본 연구를 통하여 피부 미용 소재로 알려져 있는 콜라겐 펩타이드를 섭취한 후 혈중에 나타나는 소화분해산물 중 콜라겐 특유의 아미노산인 hydroxyproline과 이것의 dipeptide 형태인 Pro-Hyp이 피부에 미치는 영향을 알아보고자 하였다. 피부 미용 소재를 경구 섭취하는 사람의 피부는 광노화 및 자연노화가 어느 정도 진행된 상태이므로, 이와 유사한 조건을 조성하기 위해 경구 투여를 시작하기 전에 hairless mouse에 7주간 주 3회 UV를 조사하여 주름을 선유발시켰다. 군 분리를 시킨 후 7주간 경구로 실험물질을 주 5회 반복 투여하면서, 주 3회 UV 조사를 병행하였으며, 시험 기간 중 임상증상, 체중변화를 관찰하였고, 3회 (0,4. 7주차) 피부주형을 채취하여 영상 분석을 실시하였으며, 2회 (0,7 주차)의 피부탄력측정 및 1회(7주차)의 TEWL측정을 병행하였다. 실험종료 후 피부조직의 병리조직학적 검사(H&E, Masson-Trichrome stain) 및 피부두께 측정을 실시하였다. 실험기간 중 이상 임상증상을 보이거나 사망한 개체는 없었으며, 체중에 대한 통계적으로 유의한 변화는 관찰할 수 없었다. 피부 육안관찰 시 NC군에 비해 UC군의 피부 주름 증가가 뚜렷하였으며, 시험군들은 UC군에 비해 주름 증가가 적었다. 경구 투여 0, 4, 7주차에 제작된 replica의 주름 지표를 통계 처리한 결과, UV/H군과 UV/P군은 DC군에 비해 감소경향은 보였으나 유의적인 차이는 없었다. 탄력측정검사 결과 UV를 쬐지 않은 NC군은 UC군에 비해 UA/UF, Ur/Ue, Ur/Uf 지표에서 유의적인 증가를 보였다. UV/H군과 UV/P군은 모두 0, 7주차 모두 유의적인 차이를 보이지 않았다. 피부 보습력 측정 결과 NC군 및 모든 실험군에서 UC군에 비해 유의적인 TEWL 감소를 관찰할 수 있었다. 피부 두께의 경우 NC군, UV/H군, UV/P군은 UC군에 비해 피부 두께가 유의적으로 감소하였다. 진피층에서의 교원질 확인을 위해 실시한 Masson-Trichrome stain의 경우 UC군에서는 상부 진피층에서의 염색상 소실이 관찰되었으나, 모든 실험군에서 진피층 손상이 거의 관찰되지 않아 실험물질에 의한 회복을 확인 할 수 있었다. 이상의 실험 결과 hydroxyproline, Pro-Hyp의 경구반복투여시 자외선에 의해 광노화가 유발된 피부에 보습 효과, 피부 두께 감소 효과, 진피층 손상 회복 효과를 유의적으로 확인할 수 있었으며, 단, 피부 주름 개선 정도는 통계적 유의성은 없이 경향성만을 확인할 수 있었다. Hydroxyproline과 이를 함유하는 dipeptide 형태의 Pro-Hyp를 경구 투여한 시험으로부터 확인한 피부 보습 및 진피층 회복 효과는 이 물질들이 유래된 콜라겐을 도포 또는 섭취했을 때 피부에 나타날 수 있는 작용들과 상통하며, 단독 미용 소재로도 활용될 수 있는 가능성을 보여주었다. 본 연구에서는 콜라겐 펩타이드를 섭취 시 체내에 흡수되어 혈액을 통해 각 조직에 운반되는 분해 산물이 피부에 미치는 영향을 동물 시험을 통해 살펴보았는데, 향후 연구에서는 관절, 혈관과 같은 다른 생체 조직들에 미치는 영향에 대해서도 검증해 볼 필요가 있다.
