Isolation and Culture of Phaseolus vulgaris L. Callus Protoplasts (강남콩(Phaseolus vulgaris L.) Callus의 원형질체 유이 및 배양)

  • The isolatin and culture of protoplasts from hypocotyl originated callus of Phaseolus vulgaris cv. Damyang were carried out. The maximum protoplast yield of 4.6$\times$105 per gram fresh callus, using the 13-day-old callus, was obtained by digeston for 6 hours in the enzyme solution. After 10 day-culture of the isolated callus protoplsts, plating efficiency was 50%. Thereafter, cell cluster medium, and followed by leading to callus formation on an agar medium after 3 weeks of the liquid culture.

Anticonvulsant potential of callus cultures of Convolvulus microphyllus Sieb.

  • Callus cultures of Convolvulus microphyllus Sieb. was induced on Murashige and Skoog's medium supplemented with 2,4-dichloro phenoxy acetic acid, 6-benzyl adenine, indole acetic acid and kinetin (1 ppm each). Methanolic extracts of whole plant, leaf, stem and leaf and stem calli were tested for anticonvulsant activity against standard drug phenytoin using maximal electroshock model on mice. It was observed that the animals treated with methanolic extracts of stem callus, leaf callus and whole plant (200 mg/kg, oral) showed significant protection against tonic convulsions induced by transcorneal electroshock. Anticonvulsant activity of methanolic extract of stem callus was comparable to that of standard drug phenytoin.

Plant Regeneration from Sliced Mature Embryo Fragments of Wheat Cultivars

  • Mature embryos were aseptically excised with a scalpel and sliced in fragments measuring 0.5 mm in diameter (sliced mature embryo fragment; 4 ${\~}$ 5 fragments/one embryo). Sliced mature embryo fragments of six wheat cultivars were cultured to develop an efficient method of callus induction and plant regeneration. Callus derived from sliced mature embryo fragments showed a good capacity to embryogenesis and regeneration. Furthermore sliced mature embryo fragments decreased contamination from fungi and bacteria. The high efficiency of callus induction were obtained Keumkangmil and Bob­white. For plant regeneration, selected embryogenic calli were transferred to two types regeneration media. An average number of green spots per callus was 4 to 5 in regeneration media after about one week. Percentage of plant regeneration showed high in regeneration medium containing 0.1 mg/l 2,4-D and 5 mg/l zeatin. Especially, Keumkangmil ($27.5\%$) and Bobwhite ($33.3\%$) showed high regeneration efficiency. This regeneration system from sliced mature embryo fragments may provide an effective and convenient explant for plant transformation studies.

Establishment of a novel plant regeneration system from suspension-derived callus in the halophytic Leymus chinensis (Trin.)

  • The establishment of cell suspension culture and plant regeneration of the halophytic Leymus chinensis (Trin.) are described in this study for the first time. Callus induction solid medium containing Murashige and Shoog (MS) basic salt, $2.0\;mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid (2,4-D), and $5.0\;mg\;l^{-1}$ L-glutamic acid with $30.0\;g\;l^{-1}$ sucrose and $4.0\;g\;l^{-1}$ gelrite for solidification induced the highest rate of cell division in Type 1 callus among calli of various types. Liquid medium with the same hormone distribution was therefore, used for cell suspension culture from Type 1 callus. Over a 30 d suspension culture at 100 rpm, great amounts of biomass were accumulated, with 71.07% average daily increment and 22.32-fold total fresh weight increment. Comparison of before and after suspension culture, the distribution of different size callus pieces and the maintenance of callus type were basically unaltered, but a slight increase in relative water contents was observed. To induce the potential of plant regeneration, the directly transferring on plant regeneration solid medium containing MS basic salt, $0.2\;mg\;l^{-1}$ $\alpha$-naphthalene acetic acid (NAA), $2.0\;mg\;l^{-1}$ kinetin (Kn), and $2.0\;g\;l^{-1}$ casamino acid and indirectly transferring were simultaneously performed. Even now growth rates of suspension-derived callus on solid medium were approximately half of those of Type 1 callus, but faster somatic embryogenesis was observed. Rooting of all regenerated shoots was successfully performed on half-strength MS medium. All plants appeared phenotypically normal.

