The Performance of NI/$MgAl_2O_4$ Coated Metal Monolith in Natural Gas Steam Reforming for Hydrogen Production (NI/$MgAl_2O_4$코팅된 금속 모노리스 촉매의 수소 생산을 위한 천연가스 수증기 개질 반응특성에 관한 연구)

  • The metal monolith catalyst coated with 15wt% Ni/$MgAl_2O_4$ is applied to the natural gas steam reforming for hydrogen production. To address the improvement of adherence between metal monolith and catalyst coating layer, the pre-calcination temperature as well as the coating conditions of $Al_2O_3$ sol are optimized. When the Fe-Cr alloy monolith is pre-calcined at $900^{\circ}C$ for 6 h, $Al_2O_3$ layer was formed uniformly on the entire surface of the metal substrate. It is seen that the formation of $Al_2O_3$ layer on the monolith surface is essential for the uniform coating of $Al_2O_3$ sol onto the monolith substrate. The monolith catalyst coated with 10wt% $Al_2O_3$ sol shows high $CH_4$ conversion and good thermal stability as compared with the monolith catalyst without $Al_2O_3$ sol coating under severe reaction conditions with high GHSV of 30,000 $h^{-1}$ at $700^{\circ}C$. In addition, the metal monolith catalyst shows higher catalytic activity and better thermal conductivity than 15wt% Ni/$MgAl_2O_4$ pellet catalyst.

  • This study was carried out investigate optimal additive content of spent medium from oyster mushroom in Volvariella volvacea cultivation. The formation of spent mushroom medium from oyster mushroom consisted of 50% pine sawdust, 30% beet pulp and 20% cotton seed hull pellet. The higher amount of spent medium from oyster mushroom resulted in higher total nitrogen content and C/N ratio of medium decreased according to increasing mixing ratio of spent medium from oyster mushroom. The fruit body yield and biological efficiency of supplemented with 50% spent medium from oyster mushroom was same level of non additive medium.

  • The enantiomeric separation of lactic acid for its absolute configuration has become important task for understanding its biological origin and metabolic process involved in the formation of lactic acid. It involves the conversion of enantiomers as diastereomeric O-pentafluoropropionylated (S)-(+)-3-methyl-2-butyl ester and the direct separation by gas chromatography-mass spectrometry on a achiral capillary column. The (R)- and (S)-lactic acids were completely separated with a high resolution of 1.9. The newly developed method showed good linearity (r ${\geq}$ 0.999), precision (% relative standard deviation = 3.4-6.2), and accuracy (% relative error = -7.7-1.4) with the detection limit of 0.011 ${\mu}g/mL$. When the method was applied to determine the absolute configuration of lactic acid in Lactobacillus delbrueckii subsp. lactis 304 (LAB 304), the composition (%) of (R)-lactic acid in the cell pellet and in the culture medium were $89.0{\pm}0.1$ and $78.2{\pm}0.4$, respectively. Thus, it was verified that the present method is useful for the identification and composition test of lactic enantiomers in microorganisms.

Reduction of Polyspermy in Porcine in vitro Fertilization by Modified Swim-UP Method

  • The high incidence of polyspermic fertilization is one of the major causes lowering the overall efficiency of porcine IVF. The common procedure for IVF involves the co-culture of both gametes in the medium drop, which increases sperm concentration and incidence of polyspermy. Therefore, the present study was carried out to increase the efficiency of porcine IVF by reducing polyspermy using a modified swim-up method. This method modifies conventional swim-up washing by placing oocytes directly at the time of washing. Sperm pellet was prepared in the tube and mature oocytes were placed on cell strainer with $70 \mu m$ pore size (Falcon 2350) at the top of the tube. After insemination, the oocytes were stained for examination. Also, the developmental potential of fertilized embryos was measured to evaluate for the feasibility of this method. While having similar penetration rates in both methods ($86.67 \pm 2.36% to 83.33 \pm 1.36%$), there was a significant reduction of polyspermy in modified swim-up method ($17.50 \pm 1.60%$) compare to the control ($44.1 \pm 3.70%$ (p<0.05). Subsequent culture showed higher rate of blastocyst formation in modified swim-up method (20.44$\pm$0.99%) than the control ($15.73 \pm 3.26%$) (P<0.05), even though there was no significant difference. These results suggest that, by controlling the number of spermatozoa reaching the oocytes, porcine oocytes might be protected from polyspermy in vitro. Also, the developmental potential of the fertilized embryos using this method could be improved by increasing the pool of spermatozoa with better quality. Further optimization of the procedure required to implicate this method in routine porcine IVF.

