Cryopreservation of Embryogenic Tissue and Plant Regeneration in Larix leptolepis (낙엽송 (Larix leptolepis) 배발생조직의 초저온보존 및 식물체 재분화)

  • The possibility for long-term preservation of Larix leptolepis embryogenic tissue was tested in this study. Higher relative increase of the tissue fresh weight was observed when embryogenic tissue was pretreated for 24 hrs in a medium containing 0.4 M sorbitol or 20% polyethyleneglycol with cooling rate of -0.33$^{\circ}C$/min. The fast cooling rate of -0.5$^{\circ}C$ and -1.$0^{\circ}C$/min appeared to be less effective in regrowth of tissues from cryopreservation. No DNA variants have been observed by PCR analysis among the embryogenic tissues recovered after 1-, 7-, and 28-day-cryopreservation. The post-thaw embryogenic tissue gave rise to mature somatic embryos which developed into plants.

Effective Concentration Method for Applying PCR to Detect Viruses in Water (수계바이러스검출에 PCR을 이용하기 위한 효과적인 농축기법)

  • In detecting pathogeuic viruses in water sample, polymerase-chain-reaction (PCR) amplification was uscd. In order lo obtain the intact viral particlc, five concentration techniques were compared and an improved procedure was developed with some modifications. Among them, adsorption-elution~EG precipitation and flocculatio~~iultracentriEugation were more efficient than others with thc detection limit of 10 PFU $ml^{-1}$. By the additional step removing inhibitory compounds for PCR reaction, the purity of the concentrated sample was improved and the detection limit was lowered by one order (to 1 PFU $ml^{-1}$. To examine the availability of the optimized procedure for field surveys, the distributions of enterovirus in Han River were estimated using the novel procedure. Seventy-five percentage (618) of sewagc samples and twenty percentage (2110) of river water samples were positive for enterovirus. These results indicate that adsorption-elutionPEG precipitation by PCR method is useful for the prompt and handy monitoring of viral contaminaiton in water environment and pathogenic viruses are widely distributed in water environments of Seoul.

  • Acyclovir(ACV) is an important antiviral drug used extensively against infections caused by herpes viruses, especially herpes simplex and varicella zoster. Because of high crystallinity and large particle size, solubility of intact ACV is very low in water(1.3 mg/ml). The goal of this work is to enhance the solubility of ACV. To make solid dispersion, Polyethyleneglycol, Hydroxyprophylmethylcelluose and Polyvinylpyrrolidone were used as polymer carriers in this work. Polymer carriers and drug were dissolved in acetic acid. And then spray drying method and freeze drying method were used as solvent extraction. Morphology, crystallization and functional group were characterized using SEM, XRD and FT-IR. The result of in vitro test showed the sample using PVP as polymer carrier had higher dissolution rate(up to 466%) than intact ACV.

  • To improve the starch fermentation ability of yeast, hybrids were introduced by protoplast fusion of S. cerevisiae and S. diastaticus. The protoplasts of parental auxotrophic cells were fused in the presence of 10 mM CaCl$_2$and 30% of polyethyleneglycol (M.W 4, 000). The frequencies of fusant formation varied depending upon the strains used and were 3.51$\times$10$^{-4}$ to 5.04$\times$10$^{-4}$ for the regenerated protoplasts. The strains capable of extensive starch hydrolysis produce only 10% to total fusants. The 4 strains were finally selected by the results of starch fermentation and genetic stability test. The DNA content and cell volume of the fusants were greater than those of the parental strains.

  • In this study, we have designed [Epoxy/PEG] polymer gel electrolyte systems by thermal curing the mixtures of epoxy, PEG, imidazole catalyst, and a plasticizer of 1:1 ethylene carbonate and propylene carbonate in the presence of $LiPF_6$ salt. In order to enhance the poor mechanical property of the Corresponding [Epoxy/PEG] gel electrolyte PVdF-HFP was incorporated into the system. The ionic conductivities of the polymer gel electrolytes were related to the amount of PVdF-HFP in blends as well as the amount of liquid electrolyte. The optimized gel system showed room-temperature conductivities of $2.56\times10^{-3}S/cm$.

  • O-carboxymethyl water-soluble chitosan (OCMCh) prepared for enhance the application of chitosan was modified with mthoxy polyethyleneglycol (mPEG) by ion-complex for long circulation in the blood. OCMCh-PEG-PLLs was prepared by forming ion-complex with OCMCh-PEG and Poly(L-Lysine) (PLL) for drug and gene delivery system. The physicochemcal characterisitcs of OCMCh-PEG-PLLs were investigated by FT-IR, $^1H$-NMR. These results showed that CMCh-PEG-PLLs were successfully syntehsized by ion-complex. Particle size distribution and zeta potential of the OCMCh-PEG-PLLs were determined using dynamic light scattering technique. Transmission electron microscopy (TEM) was also used to observe the morphology of the OCMCh-PEG-PLLs. OCMCh-PEG-PLLs have spherical shapes with particle size 290∼390 nm. OCMCh-PEG-PLLs were showed when the feeding amount of mPEG ratio was increased, particle size and zeta potential were decreased. Based on these results, it is possible to introduction of the OCMCh-PEG-PLLs into various biomedical fields such as drug and gene delivery system.

