The Evaluation of Usefulness of 99Mo-99mTc Generator Using(n,γ)99Mo Developed by Korea Atomic Energy Research ((n,γ)99Mo를 이용한 99Mo-99mTc발생기의 유용성 평가)

  • Purpose: The Molybdenum which is the raw material of $^{99}Mo-^{99m}Tc$ generator is produced from the nuclear reactor. However, output has dwindled as the two nuclear reactors supplying the bulk of radioactive material-one in Chalk River, Ontario and the other in Petten, the Netherlands-have been closed for repairs or maintenance. This resulted in the enhancement of its price. So $^{99}Mo-^{99m}Tc$ generator using$(n,{\gamma})^{99}Mo$ is developed by Korea Atomic Energy Research Institute (KAERI). Medicinal availability of this generator is evaluated in this study. Materials and Methods: The radioactivity of $^{99m}Tc$ eluted in generator 1, 2 and 3 unit developed by KAERI was measured. The quality control test of generator such as appearance test, pH test, LAL test, sterility test, chemical impurity (Al) test and radiochemical purity test were performed. Planar and SPECT/CT image sof SD rat (6 weeks, Female) at 2 hr after injection of $^{99m}Tc-HDP$ (hydroxymethylenediphosphonate) (TechneScan HDP, Malinckrodt Medical, Dutch) and $^{99m}Tc-DPD$ (diphosphono-1, 2-propanedicarboxylicacid) (TECEOS, CIS bio international, France) which were labeled with $^{99m}Tc$ eluted in KAERI and commercial generator (40.5 GBq, Malinckrodt Medical, Dutch) using SPECT/CT camera (Symbia, Siemense, Germany) were obtained respectively. Results: The mean radioactivity of $^{99m}Tc$ elution generator 1unit was 4.18 GBq (113 mCi), generator 2 unit was 4.73 GBq (128 mCi) and generator 3 unit was 3.33 GBq (90 mCi). All quality control tests were within normal limit except pyrogentest. Pyrogen test was positive. Planar and SPECT/CT images of rat injected $^{99m}Tc-HDP$ which was labeled with $^{99m}Tc$ eluted in commercial generator show increased uptake in bone, stomach and bowl. Planar images show increased uptake in liver and bone in case of $^{99m}Tc-DPD$. However, images of rat injected $^{99m}Tc-HDP$ and $^{99m}Tc-DPD$ which were labelled $^{99m}Tc$ eluted in KAERI generator show increased uptake in bone, liver and spleen. Conclusion: If shortcoming is removed such as pyrogen and liver appearance, domestic role as an alternative generator is thought to be able to fill and to secure the national medical service by supplying $^{99m}Tc$ when the supply of $^{99m}Tc$ be comes short.

Studies on the Biological Behaviors of Taxol Derivatives (Taxol 유도체들의 생물학적 거동에 관한 연구)

  • This study was designed to prospect the $^{111}In$-labelled paclitaxel as tumor imaging agent. In order to provide a taxol molecule with a functional group which is able to chelate In-111, taxol-DTPA conjugate and 2'-hemisuccinyltaxol were synthesized by esterification of taxol at C-2'on C-13 carbon with DTPA anhydride and succinic anhydride, respectively. Synthesis yield of the taxol derivatives was 34% for taxol-DTPA and 80% for 2'-hemisuccinyltaxol. Cytotoxicity of the taxol derivatives were measured by MTT method toward cell lines HT29, B16, P388, and CT26. The cytotoxic activities of the taxol derivatives were maintained, although less active than taxol. Radiolabelling of the taxol derivatives were proceeded directly with $^{111}InCl_3$ or indirectly with $^{111}In$-citrate(ligand-exchange method). The ligand-exchange method was not suitable because some precipitates appeared during the reaction. On the contrary, by direct radiolabelling method, we were able to obtain taxol-DTPA-$^{111}In$ in 100% radiochemical yield. However, 2'-hemisuccinyltaxol was not labelled by both methods. Yield and radiochemical purity of the radiolabelled com-pound were determined by HPLC, paper chromatography and instant thin layer chromatography. Taxol-DTPA-$^{111}In$ was characterized to be hydrophilic by lipophilicity test, and nearly non-adhesive to HT29, B16, P388, and CT26 by cell binding affinity test. Binding affinity of the taxol-DTPA-$^{111}In$ complex to serum proteins was also examined by protein precipitation with 30% trichloroacetic acid. The results showed that 30% of the taxol-DTPA-$^{111}In$ complex binds with serum proteins.

