효소제를 사용한 개량식고추장의 숙성과정 중 휘발성 향기성분의 특성 (Characteristics of Volatile Flavor Compounds in Kochujang Prepared with Commercial Enzyme During Fermentation)

  • 시판의 amylase와 protease효소제로 원료를 처리하여 담금한 효소제 사용 고추장 숙성과정 중 휘발성 향기성분을 purge and trap 장치로 포집하여 GC-MSD로 분석, 동정한 결과 alcohol 16종, ester 16종, acid 7종, aldehyde 4종, alkane 2종, benzene 1종, ketone 3종, alkene 1종, amino 2종, phenol 1종, 기타 1종 등 54종의 휘발성 향기성분이 동정되었다. 동정된 향기성분 수는 담금직후에 alcohol 7종, ester 6종, aldehyde 3종 등 총 23종이 검출되었으나, 30일에는 alcohol 4종, ester 1종을 비롯한 8종이 추가 검출되어 31종으로 증가되었다. 120일에는 49종으로 향기성분 수가 최대에 달하였다. 숙성 전 과정을 통하여 검출된 향기성분은 2-methyl-1-propanol, ethanol, 3-methyl-1-butanol 등 alcohol류 6종, ethyl acetate, isoamyl acetate, ethyl butyrate 등 ester류 5종, butanal, acetaldehyde 등 aldehyde 3종, 기타 6종 등 총 20종이었다. 향기성분의 면적비율(peak area%)은 숙성기간에 따라 다소 차이가 있으나 ethanol, ethyl acetate, ethyl butyrate, ethenone, 2-methyl-1-propanol, 3-methyl-1-butanol 등이 높아 이들 성분이 효소제사용 고추장의 향기 주성분으로 나타났다. 숙성시기별로는 담금직후에 ethyl acetate가, 90일에는 3-methyl-1-butane이, 이외의 기간에는 ethanol이 가장 높았다.

하천퇴적물 중의 수은 및 메틸수은 분석 연구 (Analysis of mercury and methylmercury in river sediment samples)

  • 본 연구에서는 퍼지 & 트랩 GC-MS를 이용한 퇴적물 중의 메틸수은 분석방법을 제시하고, 국내 주요 하천 퇴적물 중의 총수은 및 메틸수은 등을 분석하였다. 분석방법의 검증을 위하여 고농도 및 저농도의 인증표준물질 2종을 분석하였으며, 현장시료 81건에 대하여 EPA 1630 method 원리의 GCCVAFS 방법으로 분석한 결과와 GC-MS 방법의 분석결과를 비교하였다. 방법검출한계 (MDL)는 0.06 ng/g, 정량한계 (MQL)는 0.20 ng/g이었으며 정밀도와 정확도는 각각 11.2% 및 $102{\pm}11.4%$로 나타났다. 조사된 81건에 대한 하천 퇴적물의 총수은 및 메틸수은 농도는 $2.43{\sim}1.56{\times}10^3$ ng/g 및 N.D.~5.95 ng/g 로 나타났다. 총수은의 분포특성은 외부오염원에 의한 영향으로 인해 유기물, 황함량 및 납, 크롬, 니켈 등의 중금속 농도와 유의한 상관성이 나타났으나, 메틸수은의 경우, 총수은을 제외한 다른 인자들과의 유의한 상관성이 조사되지 않았다.