  • PHEA (hydroxyethyl-aspartamide)-mPEG (methoy-polyethyleneglycol)-$C_{16}$ (hexadecylamine)-ED (ethylenediamine) was prepared as a drug delivery carrier. The structure and molecular weight of polymers were characterized by $^1H$-NMR and gel permeation chromatography. Micelle size and shape were measured by electro-photometer light scattering and transmission electron microscope. The mean diameter of micelles was 23 nm in aqueous solution. To evaluate the potential of these polymeric micelles as a drug carrier, PSI-mPEG-$C_{16}$-ED was conjugated with Cy5.5 for Near-Infrared Fluorescent (NIRF) based optical imaging. PSI-mPEG-$C_{16}$-ED-Cy5.5 was injected intravenously into mice (n=5) and in vivo NIRF imaging was performed during 48 h after injection. The biodistribution study at 24 h after injection showed the longcirculation property of PSI-mPEG-$C_{16}$-ED-Cy5.5. Therefore, PSI-mPEG-$C_{16}$-ED micelles could be a promising drug carrier and imaging agent.

Isomeric Folate-Conjugated Polymeric Micelles Bind to Folate Receptors and Display Anticancer Effects

  • The present study aimed to prepare and evaluate polymeric micelles conjugated with folic acid through ${\alpha}$- or ${\gamma}$-carboxyl groups for antitumor efficacy. The isomeric block copolymers, ${\alpha}$- and ${\gamma}$-folate-polyethyleneglycol-distearoyl phosphatidylethanolamine (${\alpha}$- and ${\gamma}$-Fol-PEG-DSPE), were produced by solid phase peptide synthesis. Three types of doxorubicin (DOX)-loaded polymeric micelles (MPEG-DSPE-DOX and ${\alpha}$- / ${\gamma}$-Fol-PEG-DSPEDOX micelles) were prepared via the film formation method. Compared with MPEG-DSPE-DOX micelles, the ${\alpha}$- / ${\gamma}$-Fol-PEG-DSPE-DOX micelles presented a higher cellular uptake behavior in the live cell study. Cell viability percentages were 81.8%, 57.3%, 56.6% at 2 hours for MPEG-DSPE-DOX, ${\alpha}$- and ${\gamma}$-Fol-PEG-DSPE-DOX micelles, respectively (p<0.05). Using the KB xenograft tumor model, both ${\alpha}$- and ${\gamma}$-folate-conjugated micelles were found to have better antitumor effects with lower toxicity in comparison with MPEG-DSPE-DOX micelles. No difference in in vivo antitumor efficacy was found between ${\alpha}$- and ${\gamma}$-Fol-PEG-DSPE-DOX micelles. The folate-conjugated micelles might be a potentially useful strategy for tumor targeting of therapeutic agents, whether grafting with folic acid through ${\alpha}$- or ${\gamma}$-carboxyl groups.

  • Graft copolymerized polycarboxylate(PC)-type superplasticizers which have carboxylic acid with $\pi$ bond among the molecular structure and polyethyleneglycol methyl ether methacrylate(PMEM) were synthesized by free radical reaction. To investigate their chemical structures and molecular weights, PCs were analyzed by FT-IR(fourier transform spectrometer), C-NMR(nuclear magnetic resonance spectrometer) and GPC(gel permeation chromatograpy). When types of carboxylic acids(methacrylic acid, acrylic acid, maleic anhydride, and itaconic acid) and molar ratios of carboxylic acid/PMEM) were varied, adsorptive and fluid characteristics in cement paste were discussed. As the molar ratio of carboxylic acid/PMEM) was higher, amount adsorbed on the cement particles and the fluidity of cement paste by mini-slump spread testing method were increased. When main chain of PC was methacrylic acid, a larger amount was adsorbed on the cement particles. PCs with acrylic acid as main chain showed higher dispersing power. However, it was confirmed that PCs with dicarboxylic acids(maleic anhydride, itaconic acid) didn't have good adsorption and dispersibility.

Maillard Reaction in an Intermediate Moisture Model Food System (중간수분식품 모델계에서의 마이야르 반응에 관한연구)

  • An intermediate moisture model food system was used to investigate the effects of water activity (Aw), temperature, pH and polyethyleneglycol (PEG) on the Maillard reaction. The initial molar ratio of glucose to lysine was varied from one half to four. The maximum Maillard reaction was obtained from an initial glucose/lysine molar ratio of approximately three. The rate of Maillard reaction showed a maximum in the range of water activity of a normal intermediate moisture food. 'the model food system was prepared to hold water activity range of 0.47-0.84 and the samples were held at various temperatures. The maximum browning rate occurred at an Aw value of approximately 0.89 at $40^{\circ}C$ and $60^{\circ}C$, 0.74 at $30^{\circ}C$ and 0.67 at $20^{\circ}C$, respectively. The Arrhenius activiation energies for nonenzymatic browning pigment production were 18.03, 15.18 and 9.90 Kcal/mole for the sample with Aw 0.84, 0.74 and 0.67. When the pH of the model system was increased, a significant increase in the browning reaction was observed. On the inhibitive effects of PEG, the higher degree of polymerization, the more inhibition of browning reaction.