Investigation of Dental Plaque, Calculus and Gingival Inflammation in Beagle Dogs (비글견에서 치태와 치석, 치은 염증의 조사)

  • This study was performed to investigate of dental plaque, calculus and gingival inflammation in Beagle dogs. Forty adults Beagle dogs (28 male and 12 female) were used in this study. The dogs weighed 9.5 kg and were in good oral and systemic health as determined by physical examination, and all dogs had full and normal dentition. The dogs were given a commercial pellet feed during 2 years period. For all examination procedures, the dogs were premedicated with a subcutaneous injection of atropine sulfate (0.04 mg/kg). Anesthesia was induced and maintained by intravenous administration of ketamine (8 mg/kg) and xylazine (2 mg/kg). Dental plaque, calculus and gingival inflammation were assessed by Logan and Boyce clinical plaque index. Calculi covering the maxillary carnassial and first molar teeth were extensive and were accompanied by severe gingival inflammation and pocket formation. Calculi, accompanied by gingival inflammation, were clearly evident on buccal surfaces of other teeth. Calculi didn't showed on the lingual surfaces, but linguogingival inflammation formed in premolar teeth. Although the general pattern was clear, there was considerable variation among dogs in the rate of deposition of calculus and extend of gingival inflammation. This investigation suggest that feeding of the commercial dry food without dental hygiene increase plaque accumulation and may be a contributing factor in calculi formation and periodontal disease.

Study on the Fuel Decomposition Characteristics and Coke Formation by Type of Endothermic Fuel and Method of Catalyst Molding (흡열연료 종류와 촉매 성형 방법에 따른 분해특성과 코크 생성에 관한 연구)

  • This study was carried out to investigate fuel decomposition characteristics and coke formation according to types of endothermic fuels and methods of catalyst molding. Methylcyclohexane (MCH), n-dodecane, and exo-tetrahydrodipentadiene (exo-THDCP) were used as the endothermic fuels. As a catalyst, USY720 supported with platinum was used. It was manufactured by only using pressure to disk-type, or pelletized with a binder and a silica solution. The characteristics of the catalysts according to the molding method were analyzed by X-ray diffraction analysis, scanning electron microscopy, nitrogen adsorption-desorption isotherm, and ammonia temperature programmed desorption analysis. The reaction was carried out under conditions of high temperature and high pressure ($500^{\circ}C$, 50 bar) in which the fuel could exist in a supercritical state. The product was analyzed by gas chromatograph/mass spectrometer and the coke produced by the catalyst was analyzed by thermogravimetric analyzer. After the reaction, the composition of the products varied greatly depending on the structure of the fuel. In addition, the crystallinity and surface properties of the catalysts were not changed by the method of catalyst molding, but the changes of the acid sites and the pore characteristics were observed, which resulted in changes in the amount and composition of products and coke.

Submicroscopy of Forest Soils (kandiustults) Derived from Granite in Southern Part of Korea (우리나라 남부지역(南部地域) 화강암질(花崗巖質) 삼림토양(森林土壤)의 SEM과 TEM에 의한 관찰(觀察))

  • To understand the weathering processes of the soil by submicroscopic method is very important to realize the properties of the soils. In this study soil formation processes show every steps to the changes in chemical and mechanical properties and the submicroscopic characteristics of soil weathering on the profiles of forest soils derived from granite in southern part of Korea. Fecal pellets(SEM) are given a full detail of the positive activities of the forest soil animals; mainly invertebrates in the O horizon and the E horizon. External shapes of fecal pellets have been divided into five groups : spherical, ellipsoidal, cylindrical, platy and threadlike. But doughnutlike form of fecal pellets is observed in this study. The soluble and suspended materials in the soils move downwards by percolation from the A horizon to the B or the BC horizons, and result in the illuviation cutans(SEM) on the ped surface of the lower horizon and deposited stack of kaolinite. Illuviated cutans are deposited on the ped surface even in the depth of 312cm in the BC horizon as well as the Bt horizon and comprise of fine silt, coarse clay and fine clay. A lot of halloysites are observed on the cutan surface. Halloysite formation from feldspars has been well known but a lot of hallyosite formation are observed in this study. The formation were predicted by Jackson(1962), inferred by Wada and Kakuto(1983a, b) and proved evidently by Cho and Mermut(1992a, b). This also suggests that halloysites in the soils derived from granite are formed a lot from ferruginous chlorites. The release of Fe from the chlorite structure are significant pedogenic processes and newly formed Fe oxides imparted a red color to the soils. The iron oxides particles, which are ejected and recrystalized, aggregate thickly on the edge of the ferruginous chlorites, and this indicates the release of structural Fe from weathered chlorites. Hematites and goethites are frequent in the fine clay in this soils.

