Protopanaxadiol modulates LPS-induced inflammatory activity in murine macrophage RAW264.7 cells

  • Protopanaxadiol (PPD) is a mixture of protopanaxadiol type saponins with a dammarane skeleton, from Korean red ginseng (Panax ginseng C.A. Meyer; Araliaceae). Korean ginseng is well-known herb to treat almost all kinds of diseases in Oriental medicine. This herb was particularly prescribed for treatment various inflammatory diseases, including rheumatoid arthritis, atherosclerosis, and diabetes mellitus, for centuries. To understand the efficacy of ginseng against inflammatory diseases, we aimed to show anti-inflammatory activities of the PPD in murine macrophage cell line, RAW264.7 cells using nitric oxide (NO) production assay and the expressions of pro-inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), and IL-6, and monocyte chemotactic protein-1 (MCP-1). We found that PPD saponin significantly blocked LPS ($1{\mu}g/ml$)-induced NO production in a dose-dependent manner. In addition, PPD abrogated the expressions of LPS-induced pro-inflammatory cytokines, such as IL-$1{\beta}$ and MCP-1. Moreover, cyclooxygenase (COX)-2, a critical enzyme to produce prostaglandin E2 (PGE2), was significantly inhibited by PPD in LPS-activated RAW264.7 cells. Taken together, these results suggested that anti-inflammatory efficacy of Korean red ginseng on inflammatory diseases is, at least, due to the NO inhibitory activity and the inhibition of the expressional level of inflammatory cytokines and/or mediators.

Tentative identification of 20(S)-protopanaxadiol metabolites in human plasma and urine using ultra-performance liquid chromatography coupled with triple quadrupole time-of-flight mass spectrometry

  • Background: 20(S)-Protopanaxadiol (PPD), the aglycone part of 20(S)-protopanaxadiol ginsenosides, possesses antidepressant activity among many other pharmacological activities. It is currently undergoing clinical trial in China as an antidepressant. Methods: In this study, an ultra-performance liquid chromatography coupled with triple quadrupole time-of-flight mass tandem mass spectrometry method was established to identify the metabolites of PPD in human plasma and urine following oral administration in phase IIa clinical trial. Results: A total of 40 metabolites in human plasma and urine were identified using this method. Four metabolites identified were isolated from rat feces, and two of them were analyzed by NMR to elucidate the exact structures. The structures of isolated compounds were confirmed as (20S,24S)-epoxydammarane-12,23,25-triol-3-one and (20S,24S)-epoxydammarane-3,12,23,25-tetrol. Both compounds were found as metabolites in human for the first time. Upon comparing our findings with the findings of the in vitro study of PPD metabolism in human liver microsomes and human hepatocytes, metabolites with m/z 475.3783 and phase II metabolites were not found in our study whereas metabolites with m/z 505.3530, 523.3641, and 525.3788 were exclusively detected in our experiments. Conclusion: The metabolites identified using ultra-performance liquid chromatography coupled with triple quadrupole time-of-flight mass spectrometry in our study were mostly hydroxylated metabolites. This indicated that PPD was metabolized in human body mainly through phase I hepatic metabolism. The main metabolites are in 20,24-oxide form with multiple hydroxylation sites. Finally, the metabolic pathways of PPD in vivo (human) were proposed based on structural analysis.

Decreased GCF DEL-1 and increased GCF neutrophils with increasing probing pocket depth

  • Purpose: Developmental endothelial locus-1 (DEL-1) plays a role in regulating neutrophil migration within the periodontium. The objective of this study was to evaluate the levels of DEL-1 in saliva and gingival crevicular fluid (GCF), as well as the number of neutrophils in patients with periodontitis. Methods: Forty systemically healthy, non-smoking periodontitis patients participated in this study. Clinical periodontal parameters, including the plaque index, probing pocket depth (PPD), clinical attachment level, bleeding on probing, modified sulcular bleeding index, and marginal bone level, were measured. Levels of DEL-1, interleukin (IL)-1β, IL-6, and IL-8 in unstimulated saliva samples, as well as DEL-1 in the GCF of 3 teeth from each participant, were assessed. Neutrophil counts in oral rinse and GCF samples were recorded. Spearman correlation coefficients were used to examine the correlation between protein levels, clinical parameters, and neutrophil quantities. Participants were divided into 2 age groups (those under 50 years and those 50 years or older) in order to investigate potential age-related differences. Results: DEL-1 levels in the GCF showed a negative relationship with PPD (sum). Neutrophils in oral rinse samples were positively correlated with PPD, IL-8, and IL-1β levels. Neutrophils in GCF exhibited a positive correlation with PPD (sum). Salivary DEL-1 levels showed correlations with IL-8 and IL-1β, but not with the clinical parameters of periodontitis. Conclusions: The negative relationship observed between PPD and GCF DEL-1 levels is consistent with the proposed protective role of DEL-1.