Properties of Sunglass Lenses by Non-Destructive Test for the Sunglass Standards (비파괴식 검사를 통한 선글라스의 품질에 관한 연구)

  • In this study, we compared the properties of 23 high and 55 low price sunglass lenses with the standards in the ordinary optical properties, materials, coloration, UV, IR and luminous transmittance, color acceptance for traffic signal, chromaticity and contrast sensitivity. The ordinary optical properties of the lenses met comparatively the requirements of the KS standard. The HIGH-type and LOW-type lenses were primarily made by glass and acrylate, respectively. In the coloration, HIGH-type was in group around neutral color but LOW-type was distributed widely on the line between 570 nm and 485 nm. There are fails in 7% of HIGH-type and 18% of LOW-type in the stimulus purity of the luminous transmittance. Wavelength of the UV/VIS cut-off was over 350 nm for HIGH-type but 6% of LOW-type was under 350 nm. In the erythemal UV, all HIGH-type met the needs of standards but 5 LOW-type failed with DIN standard In the near UV, KS standard worked in stringency, and HIGH-type showed more failure than LOW-type. The characteristics of the IR transmittance of HIGH-type was better than that of LOW-type. In the color acceptance of traffic signal, all HIGH-type met the needs of ANSI standards but 21.8% of LOW-type failed with the standard. In the contrast sensitivity tested with various coloured sunglasses, the value increased with increasing of Land decreasing of test distance. In view of the results so far, HIGH-type met with excellent properties as compared with LOW-type.

Extraction Process of Lead and Variations of Physicochemical Properties using the Smelting Experiment of Galena (방연석 제련실험을 통한 납 추출과정 및 물리화학적 거동변화 연구)

  • This study aims at identifying the variation of physicochemical properties for galena ore in order to use it in understanding of manufacturing techniques in terms of metallurgical method and smelting or refining process for obtaining lead. The ores in the study obtained from the operating mine have been washed and pre-treated for a test. Metallic lead has been extracted by roasting process. The result displays that galena is still in present in the matt despite of exposure to high temperature, over $1000^{\circ}C$. Nearly $11.1g/cm^3$ specific gravity metallic lead has been collected from the refining test of which aim was to remove the remnant galena and a trace of concentrated silver has been identified at the grain boundary. The result suggests that at least one refining process was essential to acquire high purity metallic lead and cupellation had been executed to remove remnant silver.

Use of Microsatellite Markers Derived from Genomic and Expressed Sequence Tag (EST) Data to Identify Commercial Watermelon Cultivars (수박 시판 품종의 식별을 위한 Genomic과 Expressed Sequence Tag (EST)에서 유래된 Microsatellite Marker의 이용)

  • This study was carried out to construct a DNA profile database for 102 watermelon cultivars through the comparison of polymorphism level and genetic relatedness using genomic microsatellite (gMS) and expressed sequence tag (EST)-microsatellite (eMS) markers. Sixteen gMS and 10 eMS primers showed hyper-variability and were able to represent the genetic variation within 102 watermelon cultivars. With gMS markers, an average of 3.63 alleles per marker were detected with a polymorphism information content (PIC) value of 0.479, whereas with eMS markers, the average number of alleles per marker was 2.50 and the PIC value was 0.425, indicating that eMS detects a lower polymorphism level compared to gMS. Cluster analysis and Jaccard's genetic distance coefficients using the unweighted pair group method with arithmetic average (UPGMA) based on the gMS, eMS, and combined data sets showed that 102 commercial watermelon cultivars could be categorized into 6 to 8 major groups corresponding to phenotypic traits. Moreover, this method was sufficient to identify 78 out of 102 cultivars. Correlation analysis with Mantel tests for those clusters using 3 data sets showed high correlation ($r{\geq}0.80$). Therefore, the microsatellite markers used in this study may serve as a useful tool for germplasm evaluation, genetic purity assessment, and fingerprinting of watermelon cultivars.

