the expression level of Os-Bil decreased by the treatments of 50 and 200 mM NaCl in a concentration-dependent manner, and in Sanghaehyanghyella it rather increased slightly at 50 mM but decreased drastically at 200 mM. Comparably, IR29, a salt-sensitive cultivar, showed a reduction of the Os-Bil gene expression after the treatment of 100 mM NaCl, but Pokkali, a salt-tolerance cultivar, rather increased about two times in the level of Os-Bil transcripts. These results suggest that the BPH resistance gene may involve in the difference in the salt-tolerance at least between the two indica rice cultivars.

Change in the Plant Temperature of Tomato by Fogging and Airflow in Plastic Greenhouse (포그분사 및 공기유동에 의한 온실재배 토마토의 엽온 변화)

  • To investigate the influence of surrounding environment on the plant temperature and examine the effect of plant temperature control by fogging and airflow, plant temperature of tomato, inside and outside air temperature and relative humidity, solar radiation and wind speed were measured and analyzed under various experimental conditions in plastic greenhouse with two-fluid fogging systems and air circulation fans. According to the analysis of plant temperature and the change of inside and outside air temperature in each condition, inside air temperature and plant temperature were significantly higher than outside air temperature in the control and shading condition. However, in the fogging condition, inside air temperature was lower or slightly higher than outside air temperature. It showed that plant temperature could be kept with the temperature similar to or lower than inside air temperature in fogging and airflow condition. To derive the relationship between surrounding environmental factor and plant temperature, we did multiple regression analysis. The optimum regression equation for the temperature difference between plant and air included solar radiation, wind speed and vapor pressure deficit and RMS error was $0.8^{\circ}C$. To investigate whether the fogging and airflow contribute to reduce high temperature stress of plant, photosynthetic rate of tomato leaf was measured under the experimental conditions. Photosynthetic rate was the highest when using both fogging and airflow, and then fogging, airflow and lastly the control. So, we could assume that fogging and airflow can make better effect of plant temperature control to reduce high temperature stress of plant which can increase photosynthetic rate. It showed that the temperature difference between plant and air was highly affected by surrounding environment. Also, we could estimate plant temperature by measuring the surrounding environment, and use it for environment control to reduce the high temperature stress of plant. In addition, by using fogging and airflow, we can decrease temperature difference between plant and air, increase photosynthetic rate, and make proper environment for plants. We could conclude that both fogging and airflow are effective to reduce the high temperature stress of plant.
