An Investigation of the Control of Two-Spotted Spider Mites (Tetranychus urticae Koch) Resistant to Organo-Phosphates (유기인제저항성 점박이응애(Tetranychus urticae Koch)에 관한 연구)

  • The study involved determination of resistance levels of spider mites ta argano-phosphates using topical application and slide dip techniques; laboratory serening tests of alternative acaricides using an O/P resistant strain and a field trial of the screened materials. 1. Strains of Tetranychus were from Timaru(TR), Havelock Narth (HNR), Lincaln (LN). Germany (GR, GN). Comparisons of the resistant strains and normal strains at the LD50 and LC50 levels were as follows : (a) Using the topical application tochnique; with Parathian. resistant levels of the GR. TR and HNR strains of T. urticae were respeativuly, 1035. 484 and 452 times as resistant' as the LN strain. (b) Using the slide dip technique; with Phosdrin, resistant of GR, TR and HNR strains of T. urticae were 635, 274 and 266 times greater respeativuly, than the GN strain. 2. The laboratory sereaning tests were carried out far their contact plus stomach and residual effect to assess the toxicities of eleven alternative materials which would be used far control of O/P resistant strain of T. urticae. The acaricide groups represented were 3 organo-chlorines (Spidex, Kelthane and C 8514), 2 nitrophenyls (UC 19786 and Morocide), 2 cyclic carbonates(Eradex and Morestan). I carbamate (UC2004 7A), 1 mixture of carbamate and orano-chlorine and 2 other chemicals (C 8677 and M2527). From all acaricide tested. Kelthane and Morocide were the most effective, folowed by Spidex and M2527. Morestan, C8514. C8677 and RS 143 were intermediate, but Eradex, UC 19786 and UC 20046A were poor. 3, The number of sapmles required for estimation of the population in the field evaluation of acaricidal effects was one giving the highest practical precision. It was decided, after preliminary sampling trials. to use samples of 30 leaves per replicate which gave a $5.7\%$ standard error. 4. In the field trials, Morocide applied at the $0.05\%\;and\; 0.04\%$ a. i. conc. to black currant trees gave excellent control of O/P resistant population of T. urticae for about 12 days, but Morocide 0.025 and Kel thane $0.02\%$ a. i. cone. gave efficient control for about 6 days. In other words. first applications of Kel thane ane Moroeide gave very high degrees of control of O/P resistant population of the two-spotted spider mite. However, the results indicate that secondary application would sometimes be necessary. There was no foliage damage of black Currants and strawberries by either acaricides at the concentrations used. Identification and Physiological Characters of Intestinal Bacteria of the Black Soldier Fly, Hermetia illucens (아메리카동애등에 장내세균 동정과 생리적 특징)

  • The black soldier fly, Hermetia illucens, larvae may depend on indigenous bacteria in the intestine to feed and digest diverse food sources. To prove this hypothesis, we isolated and identified the intestinal bacteria of the black soldier fly for their digestive and antimicrobial abilities. The last instar larvae had long digestive tracts, which were about seven times longer than its body length. An individual of H. illucens larvae possessed a total of $5.0{\pm}10^6$ bacteria in the whole intestine, of which more than 98% bacteria were located in the hindgut. Three different bacterial isolates cultured on nutrient agar (NA) medium were detected in the intestine and identified as Morganella morganii, Providencia rettgeri and Bacillus halodurans by Biolog microbial identification system. Analysis of 16S rDNA sequences of the intestinal bacteria detected the additional bacteria of Proteus mirabilis, Providencia alcalifaciens, and Providencia sp. These intestinal bacteria cultured on NA medium exhibited high resistance to 4 antibiotics and inhibited growth of other microbes which are mainly plant pathogens. Also, these bacteria exhibited catalytic activities to degrade cellulose, lipid, proteins, and carbohydrates. These results suggest that H. illucens larvae possess intestinal bacteria that may play crucial roles in their digestive physiology.

Experimental Study on Engineering Performance Evaluation and Field Performance of Environmentally Friendly Functional Concrete (친환경 기능성 콘크리트의 공학적 성능평가 및 현장적용성능에 관한 실험적 연구)

  • In this study, the physical, mechanical, structural, and environmental performances based on field measured data were evaluated to check the suitability of concrete for ecological preservation and cultivation of a hydrophilic environment. More specifically, the study is focused on developing an environmentally friendly functional concrete with river ecology restoration and natural river early formation capabilities. The mechanical performance evaluation results showed that the increase in mix rate of the PVA (Poly Vinyl Alcohol) reinforcement fibers and silica fume caused an increase in the strength. The optimal mix rate was found to be 0.05 volume % PVA fiber and approximately 10% silica fume. The frost resistance evaluation showed that superior performance was gained when 0.05 volume % PVA fiber and 15% silica fume was mixed simultaneously. In the structural performance evaluation, the bending strength was improved by 47.7% compared to plain concrete when mixed with 0.05 volume % PVA fiber. The flexural toughness also saw significant improvement. The environmental monitoring of field performance showed that grasses germinated most rapidly, but the growth of red poppies, a plant that germinates in the spring, was most active with passing of time. Coverage measurements in all of the monitoring locations found favorable coverage of over 95% after 12 weeks. The study results showed that the environmentally friendly functional concrete had outstanding environmental performance.

