The Use of Cassava Chips as an Energy Source for Lactating Dairy Cows Fed with Rice Straw

  • Thirty-six crossbred (70% Holstein Friesian) cows in mid-lactation were assigned to one of four treatments. The dietary treatments were concentrate based, containing 13.5, 27.0, 40.5 and 54.0% of cassava in concentrate replacing ground maize (16.0% CP). There were curvilinear responses to intake of organic matter, non-structural carbohydrate and metabolisable energy. Cassava and corn fed in a ratio of 50:50 maximised organic matter, metabolisable energy intake; milk yield, milk protein and lactose yield. Milk fat yield was not affected by levels of inclusion. Dietary treatment did not influence ruminal pH, ammonia and volatile fatty acid concentrations or plasma glucose. The low market price for cassava resulted in a lower concentrate feed cost. The optimal level of cassava in a dairy cow diet is suggested as being between 20.0 and 30.0% of cassava in dry matter intake when fed with rice straw.

Hematological Parameters and Stress Responses of Olive Flounder Paralichthys olivaceus by Acute pH Change (pH의 급성 변화에 따른 넙치(Paralichthys olivaceus)의 혈액학적 성상 및 스트레스 반응)

  • Olive flounder Paralichthys olivaceus (total weight 216.4±14.6 g, total length 28.4±1.7 cm) were exposed to different pH levels (3, 4, 5, 6, 7, 8, and 9) for 96 h. At pH 4, hemoglobin decreased significantly, while plasma calcium, glucose, cholesterol, and ALP increased significantly. Exposure to pH 4 also induced stress responses, as evidenced by a significant decrease in heat shock protein 70 (HSP 70) and a significant increase in cortisol. The results of this study indicate that acute exposure to acidic or alkaline pH (pH 3 or 9) induced significant mortality, while exposure to pH 4significantly affected hematological parameters and stress responses in P. olivaceus.

  • The purpose of this study was to explore the relationship between life style and metabolic syndrome. The cross-sectional survey was conducted in Pingliang community in Shanghai in Jan 2003. The data was collected by questionnaire, and the results were analyzed by SPSS. It was found that the prevalence of Metabolic Syndrome (MS) was $13.4\%$ in the community, and the body mass index (BMI), waist-to-hip ratio (WHR), systolic blood pressure (SBP), diastolic blood pressure (DBP), serum triglyceride (TG), total cholesterol(TC), low density lipoprotein-cholesterol (LDL-C), and fast plasma glucose (FPG) in MS group were higher than that in non-MS group. Logistic regression analysis indicated that BMI and WHR were positively correlated to the prevalence of MS, and physical activity was negatively correlated to the prevalence of MS. People with higher education levels (${\geq}10y$) had lower BMI, SBP, DBP, LDL-C and FPG. The prevalence of MS in the higher education level group was significantly lower than that of the lower education level group. These results suggested that BMI, WHR and physical activity were important factors of MS, and education background played an important role in the occurrence of MS. Therefore, it is very important to build a healthy life style for preventing and controlling the incidence and developing of MS.

Blood Lipid Level, Nutrient Intake, Self-esteem and Physical Activity of Obese and Normal Weight Elementary School Children (비만 아동과 정상체중 아동의 혈중 지질 농도, 영양섭취, 자아존중감 및 신체활동 비교)

  • The purpose of this study was to analyze characteristics of the obese children in depth and provide basic information for a comprehensive educational program for the obese children. Physical and biochemical characteristics, nutrient intake, and self-esteem, etc. of 16 obese elementary school children (OC) and 14 normal weight elementary school children (NWC) were examined. Self-esteem was measured using Likert-type scales with description. Plasma levels of total cholesterol, LDL-cholesterol, and glucose of OC were higher than those of NWC. Dietary intake with a lunch provided more energy and vitamin B1 in OC than in NWC. Physical appearance and athletic competence among self-esteem in OC were lower than in NWC. However, there was no significant difference in physical activity between OC and NWC. In OC, atherogenic index was positively related with waist circumference, obesity index and BMI while athletic competence self-esteem was negatively related with them. It is recommended that the children should receive the comprehensive educational program for improving children' dietary behavior and their self-esteem.

