Embryos of early-ripening peaches could not achieve physiological maturation or undergo abortion before harvest. Embryo rescue is an effective strategy to rescue embryos from early-ripening peaches. Thus, the current study was carried out to determine the appropriate developmental stage and optimal medium composition for embryo rescue in early-ripening peach. Development of open-pollinated 'Yumi' fruit was investigated from 20 to 90 days after full bloom (DAFB) to explore period occurring endocarp hardening. After endocarp hardening, embryo development was observed by light microscopes. Shoot and root meristems were observed at 65 DAFB and embryo size rapidly increased at 75 DAFB. Embryos collected at 75, 80, 85, and 90 DAFB were cultured on four media based on Driver and Kuniyuki (DKW) medium. Germination rate of embryos cultured on four media gradually increased from 75 to 90 DAFB and reached 100% at 90 DAFB. Notably, M3 medium (0.5 DKW supplemented with 6-benzylaminopurine (BAP) 1.0 ㎎/L) displayed the highest germination rate at 75 and 80 DAFB stages. Growth and development of shoot and root were pronounced in plantlet cultured at 90 DAFB stage. While delayed shoot growth was evident in plantlets cultured at 75, 80, and 85 DAFB stages, this retardation could be overcome through the application of growth regulators, particularly in M3 and M4 (0.5 DKW supplemented with BAP 1.0 ㎎/L and indole-3-butyric acid 0.5 ㎎/L) media. Remarkably, roots of plantlet grown in M4 medium exhibited limited elongation. In conclusion, germination rate of embryo and growth of embryo cultured plantlet can be enhanced by collecting seeds from early-ripening 'Yumi' at the 90 DAFB stage and conducting embryo culture using the M3 medium.
Purpose: The compression strength of corrugated fiberboard containers used to package agricultural products rapidly decreases owing to various environmental factors encountered during the distribution of unitized products. The main factors affecting compression strength are moisture absorption, long-term top load, and fatigue caused by shock and vibration during transport. This study characterized the durability of corrugated fiberboard containers for packaging fruits and vegetables under simulated transportation conditions. Methods: Compression tests were done after corrugated fiberboard containers containing fruit were vibrated by an electro-dynamic vibration test system using the power spectral density of routes typically traveled to transport fruits and vegetables in South Korea. Results: To predict loss of compression strength owing to vibration fatigue, a multiple nonlinear regression equation ($r^2=0.9217$, $RMSE=0.6347$) was developed using three independent variables of initial container compression strength, namely top stacked weight, loading weight, and vibration time. To test the applicability of our model, we compared our experimental results with those obtained during a road test in which peaches were transported in corrugated containers. Conclusions: The comparison revealed a highly significant ($p{\leq}0.05$) relationship between the experimental and road-test results.
Sample presentation, which means how to set samples to an NIR instrument, is very important in Near Infrared (NIR) Spectroscopy. When sample presentation is not suitable for the samples that you use, very good spectra can not be obtained even if you use a sophisticated NIR instrument. In my presentation, various NIR sample presentations for agricultural products such as intact fruits, single grains, vegetable juice and the others will be explained. In case of peaches with thin peel, the fiber optics of Interactance can be used. However, the fiber optics are not suitable for oranges with relatively thick peel. In this case, transmittance method is useful. As for a small sample such as single grains, a specially designed cell is needed. The cell in transmittance mode has been developed and then applied to single kernels of rice and soybean. In this case we also used the fiber optics. As regards liquid type of sample, a cuvette cell made of quartz in transmittance mode is popular. However, it is time-consuming to wash and dry it. In order to compensate this disadvantage the sample presentation using normal test tubes as sample cells have been developed and applied to milk, rumen juice and urine of a milking cow. An individual test tube can be used for each sample if you use the calibration equation with sample cell compensation. The test tube cell has also been applied to spinach juice for determination of undesirable constituents. It is concluded that sample presentation is most important for NIR Spectroscopy.
