Effect of Growth Retardants on Endogenous ABA-like Substance Content and GA-like Substance Activity of Sedirea japonica Seedlings Cultured In Vitro (생장억제제 처리가 기내 배양한 나도풍란 (Sedirea japonica) 유묘의 ABA 유사물질 함량 및 GA 유사물질 활성에 미치는 영향)

  • This experiment was conducted to identify the effect of several plant growth retardants on endogenous ABA-like substance content and GA-like substance activity in seedlings of Sedirea japonica cultured in vitro. When seedlings of Sedirea japonica were treated with low concentration of 0.05 mg/L Uniconazole, 0.1 mg/L Ancymidol and 0.3 mg/L Paclobutrazol, the content of ABA-like substances of the leaf was lower than that of the control. However, the activity of GA-like substances was similar or higher in treated seedlings. In the mid and high concentrations of three kinds of growth retardants, the ABA-like substance content was increased, but GA-like substance activity was inhibited. The content of ABA-like substances in the root was lower in 0.05 and 0.2 mg/L Uniconazole, 0.2 mg/L Ancymidol and 0.1 mg/L Paclobutrazol treatments than that of the control, but in the mid and high concentration treatments, the content was increased. GA-like substance activity in low concentration was increased but in the mid and high concentration, the activity was inhibited compared with low concentration treatment.

Identification of FM001 as Plant Growth-Promoting Substance from Acremonium strictum MJN1 Culture

  • A plant growth-promoting substance, FM001, was isolated from the culture broth of Acremonium strictum MJN1. The purification steps included solvent extraction, adsorption chromatography using Diaion HP20, TLC on silica, and HLPC using a C-18 column. The purified FM001 enhanced rice seedling growth by $11.1\%\;and\;34.0\%$ of the dried weight of the shoots and roots, and also radish growth by $26.5\%\;and\;23.7\%$ of the top length and dried weight. FM001 also significantly promoted the growth of red pepper by increasing $32.7\%$ of fruit weight and $11.3\%$ as regards the height. FM001 consisted of C, H, O, N, and S, and its molecular weight was determined to be 537.78 Da. The structure of FM001 resembled brassinosteriods, and it would appear to have great potential as an effective bio-fertilizer.

Purification and Antifungal Activities of an Antibiotic Produced by Gliocladium virens G1 Against Plant Pathogens

  • This study was undertaken to separate and identify antifungla substances produced by Gilocladium virens G1, a biocontrol agent used for the control of plant diseases caused by Rhizoctonea solani. The culture of G. virens G1 effectively inhibited the growth of R. solani, Colletotrichum gloeosporioides, and Phytophthora capsici, but less that of Fusarium oxysporum. The n-hexane extract of the G. virens culture, which was used for the purification of responsible substances, strongly inhibited R. solani and C. gloeosporioides, but not P. capsici, although the n-butanol extract was effective on all of the pathogens tested. An antifungal substance was purified using the n-hexane extract by Silica gel column chromatography and HPLC. The substance was examined for purity by HPLC and for nature by UV spectrometry, which differed from known antibiotic compounds such as gliotoxin, viridin and gliovirin. The antifungal substance was very liphophilic based on its solvent-solubility and Rf values on TLC, and more inhibitory to C. gloeosporioides than other fungal pathogens tested.

Isolation and Characterization of Bacillus sp. BT182-3 for Biocontrol Against a Plant Pathogenic Pseudomonas syringae (식물병원균 Pseudomonas syringae에 대한 생물방제균 Bacillus sp. BT182-3의 분리 및 특성)

  • For a microbial control of a plant pathogenic Pseudomonas syringae, Bacillus sp. strain BT182-3 was isolated. The strain BT182-3 had a growth inhibition against P. syringae not only on agar plate but also on cultured broth. After heat treatment at $40^{\cird}C$ and $80^{\cird}C$ for 30min, the lytic substance from the strain BT182-3 had about 52% remaining activity and 17% remaining activity, respectively. The optimal pH and temperature of the lytic substance was 6.0 and $28^{\cird}C$, respectively. Germination ratio of healthy radish seeds was 87% at $25^{\cird}C$ for 5 days in 0.8% saline, and that of the radish seeds infected with P. syringae was 67%, while that of the radish seeds treated with cultured broth of the strain BT182-3 was 90%. The 5-days healthy radish seedlings were 3.90cm at high and the seedlings infected with P. syringae were 3.06cm at high, while the seedlings treated with cultured broth of the strain BT182-3 were 4.30cm at high. The growth of the radish seedlings infected with P. syringae was inhibited after cultivation for 40days on pots, while the growth of the infected radish seedlings with P. syringae was recovered at stem length, root length and total weight at the same as the healthy seedlings after treatment of a lytic substance from the strain BT182-3.