Novel Detection Technology for Glycated Hemoglobin using Gold Nanoparticles (금 나노입자를 이용한 새로운 당화혈색소의 검출 기술)

  • We report a novel detection technology for glycated hemoglobin (HbA1c) that is measured primarily to identify the three-month average plasma glucose concentration. In enzymatic measuring of glycated hemoglobin, the generated hydrogen peroxide was then used as a reducing agent of gold (III) for the synthesis of gold (0). Gold nanoparticles obtained from this novel approach were measured by optical and piezoelectric methods. In optical method, we have developed polymer based film-type sensor cartridge filled with all the reagents for glycated hemoglobin analysis and the cartridge worked very well having the detection limit of 0.53% of glycated hemoglobin. On the other hand, quartz crystal microbalance (QCM) sensors also have been developed to determine the abilities of surface modified QCM sensors at various levels of the concentration of glycated hemoglobin to bind gold nanoparticles and limit of detection was 0.90%. Finally, despite of relatively lower sensitivities of QCM sensor and film-type optical sensor than well-plate based optical detection, these two sensors were available to measure the glycated hemoglobin level for diabetes patients and normal person.

Lipocalin-2 Secreted by the Liver Regulates Neuronal Cell Function Through AKT-Dependent Signaling in Hepatic Encephalopathy Mouse Model

  • Hepatic encephalopathy (HE) associated with liver failure is accompanied by hyperammonemia, severe inflammation, depression, anxiety, and memory deficits as well as liver injury. Recent studies have focused on the liver-brain-inflammation axis to identify a therapeutic solution for patients with HE. Lipocalin-2 is an inflammation-related glycoprotein that is secreted by various organs and is involved in cellular mechanisms including iron homeostasis, glucose metabolism, cell death, neurite outgrowth, and neurogenesis. In this study, we investigated that the roles of lipocalin-2 both in the brain cortex of mice with HE and in Neuro-2a (N2A) cells. We detected elevated levels of lipocalin-2 both in the plasma and liver in a bile duct ligation mouse model of HE. We confirmed changes in cytokine expression, such as interleukin-1β, cyclooxygenase 2 expression, and iron metabolism related to gene expression through AKT-mediated signaling both in the brain cortex of mice with HE and N2A cells. Our data showed negative effects of hepatic lipocalin-2 on cell survival, iron homeostasis, and neurite outgrowth in N2A cells. Thus, we suggest that regulation of lipocalin-2 in the brain in HE may be a critical therapeutic approach to alleviate neuropathological problems focused on the liver-brain axis.

Hypoglycemic and hypolipidemic effects of Saururus chinensis Baill in streptozotocin-induced diabetic rats

  • Saururus chinensis Baill was reported to inhibit ${\alpha}-glucosidase$ in vitro and flatten postprandial increase in blood glucose in streptozotocin (STZ)-induced diabetic rats. We studied the effect of chronic consumption of S. chinensis Baill on blood glucose and lipid profile in STZ-induced diabetic male rats fed high fat diet. Male rats weighing 100-120 g were fed 30% fat diet with and without 10% freeze-dried leaves of S. chinensis Baill for 7 weeks after 1 week of adaptation. The rats were rendered diabetic by intravenous injection of STZ (60 mg/kg) after 6-week feeding of the assigned diets. At 1 week after the injection, the rats were sacrificed after an overnight fast. Plasma glucose ($380.2{\pm}14.4mg/dL$), total cholesterol ($93.9{\pm}7.9mg/dL$) and triglyceride levels ($123.6{\pm}7.5mg/dL$) of the S. chinensis Baill group were significantly lower than those of the control group ($418.1{\pm}12.0mg/dL,\;119.9{\pm}9.4mg/dL,\;152.0{\pm}10.3mg/dL$, respectively, p<0.05). Chronic consumption of S. chinesis Baill significantly decreased maltase activity of the small intestinal mucosa ($120.1{\pm}8.7U/g$) protein compared with the control group ($96.8{\pm}7.0U/g protein, p<0.05). These results suggest that S. chinensis Baill have hypoglycemic and hypolipidemic effects by inhibiting ${\alpha}-glucosidase$ activity in the animal model of diabetes mellitus.

Kimchi attenuates fatty streak formation in the aorta of low-density lipoprotein receptor knockout mice via inhibition of endoplasmic reticulum stress and apoptosis