Analysis of optimal activities according to thermal comfort in the forest: focusing on a program for the elderly at the National Forest Therapy Center

  • The purpose of this study was to scientifically activate the forest healing program activities for the elderly. The predicted mean vote (PMV) and predicted percentage of dissatisfied (PPD), which are indices of thermal comfort in the thermal environment, and degree of canopy closure were compared and analyzed. Based on this information, the study objective was to present the appropriate conditions for maintaining the best comfort for the elderly. Six deck road shelters, which are the most active locations in forest healing programs among the National Center for Forest Therapy, were selected as the study sites. The results indicated that in the case of the conditions of 1 clo (clothing insulation value) and 1 met (metabolic rate) at an air temperature of 19 to 21 degrees in September on the measurement date, the PMV values ranged between -1.85 and -0.98 at all sites, and PPD values ranged between 25.60% and 68.68%. On the other hand, in the case of 1.3 clo and 1.6 met conditions, the PMV values ranged between -0.08 and 0.23 for all sites and PPD values ranged between 5.40 and 6.18. As shown above, the difference in thermal environment comfort and satisfaction according to the condition of the amount of metabolism and the amount of clothing could be confirmed. In addition, an analysis of the relation between PPD and canopy closure suggested a significantly positive correlation between them, and it was found that canopy closure was a factor affecting thermal comfort. Studies on effects of forest thermal environmental comfort and canopy closure on forest healing program areas should be conducted extensively according to seasonal conditions to provide information that can be used for more effective forest healing programs.

우려 화학물질에 대한 필수 용도 개념 적용 - PFAS, 6PPD, Benzene 중심으로 - (Application of the Essential Uses Concept to Substances of Concern - PFAS, 6PPD and Benzene -)

  • Background: There is growing international recognition of the need for improvements to national chemical management systems for hazardous chemicals. The European Union has recently introduced the concept of 'essential uses' as a new approach to the management hazardous chemicals by limiting their uses. Objectives: This paper examines the concept of essential uses in chemical management and how to apply it through a case study of essential use. This approach is distinct from the current chemical management system, but seeks to improve its potential benefits by effectively restricting or gradually decreasing the use of hazardous substances. Methods: The concept of essential uses was introduced by reviewing the Montreal Protocol on Substances that Deplete the Ozone Layer, Cousins's three essentiality categories applied to PFAS, restriction options assessed in the PFAS restriction proposal under REACH, and the California Safer Consumer Products regulations prioritizing 6PPD in motor vehicle tires. Based on these essential uses concepts for PFAS and 6PPD, uses of benzene were classified in accordance with the essential uses approach for products using benzene in South Korea. Results: The essential use concept is able to manage the restriction and authorization of substances of concern through essential uses and non-essential uses and the feasible substitution of uses and substances. Conclusions: If the concept and methodology of essential uses are clearly established, they can be expected to shift the national chemical management paradigm from regulating substances to limiting uses under the existing substance management system.

결핵성 흉막염의 진단에서 결핵균 항원 자극에 의한 흉수 세포의 IFN-γ 분비 검사의 유용성 (The Utility of Pleural Fluid Cell IFN-γ Production Assay in the Diagnosis of Tuberculous Pleurisy)

  • 연구배경 : 결핵균 항원자극에 의한 흉수 세포의 $IFN-{\gamma}$ 분비검사가 결핵성 흉막염과 다른 원인의 흉수의 감별진단에 유용한지 알아보고자 하였다. 방 법 : 39명의 결핵성 흉막염 환자와 비결핵성 흉수를 보인 26명의 환자(부폐렴성 흉수 13명, 암성 흉수 13명)들의 흉수세포를 분리하여 배양하면서 PPD와 PHA로 24시간 자극한 후 배양상층액의 $IFN-{\gamma}$ 농도를 ELISA 방법으로 측정하였다. 결 과 : 1) 결핵성 흉수의 ADA 활성도 및 $IFN-{\gamma}$ 농도는 비결핵성 흉수보다 높았다(p < 0.01). 2) PPD로 흉수세포들을 자극하였을 때 분비되는 $IFN-{\gamma}$ 농도는 결핵성 흉수($755,266{\pm}886,636pg/ml$)에서 비결핵성 흉수($3,509{\pm}6,980pg/ml$)보다 높았다(p<0.01). 결핵성 흉막염의 진단기준을 $IFN-{\gamma}$ 농도가 10,000 pg/ml 이상인 경우로 정의하였을 경우 민감도는 97.4%, 특이도는 92.3%였다. 3) PPD 자극에 의한 흉수세포의 $IFN-{\gamma}$ 분비를 PHA로 자극하였을 때의 $IFN-{\gamma}$ 분비에 대한 비로 비교하였을 때 결핵성 흉수($59{\pm}85$)에서 비결핵성 흉수($5{\pm}18$)보다 높았다(p<0.01). 결핵성 흉막염의 진단기준을 PPD/PHA 비가 5이상인 경우로 정의하였을 경우 민감도는 76.9%, 특이도는 92.3%였다. 결 론 : 결핵균 항원 자극에 의한 흉수세포 $IFN-{\gamma}$ 분비검사는 결핵성 흉막염의 진단에 이용될 수 있을 것이다.