Use of Peristeum as a Source of Endothelial-like Cells (혈관내피유사세포 채취의 원천으로 골막의 활용)

  • Purpose: The periosteum is a well-known source of osteogenic precursor cells for tissue-engineered bone formation. However, cultured endothelial or endothelial-like cells derived from periosteum have not yet been investigated. This study focused on endothelial-like cell culture from the periosteum. Methods: Periosteal tissues were harvested from the mandible during surgical extraction of lower impacted third molars. The tissues were treated with 0.075% type I collagenase in phosphate-buffered saline (PBS) for 1 hr at $37^{\circ}C$ to release cellular fractions. The collagenase was inactivated with an equal volume of DMEM/10% fetal bovine serum (FBS) and the infranatant was centrifuged for 10 min at 2,400 rpm. The cellular pellet was filtered through a $100{\mu}m$ nylon cell strainer, and the filtered cells were centrifuged for 10 min at 2,400 rpm. The resuspended cells were plated into T25 flasks and cultured in endothelial cell basal medium (EBM)-2. Results: Among the hematopoietic markers, CD146 was more highly expressed than CD31 and CD34. The periosteal-derived cells also expressed CD90 and CD166, mesenchymal stem cell markers. Considering that the expression of CD146 was constant and that the expression of CD90 was lower at passage 5, respectively, the CD146 positive cells in passage 5 were isolated using the magnetic cell sorting (MACS) system. These CD146 sorted, periosteal-derived cells formed tube-like structures on Matrigel. The uptake of acetylated, low-density lipoprotein, labeled with 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI-Ac-LDL) was also examined in these cells. Conclusion: These results suggest that the CD146-sorted positive cells can be referred to as periosteal-derived CD146 positive endothelial-like cells. In particular, when a co-culture system with endothelial and osteoblastic cells in a three-dimensional scaffold is used, the use of periosteum as a single cell source would be strongly beneficial for bone tissue engineering.

Favorable Condition of Culture and Sclerotial Formation by Inonotus obliquus (차가버섯(Inonotus obliquus)의 배양조건과 균핵 형성)

  • Altogether twenty isolates were collected from Wakayama, Japan. The optimum mycelial growth of I. obliquus was observed in BMYA and MCM. The optimum temperature and pH for the mycelial growth were $25^{\circ}C\;and\;6.0{\sim}7.0$, respectively. The optimum carbon and nitrogen sources were dextrose and yeast extract, respectively. Similarly, the optimum mineral salt was $K_{2}HPO_{4}$. The optimum number of mycelial discs for the mycelial growth was $6{\sim}7$ per 100 ml. Similarly, the optimum culture period was $21{\sim}22$ days in liquid broth. The optimum brown rice : water ratio was 1 : 1. No difference in mycelial growth was observed in all the four types of tree stumps used.

High Temperature Oxidation Behavior of Nd-doped $UO_2$ (네오듐 고용 이산화우라늄의 고온 산화거동)

  • The phase change of $(U_{1-x}Nd_x)_3O_8$ powder produced by oxidation of Nd-doped $UO_2$ pellet at $500^{\circ}C$ was investigated by high temperature oxidation heat treatment at $900{\sim}1500^{\circ}C$ under an air atmosphere. The XRD analysis results showed that the formation of $(U_{1-y}Nd_y)O_{2+z}$ phase and $U_3O_8$ phase from metastable $(U,Nd)_3O_8$ phase initiated at a temperature of $1000^{\circ}C$. The relative integrated intensity of $(U_{1-y}Nd_y)O_{2+z}$ phase to $U_3O_8$ phase increased with increasing of the oxidation temperature from 1100 to $1500^{\circ}C$. And also, it was found from the SEM observation that the particle size of $(U_{1-y}Nd_y)O_{2+z}$ phase increased with increasing of the oxidation temperature. However, electrone probe X-ray microanalyzer (EPMA) analysis results showed that Nd contents in $(U_{1-y}Nd_y)O_{2+z}$ phase decreased with increasing of the oxidation temperature. This behavior on the ground of XRD, SEM, and EPMA analysis data could be interpreted in terms of the transportation of U ions from $U_3O_8$ phase into $(U_{1-y}Nd_y)O_{2+z}$ phase through the interface of two phases during high temperature oxidation.
