The study of quality standard of Pleurotus eryngii (큰느타리버섯의 품질기준에 관한 연구)

  • In order to make the objective quality standard of Pleurotus eryngii, we statistically analysed fruit body collected by the type of market with our own mushroom research lab's quality standard and suggested the quality standard of Pleurotus eryngii by grade. Namely, premium(extra large), first(large), second(middle), and third grade(small) fruit body were 120~130, 110~120 below, 80~110below, and 80mm and below, respectively in height. The diameters of stipe of each grades were 40~45, 30~40below, 25~30below, 25mm below, respectively. The standard of the diameter of pileus indicated ratio of the diameter of pileus/the diameter of stipe instead of the diameter of pileus only because a quality is depend on a ratio of pileus/stipe. The premium and first grade of a ratio of pileus/stipe were 1.6 below and 1.7and below respectively. The weights of premium(extra large), first(large), second(middle), and third grade(small) fruit body were 95~120, 70~95below, 50~70below, 50g below, respectively.

  • We have researched on methods which can reduce fruit-body malformations of king oyster mushroom(Pleurotus eryngii). We collected many pathogens from diseased fruit-body or malformated fruit-body and identified with chemicobiological test and microscope. The factors of fruit-body malformations and increment of contamination during the pin-heading induction time were researched with ventilation amounts in growing room. When the pathogens having high pathogenicity were inoculated in spawn running bottle and at pin-heading induction time, symptom appeared or not appeared in according to air ventilation amounts in growing room. During the pin-heading induction time, humidity degree in growing room have kept of high level and air ventilation amounts were so little that fruit-body malformations ratio was high. But, even though pathogens were inoculated at the surface of bottle factitiously, if air ventilation amounts were enough, fruit-body malformation ratio was low.

Characteristics and breeding of a new cultivar Pleurotus eryngii, Seolsong (큰느타리버섯 신품종 '설송'의 육성 및 그 특성)

  • To develop a new cultivar of king oyster mushroom(Pleurotus eryngii), G09-21 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2824-21 derived from ASI 2824(Keunneutari No. 2) and dikaryotic strain ASI 2887(Aeryni 3). The Pe21-53(G09-21-10 x ASI 2844-9) was shown the best cultural characteristics, selected to be a new cultivar and designated as 'Seolsong'. The 'Seolsong' was distinctly formed incompatibility line in the confrontation growth of parental strains Keunneutari No. 2, Aeryni 3 and ASI 2844. Analysis of the genetic characteristics of the new cultivar 'Seolsong' showed a different DNA profile as that of the control strains, Keunneutari No. 2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25{\sim}30^{\circ}C$ and pH 5~8, respectively. This new cultivar 'Seolsong' of fruiting body production per bottle was about $131.4{{\pm}}43.1$ g which is about 102% quantity compared to that of other cultivar Keunneutari No. 2. And also the stipe is thick and long, but the number of available stipe is few. Particularly, it was tolerant of high moisture above 90% during the growth period after primodia formation. We therefore expect that this new strain will save of labor and cost of cultivation by without culling work.

Optimization of Extraction Conditions for Functional Components of Roasted Pleurotus eryngii by Microwave-Assisted Extraction (볶음 새송이버섯 기능성분의 마이크로웨이브 추출조건 최적화)

  • Response surface methodology was employed to optimize extraction conditions for finding the maximizing the functional properties of roasted Pleurotus eryngii. Based on the central composite design, independent variables were ethanol concentration ($0{\sim}100%$), extraction time ($1{\sim}9$ min) and microwave power ($25{\sim}125$ W). Soluble solid content, electron donating ability and nitrite-scavenging ability were mainly affected by ethanol concentration, but ACE inhibition activity was largely affected by extraction time. The optimum ranges of extraction conditions resulting from superimposing the response surface were predicted to be ethanol concentration ($25{\sim}50%$), extraction time ($3{\sim}9$ min) and microwave power ($80{\sim}125$ W). Total protein and total phenolic compound content of optimal extracts were 45.80 mg/g and 7.42 mg/g, respectively. In phenolic compounds of roasted Pleurotus eryngii extracts, protocatechuic acid was the highest concentration at 1226.32 ${\mu}g/g$, followed by salicylic acid, catechin, p-hydroxybenzoic acid, caffeic acid, coumaric acid and hesperidin.