새 국립중앙박물관 로튠다에서의 열환경 분석 (Analysis on Thermal Environment in the Rotunda of New National Museum of Korea)

  • Thermal comfort in the Rotunda which is high wide visiting space of the new national museum of Korea has been numerically investigated in this paper. To evaluate thor-mal comfort of the Rotunda, well-known indices, PMV and PPD were introduced. The results of present investigation show that thermal comfort is satisfied at the breathing zone of the visiting space. However a thermal stratification with $9^{\circ}C$ of temperature difference occurs along the height of the Rotunda which makes the thermal environment worse. For example, the PPD value reaches up to 50% in the 6th floor connection passage. Consequently, additional HVAC design factors should be considered in order to reduce the large thermal stratification.

장조장 XLPE 케이블 절연 파괴 원인 규명을 위한 전구차단 장치의 응용 (The Application of Pre-Breakdown Partial Discharge Detection Method for Breakdown of Investigation of Long Distance XLPE Cable)

  • This paper describes AC test for long delivered drum length of cable using pre-breakdown partial discharge detection(PPD) method, which can be applied to observe the causes of electrical breakdown of EHV XLPE insulation. This method is to de-energize applied voltage to cable rapidly using pre-breakdown partial discharge, and is able to prevent complete breakdown. By this effect, defects such as voids, metal particles and protrusion in cable can be observed clearly. AC resonant test set was used for PPD method. Localization of partial discharge was performed by time difference detection of the traveling wave. Accuracy of localization was 1% to the length of test cable. Through several experiments with XLPE cable, successful results of PPD method were obtained.

  • 상당히 성공적인 사람병원체인 결핵균을 Robert Koch가 발견한 이래, 결핵 방제의 상당한 진보에도 불구하고, 결핵의 발생은 현재까지 계속되어오고 있다(Tiruviluamala and Reichman 2002). 그러므로, 완전한 민감성과 특이성을 갖는 신속한 진단방법이 결핵과 결핵의 잠복감염을 진단하는데 요구되어지며(Tiruviluamala and Reichman 2002), 계속적으로 결핵균과 BCG의 유전체와 단백질체 연구를 통하여 결핵 방제수단이 발전되어질 것이다 (Jungblut et al. 1999). 본 연구는 PPD가 결핵을 진단하는 특이한 항원만으로 정확하게 조화를 이루고 있지 않다는 것을 보여주었으며, 그것의 역가는 결론적으로 생물학적으로 평가되어져야 하지만, PPD의 제조는 단백질체 분석 기법에 의해 분석되고 표준화될 수 있음을 보여주었다. 이를 통하여 결핵 피내진단의 일괄된 결과를 위해 PPD 사용 전에 이와같은 단백질체 분석기법을 적용할 수 있을 것으로 사료되며, 본 연구에서 밝혀진 특이 항원은 새로운 피내진단항원으로 적용될 수 있을 것으로 사료된다.

비흡수성 차폐막을 이용한 치조골재생술의 임상적 효과 (A Retrospective Study of the Clinical Outcome of Guided Tissue Regeneration in infrabony defects)

  • The purpose of this study was to evaluate the extent and predictability of periodontal regeneration with barrier membranes in deep infrabony defects. 25 patients(40% smokers) were included in this study. Fourty-one deep infrabony defects treated with membranes(PPD>6mm) were evaluated 1 year postoperatively following a plaque control regimen. Probing pocket depth(PPD), gingival recession(REC), and probing attachment level(PAL) were evaluated at baseline and postoperative 1 year. Plaque score at baseline was 16.2 and plaque score at 1 year was 9.9 A PAL gain of $4.1{\pm}2.5mm$ along with a PPD reduction of $5.0{\pm}2.3mm$ were observed. A PAL gain of $4.1{\pm}2.5mm$ was observed at the smoking group and a PAL gain of $4.0{\pm}2.5mm$ was observed at the non-smoking sroup. It was concluded that periodontal regeneration with membrane represented the predictable and effective treatment modality in the deep infrabony defects.