In order to produce vinegar using onions, 12 acetic acid bacteria were screened from the juice of fallen peaches, and a strain showing the highest acetic acid productivity among them was selected and identified as Acetobacter tropicalis No. 22. The culture broth containing 2.5%(w/v) of initial sugar concentration showed maximum acetic production after 10 days of cultivation, and the acetic acid was produced at the highest rate and reached the maximum acidity after 2 to 6 days of cultivation when the residual sugar and the ethanol concentration were in the range of 1.6 to 2%(w/v) and 0.6 to 1.8%(v/v), respectively. Also optimum conditions for acetic acid production by response surface method using the fractional factorial design with 3 variables and 5 levels were involved with initial ethanol content of 4.67%(v/v), initial acidity of 0.03%, and initial glucose concentration of 2.35%(w/v) and predicted level of acetic acid production at these conditions was 3.77%.

Okinawa Prefecture is located in the southernmost part of Japan and has a subtropical climate. A lot of tropical fruits such as pineapples, mangoes and papayas are produced. Pineapples were mainly supplied to the canning industry. Since April 1990, the Japanese Government released the foreign trade restriction of canned pineapples and pineapple juice in accordance with GATT. Okinawan Farmers have been expanding the customers from the processing industry to the fresh market. During the recent year, fully mature, high quality pineapples which have 15 Brix in sugar content are produced in green houses on Okinawa. Prices are 2 to 3 times those of the imported pineapples. At present, they are sorted manually, and this sorting might cause the pineapples to lose their reputation. NIR applied fruit sorters have been developed and operated for apples and peaches in Japan . NIR applied tests were carried out to consider the feasibility into the NIR technique for measurement of the NIR a alysis suggests adequate correlations between sugar content and NIR reflectance properties of pineapples and mangoes.

To detect Xanthomonas arboricola pv. pruni, a loopmediated isothermal amplification (LAMP) detection method were developed. The LAMP assay was designed to test crude plant tissue without pre-extraction, or heating incubation, and without advanced analysis equipment. The LAMP primers were designed by targeting an ABC transporter ATP-binding protein, this primer set was tested using the genomic DNA of Xanthomonas and non-Xanthomonas strains, and a ladder product was generated from the genomic DNA of X. arboricola pv. pruni strain but not from 12 other Xanthomonas species strains and 6 strains of other genera. The LAMP conditions were checked with the healthy leaves of 31 peach varieties, and no reaction was detected using either the peach leaves or the peach DNA as a template. Furthermore, the high diagnostic accuracy of the LAMP method was confirmed with 13 X. arboricola pv. pruni strains isolated from various regions in Korea, with all samples exhibiting a positive reaction in LAMP assays. In particular, the LAMP method successfully detected the pathogen in diseased peach leaves and fruit in the field, and the LAMP conditions were proven to be a reliable diagnostic method for the specific detection and identification of X. arboricola pv. pruni in peach orchards.
This study was attempted to determine the pectin contents of various fruits(Cherry, strawberry, plum, peach, apricot, apple, crabapple) and to investigate the changes in the properties of pectin and pectin gel during crabapple ripening. Methoxyl content, molecular weight and intrinsic viscosity were measured in ripened and unripened crabapples. In order to determine the characteristics of pectin gel, percent sag was measured by exchange Ridgelimeter, hardness and textural characteristics of jellies were measured by a penetrometer and a texturometer. The experimental results are summarized as follows: 1. The pectin contents of various fruits were 1.16% for cherry, 3.17% fir strawberry, 4.29% for plum, 5.51% for apricot, and pectin contents of peaches and apples varies depends on varieties from 4.54% to 5.61% to 6.52, respectively. The pectin contents of unripened and ripened crabapple wee 9.21% and 9.84% respectively, indicating that pectin content increased with fruit maturity. 2. The methoxyl contents, which was 6.61%~6.95%, was very low and it increased significantly during fruit ripening. Further, a decrease in acetyl content, intrinsic viscosity, molecular weight occurred during the ripening of crabapples. 3. The percent sag of crabapple jellies increased with fruit maturity from 5.26% to 5.79%, and the corresponding jelly grade decreased in ripened crabapple. 4. The hardness of jellies determined by a penetrometer was 79.15 pp.U. for ripened crabapple jelly, 90.62 pp.U. for unripe ned crabapple jelly. 5. In the textural characteristics of gels measured by a texturometer, the gels made from unripened crabapple were shown to have the highest values in hardness, cohesiveness, adhesiveness and gumminess.