Saccharomyces와 Hansenula의 혼합배양에 의해 제조한 민속증류수의 향미특성 (Flavor Characteristics of Korean Traditional Distilled Liquors Produced by the Co-culture of Saccharomyces and Hansenula)

  • Andong Soju is a Korean traditional distilled liquor brewed with Nuruk which is cultured with wild microorganisms. To provide useful information for scientific production and systematic quality control of traditional distilled liquor, the effects of mixed culture of the alcoholic yeasts and saccharifying molds isolated from the Nuruk, and mashes on the flavor and sensory characteristics were investigated. Distillate from mashes cocultured with Saccharomyces cerevisiae and Hansenula anomala using Mucor Nuruk was compared with distillate from mashes brewed with Andong Nuruk and with distillate from plant fermented mashes to analyze their flavor characteristics. The volatile flavor compounds in distillates were analyzed by GC and GC-MS using direct injection, solvent extraction, and purge & trap methods. Alcohols such as 3-methyl-1-butanol, 2-methyl-1-propanol, 1-propanol, and 2-phenyl ethanol; aldehydes such as acetaldehyde and 2-furancar-boxaldehyde; esters such as ethyl ester of acetic acid, hexanoic acid, octanoic acid, decanoic acid; alkanes, alkenes, ketone, sulfur, and pyrone compounds were detected. Alcohols were chief components of flavor compounds. No significant difference in overall acceptability test was shown among three experimental groups(p<0.05), but Nuruk-like aroma, Kaoliangchiew-like aroma, sweet taste, and well rounded mouthfeel showed significant differences among them(p<0.05).

Quality Properties of Seasoned-Dried Pacific Saury Treated with Liquid Smoke -1. Volatile Flavor Compounds in Commercial Liquid Smokes-

  • In order to identify of volatile flavor compounds and polycyclic aromatic hydrocarbons (PAHs) in commercial liquid smokes, this study was conducted to analyze volatile flavor compounds by solvent extraction and/or Purge & Trap method/GC/MSD. A total of 156 volatile flavor compounds were detected in 6 commercial liquid smokes, and these compounds were composed mainly of 12 aldehydes, 60 ketones, 7 alcohols, 14 acids, 20 esters, 24 aromatic compounds, 7 furans and 12 miscellaneous compounds. Ketones $(806.6-7,573.9\mu g/mL)$ and aromatic compounds $(282.6-7,896.3 \mu g/mL)$ were more abundant than others. The PAHs known as carcinogen have not been detected in this study. The acids $(422.9-4,903.1\mu g/mL)$ was identified in relatively high concentration compared to other groups. Phenol and its derivatives among aromatic compounds were in relatively high concentration. Especially, the phenol and its derivatives including o-cresol, guaiacol, 4-ethylguaiacol and syringol were in higher concentration.

Headspace Volatile Compounds of Steamed Liriopis Tuber Tea Affected by Steaming Frequency

  • Flavor quality of Liriopis tuber tea that was made using a steaming process was studied by measuring changes in headspace volatile compounds. Headspace volatile compounds of the prepared samples were isolated, separated and identified by the combined system of purge & trap, automatic thermal desorber, gas chromatography, and mass selective detector. As steaming frequencies were increased, the area percent of aldehydes decreased from 32.01% to 3.39% at 1 and 9 steaming frequency times, respectively. However, furans and ketones increased from 18.67% to 33.86% and from 9.60% to 17.40% at 1 and 9 times, respectively. The savory flavor of Liriopis tuber tea was due to a decrease in aldehydes contributing a fresh flavor at the 1st steaming process and newly generated furans from nonenzymatic browning with repeated steaming frequencies. These results will provide basic information for quality control of the newly developed Liriopis tuber tea.

  • Even if odorous compounds remained very low concentration in water, it caused strong odor. Because Geosmin and most of odorous compound had very vaporization, those were difficult to analyze with GC/MSD and Purge & Trap. So, we needed pre-treatment method for decreasing amounts of extracting solvents, improving recovery efficiencies and increasing analytical efficiencies. This study developed efficient technology for analyzing odorous compounds, using various adsorbents and extracting solvents. The optimum adsorbent was XAD resins. Especially, XAD-2, XAD-7 and XAD-2010 were superior, but XAD-2 of these and MTBE was the optimum extraction solvent. Other extraction solvent's efficiency was in order of MTBE>Dichloromethane>n-Hexane>Diethylether. The optimum NaCl dosage for increasing efficiency was 5g in liquid-liquid extraction method. The shaking time(0∼24hr) had no concern with adsorption efficiency. The optimum adsorbent was XAD-2 resin and extraction solvent was MTBE. Dosing NaCl, adsorption efficiency was increased in liquid-liquid extraction method, but NaCl has no effect on liquid-solid extraction method. In this experimental results, this method will apply to not only Geosmin but other well-known odorous compounds (2-MIB, IBMP, IPMP, TCA) and algae toxins (Mycrocystin, Anatoxin etc)

