Spectrophotometric and Kinetic Determination of Some Sulphur Containing Drugs in Bulk and Drug Formulations

  • Two simple and sensitive spectrophotometric methods were developed for the determination of carbocisteine, penicillamine, ethionamide and thioctic acid in bulk and in their pharmaceutical preparations using alkaline potassium permanganate as an oxidizing agent. The first one involves determination of ethionamide and thioctic acid by spectrophotometric investigation of the oxidation reaction of the two drugs. The second method involves determination of carbocisteine and penicillamine by kinetic studies of the oxidation reaction of these two drugs at room temperature for a fixed time of 20 minutes. The absorbance of the colored manganate ions was measured at 610 nm in both methods. 1-10 ${\mu}$g/mL of ethionamide and thioctic acid could be etermined by the spectrophotometric method with detection limits of 0.11 and 0.089 ${\mu}$g/mL for the two drugs respectively. 2-10 ${\mu}$g/mL of carbocisteine and penicillamine could be determined by the kinetic method with detection limits of 0.14 and 0.21 ${\mu}$g/mL respectively. The two methods were successfully applied for the determination of these drugs in their dosage forms.

Cytotoxicity of Vibrio vulnificus Cytolysin on Rat Neutrophils

  • Cytolysin produced by Vibrio vulnificus has been known to be lethal to mice by increasing vascular permeability and neutrophil sequestration in the lung. In the present study, a cytotoxic mechanism of V. vulnificus cytolysin on the neutrophil was investigated. Cytolysin rapidly bound to neutrophils and induced cell death, as determined by the trypan blue exclusion test. V. vulnificus cytolysin caused the depletion of cellular ATP without the release of ATP or lactate dehydrogenase. Formation of transmembrane pores was evidenced by the rapid efflux of potassium and 2-deoxy-D-[$^3H$]glucose from cytolysin-treated neutrophils. It was further confirmed by the rapid flow of monovalent ions in the patch clamp of cytolysin-treated neutrophil membrane. The pore formation was accompanied by the oligomerization of cytolysin monomers on the neutrophil membrane as demonstrated by immunoblot, which exhibited a 210 kDa band corresponding to a tetramer of the native cytolysin of $M_r$ 51,000. These findings indicate that V. vulnificus cytolysin rapidly binds to the neutrophil membrane and oligomerizes to form small transmembrane pores, which induce the efflux of potassium and the depletion of cellular ATP leading to cell death without cytolysis.

Synthesis of Lithium Titanate Whisker Using Ion-Exchange of Acid Treatment

  • Lithium titanate whiske($Li_{x}Ti_{4}O_9$) was prepared by an ion-exchange reaction. To this end, the initial material, potassium tetratitanate ($K_{2}Ti_{4}O_9{\cdot}nH_{2}O$) was prepared by calcination of a mixture of $K_{2}CO_3\;and\;TiO_2$ with a molar ratio of 2.8 at $1050^{\circ}C$ for 3 h, followed by boiling water treatment of the calcined products for 10 h. Fibrous potassium tetratitanate could be transformed into layered hydrous titanium dioxide ($H_{2}Ti_{4}O_9{\cdot}nH_{2}O$) through an exchange of $K^{+}\;with\;H^{+}$ using 0.075 M HCl. Also, lithium titanate whisker was finally prepared as $Li^{+}\;and\;H^{+}$ ions were exchanged by adding 20 mL of a mixture solution of LiOH and $LiNO_3$ to 1g whisker and stirring for $5\~15$ days. The average length and diameter of the $Li_{x}Ti_{4}O_9$ whiskers were $10\~20{\mu}m\;and\;1\~3{\mu}m$, respectively.

ESTIMATION FOR DEWATERABILITY ON INTERACTION BETWEEN CATIONIC FLOCCULANTS AND IONIC MATERIALS IN DISSOLVING WATER

  • Commonly, the flocculant is dissolved in process or recycle water in industrial plant which has many ionic materials. Therefore, the polymer degradation in aqueous solution by chemical, mechanical or bacteriological may occur, sometimes rapidly. Even if the same flocculant is dissolved, the flocculation characteristics and the properties of dissolving polymer varied with the kind of dissolving water. In this study, we try to estimate the interaction between flocculants and ionic materials in dissolving water using self inversing emulsion polymer; polyacrylamide-co-trimethyl ammonium ethyl acrylate chloride flocculants which have varying molecular weights and structures at a several conditions. The polymeric flocculant is dissolved in artificial dissolving water with Potassium Chloride (PC), Calcium Chloride anhydrous (CC), Potassium Hydroxide (PH), Sodium Chloride (SC), Sodium Bromate (SB) and Iron (II) Sulfate Heptahydrate (IS) as ionic sources. Experimental results indicate that the cationic and anionic ions in dissolving water induce the hydrolysis, degradation of cationic functional group and uncoiling of polymeric flocculants, therefore, the flocculation efficiency decreased by undesired polymer. According that result, it is important to estimate not only its structures and physical properties but also the qualities of dissolving water to optimize the efficiency.

Minimum Pollution of Silicate Oxide in the CMP Process (CMP공정에 의한 실리케이트 산화막의 오염 최소화)

  • We have investigated the CMP slurry properties of silicate oxide thin films surface on CMP cleaning process. The metallic contaminations by CMP slurry were evaluated in four different oxide films, such as plasma enhanced tetra-ethyl-ortho-silicate glass(PE-TEOS), $O_3$ boro-phospho silicate giass( $O_3$-BPSG), PE-BPSG, and phospho-silicate glass(PSG). All films were polished with KOH-based slurry prior to entering the post-CMP cleaner. The Total X-Ray Fluorescence(TXRF) measurements showed that all oxide surfaces are heavily contaminated by potassium and calcium during polishing, which is due to a CMP slurry. The polished $O_3$-BPSG films presented higher potassium and calcium contaminations compared to PE-TEOS because of a mobile ions gettering ability of phosphorus. For PSG oxides, the slurry induced mobile ion contamination increased with an increase of phosphorus contents.

