Effect of Moutan Cortex Radicis on Gene Expression Profile of Differentiated PC12 Rat Cells Oxidative-stressed with Hydrogen Peroxide (모단피의 PC12 cell 항산화 효과와 관련 HO, MIF, COMT 유전자 발현에 미치는 영향)

  • Oriental medicine explains aging as the weakening of Kidney-ai, and Kidney-strengthening herbal medicines such as Yukmijihwang-tang have been studied for anti-aging effects. In Western Medicine, the hypothesis that reactive oxidant species(ROS) contribute to the aging process is generally accepted. It has been reported that Moutan Cortex Radicis extract (MCR) was the most effective constituent of Yukmijihwang-tang in decreasing ROS production in oxidative-stressed cells. The purpose of this study is to confirm the anti-oxidant effect of MCR on PC12 cells, the expression of Heme oxygenase (HO), Macrophage migradon inhibitory factor (MIF), Catechol-O-methyltransferase (COMT) using real time RT PCR. PC12 cells were treated without or with hydrogen peroxide in the presence or absence of MCR using MTS assay. Hydrogen peroxide decreased the viability of PC12 cells by 53% and MCR did not influence that of stressed PC 12 cells irrespective of dose or incubation period. However, MCR showed an inhibitory effect on production of ROS in stressed cells, both dose and incubation time dependently. In particular, 1 ㎎/㎖ of MCR for 24 h culture almost returned to normal level. In the quantiation of anti-aging related gene expression, MCR at 1 ㎎/㎖ increased the expression of HO by 370%, MIF by 180% and COMT by 280% through real time RT PCR. In conclusion, MCR treatment protected PC12 cells from hydrogen peroxide and decreased ROS production and enhanced anti-oxidative gene expression such as HO, COMT and MIF, which suggests that MCR is involved in controlling anti-aging of nerve cells through elimination of cytotoxic stimuli.

  • LED is just a semiconductor which can produce light. Currently, there are active research works on LED lighting technologies according to the growth of energy-saving environmental industry. Especially, LED communication is one of the active research works in these fields. In this paper, we design a LED communication-based PC-PC transmission system. A transmission circuit system(transmitter) using LED and a receiving circuit system(receiver) using PD(photo detector) and Op-amp are designed, respectively. The experiments for the designed system are performed as follows. One computer is connected at the end of transmitter and receiver, respectively, and text files are transmitted by using text transmission programming. In this experiment, we test the performance with various baud rates, LED colors, transmission ranges.

Glycyrrhizin Attenuates MPTP Neurotoxicity in Mouse and $MPP^+$-Induced Cell Death in PC12 Cells

  • The present study examined the inhibitory effect of licorice compounds glycyrrhizin and a metabolite $18{\beta}$-glycyrrhetinic acid on the neurotoxicity of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in the mouse and on the 1-methyl-4-phenylpyridinium ($MPP^+$)-induced cell death in differentiated PC12 cells. MPTP treatment increased the activities of total superoxide dismutase, catalase and glutathione peroxidase and the levels of malondialdehyde and carbonyls in the brain compared to control mouse brain. Co-administration of glycyrrhizin (16.8 mg/kg) attenuated the MPTP effect on the enzyme activities and formation of tissue peroxidation products. In vitro assay, licorice compounds attenuated the $MPP^+$-induced cell death and caspase-3 activation in PC12 cells. Glycyrrhizin up to $100{\mu}M$ significantly attenuated the toxicity of $MPP^+$. Meanwhile, $18{\beta}$-glycyrrhetinic acid showed a maximum inhibitory effect at $10{\mu}M$; beyond this concentration the inhibitory effect declined. Glycyrrhizin and $18{\beta}$-glycyrrhetinic acid attenuated the hydrogen peroxide- or nitrogen species-induced cell death. Results from this study indicate that glycyrrhizin may attenuate brain tissue damage in mice treated with MPTP through inhibitory effect on oxidative tissue damage. Glycyrrhizin and $18{\beta}$-glycyrrhetinic acid may reduce the $MPP^+$ toxicity in PC12 cells by suppressing caspase-3 activation. The effect seems to be ascribed to the antioxidant effect.

Effects of (+)-Eudesmin from the Stem Bark of Magnolia kobus DC. var. borealis Sarg. on Neurite Outgrowth in PC12 Cells

  • (+)-Eudesmin [4,8-bis(3,4-dimethoxyphenyl)-3,7 -dioxabicyclo[3.3.0]octane] was isolated from the stem bark of Magnolia kobus DC. var. borealis Sarg. and found to have neuritogenic activity. $50\;{\mu}M$ (+)-eudesmin induced neurite outgrowth and enhanced nerve growth factor (NGF)-mediated neurite outgrowth from PC12 cells. At this concentration, (+)-eudesmin also enhanced NGF-induced neurite-bearing activity and this activity was partially blocked by various protein kinase inhibitors. These included PD98059, a mitogen-activated protein kinase (MAPK) kinase inhibitor. GF109203X, a protein kinase C (PKC) inhibitor and H89, a protein kinase A (PKA) inhibitor. These results suggest that (+)-eudesmin can induce neurite outgrowth from PC12 cells by stimulating up-stream MAPK, PKC and PKA pathways.