Production of Methane from Anaerobic Fermentation of Marine Macro-algae (해조류의 혐기성 발효를 이용한 메탄 생산)

  • Methane was produced from the anaerobic digestion of marine macro-algae. Elemental analysis was first performed to estimate the theoretical methane production of three macro-algae (Undaria pinnatifida, Laminaria japonica, Hizikia fusiformis). Three algae were found to contain C 34 ~ 36%, H 5%, O 37 ~ 43%, N 2 ~ 4%, S 0.4 ~ 0.7%, and ash 14~21%, and the theoretical methane content was in the range of 56 ~ 60%, which can produce 442 ~ 568 mL $CH_4$ per g of volatile solid (VS). Using the biological methane potential (BMP) test, we found that L. japonica resulted in the highest yield of methane (52%). Moreover, various operational conditions, such as algae amount, pH, salinity, particle size, and pre-treatment, were investigated in order to find an optimal condition of anaerobic digestion. At pH 8.0, the autoclaved L. japonica (5g VS/200 mL), when used without washing salt, produced 268.5 mL/g VS which is 65% of the theoretical methane productions. Furthermore, using a CSTR (with the working volume of 7 L out of the total volume of 10 L), we have successfully operated the reactor for 65 days and obtained maximum methane production rate of 1.4 L/day with purity of 70%.

Purification and Characteristics of Tyrosinase Inhibitor Produced by Actinomycetes F-97 (방선균 F-97이 생산하는 Tyrosinase 저해제의 정제 및 특징)

  • An actinomycetes F-97 producing tyrosinase inhibitor was isolated from soil samples. Isolation and purification of tyrosinase inhibitor produced by F-97 was performed as follows: IRC-120 ($NH_4^+$ type) column chromatography, silica gel column chromatography, $C_{18}$ column chromatography and Sephadex LH-20 column chromatography were used successively after the centrifuged supernatant was adjusted to pH 4.0. To identify the purity of the inhibitor, octadecylsilyl(ODS) HPLC was carried out with 5% methanol as a mobile phase. Finally, the purification yield of a tyrosinase inhibitor was 5.24%. The inhibitor was very soluble in water, methanol and ethanol but insoluble in acetone, butanol, ethylacetate and chloroform. The ${\lambda}_{max}$ value of this inhibitor in water was 194nm under UV light. The biochemical test of the inhibitor was positive in Molish, Benedict, cone. $H_2SO_4$, and $KMnO_4$ tests but negative in iodine, ninhydrin, Million, Sakaguchi, xanthoproteic and Emerson tests. The tyrosinase inhibitor was stable against heat treatment of $100^{\circ}C$ for 50 minutes and pH $4{\sim}9$. The $IC_{50}$ value of this inhibitor was $19.2{\mu}g/ml$ for mushroom tyrosinase. In $1,000{\mu}g/ml$ inhibitor concentration, inhibition zone was 27 mm for Streptomyces bikiniensis NRRL B-1049. The inhibition of F-97 against mushroom tyrosinase was competitive with tyrosine.

Synthesis and Evaluation of F-18 Labeled 2'-Deoxy-2'-fluoro-5-methyl-1-β-L-arabinofuranosyluracil (L-[18F]FMAU)

  • L-[18F]FMAU ([18F]1b) was prepared from the precursor 2-O-[(trifluoromethyl)-sulfonyl]-1,3,5-tri-Obenzoyl- α-L-ribofuranose, by coupling the radioactive fluoro-sugar with the corresponding silylated thymine in 4 steps. The final products, including the α and β anomers, were purified using reverse phase HPLC with an appropriate solvent (5% CH3CN/H2O) at a flow rate of 3.0 mL/min. The total elapsed time of synthesis was about 180-200 min from EOB. The α/β anomeric ratio of the compounds was about 1:9, and the radiochemical purity of the product (β-form) was >98% with decay-corrected yields of 25-35%. All radioactive samples were confirmed using co-injection with pure non-radioactive analogues in every step. In the cellular uptake in vitro test of herpes simplex virus-thymidine kinase (HSV1-TK) gene expressed cells, the percent uptake of injected dose (%ID) of L- and D-FMAU was 37.28 and 65.86, respectively after 240 min incubation. However, the relative uptake (MCA-TK/MCA cellular uptake ratio) of L-FMAU was higher than that of D-FMAU (%ID of L-FMAU, 0.36 and D-FMAU, 0.93 after 240 min incubation in MCA cells). This means that L-FMAU will show better specific HSV1-TK gene expressed cell uptake for selective HSV1-TK gene imaging.