Effects of Pleurotus eryngii on Lipid Levels and Enzyme Activity in Male Rats Fed High Cholesterol Diets (콜레스테롤 식이를 섭취한 흰쥐에 새송이버섯이 지질대사 및 효소활성에 미치는 영향)

  • Effects of dietary supplementation of Pleurotus eryngii(PE) to a diet containig the cholesterol on lipid levels in the liver and serum, and enzyme activities of rats were studied by feeding 8-weeks old male rats with the four diets for 4 weeks, respectively: normal diet, control diet (10% fat + 0.5% cholesterol), 3% and 5% PE diets (control diet + 3% and 5% PE dry powder). The body weight gains and food efficiency ratios of the 3% and 5% PE diet groups were significantly lower than those of the control group. The liver and epididymal fat pad weights, and hepatic triglyceride Levels of the 5% PE diet group were more significantly decreased than those of the control group. The concentrations in serum triglyceride, total cholesterol, LDL+VLDL-cholesterol, and atherogenic index ratios were significantly lower in the 3% or 5% PE groups compared to those of the control group. Ratio of HDL-cholesterol to total-cholesterol was significantly higher in the 3% and 5% PE groups compared to that of the control group. The fecal total lipid excretion of the 5% PE diet group was more significantly decreased than that of the control group. There was no significant difference in the hepatic cholesterol, and the serum HDL-cholesterol concentrations, and the GOT, GPT and ${\gamma}-GTP$ activities among the experimental groups. These results showed that the Pleurotus eryngii powder feeding decreased the total cholesterol, LDL-cholesterol and atherogenic index, and increased the ratio of HDL-cholesterol to total-cholesterol in serum of the rats.

Establishment of Microwave-Assisted Extraction Conditions for Antioxidative Extracts from Pleurotus eryngii (새송이버섯의 항산화성 추출물 제조를 위한 마이크로웨이브 추출조건 설정)

  • Through the application of a microwave-assisted extraction, the aim of study was to investigate the extraction conditions for the total yield, electron donating ability, total polyphenol contents and nitrite scavenging effect from Pleurotus eryngii. This experiment was conducted under the following extraction conditions: the microwave power (60, 90, 120 W), sample to solvent ratio (1:50, 1:20, 1:10), extraction time (1, 5, 10, 15 mins), and extractant like water, $50\%$ EtOH and $99\%$ EtOH. During the increase of microwave power ranging from 60 to 120 W, the maximized effective components were obtained when the microwave power was set at 60 W. By decreasing the ratio down to 1:10 (g/mL), the extraction of effective components but not total yield was the highest at 1:10 (g/mL). As going up to 15 min, the maximum extraction of effective components was achieved at 1 min for the extraction time and was little affected at over 5 min. In conclusion, the most appropriate conditions for the extraction of effective compounds from Pleurotus eryngii were as follows: the microwave power at 60 W, 1:10 of the sample to solvent ratio, extraction time at 1 min and water as the extractant.