Effect of Plant Growth Regulators on Calls Initiation and Organogenesis from Tissue Culture of Arabidopsis thaliana Stem (애기장대 줄기 조직배양에 있어서 식물생장조절제가 캘러스 형성과 기관분화에 미치는 영향)

  • This experiment was carried out to investigate the effects of plant growth regulators on the organogenesis from the tissue culture of Arabidopsis thaliana stem, and the origin of the callus development. When the stem segments were cultured on medium with 2mg/L IAA or NAA, adventitious roots and trichomes were differentiated after 11 days of culture. Callus vigorously formed on medium with 2/L2,4 after 7 days of culture, but adventitious roots and trichomes were not differentiated from callus after 10 days of culture. This results suggesting that picloram is very effective auxin for the callus formation and organogenesis. Callus weakly formed on 0.05mg/L kinetin, and formed on combination of auxins(2mg/L) with 0.05mg/L kinetin. But the effect of combination of auxins and kinetin the callus formation was less than 2,4-D or picloram alone. A histological examination of callus formed on picloram showed that phloram showed that phloem parenchyma cells were divided and enlarged after 2 days of culture. Cortex parenchyma cells were divided and meristematic nodules were developed from these cells after 4 days of culture. Finally, callus formed on outside of cortex and epidermis by division of meristematic nodules after 7 days of culture.

  • aThis experiment was carried out to obtain the basic information on some factors affecting callus formation and plant regeneration in seed culture of orchardgrass. Varietal difference in callus growth and plant regeneration was obvious among four varieties of orchardgrass. "Amba" showed a relatively high capacity for plant regeneration. The $N_6$ medium was superior to MS and $B_5$ in callus formation and plant regeneration. The fresh weight of callus was promoted by the increase of dicamba concentration. However, the maximum frequency (30.0%) of plant regeneration was obtained from the callus formed in presence of 3 mg/L dicamba.

  • The experiment was conducted to investigate optimal condition for callus induction and plant regeneration for transformation system of gerbera. Callus induction was more effective in 'white day' then other two cultivar 'Songsongee' and 'Love Song' The optimized plant growth regulators concentration on callus induction, was MS basal medium with NAA 0.1 mg/L+ TDZ 0.5 mg/L. The optimized plant growth regulators concentration on plant regeneration, which was used MS basal medium was IAA 1.0 mg/L + BA 1.0 mg/L + Zeatin 0.1 mg/L. The optimized petiole age for more effective plant regeneration was 32 days petiole after in vitro subculture and MS basal medium strength was 1/2 MS strength.

  • This study was camed out in order to establish plant regeneration via seed-derived callus of perennialryegrass.Varietal difference in callus growth and plant regeneration was obvious between two cultivars of perennialryegrass. "Reveille" showed a relatively high capacity for plant regeneration. The MS medium was superiorto SH or Bj in callus formation and plant regeneration. The highest regeneration frequency (60%) from calluswas obtained in presence of 5 mg/ l 2,4-D and 1 mg/ t kinetin. Regeneration response varied among calluscultures initiated from the same cultivar. Regeneration frequency was the most effective in 6 weeks-old calliafter initiation and lost their regeneration capacity gradually over a period of 12 weeks.(Key words : Perennial ryegrass (Lolium perenne L.), Plant regeneration)e L.), Plant regeneration)

  • The callus of Canavalia lineata was induced from leaf tissues in MS medium supplemented with 10-5 M kinetin and 10-6 M IAA and was subcultured in Miller's medium supplemented with 10-5 M BAP and 10-6 M 2,4-D. When free amino acids of callus were analysed by HPLC, canavanine was not detected in the callus cultured either in the dark or light. But exogenously supplied canacanine was accumulated or consumned in the callus of Canavalia lineata.

  • Explants from stem, leaf and root tissue of Cissus juttae were plased on Ms medium supplemented ifferent combination of 2.4-D, NAA, Kinetin and BA in a actorial experiment. some callus formed in every treatment, but the best callus growth was on media containing $1.0{\;}mg/{\ell}$ NAA and $1.0{\;}mg/{\ell}$ BA. Formation of advantitious emboyos was evident in callus to MS medium supplemented with $0.1{\;}mg/{\ell}$.
