Development of Tobacco Mosaic Virus (TMV) resistant Flue-cured $\textrm{F}_1$ Hybrid, KF118

  • Tobacco mosaic virus (TMV), TMV-common strain, causes severe damage at growth, yield and leaf quality on flue-cured tobacco in Korea. Controlling TMV outbreak with cultivating practices and chemicals are limited, thus, the best way would be developing TMV resistant variety. A new flue-cured hybrid, KF118, was developed from a cross of maternal parent MSTC175 and TC853 at the Agro-tech. Research Group, KT&G Central Research Institute. KF118 exhibited more vigorous growth characteristics in early stage than that of NC82, standard variety in Korea, while other characters were favorable to those of NC82, It is highly resistant to TMV and bacterial wilt(Ral-stonia solanacerum), and black shank(phytophthora parasitica var. nicotianae) is comparable to that of NC82. It should adapt well to the flue-cured production area, and can reduce premature flowering under unfavorable weather conditions. Yield of KF118 is 1 to 2% higher, and leaf quality is 4% higher than NC82. Fl hybrid, KF118, met acceptable standards for chemical and physical characteristics of cured leaf, and ranked high in good smoking taste by panel members in KT&G.

Influence of degree of maturity and duration of cold treatment on the growth and flowering of L. elegans 'Connecticut King' bulb (Lilium elegans 'Connecticut King'의 촉성재배(促成栽培)에 있어서 구근(球根)의 숙도(熟度) 및 저온처리기간(低溫處理期間)이 생육(生育) 및 개화(開花)에 미치는 영향(影響))

  • Effects of harvest date, number of the leaves remained on stump after flower cutting, duration of cold treatment on the growth and flowering of L. elegans 'Connecticut King' bulb were tested. Shoot emerging rate increased over 90% by delayed cold treatment, late harvesting time and more leaves remained. Flowering rate increased over 90% when conducted cold treatment of 90 days to the premature bulb at flowering date, the bulb matured for 50 days with 10 leaves and the bulb matured for over 30 days with 20 leaves, and when conducted cold treatment of 50 days to the bulb matured for over 30 days with all leaves. Delayed cold treatment duration shortened days needed to shoot emergence and flowering. Especially days to flower of the premature bulb at flowering date were the shortest of all treatments, and can be flowered within 100 days after planting. In generally, plant height, number of flowers per plant and stem diameter generally increased by longer bulb maturing period after flower cuting, and the premature bulb harvested at flowering date showed poor growing and flowering responses. But, if used the larger size bulb, it is believed that the prematured bulb can be forced to flower normally in current year without bulb maturing period.

Effects of Polyamine on Flowering in Lemna gibba G3 (좀개구리밥(Lemna gibba G3)의 개화에 미치는 Polyamine의 영향)

  • The flowering in Lemna gibba G3, a long-day plant, was promoted under continuous light by agmatine, putrescine, spermidine and spermine present in the culture medium. Methylglyoxal-bis (guanylhydrazone) (MGBG) and cyc10hexylamine (CHA), inhibitors of polyamine biosynthesis, were found to suppress the flowering in the plants. The vegetative grov.1h rate was kept constant while the flowering was being promoted by the pOlyamines, and the inhibitors with depressive effect on flowering showed stimulatory effect on vegetative grov.1h. The pattern of vegetative growth during floral promotion or depression was an indication that the promotive action of the pOlyamines and the suppressive effect of the inhibitors may be outcome of their possible involvement specifically in the flowering process rather than in broad spectrum of growth of L. gibba G3. The degree of promotive action of spermdine and spermine could not be altered (or lessened) by simultaneous application of their inhibitors to the medium. This phenomenon indicates that the flowering process in L. gibba G3 may largely be dependent to the status of endogenous spermidine and spermine. Endogenous level of spermidine in florally induced Lemna, was found to rapidly increase. In 24 h of floral induction, the content reached at the level 2 times higher than that in non-induced plants. The elevated level of spermidine provides an additional, though premature, evidence supporting the postulation that endogenous polyamine status might play an important role in the very early stage of floral induction in L. gibba G3.bba G3.