열처리가 마늘과 김치에서 발생되는 휘발성 유기화합물에 미치는 영향 (Heat Effects for the Volatile Organic Compounds emitted from Garlic and Kimchee)

  • 생마늘과 김치에서 발생되는 휘발성 유기화합물에 대해 열흡탈착 혹은 퍼지/트랩을 이용한 가스크로마토그래프/질량선택성 검출기법에 의해 분석하였다. 메틸알릴 황, 이메틸 황, 이메틸 이황, 이알릴 황, 이알릴 이황, 메틸알릴 이황, 이알릴 이황 및 이메틸 삼황과 같은 대단히 악취가 강한 성분이 검출되었으며 이들 중 이메틸 이황과 이메틸 삼황은 김치의 숙성과정이나 마늘이 아닌 김치의 재료에서 발생된 것으로 추정되었다. 마늘이나 김치에서 발생되는 악취성분은 우리나라 사람의 입김이나 김치를 보관하는 냉장고 안의 공기 중에서도 검출되었다. 한국인의 입냄새나 냉장고 안의 냄새의 원인은 마늘이나 김치에서 발생되는 이황 화합물이 주성분으로 판단되었다. 이러한 냄새를 없애주는 간단한 방법으로 마늘이나 김치를 열처리하는 것이 대단히 효과가 있음이 밝혀졌다.

SPME를 이용한 수용액중의 휘발성 유기화합물 분석 (Determination of Volatile Organic Compounds (VOCs) in Drinking Water using Solid Phase Microextraction (SPME))

  • 16종의 휘발성 유기화합물을 blank water에 첨가하고 고정상으로 $100{\mu}m$ polydimethyl siloxane을 입힌 solid phase microextraction(SPME) fiber를 사용하는 headspace SPME방법으로 추출하여 gas chromatography/mass spectrometry (GC/MS)로 분석하였다. 이 방법의 평균회수율은 97%, 평균상대표준편차는 4.7%, 그리고 검출한계는 $0.01-0.5{\mu}g/l$를 나타냈다. 즉 SPME 방법을 이용한 수용액 중의 휘발성 유기화합물의 결과는 현재 많이 사용되고 있는 기존의 다른 방법보다 우수하고 편리한 방법으로 나타났다.

A Simple and Simultaneous Analysis of Volatile Halogenated Hydrocarbons in Indoor Air Using Personal Sampler

  • To understand the human exposure levels of volatile halogenated hydrocar-bons in ambient air, a new rapid and convenient analytical method for determination of the compounds in gaseous phase was evaluated and established. The method is based upon passsive diffusion to personal sampler containing adsorbent and solvent extraction followed by purge trap/ on-column cryof-ocusing method. A new method needs no special instrumentation for gas collection because it is based upon the passive diffusion principle. The typical chromatogram obtained in this study proved that rapid and simultaneous determination of target analytes was possible with good resolution. The developed method was successfully applied to determine the volatile halogenated hydrocarbons in indoor air and the values obtained by this new method were compared with those by direct suction method. The concentration of carbon tetrachloride, 1,1,2-trichloroethylene, chloroform showed the values below 400$\mug/m^3$ except the maximum of 1,513$\mug/m^3$ of chloroform. 1,1,1-Trichloroethane showed approximately 1,000 to 5,000$\mug/m^3$ range of diurnal fluctuation in indoor air.