The Durability of the Concrete Using Bottom Ash as Fine Aggregate (바텀애시를 잔골재로 사용한 콘크리트의 내구성능에 관한 연구)

  • This study is about the reuse of bottom ash, which is released as a necessity in thermal power plant. In general, coal-ash are classified as fly-ash, bottom-ash, cinder-ash. Of these, a large amount of fly ash is being recycled as cement substitutes. While, recycling rates of bottom ash are the lowest due to its porosity and high absorption. In this study, the durability of the concrete using bottom ash as a concrete fine aggregate was evaluated. The using level of the bottom ash ranges to step-by-step from 0% to 30%. According to the result of the durability test, regardless of the presence of the bottom ash, freeze-thaw durability could be secured by air entrainment. In case of the resistance to chloride ions penetration, the length change, and the effects on heavy metals, the replacement of bottom ash as fine aggregate was not critical. Although carbonation penetration was higher as the replacement level of bottom ash increased, the experiment showed that it could be possible to use bottom ash as concrete fine aggregate with proper mix design.

Preliminary study on absorption characteristic of a human body according to the amount of clothing worn for developing standard test dummy (표준더미 개발을 위한 착의량에 따른 인체의 흡음특성 기초연구)

  • The purpose of this study is to evaluate the sound absorption characteristics of a human body according to the amount of clothing worn by using reverberation method measurement method for developing standard test dummy, which can be used for testing absorption of occupied audience chair. Test method was based on the previous study (Conti et al., 2004), each experiment is conducted in the reverberation room and a subject wearing clothes is standing in that chamber. In this experiment, the sound absorption area of each frequency band according to the wearing of various material clothing was measured. As a result of measurements, the average sound absorption area of the whole frequency band was $0.25m^2-0.48m^2$ in case of a subject not wearing outer clothes, and $0.38m^2-0.98m^2$ in case of wearing of outerwear. Polyester tops by showing the maximum value, the highest characteristics in the 800 Hz to 1 kHz band among the rest of fiber materials. The outer jacket made of the wool and cotton materials show a higher absorption area as the frequency increases to the higher frequency band. The change of the sound absorption area according to the clothing amount was divided by the thermal resistance (clo) of the worn clothes and the weight per body surface area.

Essential Oil Isolated from Iranian Yarrow as a Bio-rational Agent to the Management of Saw-toothed Grain Beetle, Oryzaephilus surinamensis (L.) (머리대장가는납작벌레의 합리적 방제 물질로 이란 서양가새풀 정유의 살충효과 평가)

  • Overuse of synthetic pesticides caused negative side-effects such as environmental contamination, development of insect pests' resistance, and effects on non-target organisms. Plant origin substances without/or with low mammalian toxicity have been considered as promising alternatives to the synthetic pesticides. Fumigant toxicity of the essential oil of Iranian Yarrow, Achillea millefolium L., was investigated against a cosmopolitan stored-product insect pest: saw-toothed grain beetle (Oryzaephilus surinamensis L.). Chemical profile of this essential oil was studied by Gas Chromatography-Mass Spectrometry. Tested concentrations were significantly effective to the mortality of insect pest. A positive correlation between essential oil concentrations and pest mortality were realized. LC50 value (lethal concentration needed to 50% mortality) was achieved as $17.977(16.195{\pm}20.433){\mu}l/l$ air. The main components were 1,8-Cineole (13.17%), nerolidol (12.87%), ${\alpha}$-cubebene (12.35%), artemisia ketone (6.69%), ${\alpha}$-terpineol (5.27%), alloaromadendrene oxide (4.71%) and borneol (3.99%). Terpenic compounds including monoterpene hydrocarbons (8.19%), monoterpenoids (44.23%), sesquiterpene hydrocarbons (21.69%) and sesquiterpenoids (22.24%) were 96.35% of the total identified compounds. Results indicated that the terpene-rich A. millefolium essential oil may be considered as a safe bio-agent in the O. surinamensis management.