Consecutive automated production of carbon-11 labeled radiopharmaceuticals by sharing 11C-methylation reagent from one 11C-synthetic module

  • Increasing clinical demand for carbon-11 labeled radiopharmaceuticals has triggered technological advances in fields of radiochemistry and automated modules. Even though carbon-11 has a short half-life ($t_{1/2}=20.4min$), the consecutive second production of carbon-11 labeled radiopharmaceutical in one $^{11}C$-synthetic module should be delayed at least over 4 h to avoid the high radiation exposure. We herein aimed to produce two different carbon-11 labeled radiopharmaceuticals ([$^{11}C$]PIB and [$^{11}C$]methionine) by sharing of [$^{11}C$]methylation source in one $^{11}C$-synthetic module. The synthesis of $^{11}C$-labeling reagents ($[^{11}C]CH_3I$ or $[^{11}C]CH_3OTf$) is fully automated using the commercial TRACERlab $FX_{C-pro}$ module and is readily adaptable to $^{11}C$-labeling reactor for [$^{11}C$]PIB as well as another $^{11}C$-labeling apparatus for [$^{11}C$]methionine via the three-way valve. After completing the [$^{11}C$]PIB production, the re-synthesized $[^{11}C]CH_3I$ was passed through the three-way valve connected the polyetheretherketone (PEEK) line and loaded into the C18 Sep-Pak cartridge including the methionine precursor. The labeled product [^${11}C$]methionine was purified by a simple cartridge separation and reformulated into saline. The radiochemical yield of [$^{11}C$]PIB and [$^{11}C$]methionine were $5.3{\pm}0.6%$ and $18.7{\pm}0.8%$ (n.d.c.), respectively, with over 97% of radiochemical purity. The specific activity of [$^{11}C$]PIB was over $110GBq/{\mu}mol$. Total production time of two radiopharmaceuticals needs about 2 h from $1^{st}$ beam irradiation including quality control tests. Final [$^{11}C$]PIB and [$^{11}C$]methionine were satisfied all quality control test standards.

Single Oral Dose Toxicity Study and Aconitine Content Analysis of Raw Aconiti Tuber and Sambu-tang (생부자(生附子) 및 삼부탕(蔘附湯)의 aconitine 함량 분석 및 삼부탕 단회투여 독성시험)

  • Objectives: This study investigated the aconitine contents analysis of Buja extracts (raw material of Buja, hot water extract of Buja, and hot water extract of Sambu-tang) and the single oral dose toxicity of Sambu-tang-R in six-week-old Sprague-Dawley rats in order to compare the toxicity of Buja extracts.Methods: Aconitine content analysis is that Buja extracts were hold purity test. To detect single oral dose toxicity, six-week-old Sprague-Dawley rats were divided into two groups, a normal control group and a sambutang-R (2,000 mg/kg) group. For 14 days of treatment, clinical signs, body weight, clinical chemistry, necropsy, and histopathology were examined.Results: The aconitine contents of the Buja extracts were Buja-RH (0.1738%), Buja-RD (0.1746%), and Sambu-tang-R (0.0961%). There were no cases of death in either the control group or the experimental group. Nor was there any disorder to the clinical signs or any significant change in body weight in either group. There was no significant change of clinical chemistry or disorder of necropsy findings in either the control or the experimental group. And there was no difference in histopathological findings in comparing the control group with the experimental group.Conclusions: These results suggest that the aconitine content of the hot water extract of Buja was similar to the raw material of Buja, but the hot water extract of Sambu-tang had greatly decreased aconitine content. These results also suggest that a single oral lethal dose of Sambu-tang-R for Sprague-Dawley rats exceeds 2,000 mg/kg for both female and male rats.
