The Inhibitory Effect of Polyphenol Oxidase on Polyphenol-Induced DNA Damage of Bacillus subtilis (고초균에서 폴리페놀로 유도된 DNA 손상에 대한 폴리페놀산화효소의 억제효과)

  • Antimutagenic activity of the enzymatic browning reaction products (EBRPs) was investigated by using the spore rec-assay with Bacillus subtilis strains H17 $(rec^+)\;and\;M45 (rec^-)$. The EBRPs tested were prepared from the reactions of five different kinds of polyphenols with polyphenol oxidase isolated from the leaves Perilla frutescens. In the spore rec-assay, most of the polyphenolic compounds tested showed positive, whereas only their tested compound showed negative respectively. In addition of polyphenol oxidase inhibitors such as cysteine, glutathione and ascorbic acid to the reaction mixtures consisted with the polyphenol oxidase and polyphenols, the mutagenic effects were increased in the spore recassay. These results show that the activity of polyphenol oxidase may play an important role in the reduction of mutagenicity of polyphenols.

  • Doxorubicin induces oxidative stress leading to cardiotoxicity causing electrocardiogram abnormalities and increases in biomarkers associated with toxicity. Green tea extract (GTE) is reported to possess antioxidant activity mainly via its polyphenolic constituent, catechins. This study was intended to determine the effect of various doses of GTE (25, 50 and 100 mg/kg/day p.o. for 30 days) on doxorubicin-induced electrocardiographic and biochemical changes in rat heart. The latter included lactate dehydrogenase, creatine kinase, and glutamic oxaloacetate transaminase in serum and superoxide dismutase, catalase, and reduced glutathione, as well as membrane bound enzymes like $Na^+K^+ATPase,\;Ca^{2+}ATPase,\;Mg^{2+}ATPase$ and decreased lipid peroxidation in heart tissue Results demonstrated that rats which received GTE were less susceptible to such changes indicating protection afforded by GTE.

Antioxidative Activities of Korean Apple Polyphenols

  • The antioxidative activity and the polyphenolic composition were examined in four different cultivars of apple (Malus domestica), 'Fuji', 'Tsugaru', 'Hongro' and 'Kogetsu', and their parts (peel, core, pulp and juice). The total phenolics, flavonoids and anthocyanins differed among the tested cultivars and parts. Peel parts had the highest total phenolics and anthocyanin content. Contributions of those phenolics to total antioxidative activity were determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assays, and the linoleic acid oxidation assay. Concentration of phenolics contributes significantly to the total antioxidative activity of apples. Clearly, apple peels, especially from Hongros and Kogetsus, possess high levels of phenolic compounds and antioxidants. Therefore, apple peels may potentially function as a value-added ingredient.

  • Resveratrol (trans-3,4',5-trihydroxystilbene), which is a polyphenolic compound found in a variety of plants such as grapes and wine, has been reported to have a variety of anti-inflammatory, anti-platelet, and anti-carcinogenic effects. Recently resveratrol of was reported to serve as an estrogen agonist in MCF-7 cells Based on its structural similarity to diethylstilbestrol, a synthetic estrogen, we examined whether resveratrol and its derivatives might be estrogenic using stable MCF-7-ERE cells. Resveratrol functioned as a superagonist at high concentrations (i.e., produced a greater maximal transcriptional response than estradiol) Among the resveratrol derivatives, 10 compounds showed significant estrogenic activity.

  • Antimutagenicity profiles of the enzymatic browning reaction products(EBRP) were investigated. The rec-assay with Bacillus subtilis strains $H17(rec^+)$ and $M45(rec^-)$ was carried out using their spores. The biological activities were evaluated for seven different enzymatic browning reaction products, which resulted from the reactions of seven polyphenols with polyphenol oxidase isolated from Ginkgo biloba leaves. In the spore $rec^-$ assay, most of the polyphenolic compounds tested were positive, whereas their enzymatic browning reaction products were tested negative. The mutagenicity of enzymic browning mixtures of the polyphenols and the enzymes obtained from Ginkgo biloba leaves showed negative results in the mutagenicity test using Bacillus subtilis strains $H17(rec^+)$ and $M45(rec^-)$. In the case where polyphenol oxidase inhibitors were added in the enzymatic reaction mixtures with polyphenols, the polyphenols showed mutagenic effect in the spore $rec^-$ assay. This suggests that the activity of polyphenol oxidase is decreased.

Studies on the Epiphytic Yeast in Seaweeds (해조류상에 부착한 효모에 관한 연구)

  • The yeast population on 14 species of seaweeds and in water estimated by cultural mothods over a 5-month period in south-west in Korea, Nine species of yeasts, comprising unidentified one, and one of yeast-like fungi were identified. Fifty phycase were attributed to the difference of cultural method between the present work and earlier repoeters and, to some exent, the higher number of Rhodotorula glutinis which had a prior adaptation to the release of inhibitory polyphenolic materials. Although, to what extent, all division of algae showed a similar variation in yeast population, correlated with month, the rapid decrease of yeast population in August seems to be the cause of exposure of heat irradiation in this month. The cultural estimate of per se fltration without double filter and one of unidentified species are discussed.

  • From phytochemical screening of the seven species of Caryophyllaceae, that is, one species of Melandrium, and Dianthus and Pseudostellaria, two species of Stellaria and Cerastium the following compounds were identified. 1) aldehyde, sugars, other reducing compounds and glycosides from the above mentioned seven species of Caryophyllaceae were identified by thin layer chromatography method and comparison of Rf values with the standard substances. 2) Saponins, steroids and terpenoids showed positive reaction with the Froth testing in all of seven plants. Five species of plant showed positive reaction with Liebermann-Buchard test. 3) Flavonoid and polyphenolic compounds were identified by the spots in TLC-test and chlorogenic acid showed the same Rf values with the standard substances. 4) The spots of alkaloid were shown at Rf=0.68 (Stellaria aquatica) and at Rf=0.71 (Pseudostellaria palibiniana) and at Rf=0.70, and 0.44 (Dianthus sinensis).

Butein-Induced Apoptosis in Human T Lymphoma Jurkat Cells (Butein의 Jurkat T 림포마 세포에서 발현되는 세포괴사 효과)

  • Butein is a one of polyphenolic compound widely available in numerous plants. It has broad biological activities including antioxidant and anti-inflammatory activities, which contributed to its protective effects against cancer. Evidences that butein influence proliferation of tumor cells make it important to determine how butein affects cell death of various cancers. In this study, we show that butein, a phenolic compound, induces apoptosis in human T lymphoma jurkat cells. We found that treatment of cells with butein increased apoptosis in a dose- and time- dependent manner as determined by staining cells with Annexin V and 7AAD. There was no significant apoptotic cell death when normal lymphocytes and monocytes from healthy donor were treated with butein. We also found caspase-3 activity was increased during butein-induced apoptosis. The buteininduced apoptotic cell death was blocked by the treatment of cells with caspase-3 inhibitor. These results indicate that butein has the potential to provide an effective strategy against cancer with the advantage of being widely avalible.

  • Silymarin, a polyphenolic flavonoid antiodant, has been shown to have anti-inflammatory, hepatoprotective, and anticarcinogenic effects. In the present study, we report the inhibitory effect of silymarin on nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) mRNA expression in macrophages.
