상황버섯으로부터 분리한 고분자 다당류의 특성 (Characteristics of the Polysaccharide extracted from Phellinus linteus)

  • 상황버섯 자실체의 열수 및 알카리 추출 다당류에 대한 성분 분석을 조사하여 다음과 같은 결과를 얻었다. 1. 상황버섯 자실체를 열수 및 알칼리 추출시 수율은 각각 1.1%와 4.4% 였으며, 당은 각각 81.1%와 37.4%를 함유하고 있었다. 2. 구성당은 glucose와 galactose가 주를 이루고, 아미노산은 열수 추출분획은 Tre와 Ser을 다량 함유하고 있었으며, 알칼리 추출분획은 Asp와 Ala가 주를 이루는 것으로 조사되었다.

수종의 버섯 추출물이 티로시나아제 프로모터에 미치는 효과 (Effect of Mushroom Extracts on Tyrosinase Promoter)

  • To estimate the inhibitory effect of some mushroom extract on melanin biosynthesis, we tested its inhibitory effects of five mushroom extracts on tyrosinase promoter in Bl6 mouse melanoma cells. In five mushroom extracts, Cordyceps militaris and Poria cocos exhibited low repression effect on tyrosinase promoter. However, Ganoderma lucidum, Paecilomyces japonicus, Phellinus linteus showed high repression effect, Especially, Paecilomyces japonicus and Phellinus linteus extracts had very higher repression effect approximately $81{\sim}83%\;at\;100{\mu}g/mL$. In the MTT assay, Paecilomyces japonicus and Phellinus linteus extracts exhibited high cytotoxicity. Therefore, repression effect of the extracts were closely connected with cytotoxicity.

  • Fraction (PMEx-AH-${\beta}$) of water-soluble polysaccharide, showing stimulating activity against macrophages, was isolated from mycelia of solid cultured Phellinus linteus by hot-water extraction, ethanol precipitation, and chromatography. Chemical characteristics of PMEx-AH-${\beta}$ were as follows: carbohydrate content, 71%; monosaccharide composition, Man:Glu:Gal (9:64:27); molecular weight, $1-7{\times}10^4$; uronic acid content, 6.8%. Fundamental structure of PMEx-AH-${\beta}$ is deduced as ${\beta}$-($1{$\rightarrow}6$)-D-glucan with ${\beta}$-($1{\rightarow}3$)-D-glucosidic side chains based on methylation analysis.

Antitumor and Antioxidant Activities of the Extracts from Fruiting Body of Phellinus linteus

  • Fruiting bodies of Phellinus linteus were extracted by hot water and alkali methods. Sugar contents of PL-H (hot water extract) and PL-A (alkali water extract) were 81.1%, 37.4% and protein contents were 6.2%, 21.8%, respectively. Amino acid pattern showed that two extracts contained large amount of aspartic acid and alanine. Two extracts showed characteristic IR absorption pattern for glycosidic bond at $890\;cm^{-1}$. PL-H was divided two fractions by gel filtration chromatography and the molecular weights of each fraction were estimated to be about 10 kD and 225 kD, respectively and also PL-A was estimated 10 kD. Two extracts showed strong antitumor, immunomodulating and antioxidant activities, and were compared with commercialized glycopeptide anticancer drugs.

신규 목질진흙버섯 KACC93057P의 배양적, 형태적 특성 (Morphological and cultural characteristics of a novel Phellinus linteus KACC93057P)

  • 국내에서 분리한 신규 P linteus KACC93057P의 자실체의 형태적 특성을 육안적 으로 관찰한 결과 대가없이 기주부착형으로 자실체표면은 단단한 목질형태 (hardy woody) 로 동심원으로 윤층대 (zonate)와 홈을 형성하고 담갈색을 띠고 있었다. 자실체 뒷면 테두리는 진황색, 내부는 담황갈색을 띠며 현미경 미세구조를 관찰한 결과 관공은 원형또는 각진형이며, 관공구는 5-7 per mm크기로 나타났다. hyphal system은 2분지형이며 포자는 $4.5-6{\times}4-5{\mu}m$크기로 난형또는 유구형 담갈색을 띠어 P linteus와 유사한 특성을 보였다. YGMA배지상에서 KACC93057P는 $25-30^{\circ}C$가 최적온도로 균사체 성장률이 장수상황과 고려상황에 비하여 10-25% 높았으며 균사밀도가 다른 균주보다 현저히 높게 나타났다. ITS-rDNA염기서열비교결과 96% 이상의 상동성으로 I. lonicericola, I. linteus, I. baumii와 근연관계를 형성 하였다.

  • 참나무 단목원목으로 재배한 P. linteus KACC93057P(PLHS)2년산 자실체 60%에탄올 추출물의 총 페놀함량은 $19.05{\pm}0.32mg\;GAE/g$로 다른 비교구 버섯 종에 비하여 4배에서 10배이상의 높게 나타났다. $125{\mu}g/mL$농도에서 DPPH와 ABTS 라디칼 소거능은 62%와 100%를 나타냈으며 FRAP 환원력은 $0.7{\mu}MFeSO_4/g$으로 비교 구 버섯종 보다 2에서 10배이상의 유효활성이 검출 되었다. PLHS 자실체 ethyl acetate분배 추출물의 HPLC분석에서 styrylpyrone 계열의 화합물 davallialactone, hispidin, hypholomine B와 phenylpropanoid 계열의 화합물인 caffeic acid가 동정 되었다.