  • Flowering is one of the most important development traits related to the production of Brassica rapa crops. After planting, a sudden low temperature triggers premature flowering, which leads to a reduction in the yield and quality of harvested production. Therefore, understanding the mechanism of flowering control is important in the agricultural productivity for preventing Brassica rapa crops. Vernalization is generally known as the main factor of flowering in the Brassica plant. However, in the subspecies of Brassica rapa, some accession such as Yellow sarson and Komatsuna display the flowering phenotype without vernalization. Circadian genes, which diurnally regulate plant physiology, have a role for photoperiodic flowering but are related to the regulation of the vernalizarion mechanism. In this report, the 22 B. rapa accession were divided into two groups, vernalization and non-vernalization, and the sequenced circadian gene, BrPRR1s. Among them, the BrPRR1b gene was found to have deletion regions, which could classify the two groups. The PCR primer was designed to amplify a short band of 422bp in the vernalization type and a long band of 451bp in the non-vernalization type. This primer set was applied to distinguish the flowering types in the 43 B. rapa accession and 4 Brassica genus crop, Broccoli, cabbage, mustard, and rape. The PCR analysis results and flowering time information of each crop demonstrated that the primer set can be used as marker to discern the flowering type in Brassica crops. This marker system can be applied to the B. rapa breeding when selecting the flowering character of new progenies or introducing varieties at an early stage. In addition, these results displayed that the circadian clock genes can be a good strategy for the flowering control of B. rapa crops.

Comparison of Diploid and Haploid Plants for Cool Temperature and Short - day in Nicotiana tabacum L. (연초의 이배체와 반수체의 저온단일 감응성 비교)

  • Response of two ploidy levels to cool temperature and short day treatment were investigated under controlled conditione of Phytotron. The haploid and diploid of seven genotypes were started and grown to the 8- leaf stage in the greenhouse. They were treated during 15 and 20 days to 8- hour photoperiods at 18$^{\circ}C$ in controlled - environmental room to induce premature flowering, respectively. Diploid plants of seven genotypes flower later than their haploid plants at 20 days treatment. But under 15 days treatment, diploid plants of NC82, Hicks, BY4, NC2326 and Coker86 were not different from their haploid plants for days to flower. Diploid plants of seven genotypes developed the same number of leaves as their haploid plants at 20 days treatment. Under 15 days treatment, diploid plants of Coker347 and NC95 developed more leaves per plant than their haploid plants. Correlation coefficient between the ranks of leaves per plant of seven genotypes at two ploidy levels was 0.964 and 0.929 at 15 and 20 days treatment, respectively.

Development of a New Flue - cured Tobacco Variety KFl13 by Nicotiana africana Method (Nicotiana africana 방법에 의한 황색종 연초 신품종 KF113 육성)

  • A new flue - cured tobacco variety KF113 was developed by Nicotiana africana method from a cross of NC82 and Coker 347 at the Suwon Experiment Station. It was tested in the 0fficial Variety Test in 1991-1992 and the flue - cured Regional Farm Test in 1992. KFl13 flowers 4 days later than WC82 (standard variety in Korea) and its harvestable leaves are 2 more than those of NC82. The leaf type and shape of KFl13 resembles Coker 347. It has high resistance to bacterial wilt (Pseudomonas solanacearum) and black shank (Phytophthora parasitica var. nicotianae), and is susceptible to tobacco mosaic virus. It should adapt well to the flue - cured production area and can reduce premature flowering under unfavorable weather conditions. Yield of KFl13 is 5% higher than that of NC82, and nearly equal in value per kg compare with NC82. This variety met acceptable standards for chemical and physical characteristics of cured leaf and for smoking taste evaluated by panel members in Korea Ginseng & Tobacco Research Institute.