Using the big data analysis of the Choseonwangjosilrok, this research aimed to figure out the fruits' types, prevalence, seasonal appearances as well as the royalty's perspective on fruits during Choseon period. Choseonwangjosilrok included nineteen kinds of fruits and five kinds of nuts, totaling 1,601 cases at 72.8% and 533 cases at 24.2% respectively. The text recorded fruits being used as: tributes for kings, gifts from kings to palace officials, tomb offerings, county specialties, trade goods or gifts to the foreign ambassadors, and medicine ingredients in oriental pharmacy. Seasonally the fruits appeared demonstrating an even distribution. Periodic characteristics were observed in decreasing quantity chronologically. From fifteenth century to nineteenth century, the fruits with timely features were seen: 804 times at 36.6%, 578 times at 26.3%, 490 times at 22.3%, 248 times at 11.3%, and 78 times at 3.5% respectively. In fifteenth century: citrons, quinces, pomegranates, cherries, permissions, watermelons, Korean melons, omija, walnuts, chestnuts, and pine nuts appeared most frequently. In sixteenth century: pears, grapes, apricots, peaches, and hazelnuts appeared most frequently. In seventeenth century: tangerines and dates appeared most frequently. In eighteenth century, trifoliate orange was the most frequently mentioned fruit.
According to the report of the Korea Agro-Fisheries and Food Trade Corporation (2019), the size of the HMR food market in 2019 exceeded 4 trillion won. On the other hand, the consumption of agricultural products continued to decrease. As reported by Woo (2020) who analyzed consumer panel survey data, when the 2010 agricultural product purchase index was 100, the crop purchase index in 2019 was 72 and the vegetable and special crop purchase index was 69. Therefore, the Rural Development Administration promoted the research projects to develop the technologies for producing stably raw materials of HMR and for optimizing local foods as HMR ingredients. Researches on the development of HMR using local agricultural products has been being promoted in Cheonan, Chungju, and Jeju in 2020. In the case of Jeju, it has been prepared and realized the B2B commercialization of Jeju agricultural products in connection with the convenience store ingredients supplier in the province. In the case of Cheonan, development of HMR foods like as Sundae, dumpling, sausage, and Tteokgalbi using willd chive, cherry tomatoes, shiitake mushroom, fortified perilla, and licopene fortified watermelon has been being promoted. Lastly, in the case of Chungju, a diverse HMR foods have been developed using beans, centella asatica, Japanese lady bell, apples and peaches such as blended juices, frozen boiled rice, and tofu kit. In the future, in connection with the regional specialized crop cultivation project promoted by the Rural Development Administration, we intend to support the convergence commercialization of R&D technology based on regional characteristics.
In this study, the effect of orchard expansion, structural improvement, and potential demand of the orchard scale-up project implemented since 2004 to strengthen the competitiveness of orchard farmers was examined. The results of the analysis are summarized as follows. First, looking at the effect of scale-up by checking the standard income data of the Rural Development Administration, it was possible to confirm the effect of scale-up in that the income per unit area increases as the scale increases. Second, through the scale-up project, the effect of structural improvement in transferring the orchards of the aged farm households to the younger farm households was confirmed to be 13 years old on average in the case of a sale business and 16 years old in the case of a lease business. Third, it was found that income increased at a statistically significant level after participating in the orchard scale-up project for major fruit crops such as apples, pears, and peaches. Fourth, it was found that age and cultivated area had a statistical effect on the probability of participating in the orchard scale-up project for farmers who did not participate in the project. The potential business demand using the estimated results was found to be between 25,203 and 37,089.