Objectives: To investigate anti-inflammatory and anti-arthritic effects of Gyulpidaehwangbakcho-tang (GDBT) extract in a murine model of rheumatoid arthritis. Methods: The mice received $100{\mu}g$ of bovine type II collagen in Freund's complete adjuvant by intradermal injection at the base of the tail on day 0 and a booster injection on day 21. The mice were orally administered with GDBT (200 or 50mg/kg dissolved in distilled water) daily from day 1 to day 21 after arthritis incidence, and monitored for disease incidence and the severity of arthritis up to day 21. In order to evaluate the effect of GDBT on disease progression, we examined pro-inflammatory cytokines including IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II. Results: GDBT produced a significant and dose dependent inhibition of arthritis and inflammation during the entire duration of the study. This action was characterized by the decreased production of IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2, and NOS-II in vivo. Conclusion: We believe that the anti-arthritic activity of GDBT is due to its modulatory effect on the expression of pro-inflammatory cytokine in the synovium. Our results contribute towards validation of the traditional use of GDBT in the treatment of RA and other inflammatory joint disorders.
This study was performed to investigate the effects of high pressure/high temperature (HPHT) treatment on the recovery efficiency and characteristics of porcine placenta hydrolysates. The placenta hydrolysates were characterized by solubility, free amino acid contents, gel electrophoresis, gel permeation chromatography (GPC) and amino acid composition. Placenta was treated at 37.5 MPa of pressure combined with various temperatures (150, 170, and $200^{\circ}C$) or various holding times (0, 30, and 60 min at $170^{\circ}C$). Insoluble raw placenta collagen was partially solubilized (> 60% solubility) by the HPHT treatment. Free amino group content of placenta collagen was increased from 0.1 mM/g collagen to > 0.3 mM/g collagen after HPHT treatment, reflecting partial hydrolysis of collagen. The molecular weight ($M_w$) distribution showed evidence of collagen hydrolysis by shifting of $M_w$ peaks toward low molecular weight when treated temperature or holding time was increased. Alanine (Ala), glycine (Gly), hydroxyproline (Hyp), and proline (Pro) contents increased after the HPHT treatments compared to a decrease in the others. In particular, the increase in Gly was obvious, followed by Hyp and Pro, reflecting that placenta hydrolysates were mainly composed of these amino acids. However, increasing temperature or holding time hardly affected the amino acid compositions. These results indicate that the HPHT treatment is advantageous to hydrolyze collagen derived from animal by-products.
Membrane-type 1 matrix metalloproteinase (MT1-MMP) is a zinc-dependent proteinase found in cholesterol-rich lipid rafts on the plasma membrane. MT1-MMP hydrolyzes extracellular matrix (ECM) proteins, activates pro-matrix metalloproteinase-2 (proMMP-2) and plays an important role in ECM remodeling, cancer cell migration and metastasis. The role of caveolin-1, an integral protein of caveolae, in the activation of MT1-MMP remains largely unknown. Here, we show that the expression of caveolin-1 attenuates the activation of proMMP-2, reduces proteolytic cleavage of ECM and inhibits cell migration. We utilized the cytoplasmic tail domain deletion (${\Delta}CT$) or the E240A mutant of MT1-MMP. Co-expression of caveolin-1 with the wild-type or the ${\Delta}CT$ MT1-MMP decreased the proMMP-2 activation and inhibited collagen degradation and cell migration. Caveolin-1 had no effect on the catalytically inert E240A MT1-MMP. Our findings suggest that caveolin-1 is essential in the down-regulation of MT1-MMP activity by promoting internalization from the cell surface.
