Screening of Chitin Synthase II Inhibitors from Medicinal and Wild Plants (고등식물로부터 Chitin Synthase II 활성 저해물질의 탐색)

  • Chitin is an important structural component of fungal cell wall and is synthesized by chitin synthase I, II, and III. The chitin synthase II is an essential enzyme for the formation of primary septum in Saccharomyces cerevisiae. Therefore, specific inhibitors of this enzyme might block the formation of fungal cell wall and could be used as effective antifungal agents. To search chitin synthase IIinhibitors from natural products, 67 plants were extracted with methanol and examined for the inhibitory activities against chitin synthase II of S. cerevisiae by our cell free assay system. As a result, the extracts from 16 plants showed more than 70% inhibition at the concentration of $280{\;}\mu\textrm{g}/ml$. Of note, Laurus nobilis (81.4%), Lonicera maackii (81.5%), Berchemia berchemiaefolia (82.9%), Koelreuteria paniculata (87.9%), Chamaecyparis pisifera (86%) and Taxus cuspidata (83.9%) inhibited strogly the chitin synthase IIactivity.

Primary Squamous Cell Carcinoma of the Posterior Glottis (성문후부(Posterior Glottis)에서 원발한 편평상피세포암종 1례)

  • The glottis consists of two parts : The intermembranous portion or the anterior glottis, and the posterior glottis. The posterior glottis has been described by various inappropriate terms such as posterior commissure and interarytenoid region. The structure surrounding the posterior glottis consists of three portions ; The cartilaginous portion of the vocal fold, the lateral wall of the posterior glottis, and the posterior wall of the glottis. The posterior glottis may be a very rare site for primary squamous cell carcinoma of the larynx because of its embriologic and histologic differences from the anterior glottis. Recently we have experienced a case of a 81-year-old woman who had been presented with throat discomfort and voice change for 10months. She had the smoking history of 40 pack-year. The physical examination revealed poorly demarcated, papillary, whitish-pink colored mass on the posterior glottis. The vocal cord mobility was not affected. Biopsy under the suspension larygoscope showed moderately differentiated squamous cell carcinoma. No regional and distant metastasis was found. She was irradiated with 7000cGy over 7 weeks at a daily dose of 200cGy. No evidence of recurrence was found by the fifteenth month following radiation therapy. We report this case with a review of literature.

Isolation of Actinomycetes Producing Extracellular Adenosin Deaminase (세포외 Adenosine Deaminase를 생산하는 방선균의 분리)

  • Two strains of actinomycetes producing extracellular adenosine deaminase, strain J-845S and strain J-326TK, were isolated from soil. Strain J-845S was gram-positive and non-acid-fast. This strain formed whitish, rod-shaped, smooth and non-motile spores on the aerial mycelium, and the spore chain was spiral. The hyphae of the mycelium branched abundantly. Cell wall chemotypes of the strain were of type I containing LL-diaminopimelic acids, and of phospholipid type II, and then strain J-845S was designated as Streptomyces sp.. Strain J-326TK was gram-positive and non-acid-fast. The hyphae of primary and aerial mycelium fragmented into irregular rod of coccus-like elements. The aerial mycelium either did not branch or sparsely branched. Cell wall composition was of type I and phospholipid type I. Thus, strain J-326TK was identified as Nocardioides sp.

Effects of Reynolds Number on Flow and Heat/Mass Characteristics Inside the Wavy Duct (Reynolds 수에 따른 꺾어진 덕트에서 열/물질전달 특성 고찰)

  • The present study investigates effects of flow velocity on the convective heat/mass transfer characteristics in wavy ducts of a primary surface heat exchanger application. Local heat/mass transfer coefficients on the wavy duct sidewall are determined by using a naphthalene sublimation technique. The flow visualization technique is used to understand the overall flow structures inside the duct. The aspect ratio and corrugation angle of the wavy duct is fixed at 7.3 and 145$^{\circ}$ respectively, and the Reynolds numbers, based on the duct hydraulic diameter, vary from 100 to 5,000. The results show that there exist complex secondary flows and transfer processes resulting in non-uniform distributions of the heat/mass transfer coefficients on the duct side walls. At low Re (Re<1000), relatively high heat/mass transfer regions like cell shape appear on both pressure and suction side wall due to the secondary vortex flows called Taylor-Gortler vortices perpendicular to the main flow direction. However, at high Re (Re>1000), these secondary flow cells disappear and boundary layer type flow characteristics are observed on pressure side wall and high heat/mass transfer region by the flow reattachment appears on the suction side wall. The average heat/mass transfer coefficients are higher than those of the smooth circular duct due to the secondary flows inside wavy duct. And also friction factors are about two times greater than those of the smooth circular duct.