  • Tobacco mosaic virus (TMV), TMV-common strain, causes severe damage on flue-cured tobacco in Korea. As the results, breeding programs were conducted to transfer the TMV resistance from NC567 to other cultivars. Advantage of F1, hybrid may not lie only in their heterotic value but also in directly combining of valuable dominant traits, e.g. for disease resistance, either in pairs or as groups already assembled in the two parents. A new TMV resistant flue-cured tobacco variety KF114 was developed from a cross as maternal parent MSNC567 and NC82 at the Suwon Experiment Station. KF114 was more vigorous growth characteristics in early stage than that of NC82, standard variety in Korea, and other characters were similar to those of NC82. It os high resistance to TMV, but bacterial wilt(Pseudomonas solanacerum) and black shank (Phytophthora parasitica var. nicotianae) is comparable to NC82. It should adapt well to the flue-cured production area and can reduce premature flowering under unfavorable weather conditions. Yield of KF114 is 1% to 5% higher, and nearly equal in value per KG compared with NC82. This variety, F1 hybrid, met acceptable standards for chemical and physical characteristics of cured leaf and high ranked good smoking taste by panel members in Korea Ginseng and Tobacco Research Institute.

Physiological and molecular characterization of two inbred radish lines with different bolting times (추대시기가 서로 다른 무 계통간 생리학적, 분자생물학적 개화 특성 규명)

  • The radish (Raphanus sativus L.) is an important Brassicaceae root vegetable crop worldwide. Several studies have been conducted concerning radish breeding. There are major challenges to prevent premature bolting in spring plantings. Here, we performed the characterization of two inbred radish lines which vary in bolting time. "Late bolting radish" (NH-JS1) and "early bolting radish" (NH-JS2) were generated by a conventional breeding approach. The two inbred lines showed different bolting phenotypes depending on vernalization time at $4^{\circ}C$. NH-JS1, the late bolting radish, was less sensitive to cold treatment and the less sensitivity was inversely proportional to the duration of the vernalization. We also measured gene expression levels of the major bolting time related genes in the NH-JS1 and NH-JS2 lines. RsFLC1 plays a central role in the timing of flowering initiation. It is a strong repressor and it's transcript is highly expressed in NH-JS1 compared to NH-JS2 under no treatment and vernalization conditions. RsFRI, a positive regulator of RsFLC, is also highly expressed in NH-JS1 compared to NH-JS2 regardless of vernalization. In contrast, RsSOC1, suppressed by FLC as a floral integrator gene, showed the most difference, a 5-fold increase, between NH-JS1 and NH-JS2 under vernalization conditions. From these results, we conclude that NH-JS1 showed a late flowering phenotype after cold treatment due to the expression differences of flowering time regulator genes rather than difference sensitivity to cold. These results may be useful to understand the control mechanisms of flowering time and may help identify molecular markers for selecting late bolting trait in radish.

The effect of potash on the growth and yields of soybean at different level of soil fertility and application of fertilizer (토양(土壤)과 시비(施肥)를 달리할때 대두생육(大豆生育)에 미치는 가리(加里)의 영향(影響))

  • In order to study the effect of potash on the growth and yields of Soybean at different level of soil fertility and application of fertilizer (nitrogen, phosphate and calcium), $2^3$factorical experiment was carried out by pat culture with variety 'Chang-dan-baec-muc' which is most spreaded variety in Korea. The experiment consisted of five replications in a randomized block experiment with three factors (soil, fertilization and potash). Treatment were at two levels; infertile and fertile soil, none and some of fertilization and potash. Thus, the experiment comprised eight treatment combinations which consisted of all combinations. The results of this experiment are as follows: 1. No effect of each of three factors on flowering date was found. 2. Leaf-yellowing and maturing date was quickened on the fertile soil but no effect of fertilization and potash was found. 3. More premature leaf-yellowing was found on the fertile soil. 4. Deeper leaf colour cuss showed on the fertile soil and in the case of fertilization but no effect of potash was found. 5. Increasing tendency of following character: length and width of leaf, height and dia of stem, number of branches and pods; was most remarkable on the fertile soil. Application of fertilizer showed also remarkable tendency of increasing, while increasing tendency of potash was the least. 6. Same tendency was found with following charactors; weight of total plant. stem and shell, and commercial grains, weight of 100 grain and number of commercial grains. 7. As the results of analysis of variance for weight of commercial grain it, was found the teach of the three factors increased soybean yields significantly (weight of commercial grain) but the effect of potash was less than the other two factors. No significant interaction was found among three factors. 8. Greater effect of potash on increasing soybean yields was found on the fertile soil, and in the case of fertilization.
