The control system of sludge amount inspection and discharge materials of outlet water and affiliated water-purification tank (오수/합병정화조의 배출물 제어시스템 연구)

  • The individual rotten water purification tank recently discharges wastewater and sewage through the outlet without purification ability. The outlet water and affiliated water purification tank with microorganism cultivator tank cultivates microorganism and then drops the value BOD, COD of sewage and discharges the quality of water into the outlet. The blower and water pump operating continuously prompts the waste of energy and deterioration of equipment. Each room of deposition tank, foaming tank, microorganism cultivator tank is equipment with the sludge detection senses so it can detect the density of each room. The power-drive plant of the blower and water pump which ate the system cultivating the microorganism must be made as fuzzy controlization (If the sludge amount of each room become higher, the rate of operation of blower and water pump must heighten, on the contrary, in case of row sludge amount, the total handling amount and microorganism handling amount of each room of control. Tank reducing the rate of operation must be DB. At present, the blower amount in proportion to the sludge and oxygen demanding amount has to control. Each mom must be checked outlet level of the outlet, also each room must flow backward discharge materials, and must operate feed-back control until we want to be come as a below value of BOD/COD(10PPM ; KS).

Purification and Properties of Bacteriocin Produced by Lactococcus sp. 1112-1 (Lactococcus sp. 1112-1 균주가 생산하는 Bacteriocin의 정제 및 성질)

  • Purification of the bacteriocin from Lactococcus sp. 1112-1 was achieved by successive column chromatography on CM-Sephadex C-25 and Sephadex G-50, starting from cell disruption broth. 16.2% of the initial activity was recovered after this purification step and it was shown 123-fold increase in purification. Purified bacteriocin was shown a single band on SDS-polyacrylamide gel electrophoresis. This substance was rather stable at heat treatment and alkaline pH relatively. The residual antimicrobial activity was 38% when the bacteriocin was treated by heat at $100^{\circ}C$ for 60 min. And 23% of the activity remained at pH 8.0 after standing for 48 hr. The amino acid composition of purified bacteriocin was made up 26 residues.

Effective Purification of Ginsenosides from Cultured Wild Ginseng Roots, Red Ginseng, and White Ginseng with Macroporous Resins

  • This study was aimed (i) to develop an effective method for the purification of ginsenosides for industrial use and (ii) to compare the distribution of ginsenosides in cultured wild ginseng roots (adventitious root culture of Panax ginseng) with those of red ginseng (steamed ginseng) and white ginseng (air-dried ginseng). The crude extracts of cultured wild ginseng roots, red ginseng, and white ginseng were obtained by using a 75% ethanol extraction combined with ultrasonication. This was followed sequentially by AB-8 macroporous adsorption chromatography, Amberlite IRA 900 Cl anion-exchange chromatography, and Amberlite XAD16 adsorption chromatography for further purification. The contents of total ginsenosides were increased from 4.1%, 12.1%, and 11.3% in the crude extracts of cultured wild ginseng roots, red ginseng, and white ginseng to 79.4%, 71.7%, and 72.5% in the final products, respectively. HPLC analysis demonstrated that ginsenosides in cultured wild ginseng roots were distributed in a different ratio compared with red ginseng and white ginseng.

  • When the costal zone surrounded by a breakwater has a narrow vertical opening, currents in the vicinity of a narrow entrance can result in a jet flow, coinciding with the tide. Such a Tidal-Jet Generator(TJG) can change the water mass distribution and transport processes in the domain of influence of the jet. Also, it can decrease the residual time of them. In the present study, the water purification utilizing tidal jets in the coastal zone over constant bathymetry are estimated numerically, using a finite-difference numerical scheme, named the NS-MAC-TIDE method, which isbased on the fully 3D Navier-stokes(NS) equations. The shear velocity near the inlet of the TJG are predicted from the flow field simulation, and are assessed qualitatively with the development of scouring or sediment that is caused by the change of diffusion or sweeping flowup from the seabed of sediment particles. Finally, through solving a transport equation of concentration, the residual time related on mass transport processes and the flushing mechanism for water purification are investigated.

  • Cold shock proteins (Csps) are a subgroup of the cold-induced proteins on reduction of the growth temperature below the physiological temperature. They preferentially bind to single-stranded nucleic acids to translational regulation via RNA chaperoning. Csp plays important role in cold adaptations for the psychrophilic microorganism. Recently, Cold shock protein from psychrophilic bacteria, Colwellia psychrerythraea (CpCsp) has been identified. Three dimensional structures of a number of Csps from various microorganisms have been solved by NMR spectroscopy or X-ray crystallography, but structures of psychrophilic Csps were not studied yet. Therefore, cloning and purification protocols for further structural study of psychrophilic Csp have been optimized in this study. CpCsp was expressed in E. coli with pET-11a vector system and purified by ion exchange, size exclusion, and reverse phase chromatography. Expression and purification of CpCsp in M9 minimal media was carried out and $^{15}N$-labeled proteins with high purity over 90% was obtained. Further study will be carried out to investigate the tertiary structure and dynamics of CpCsp.