Characteristics of Water Soluble Fractions of Wheat Bran Treated with Various Thermal Processes (열처리 밀기울의 수용성 분획의 특징)

  • Water soluble fractions (WSF) of wheat bran treated with thermal processes such as autoclaving, microwaving and extrusion were characterized to investigate the structural response of plant cell wall to thermal and mechanical energy. From the chemical analysis and gel filtration chromatography of WSF, gelatinization of starch was found to be the primary solubilizing mechanism of wheat bran, followed by the structural disintegration of fibrous non-starch cell wall materials. It was also found that extrusion process resulted in degrading relatively higher molecular weight non-starch polysaccharides from the cell wall. GC analysis of water soluble non-starch polysaccharides indicates that the arabinoxylan residues of cell wall are the most susceptible site to thermal treatments studied. In particular, the degrading degree of cell wall of wheat bran is the most significant for extrusion accompanying both high temperature and high shear.

  • A primary giant cell tumor of the rib is very rare. The most common site of a giant cell tumor arising from the rib is the posterior arc. A giant cell tumor arising from the anterior arc of the rib is extremely rare. The treatment of a giant cell tumor of the rib is not well defined. Generally, a complete surgical resection is performed in a patient with a primary giant cell tumor of the rib. We report a case of a giant cell tumor arising from the anterior arc of the rib that was treated with a wide excision and chest wall reconstruction.

  • Structural changes of the endosperm of Panax ginseng C.A. Meyer from fertilization to germination were investigated by light microscope. The endosperm of the ginseng seed is cellular type. Since endosperm cells adjacent embryo continuously breakdown and disappear with the elongation of embryo, the real of endosperm is gradually decreased. As the anatropous ovules of immature seed with green seed coat developes more and more, ovary cells adjacent ovary cavity become abundant by the periclinal division, their size is decreased, hypotrophy of cell wall discern, and they are gradually differentiated in seed coat. Though embryo responds strongly to basic dye at the stage of completion of endosperm formation, tissue of endosperm responds to acidic dye positively Cell wall of embryo and endosperm are composed of primary cell wall not lignified. Endosperm cells adjacent embryo begin to breakdown in the endosperm tissue of indehiscent seed before the beginning of the after-ripening. Dehiscent seed of which seed coat is opened through after-ripening represent the form as a seedling in the result of embryo developments with the formation of organs; radicle, cotyledon, plumule. Umbilifom layer represents strong positive response to the toluidine blue and the basic function. Umbiliform layer that endosperm cells breakdown and disappear is observed clearly at the periphery of the embryo cotylemon, while slightly at the periphery of the radicle.

The Biology of Phenolic Containing Vesicles

  • Phenolic compounds play a major role in the interaction of plants with their environment. They are thought to have been a feature of higher plants since early colonization of the land. Phenolics are crucial for many important aspects of plant life. They can play structural roles in different supporting or protective tissues, for example in cell walls, they can be involved in defence strategies, and signalling properties particularly in the interactions between plants and their environment. In brown algae, phenolic compounds are contained within membrane bound vesicles known as physodes, and their roles in algae are thought to be similar to those of higher plant phenolics. They can be stained using various histochemical stains, however, none of these stains are phenolic specific so care must be taken during interpretation of such results. Many, but not all phenolics are also autofluorescent under UV or violet light. Physodes are involved in cell wall construction, both in primary and secondary walls in brown algae. They bind together with other wall components to make a tough wall. They have also been found to play a role at fertilization, in blocking polyspermy in some species. Sperm are very quickly rendered immobile after phenolic release from newly fertilized zygotes seconds after fertilization. Phenolic compounds are thought to be important herbivore deterrents in some species due to their astringent nature. Phenolic compounds also offer effective UV protection in the early life stages and also the adults of many algal species. In the future, this factor may also make them an important player in the pharmaceutical and skincare industries.

  • Primary cardiac tumors are very uncommon. Seventy five percent of them are benign tumors and these are mostly myxomas. The malignant cardiac tumors, the majority of which are undifferentiated sarcomas, comprise up to 25% of all cardiac tumors. A primary malignant sarcoma of the myocardium is exceedingly rare. Thus, there have been very few such cases reported in the literature. We present here a case of a 15 year old man who had complaints of orthopnea and increasing exercise intolerance over a one month period. Transthoracic echocardiography demonstrated a well demarcated huge mass with left ventricular inflow obstruction on the posterior wall of the left atrium. The patient's symptoms were relieved by surgery. The histological diagnosis was an unclassified spindle cell sarcoma.

  • The objective of this investigation is to examine the influence of the micro catheters, which mimic the intravascular micro active endoscopes, on local pressure changes and flow rate in an arterial branch model similar to the femoral artery of human. The effects of branch to main lumen flow rate ratios and the locations of a catheter tip were found to be significant on the local pressure changes. Relatively large pressure drops and an increase in shear stress due to the obstruction effects may induce an endothelial cell damage and a change in arterial wall permeability, which have been reported to be the primary cause of the initiation of the atherosclerosis and other major vascular diseases.