Changes in antioxidant activity of processed edible mushrooms stored at room temperature and low temperature (주요 식용버섯 가공원료의 상온 및 저온 저장에 따른 항산화 활성 변화)

  • This study investigated the changes in the antioxidant activity, nitrite scavenging activity, and ��-glucan content of processed raw materials (Pleurotus eryngii, Pleurotus ostreatus, Lentinula edodes, and Flammulina velutipes) brought about by storage at room temperature (20-25℃) and low temperature (4℃). The results indicated that DPPH free radical scavenging activity was the lowest in air-dried and roasted samples that were stored at room temperature, k with the exception of the air-dried samples of P. eryngii and L. edodes. For total polyphenol contents, all roasted samples of the edible mushrooms stored at room and low temperature decreased compared with the samples pre-storage, except for the air-dried samples of P. eryngii, P. ostreatus, and L. edodes. Furthermore, the ferric reducing antioxidant power and reducing power of the air-dried and roasted samples stored at room temperature and low temperature tended to increase compared to that before storage. Moreover, the ��-glucan content in the air-dried and roasted samples stored at room temperature was significantly lower compared to that before storage, as well as to that in the samples stored at low temperature (p<0.05). The results of this study may help predict the degree to which biological activities in processed edible mushrooms change when stored at room temperature and/or low temperature conditions.

Studies for alternative material of media for Pleurotus eryngii (큰느타리버섯 배지재료의 다변화를 위한 연구)

  • This study was performed to investigate the effective components of media of Pleurotus eryngii for bottle cultivation and several alternative material for reducing cost and smooth supply. The results show that Potato starch, Cocoa shell, Corn gluten feed, Soybean flour(solvent extracted), Cottonseed hull, Poultry manure can be replaced in Bean curd dregs of basal substrates. Especially addition of Soybean flour at 2~8% and Poulty manure at 2% in place of Bean curd dregs is effective for mycelial growth of P. eryngii, Also Pine sawdust appears that can be replaced by Softwood sawdust and addition of softwood sawdust at 25.2% is most effective. Addition of $CaCO_3$, at 2~3%, $K_2CO_3$, at 0.2%, $MgSO_4{\cdot}7H_2O$ in 0.1%, $MnSO_4{\cdot}5H_2O$ in 0.05% is effective for mycelial growth of P. eryngii, Activation materials used in this study have not enough benefit for mycelial growth in the financial aspect.

Effects of medium supplemented with persimmon peels on antioxidant activity of Pleurotus eryngii (감과피 첨가배지가 큰느타리 버섯의 항산화활성에 미치는 영향)

  • In this study, we investigate the potential use of persimmon peels (PP) in mushroom culture medium for the production of functional mushrooms. Pleurotus eryngii was cultivated in medium supplemented with PP (SMPP) at the following concentrations: 0% SMPP (control), 5% SMPP, 10% SMPP, 15% SMPP, 20% SMPP, or 30% SMPP. The total polyphenol content, DPPH radical scavenging ability, ABTS cation scavenging ability, and reducing power of P. eryngii cultivated in SMPP were investigated. P. eryngii cultured in 20% SMPP produced the highest values for all four measurements. Total polyphenol content, DPPH radical scavenging activity, ABTS cation scavenging ability, and reducing power all increased upon the addition of PP. Based on our results, we can conclude that persimmon peels are a highly valuable supplement for functional mushroom culture medium.

Nutritional contents and physiological activity of Pleurotus eryngii by extraction solvents (추출용매에 따른 큰느타리 버섯의 영양성분 및 생리활성)

  • Physiological activities of 70% methanol, fermented ethanol and hot-water extracts of Pleurotus eryngii were investigated. Free radical scavenging activities of P. eryngii extracts were determined according to the elimination of DPPH radicals. Their nitrite scavenging activity and total polyphenol content were also determined. Amino acid analysis showed that phenylalanine (Phe) and glutamic acid (Glu) are most abundant essential and non-essential amino acids in the analyzed extracts. The hot-water extract of ASI 2394 represented the highest antioxidant activity with the DPPH radical scavenging rate value of $40.97{\pm}1.65%$. ASI 2820 displayed the superior capacity to eliminate nitrate regardless of extraction solvents. The hot-water extract of ASI 2887 had the highest content of polyphenol. Our results showed that P. eryngii is well qualified as a functional food.