Type I and type II collagens are considered the major collagens of bone and cartilage respectively. Monitoring the patterns of those gene and protein expressions during development will provide a basis for the understanding of the normal and abnormal growths. This study was undertaken to investigate the expression of collagen genes and proteins involved in the developing human mandible. Fifty embryos and fetuses were studied with Alcian blue-PAS, Masson's Trichrome, reverse transcription polymerase chain reaction (RT-PCR), Western blot analysis, and Southern blot analysis. Our results showed that $pro-{\alpha}1(II)$ collagen gene expression begins in the 5th week. Type II collagen is synthesized in mesenchymal cells in advance: of overt chondrogenesis. The gene expression for type II collagen was highest during the appearance of Meckel's cartilage. There was a switch in collagen protein expression from type I to type II during the appearance stage of Meckel's cartilage. The distribution of the mRNA for type II collagen corresponded well with the pattern of type II collagen protein. The endochondral ossification was observed where there was direct replacement of cartilage by bone.
Objectives: Although increased reactive oxygen species (ROS) is caused by stress accelerates collagen degradation, there was no data on the relationship between stress and urinary hydroxyproline (Hyp) and proline (Pro), a good marker of collagen degradation. The purpose of this study was to evaluate the relationship between depression, anxiety, and stress (DAS) and concentrations of urinary Hyp and Pro. Methods: 97 hospital employees aged 20 to 58 were asked to fill out comprehensive self-administrated questionnaires containing information about their medical history, lifestyle, length of the work year, shit-work and DAS. Depression Anxiety Stress Scale (DASS) was applied to evaluate chronic mental disorders. Urine samples were analyzed using High Performance Liquid Chromatography (HPLC) with double derivatization for the assay of hydroxyproline and proline. Results: The mean value of Hyp and Pro concenturation in all subjects was $194.1{\pm}113.4\;{\mu}mol/g$ and $568.2{\pm}310.7\;{\mu}mol/g$. DASS values and urinary Pro concentrations were differentiated by sex (female > male, p < 0.05) and type of job (nurse > others, p < 0.05). In the stepwise multiple linear regressions, urinary Hyp and Pro concentrations were influenced by stress (Adjusted $r^2$ = 0.051) and anxiety and job (Adjusted $r^2$ = 0.199), respectively. Conclusions: We found that stress and anxiety were correlated with urinary Hyp and Pro concentrations. To identifying a definite correlation, further study in large populations will be needed.
본 연구에서는 주박의 기능성 화장품 소재로서의 활용 가능성을 평가하기 위해 분획물에 따른 항산화 활성평가와 인간섬유아세포인 CCD-986sk 세포에서의 주름효능 평가를 진행하였다. ABTS+ radical 소거능 측정을 통해 항산화 활성을 확인한 결과, 주박 Ethyl Acetate 분획물 1,000 ㎍/ml 농도에서 75.5%로 나타나 분획 용매 중 가장 높은 항산화 활성을 보였다. Elastase, Collagenase 저해 활성 측정을 통해 주름 개선 효과를 확인하였으며, 두 실험 결과 모두 주박 Ethyl Acetate 분획물이 1,000 ㎍/ml 농도에서 가장 높은 효능을 나타내었다. UVB로 유도된 CCD-986sk 세포 내 pro-collagen type I의 합성율을 측정한 결과 주박은 동일농도에서 20 ㎍/ml에서 Ethyl Acetate, Water, Acetonitrile, Hexan 분획물 순으로 높은 효능을 보였다. 콜라겐 분해효소인 MMP-1의 저해률을 측정한 결과 4가지 용매 분획몰은 20 ㎍/ml에서 모두 70% 이상의 효능을 나타내었다. Real time PCR 실험에서 pro-collagen type I, MMP-1 및 MMP-3의 mRNA 발현량을 측정한 결과 UVB 단독군 대비 Jubak 추출물을 함께 처리했을 때 pro-collagen type I의 단백질 발현량이 증가되었고, MMP-1과 MMP-3의 mRNA 발현량은 감소하는 것을 확인하였으며 대조군(EGCG) 다음으로 Ethyl Acetate 추출물이 주름 개선에 가장 효능이 우수하였다. 이상의 결과로 주박의 용매 분획물 중 Ethyl Acetate 분획물은 UVB 자극에 의한 광노화에 대한 항주름 효과가 있음을 확인하여 화장품의 천연물 소재로서의 이용이 기대된다.
