Production and bioactivity of recombinant tilapia IL-$1\beta$

  • To study the biological activity of interleukin-$1\beta$(IL-$1\beta$), a proinflammatory cytokine, in nile tilapia, Oreochromis niliticus, the recombinant tilapia IL-$1\beta$ was produced in E. coli cells based on pQE vector. Ni-NTA (nitriloacetic acid) metal affinity chromatography was used to purify recombinant protein. The eluted fractions exhibited a single band of protein with a molecular weight of about 25kDa, which is in close agreement with 25.4 kDa predicted by the cDNA sequence. The biological activity of the purified recombinant tilapia IL-$1\beta$ was tested through its effects on IL-$1\beta$ gene expression, which are known as IL-$1\beta$ inducible genes in mammals and fishes. IL-$1\beta$ gene expression induced by poly I:C, a synthetic double stranded RNA, was also assessed in tilapia head kidney cells. IL-$1\beta$ gene expression was analysed using QPCR (quantitative polymerase chain reaction). The ratio of the indicated gene expression was expressed as the relative mRNA level to $\beta$-actin mRNA level, which is constitutively expressed in macrophages. Consequently, head kidney cells incubated for three hours with rIL-$1\beta$(10, 2, 1 $\mu{g}$/ml) showed a dose dependent increase in IL-$1\beta$ mRNA levels and 1 $\mu{g}$/ml of poly I:C was also able to induce IL-$1\beta$ gene expression in head kidney in tilapia.

  • 본 논문에서는 0.25$\mu\textrm{m}$ 1-poly 5-metal CMOS n-well 공정 을 이 용하여 나선형 인덕터와 블루투스 수신기에 응용할 수 있는 전압제어 발진기를 제안하였다. 제안된 인덕터는다층 메탈을 이용하여 인덕터의 저항 성분을 감소시켜 블루투스 주파수 대역에서 Q값을 향상시켰다 또한 Q값이 향상된 나선형 인덕터를 이용하여 부성저항 전압제어 발진기를 설계하였다. 설계된 부성저항 발진기의 시뮬레이션 결과는 외부의 커패시턴스가 2pF에서 14pF:까지 변화할 때 발진 주파수대역은 2.33GHz에서 2.58GHz이고, 발진 출력은 0dBm 이상이었다.

  • Resonant frequency tuning of micro devices is essential to achieve performance uniformity and high sensitivity. Previously reported frequency tuning methods using electrostatic force or mass deposition are not directly applicable to non-conducting polymer devices and have limitations such as dielectric breakdown or low tunable bandwidth. In this paper, thermally frequency-tunable microactuators with poly-dimethylsiloxane membranes are proposed. Permanent and/or nonpermanent frequency tunings are possible using a simple temperature control of the device. Resonant frequency and Q-factor variations of devices according to temperature change were studied using a micro heater and laser Doppler vibrometer. The initial resonant frequencies determined by polymer curing and hardening temperatures are reversibly tuned by thermal cycles. The measured resonant frequency of 9.7 kHz was tuned up by ${\sim}25%$ and Q-factor was increased from 14.5 to 27 as the micro heater voltage increased from 0 to 70 V.

Pyrromethene 597 고체 색소 레이저의 제작 및 출력 특성 (Fabrication and lasing performance of Pyrromethene 597 dye in solid-state host media)

  • PMMA에 pyrromethene 597(PM597) 색소를 주입하여 고체 색소를 제작한 후 그 레이저 출력특성을 분석하였다. 펌핑 광원으로 Q-스위치 Nd : YAG 레이저의 제 2 고조파를 사용하여 발진 중심파장 585 nm에서 최대 광 변환효율 $76\%$를 얻었다. 광안정성을 증가시키기 위해 단일항 에너지 상태의 산소 제거제인 DABCO(1,4-diazabicyclo [2,2,2]octane)를 주입하여 반복율 4 Hz, 출력에너지 250 mJ, 100,000번 이상의 펄스 출력을 얻었다.