수종 톱밥에서 목질진흙버섯균 Phellinus linteus의 균사생장 (Mycelial Growth of Phellinus linteus with Various Sawdusts)

  • 목질진흙버섯은 항암활성이 우수한 버섯으로 잘 알려져 있으나 아직까지 자실체 형성 및 발육에 관해 체계화된 연구결과는 보고되지 않고 있으므로 본 연구는 목질진흙버섯균의 인공재배를 위한 기초 자료를 얻고자 수종별 톱밥, 첨가제 및 첨가수준, 배지의 수분함량 등에 따른 균사생장 정도를 비교 분석한 결과이다. 수종별 톱밥에서의 균사생장은 참나무, 뽕나무, 복숭아나무 톱밥에서 양호하였다. 첨가제 및 첨가수준별 균사생장은 쌀겨, 밀기울, 옥수수피를 참나무 톱밥에 $20{\sim}30%$ (V/V) 혼합하였을 때 양호하였다. 참나무 톱밥과 쌀겨를 80:20으로 혼합된 배지의 수분함량은 $60{\sim}70%$에서 균사생장이 양호하였다. 참나무 톱밥과 쌀겨를 80:20으로 혼합하여 polypropylene 850 cc병에 종균 배양기간은 $25^{\circ}C$에서 58일이 소요되었다.

β-Glucuronidase 저해 활성이 우수한 결명자를 첨가한 상황 균사체 배양액의 생리활성 (Biological Activities of Phellinus linteus Mycelium Culture with Cassiae Semen Extract on β-Glucuronidase Inhibitory Activity)

  • We examined the effects of biological activity Phellinus linteus mycelium culture with cassiae semen extract. Firstly, the optimal temperature, initial pH and culture period for mycelial growth in a liquid culture of P. linteus were determined, and they were $30^{\circ}C$, pH 5.0 and 8 days respectively. The five herbal materials were examined against several health functional efficacies, and, as a result, Cassiae semen was chosen, with its superior inhibitory effects in ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities(95.3%, 80.9%, 96.1 and 24.2%, respectively). P. linteus fruit body was investigated on ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities, and they were 54.7%, 81.9%, 30.0% and 20.1%, respectively. Accordingly, C. semen was used in the following experiment, to give an additive functional effect on the P. linteus. As the amount of C. semen in the cultural media increased, mycelial weight and ${\beta}$-glucan contents also increased, but final pH was not influenced. In addition, the ${\beta}$-glucuronidase inhibitory activity, electron donating activity, and ${\alpha}$-glucosidase inhibitory activity increased. P. linteus mycelium culture showed higher activities in the other three tests above, except for electron donating activity, when C. semen was added to the medium before cultivation.

Phenolic Compounds from the Fruit Body of Phellinus linteus Increase Alkaline Phosphatase (ALP) Activity of Human Osteoblast-like Cells

  • Secondary metabolites from the fruit body of Phellinus linteus were evaluated for their proliferative effect on human osteoblast-like cells. 3-[4,5-Dimethylthiazole-2-y1]-2,5-diphenyl-tetraxolium bromide (MTT) assay and alkaline phosphatase (ALP) activity assay were used to assess the effect those isolates on the human osteoblast-like cell line (Saos-2). Activity-guided fractionation led to the isolation of ALP-activating phenolic compounds through the extraction of P. linteus, solvent partitioning, and repeated silica gel and octadecyl silica gel (ODS) column chromatographic separations. From the result of spectroscopic data including nuclear magnetic resonance (NMR), mass spectrometry (MS), and infrared spectroscopy (IR), the chemical structures of the compounds were determined as 4-(4-hydroxyphenyl)-3-buten-2-one(1), 2-(3',4'-dihydroxyphenyl)-1,3-benzodioxole-5-aldehyde (2), 4-(3,4-dihydroxyphenyl)-3-buten-2-one (3), 3,4-dihydroxybenzaldehyde (4), and protocatechuic acid methyl ester (5), respectively. This study reports the first isolation of compounds 1-3 and 5 from P. linteus. In addition, all phenolic compounds stimulated proliferation of the osteoblast-like cells and increased their ALP activity in a dose-dependent manner ($10^{-8}$ to $10^{-1}\;mg/mL$). The present data demonstrate that phenolic compounds in P. linteus stimulated mineralization in bone formation caused by osteoporosis. The bone-formation effect of P. linteus seems to be mediated, at least partly, by the stimulating effect of the phenolic compounds on the growth of osteoblasts.

Optimal Extraction Conditions of Anti-obesity Lipase Inhibitor from Phellinus linteus and Nutritional Characteristics of the Extracts

  • In an effort to develop novel mushroom-derived anti-obesity nutraceuticals, water and ethanol extracts containing the lipaseinhibitory compound from Phellinus linteus were prepared, and their nutritional components were determined. The optimal conditions for the extraction of P. linteus lipase inhibitor involved the treatment of the fruiting bodies with distilled water at $80^{\circ}C$ for 72 hr and 80% ethanol at $100^{\circ}C$ for 60 hr, respectively. The distilled water extract and ethanol extract contained 10.9% and 6.11% of crude protein, and 0.96% and 15.86% of crude fat, respectively. Additionally, the distilled water extract contained a large quantity of minerals, including 239.5 mg of K, 39.3 mg of Mg, and 39.3 mg of Na. The free amino acid content of the distilled water extracts was also higher than that of the ethanol extracts, and in particular, the distilled water extracts contained 5,139 mg of asparagine, 3,891 mg of tryptophan, 2,598 mg of alanine, and 2,066 mg of serine in 100 g of the distilled water extracts. 100 g of the distilled water and ethanol extracts were found to contain 12.31 g and 8.16 g of malic acid, respectively.