  • The purpose of this study is to find out the effects of Poria extract on rat's nephrotoxicity induced by CsA. The experimental animals were divided into 3 groups and treated as follows; Nothing was given to Sample A, Sample B was given normal saline after IV injection of CsA and Sample C was given Poria extract after CsA injection. After precription of medicine, serum BUN, creatinine, total protein, sodium, potassium, chloride ions were measured. 1. Changes in serum level. ALT, BUN, chloride ion were significantly reduced in experimental group as compaired to control group, and total protein showed significant elevation in experimental group. AST was reduced, but not significant and creatine level was below the normal range. K level showed mild elevation initially and later showed mild decrease, but no significance is noted. There were no significant differance in the sodium level. 2. Changes in Urine level. Urinary specific gravity showed significant increase in experimental group compaired to control group. Urinary creatinine level initially increased, and later decreased but showed no significance. To conclude, it can be inferred that Poria may improve nephrotoxicity and hepatotoxicity in rat induced by Cyclosporin A.

  • In counting chromosome number and karyotype study, it is necessary to let chromosomes on metaphase by pretreatment before fixation. For this purpose, colchicine, or 8-oxyquinoline are generally used. The author found out that chromosomes could be scattered by illuminating cyanoferrate complex solution in which root-tips were sunk. As materials, 8 sorts of plant such as Allium fisturosum, allium tuberosum Rottler, Triticum vulgare were used. Their root-tips were sunk on the bottom of beaker in potassium ferricyanide solution $3{\times}10-4M$ and illuminated through the solution by sterilizing lamp for 1~2 hours in dark room, keeping 10 cm distance from light source to the surface of solution and 2cm depth of solution. Then again, they were illuminated to the light which was somewhat weaker intensity than the former (distance, 16cm; depth, 3cm) for 1.5~2 hours after immersed in 1/100N-HCl and washed in water for each 5minutes. By such methods chromosomes could be scattered. About the mechanism of scattering, it is supposed that CN and Fe(CN)x ions $(x {\leq}5)$ which were gradually produced in the process of photodissociation acted together on the scattering of chromosomes.

A Study on the pH-, pNa- and pK-Sensing Properties of K and Al Coimplanted SiO$_2$ Thin Films (K 및 Al 이중이온주입된 SiO$_2$ 박막의 pH, pNa 및 pK 농도 감지특성에 관한 연구)

  • Silicon dioxide (SiO$_2$) layers were fabricated on Si$_3$N$_4$/SiO$_2$/Si layer structures by low pressure chemical vapor deposition (LPCVD). Potassium and aluminum were then coimplanted by implanting potassium ions with the energy of 100 [keY] and dose of 5x10$^{16}$ [cm ̄$^2$] and 1x10$^{17}$ [cm ̄$^2$] into an aluminum buffer layer on the SiO$_2$Si$_3$N4/SiO$_2$/Si structure. The pH, pNa, and pK ion sensitivities of the resulting layers were investigated and compared to those of as-deposited silicon dioxide layer. The pK-sensitivity of the silicon dioxide was enhanced by the K and Al coimplantation. On the contrary, the pH and pNa-sensitivities of the coimplanted silicon dioxides were quite lower than that of the as-deposited silicon dioxide.

CMP Slurry Induction Properties of Silicate Oxides Deposited on Silicon Wafer (실리콘 웨이퍼위에 증착된 실리케이트 산화막의 CMP 슬러리 오염 특성)

  • We have investigated the slurry induced metallic contaminations of undoped and doped silicate oxides surface on CMP cleaning process. The metallic contaminations by CMP slurry were evaluated in four different oxide films, such as plasma enhanced tetra-ethyl-orthyo-silicate glass(PE-TEOS), O3 boro-phos-pho-silicate glass(O3-BPSG), PE-BPSG, and phospho-silicate glass(PSG). All films were polished with KOH-based slurry prior to entering the post-CMP cleaner. The Total X-Ray fluorescence(TXRF) measurements showed that all oxide surfaces are heavily contaminated by potassium and calcium during polishing which is due to a CMP slurry. The polished O3-BPSG films presented higher potassium and calcium contaminations compared to PE-TEOS because of a mobile ions gettering ability of phosphorus. For PSG oxides, the slurry induced mobile ion contamination increased with an increase of phosphorus contents. In addition, the polishing removal rate of PSG oxides had a linear relationship as a function of phosphorus contents.

Immobilization of potassium copper hexacyanoferrate in doubly crosslinked magnetic polymer bead for highly effective Cs+ removal and facile recovery

  • A potassium copper hexacyanoferrate (KCuHCF) embedded magnetic hydrogel bead (HCF-Mbead) was synthesized via a facile double crosslinking methods of $Fe^{3+}$ ionic binding and freeze-thaw for effective $Cs^+$ removal. The HCF-Mbead had a hierarchical porous structure facilitating fast access of $Cs^+$ ions to embedded active sites. The adsorbent showed enhanced $Cs^+$ removal properties in terms of capacity (69.2 mg/g), selectivity ($K_d=4{\times}10^4mL/g$, 1 ppm $Cs^+$ in seawater) and stability (>99.5% removal in pH 3~11) with rapid magnetic separation. This study further opens the possibility to develop an efficient material that links the integration of adsorption and recovery.