Objectives : This study was carried out to investigate the anti-inflammation, anti-development and curative effects of Oyaksunki-sangamibang (OSKM) on collagen-induced arthritis in Wistar rats and ICR mice. Materials & Methods : D experiment part II, the inhibitory effects of nitric oxide synthesis, pro-inflammatory cytokines, and cyclooxygenase were studied. In experiment part II, paw eduma volume and thickness of ankle joint were measured at 0, 10, 15, and 20 days after immunization. The incidence and arthritis score were evaluated 14 days after immunization, At 15 days after immunization, serum $TNF-\alpha$ was analyzed. In experiment part III paw edema volume and thickness of ankle joint were measured at 0, 10, and 15days after treatment. At 15 days after treatment, serum $TNF-\alpha$ was analyzed. Results : In experiment part I: 1. Nitric oxide synthesis ·md pro-inflammatory cytokines were inhibited significantly by OSKM extract. 2. Cyclooxygenase 2 (COX-2) was inhibited by OSKM extract. In experiment part II: Paw edema volume, thickness of ankle joint and serum $TNF-\alpha$ level of the teated group were significantly decreased compared with the control group at 20 days after immunization. In experiment part III: Incidence of arthritis was $70\%$. OSKM-treated group had no significant change on paw edema volume, thickness of ankle joint and serum $TNF-\alpha$ level. Conclusions : These results indicated that OSKM has anti-inflammation effects on the ICR mouse, and higher inhibitory effects on the onset but lower inhibitory effects on the progression of collagen-induced arthritis in rats.
A series of 5-oxaproline peptide derivatives was synthesized and evaluated for its ability to inhibit the prolyl 4-hydroxylase in vitro. Structure-activity studies show that the 5-oxaproline sequences, prepared by the 1,3-dipolar cycloaddition of the C-methoxycarbonyl-N-mannosyl nitrone in the presence of the ethylene, are more active than the corresponding proline derivatives. Prolyl 4-hydroxylase belongs to a family of $Fe^{2+}-dependent$ dioxygenase, which catalyzes the formation of 4-hydroxyproline in collagens by the hydroxylation of proline residues in -Gly-Xaa-Pro-Gly- of procollagen chains. In this paper we discover the more selective N-Cbz-Gly-Phe-Pro-Gly-OEt $(K_m\;=\;520\;{\mu}M)$ sequences which are showed stronger binding than others in vitro. Therefore, we set out to investigate constrained tetrapeptide that was designed to mimic the proline structure of pep tides for the development of prolyl 4-hydroxylase inhibitor. From this result, we found that the most potent inhibitor is N-Dansyl-Gly-Phe-5-oxaPro-Gly-OEt $(K_i\;=\;1.6\;{\mu}M)$. This has prompted attempts to develop drugs which inhibit collagen synthesis. Prolyl 4-hydroxylase would seem a particularly suitable target for antifibrotic therapy.
본 연구는 새로운 기능성 화장품 소재 개발을 위해 복분자 씨앗의 구성성분인 casuarictin을 이용하여 섬유아세포내에서의 항노화 효과를 확인 하고자 하였다. Casuarictin의 항노화 효과를 CCD-986sk 세포를 이용하여 측정한 결과, casuarictin은 UVB에 의해 증가된 ROS 발현을 억제하였으며, pro-collagen의 발현을 증가시키고 MMP-1의 발현을 억제하는 것으로 나타났다. 이는 노화 인자인 활성산소종의 생성 억제와 주름과 밀접한 관련이 있는 콜라겐의 합성 및 분해억제를 통해 항산화 및 주름개선 효과가 있는 것으로 판단되며, 따라서 casuarictin은 화장품 분야에서 항노화 소재로서의 활용가치가 있을 것으로 사료된다.