  • BACKGROUND/OBJECTIVES: Endoplasmic reticulum (ER) stress is positively associated with atherosclerosis via elevating macrophage cell death and plaque formation, in which oxidative stress plays a pivotal role. Antioxidative, lipid-lowering, and anti-atherogenic effects of kimchi, a Korean fermented vegetable, have been established, wherein capsaicin, ascorbic acid, quercetin, 3-(4'-hydroxyl-3',5'-dimethoxyphenyl)propionic acid, and lactic acids were identified. In this study, mechanisms of action of kimchi methanol extracts (KME) on fatty streak formation via suppression of ER stress and apoptosis in aorta were examined in low-density lipoprotein receptor knockout mice. MATERIALS AND METHODS: Mice fed a high cholesterol diet with an oral administration of KME (KME group, $200 mg{\cdot}kg-bw^{-1}{\cdot}day^{-1}$) or distilled water (control group) for 8 weeks (n = 20 for group). Plasma lipid and oxidative stress levels were evaluated. Protein expression was measured by western blot assay. Fatty streak lesion size and the degree of apoptosis were examined in the aorta. RESULTS: Compared to the control group, in the KME group, plasma lipids levels were decreased and oxidative stress was alleviated (P < 0.05). Protein expression levels of nuclear factor (erythroid-derived 2)-like 2-mediated antioxidants in aorta were increased whereas those for ER stress markers, glucose regulated protein 78, phospho-protein kinase RNA-like ER kinase, phospho-eukaryotic initiation factor 2 subunit ${\alpha}$, X-box binding protein 1, and C/EBP homologous protein were decreased in the KME group (P < 0.05). Moreover, apoptosis was suppressed via downregulation of phospho-c-Jun N-terminal kinase, bcl-2-associated X protein, caspases-9, and -3 with a concomitant upregulation of anti-apoptotic protein, B-cell lymphoma 2 (P < 0.05). Fatty streak lesion size was reduced and the degree of apoptosis was less severe in the KME group (P < 0.05). CONCLUSIONS: In conclusion, antioxidant activity of KME might prevent fatty streak formation through, in part, inhibition of ER stress and apoptosis in aortic sinus where macrophages are harbored.

Effect of Diet Containing Whole Wheat Bread with Capsosiphon fulvescens and Lindera obtusiloba Ethanol Extracts on Plasma Glucose and Lipid Levels in Rats (매생이와 생강나무잎 에탄올 추출물이 첨가된 통밀빵을 섭취한 랫트의 혈당과 지질수준에 미치는 영향)

  • The present study was conducted to investigate the effect of whole wheat bread with added Lindera obtusiloba (LO) and Capsosiphon fulvescens (CF) ethanol extracts on serum glucose and lipid levels in Sprague Dawley rats. Rats were divided into five groups depending on the diet administered: normal bread (NC), whole wheat bread (W), whole wheat bread with LO leaves extract (WL), whole wheat bread with CF extract (WC), and whole wheat bread with freeze-drying CF (WDC). After 4 weeks of consuming the experimental diet, the blood glucose level and hemoglobin A1c contents were found to be significantly lower in the W, WL, WC, and WDC groups than in the NC group. The high-density lipoprotein-cholesterol levels increased in the WL group when compared to those in the NC group and triglycerides levels decreased in all wheat groups compared to those in the NC group. These results suggest that wheat breads containing LO and CF extracts are effective for preventing hypercholesterolemia and obesity.

Effect of submerged culture of Ceriporia lacerata mycelium on GLUT4 protein in db/db mouse (db/db 마우스에서 Ceriporia lacerata 균사체 배양액이 GLUT4 발현에 미치는 영향)

  • In this study, we evaluated the antidiabetic effect of a submerged culture of Ceriporia lacerata mycelium (CL01) on hematological indices, as well as protein and mRNA expression of the insulin-signaling pathway, in db/db mice. After CL01 was administrated for 4 weeks, blood glucose levels decreased consistently, and plasma insulin and c-peptide levels each decreased by roughly 55.8%, 40% of those in the negative control (p<0.05). With regard to HOMA-IR, an insulin resistance index, insulin resistance of the CL01-fed group improved over that of the negative control group by about 62% (p<0.05). In addition, we demonstrated that the protein expression levels of pIR, pAkt, pAMPK, and GLUT4 and the mRNA expression levels of Akt2, IRS1, and GLUT4 in the muscle cells of db/db mice increased in the CL01-fed group compared to the corresponding levels in the control group. These results demonstrate that CL01 affects glucose metabolism, upregulates protein and gene expression in the insulin-signaling pathway, and decreases blood glucose levels effectively by improving insulin sensitivity. More than 90% of those who suffer from type 2 diabetes are more likely to suffer from hyperinsulinemia, hypertension, obesity, and other comorbidities because of insulin resistance. Therefore, it is possible that CL01 intake could be used as a fundamental treatment for type 2 diabetes by lowering insulin resistance, and these results may prove be useful as basic evidence for further research into the mechanisms of a cure for type 2 diabetes.