  • In this paper, the reliability (NBTI degradation: ${\Delta}V_{th}$) and device characteristic of nano-scale PMOSFET with plasma nitrided oxide (PNO) is characterized in depth by comparing those with thermally nitrided oxide (TNO). PNO case shows the reduction of gate leakage current and interface state density compared to TNO with no change of the $I_{D.sat}\;vs.\;I_{OFF}$ characteristics. Gate oxide capacitance (Cox) of PNO is larger than TNO and it increases as the N concentration increases in PNO. PNO also shows the improvement of NBTI characteristics because the nitrogen peak layer is located near the $Poly/SiO_2$ interface. However, if the nitrogen concentration in PNO oxide increases, threshold voltage degradation $({\Delta}V_{th})$ becomes more degraded by NBT stress due to the enhanced generation of the fixed oxide charges.

  • An obligate methanol-oxidizing bacterium, Methylobacillus sp. strain SK1, which grows only on methanol was isolated from soil. The isolate was nonmotile Gram-negtive rod. It does not have internal membrane system. The colonies were small, whitish-yellow, and smooth. The guanine plus cytosine content of the DNA was 48 mol%. Cellular fatty acids consisted predominantly of large amounts of straight-chain saturated $C_{16:0}$ acid and unsaturated $C_{16:1}$ acid. The major ubiquinone was Q-8, and Q-10 was present as minor component. The cell was obligately aerobic and exhibited catalase, but no oxidase, activity. Poly-.betha.-hydroxybutyrate, endospores, or cysts were not observed. the isolate could grow only on methanol in mineral medium. Growth factors were not required. The isolate was unable to use methane, formaldehyde, formate, methylamine, and several other organic compounds tested as a sole source of carbon and energy. Growth was optimal at 35.deg.C and pH 7.5. It could not grow at 42.deg.C. The doubling time was 1.2h at 30.deg.C when grown with 1.0%(v/v) methanol. The growth was not affected by antibiotics inhibiting cell wall synthesis and carbon monoxide but was completely suppressed by those inhibiting protein synthesis. Methanol was found to be assimilated through the ribulose monophosphate pathway. Cytochromes of b-, c-, and o- types were found. Cell-free extracts contained a phenazine methosulfate-linked methanol dehydrogenase activity, which required ammonium ions as an activator. Cells harvested after the late exponential phase seemed to contain blue protein.ein.

무감독분류 기법에 의한 부분방전 데이터 분석 (Partial Discharge Data Analysis with Unsupervised Classification)

  • This study described partial discharge(PD) distribution analysis between the XLPE(Cross-Linked PolyEthylene)and EPDM(Ethylene Propylene Diene Monomer) interface with unsupervised classification. The ${\phi}-q-n$ patterns were analyzed using phase resolved partial discharge(PRPD). K-means cluster analysis forms a cluster based on similarities and distances among scattered individuals, and analyzes the characteristics of the formed clusters, dividing the multivariate data into several groups according to the similarity of each characteristic, Is a statistical analysis that makes it easier to navigate. It was confirmed that the phase angle of the cluster with the maximum discharge charge was concentrated around $0^{\circ}$ and $180^{\circ}$ at 30 kV after the initial phase distribution localized around $90^{\circ}$ and $300^{\circ}$ expanded to the whole phase angle according to the voltage rise. The Euclidean distance between the center of gravity and the discharge charge in the ${\Phi}-q$ cluster increased with increasing applied voltage.

무기첨가제 규소텅스텐산이 음이온교환 복합막 특성에 미치는 영향 (Effect of Silicotungstic Acid as Inorganic Filler on the Properties of Anion Exchange Composite Membranes)

  • In this study, we synthesized a poly(pheneylene oxide) (PPO)-based organic/inorganic composite membrane having silicotungstic acid (STA) for the development of an anion exchange membrane with excellent ionic conductivity and physicochemical stability. The organic/inorganic composite membranes were prepared by introducing different STA contents (0 wt%, 10 wt%, 30 wt%, and 50 wt%) into the quaternizaed(Q)-PPO matrix. The prepared anion exchange membranes were subjected to structural analysis by proton neclear magnetic resonance and Fourier transform infrared, and thermal behavior of membranes was confirmed by thermogravimetric analysis. Among the prepared composite membranes, the ion conductivity of Q-PPO/STA-50 (40.5 mS cm-1) showed 1.46 times compared to that of the pristine membrane (27.6 mS cm-1). Therefore, these results demonstrated that organic/inorganic composite membranes are promising candidates for application of anion exchange membranes.