Effect of Uniconazole and Silver Thiosulfate Treatment on Reduction of Ozone Injury in Snap Bean Plants (Uniconazole과 Silver Thiosulfate 처리(處理)가 강남콩의 오존피해(被害) 경감(輕減)에 미치는 효과(效果))

  • Studies were conducted to examine the effects of single or combined treatment of uniconazole [(E)-1-(4-chlorophenyl)-4, 4-dimethyl 2(1, 2,-4-triazol-1-yl)-1-penten-3-ol)] and silver thiosulfate (STS) on reducing ozone injury to snap beans (Phaseolus vulgaris L. 'Strike'). Two weeks after seeding, plants were given a soil drench of uniconazole(XE-1019) solution at concentrations of 0.001, 0.005 and 0.025 mg/pot, and then two days prior to ozone fumigation, 0.3 and 0.6 mM STS containing 0.01% Tween-20 were also sprayed. Uniconazole was effective in providing protection against ozone injury through increase activities of free radical scavengers such as superoxide dismutase (SOD) and peroxidase (POD) as well as the increase of chlorophyll content and stomatal resistance resulted from plant growth retardation. The phytoprotective effects of STS seemed to be related to its properly of blocking the ethylene action and increasing activities of SOD and POD. Even at low concentrations, a combined treatment with uniconazole drench, STS spray significantly reduced ozone injury compared to single application.

Isolation and Characterization of Insoluble Phosphate-Solubilizing Bacteria with Antifungal Activity (항진균능을 가진 불용성 인산 가용화 세균의 분리 및 특성)

  • To develop multifunctional microbial inoculant, an insluble phosphate-solubilizing bacterium with antifungal activity was isolated from plant rhizospheric soil. On the basis of its morphological, cultural and physiological characteristics and Biolog analysis, this bacterium was identified as Pseudomonas fluorescens RAF15. P. fluorescens RAF15 showed antifungal activities against phytopathogenic fungi Botrytis cinerea and Rhizoctonia solani. The optimal medium composition and cultural conditions for the solubilization of insoluble phosphate by P. fluorescens RAF15 were 1.5% of glucose, 0.005% of urea, 0.3% $MgCl_2{\cdot}6H_2\;0.01%\;of\;MgSO_4{\cdot}7H_2O\;0.01%,\;of\;CaCl_2{\cdot}2H_2O$, and 0.05% of NaCl along with initial pH 7.0 at $30^{\circ}C$. The soluble phosphate production under optimum condition was 863 mg/L after 5 days of cultivation. The solubilization of insoluble phosphates was associated with a drop in the pH of the culture medium. P. fluorescens RAF15 showed resistance against different environmental stresses like $10-35^{\circ}C$ temperature, 1-4% salt concentration and pH 2-11 range. The strain produced soluble phosphate to the culture broth with the concentrations of 971-1121 mg/L against $CaHPO_4$, 791-908 mg/L against $Ca_3(PO_4){_2}$, and 844 mg/L against hydroxyapatite, respectively. However, the strain produced soluble phosphate to the culture broth with the concentrations of 15 mg/L against $FePO_4$, and 5 mg/L against $AlPO_4$, respectively.

  • Formaldehyde responsive protein(FRP) 1 belongs to the family of universal stress protein(USP) and is known to respond to stress caused by fumigation of gaseous volatile organic compounds(VOCs) such as formaldehyde and toluene. However, the molecular function of this protein is not well understood at cellular and molecular level. In this study, loss of function mutant of FRP1 generated by T-DNA insertion(frp1-4) has been isolated from Arabidopsis thaliana and the function of FRP1 was characterized. The loss-of-function mutant of FRP1 appeared slight growth defects with shorter stem and rosette leaves compared to wild type. In addition, the damage caused by exogenous VOCs was more severe in frp1-4 than in control. Therefore, Arabidopsis FRP1 seems to be the protein involved not only in the growth and development of plant but also the stress resistance against toxic volatile organic compounds.

The Influence of Temperature and Water Stress on the Varietal Reactions of Rice to the Inoculum of the Blast Infected Leaves (BIL) (도열병 이병엽접종원에 대한 수도 품종의 반응에 미치는 온도 및 Water Stress의 영향)

  • Using the milled, blast infected leaves (BIL) as an inoculum source on the screening for the resistance to blast of rice plant was a simple and useful technique. The temperature with high (25^\circ C\sim 35^\circ C) and low (15^\circ C\sim 28^\circ C) and the water stressed or not, was conditioned of to the inoculation with the BIL to the test varieties in seedling stage. In low temperature, most of the varieties were more infected with blast, however the Indica-Japonica hybrids were more infected in high temperature conditions. The water stressed was more infected with blast than the not stressed. The interaction of variety with water stress was not so much as that of variety with temperature. Resistant reaction to blast (BIL) was not affected by the temperature and water stress, but the moderately resistant or susceptible one was much affected by them. Inoculum of BIL was virulent to the newly bred Indica-Japonica hybrid cultivars such as Tongil, Nopung, etc, but not virulent to the Japonica cultivars such as Nongbaek, Jinheung, etc. The discrete, mixed or variable lesions were observed mainly in the moderately resistant or susceptible cultivars such as Kanto 51, Yashiromochi, Ishikari-shiroke, etc.