  • Biogas is a renewable fuel from anaerobic digestion of organic matters such as sewage sludge, manure and food waste. Raw biogas consists mainly of methane, carbon dioxide, hydrogen sulfide, and water. Biogas may also contain other impurities such as siloxanes, halogenated hydrocarbons, aromatic hydrocarbons. Efficient power technologies such as fuel cell demand ultra-low concentration of containments in the biogas feed, imposing stringent requirements on fuel purification technology. Biogas is upgraded from pressure swing adsorption after biogas purification process which consists of water, $H_2S$ and siloxane removal. A polymer electrolyte membrane fuel cell power plant is designed to operate on reformate produced from upgraded biogas by steam reformer.

Physical·Mechanical and Temperature Properties of Fiber Reinforced Porous Green Roof Hwang-toh Concrete (섬유보강 다공성 옥상녹화 황토콘크리트의 물리·역학적 및 온도변화 특성 평가)

  • The physical, mechanical, water purification and temperature properties of fiber reinforced porous hwang-toh green roof concrete have been evaluated in this study. The effect of the depending on replacement ratio of blast furnace slag to cement was investigated such that the replacement ratio is varied to 0 % and 30 %. Also, the replacement ratios of hwang-toh were 0, 20 and 30 %. The polyvinyl alcohol fiber was used for the reinforcing fiber. A series of pH test, unit weight, void ratio, compressive strength, after purification and variation of temperature test have been performed to evaluate the performance, water purification effect and temperature properties of the fiber reinforced porous hwang-toh green roof concrete. The test results indicate that the physical and mechanical properties of fiber reinforced porous hwang-toh green roof concrete is affected by the replacement ratio of the blast furnace slag and hwang-toh contents. Results of purifying water showed that the water purification effect of porous hwang-toh green roof concrete is about 40 %. Also, the temperature properties test results indicate the green roof blocks using fiber reinforced porous hwang-toh green roof concrete have insulation and temperature reduction effect.

A Rapid and Simple Homogenizing Method for the Purification of Single-walled Carbon Nanotubes

  • We developed a simple and effective purification method to obtain high-purity single-walled carbon nanotubes (SWCNTs) with low surface damage. The purification process consists of oxidization at $430^{\circ}C$ for 1 h in a furnace system of air atmosphere and homogenization in dilute hydrochloric acid solution for extremely short time. The role of homogenizer was examined during purification process in terms of purity and quality of purified SWCNTs. High-purity and low surface damage of SWCNT products was obtained using homogenizer which was operated at 8500 rpm for 10 min in the environment of 7 % HCI solution. From XRD spectra, we observed that metal catalysts were thoroughly removed. Raman spectra showed that the intensity values of crystallization ($I_{G}/I_{D}$) of purified SWCNTs were very similar with that of pristine SWCNTs. Moreover, the structure damage of purified SWCNTs was hard to find from electron microscopy. Consequently, homogenizing, which is a quick and simple manner, can be promising method for obtaining final SWCNTs with clearly high purity and crystallinity.

  • All over the world, the existing process of water purification needs more flocculants and chlorine due to a gradual decline in the quality of source water. Therefore, the problem of the remaining aluminium and DBPs in purified water is on the rise. To solve this problem, the process of membrane filter has recently come into the spotight. This study reaches the following conclusions concerning TMP variation in order to solve the dropping of flux throgh a membrane filter when operating a membrane filter system in the process of water purification. 1. In case that a cohesion-precipitation process was introduced to pre-treatment of a membrane filter, initial TMP was very satisfactory(0.27kg/cm) in producing the constantly safe quality of water, $0.04{\sim}0.1$(mean 0.05) NTU by pouring 2mg/l of PACI(10% $Al_2O_3$) used for the existing process of water purification in high-density turbidity at a dry or flood season and at occurrence of high algae. 2. As flux increased at 0.5m/day.m, TMP increased 0.05 kgf/cm. 3. As filtering, operation mode of PVDF MF membrane filtering was 48 minutes and 1 cycle of back washing was 42 minutes, flux was increased 1.5m/day.m and TMP increased $0.25{\sim}0.27kgf/cm$. Without back washing, TMP increased 0.03 kgf/cm per a cycle.

Optimized purification and characterization of expressed hMC4R-TM2

  • Human melanocortin-4 receptor (hMC4R) among MC-Rs, expressed in the brain, is in charge of the control on energy homeostasis and food intake. The structure and function of human MC4R have been studied to understand their essential function and roles. To investigate the structure and function, it is necessary to prepare sufficient amounts of proteins. However, their expression and purification is demanding and time-consuming due to their innate insoluble and toxic properties. The heterozygous mutations of hMC4R, exchange of Asp 90 to Asn located in second transmembrane, cause severe obesity in human. To obtain purified hMC4R wt-TM2 for structural studies, it was first over-expressed and purified by fast protein liquid chromatography (FPLC) and then solution NMR studies were performed to get high-resolution spectra. In here, we established optimized purification scheme to get more purified target peptide.