Poly-imide 기판에서 제조된 flexible CIGS 태양전지의 Mo strain 개선을 통한 효율 향상 연구

  • Cu(In,Ga)Se2 (CIGS) 박막 태양전지는 높은 효율과 낮은 제조비용, 높은 신뢰성으로 인해 박막 태양전지 중 가장 각광받고 있다. 특히 유리기판 대신 가볍고 유연한 철강소재나 플라스틱 소재를 이용하여 발전분야 외에 건물일체형, 수송용, 휴대용등 다양한 분야에 적용이 가능하다. 이러한 유연 기판을 이용한 CIGS 태양전지의 개발을 위해서는 기판의 특성에 따른 다양한 공정개발이 선행되어야 한다. Poly-imide와 같은 유연기판은 공정온도가 $400^{\circ}C$이하로 낮고 기판이 매우 얇아 기존 Mo 공정을 개선하여야한다. 이러한 유연기판의 특성을 고려하여 본 연구에서는 기존 bi-layer Mo의 bottom layer의 두께를 조절하여 박막의 strain을 조절하였다. 유연기판으로는 SKC KOLON에서 제조된 GL type의 기판을 사용하였다. 기판의 두께는 50um이다. 먼저 Mo의 bottom layer 두께 비율을 기존 12.5%에서 50%로 증가 시켰으며 전체 박막의 두께 역시 900nm에서 500nm로 두께를 감소시키며 실험을 실시하였다. 그 후 흡수층은 Co-Evaporation 방법을 이용하여 제조하였으며 이때 공정온도는 기존 공정온도에서 450, $400^{\circ}C$로 낮추어 흡수층을 제조하였다. 소자 제조 후 초기 Mo의 strain 개선과 저온공정이 적용되지 않은 경우 4.4%에서 공정 최적화 후 13%로 효율이 증가하였다. 제조된 흡수층은 SEM, XRF, XRD등을 이용하여 분석하였으며 그 외 일반적인 방법을 이용하여 Mo, CdS, TCO, Al grid를 제조하였다. AR 코팅은 제외 하였으며 제조된 소자는 솔라 시뮬레이터를 이용하여 효율 특성 분석을 실시하였으며 Q.E. 분석을 실시하였다.

조피볼락(Sebastes schlegelii) Interferon Regulatory Factor 8 (IRF8)의 분자유전학적 특성 및 발현 분석 (Molecular Characterization and Expression Analysis of Interferon Regulatory Factor 8 (IRF8) in the Black Rockfish Sebastes schlegelii)

  • Interferon regulatory factor 8 (IRF8) is essential for the development of B and T cells, as well as for the activity of dendritic cells and macrophages. We performed molecular characterization of IRF8 from rock fish, Sebastes schlegelii (Ss), and investigated the spatial and temporal profile of mRNA expression after challenge with lipopolysaccharide (LPS), polyinosinic:polycytidylic acid (poly I:C), or Streptococcus iniae. The full-length cDNA sequence of SsIRF8 was 1,657 bp, containing an ORF of 1,266 bp. The gene had a predicted molecular mass of 47.7 kDa and an isoelectric point of 5.99. The amino acid sequence coded by this gene showed the highest degree of identity (90.8%) and similarity (96.2%) with IRF8 from Oplegnathus fasciatus. The SsIRF8 mRNA was expressed ubiquitously, at varying levels, with the highest level of expression observed in the spleen. To confirm the role of SsIRF8 in mediating the immune response, we measured SsIRF8 mRNA expression in the splenic tissue at different time points after injection with LPS, poly I:C, or S. iniae. The qRT-PCR results showed that SsIRF8 mRNA expression in the poly I:C-injected group was highly upregulated 6 hr after exposure (P<0.05). Expression of SsIRF8 mRNA in the S. iniae-injected group peaked at 24 hr. These results suggest that SsIRF8 might be important in regulating the strength of the rockfish immune response to immunostimulatory agents.