  • Bacillus lichemiformis Kll, a plant growth promoting rhizobacterium was reported as a producer of auxin, siderophore, as well as antifungal cellulase under some culture conditions. In vitro test, B. licheniformis Kll represented excellent antagonistic ability against Fusarium oxyspoum (KACC 40037), and showed broad spectrum against other phytopathogenic fungi. B. licheniformis Kll had cellulolytic activity toward not only carboxymethyl-cellulose (CMC) but also insoluble cellulose, such as fungal cell wall cellulose, filter paper (Whatman No. 1), and Avicel. In addition, we confirmed antifungal substance production by butanol-extract methods. The strain produced optimally the antifungal substance when it was cultivated at pH 9.0, 30${\circ}$C for 4 days on nutrient medium. The biological control mechanisms of B. lichemiformis Kll were caused by antifungal substance, cellulase and siderophore against phytopathogenic fungi.

Effect of Growth and Culture Conditions of Paecilomyces japonica and Cordyceps militaris on the Formation of Bioactive Substance

  • To investigated the effect of liquid culture conditions and nutrient sources on the formation with bioactive substance of Paeilomyces japonica and Cordyceps militaris cultivated in the country, the result are as follow; The growth temperature of two mycelia is 25$^{\circ}C$ and the proper temperature for cordycepin growth is around 20$^{\circ}C$. The formation amount of bioactive substance by nutrient sources reached its peak with using 2% glucose and 1% galactose in case of carbon sources and 0.4% inorganic compound in case of nitrogen sources. Also, the ratio of C/N was optimal with 3% glucose 1% peptone. For a natural medium, most grains were sufficient but the soybean oil was superb. The formation amount of protein-binding polysaccharide that are used for anticancer substance was in proportion to the growth rate of mycelium, had lots of aeration and showed a trend of increasing when the acidity lower. and the content of structural protein showed a trend of increasing when the acidity lower. However, the content of the structural hexosamin did not get a great the effect of culture conditions and nutrient sources. The constitution of monosaccharide that organizes a protein-binding polysaccharide greatly changed in proportion to carbon sources. When Paecilomyces japonica cultured in a silkworm larvae for 30 days, the content of cordycepin was 204.5 mg/100mL as a dry weight in the fruiting body, 41.8 mg/100mL in mycelium and larva, and the content for each bottle was average 29.5 mg/100mL. In case of Cordyceps militaris for 45days, the fruiting body was 563.5 mg/100mL, the larva and the mycelium was 86.1 mg/100mL, and the content for each bottle was average 65.0 mg/100mL.

Enhancement of Biocontrol Activity of Serratia plymuthica A21 -4 Toward Phytophthora Blight of Pepper by Amendment of Nutritional Condition

  • Serratia plymuthim A21-4 strongly inhibits the mycelial growth, zoospore formation, and cystospore germination of Phytophthor spp and Pythium species. The bacterial isolate produced antifungal substance and chitinase. The bacteria also enhanced to plant growth remarkably in low nutritional condition. The application of cell suspension of A21-4 to pepper seedlings in greenhouse experiments and soil drenching in farmer's field was proved successfully to control the phythophthora blight of pepper. For the effective control, however, relatively high density of cell number(10$\^$9/cfu/$m\ell$) is required. Density effect was similar in plant growth promoting activity of A21-4. Though this investigation we improved the problem with changes of culture condition of bacteria and some nutritional amendment.

The effect of growth substance and stem hardness on rooting of mature aloe plant (생장조절제 및 경경도가 성숙Aloe 발근에 미치는 영향)

  • Experiments were conducted to study the effect of IBA, mineral stone and stem hardness on rooting characteristics in mature Aloe vera plant. The rooting ability in matured Aloe vera was of highly significance when the amount of IBA treatment was increased to 5.000mg/l. The treatment of IBA 5.000mg/l, together with mineral stone(v/w 1.5 times), gave more root number, the length and the dry weight of root were, also, higher in semihard wood cutting than soft wood cutting in matured Aloe vera.