Inhibition of Calcium Transport by $(1R,9S)-\beta-Hydrastine$ Hydrochloride in PC12 Cells

  • The effects of $(1R,9S)-\beta-hydrastine$ hydrochloride (BHSH) on $Ca^{2+}$ transport in rat pheochromocytoma PC12 cells were investigated. In the presence of external $Ca^{2+}$, BHSH at $100{\mu}M$ inhibited $K^+$ (56mM)-induced dopamine release, and $K^+-induced$ $Ca^{2+}$ influx and a sustained rise of $[Ca^{2+}]_i$. In addition, BHSH at 100 f.!M reduced the sustained rise of $[Ca^{2+}]_i$ elicited by 20 mM caffeine, but not by $1{\mu}M$ thapsigargin, in presence of external $Ca^{2+}$. These results suggest that BHSH inhibited $K^+-induced$ dopamine release and $[Ca^{2+}]_i$ influx, and store-operated $Ca^{2+}$ channels activated by caffeine, but not by thapsigargin, in PC12 cells.

Induction of Neurite Outgrowth by (-)-(7R, 8S)-Dihydrodehyd­rodiconiferyl Alcohol from PC12 Cells

  • A lignan derivative, (-)-(7R, 8S)-dihydrodehydrodiconiferyl alcohol (DHDA), was isolated from Kalopanax septemlobus L. and was observed to have neuritogenic activity. DHDA at 50 $\mu$M caused a marked induction of neurite outgrowth and an enhancement of nerve growth factor (NGF)-mediated neurite outgrowth from PC12 cells. However, it did not exhibit any neurotrophic action. At 50 $\mu$M, DHDA enhanced NGF-induced neurite-bearing activity. This activity was partially blocked by the mitogen-activated protein kinase (MAPK) inhibitor PD98059 and by GF109203X, a protein kinase C (PKC) inhibitor. These results suggest that DHDA can induce neurite outgrowth and enhance NGF-induced neurite outgrowth from PC12 cells by amplifying up-stream steps such as MAPK and PKC.

Compatibility Studies of Blends of Engineering polymers and Thermotropic Liquid Crystalline Polymers (엔지니어링 고분자와 열방성 액정고분자 블렌드의 상용성에 관한연구)

  • 열방성 액정고분자(LCP)와 polycarbonate(PC) poly(ether imide) (PEI) poly(PEEK), polysulfone(PSF), 그리고 polyarylsulfone(PAS)과의 블렌드에 대한 상용성을 연구하였다. 제조된 블렌드의 상거동에서 액정고분자가 PC-, PEI-, PEEK-, PSF-, 그리고 PAS-rich 상 에 녹아 들어가는 양이 PC, PEI, PEEK, PSF, 그리고 PAS가 액정 고분자 -rich상에 녹아들 어가는 양보다 많음을 알수 있었다. 측정된 블렌드의 유리전이온도 결과로부터 PC, PEI, PEEK와 액정고분자 사이의 상용성이 PSF, PAS와 액정 고분자 사이의 상용성에 비하여 더 좋음을 알수 있었다. 액정 고분자의 이방성을 고려하여 고분자-고분자 상호작용계수($\chi$12)를 결정하였으며, PC, PEI, PEEK, PSF, 그리고 PAS를 포함한 액정 고분자 블렌드에서 $\chi$12는 0.078-0.183으로 나타났다.

Intercalation of Polycarbonate/Montmorillonite Nanocomposites (폴리카보네이트와 몬모릴로나이트 나노복합체의 층간삽입)

  • Polycarbonate(PC)/montmorillonite (MMT) nanocomposites were prepared by solution and melt mixing methods. A d-spacing of the nanocomposites was measured by an X-ray diffractometer. Neat montmorillonite (MMT-Na) and MMTs modified by dodecyl ammonium (MMT-DA) or dimethyl hydrogenated tallow 2-ethylhexyl ammonium (MMT-25A) were used. The d-spacing value of PC/MMT-25A and PC/MMT-DA was higher than that of PC/MMT-Na. The d-spacing increased from around 12 to $37AA$ depending on the mixing method. PC was more readily introduced to the gallery of MMT as the molecular weight of PC reduced and the mixing time increased. PC/MMT-25A showed higher thermal stability by thermogravimetric analysis (TGA) than PC/MMT-DA and PC/MMT-Na.

Life Cycle Assessment of Mobile Phone Charger Containing Recycled Plastics (재생 플라스틱을 적용한 휴대폰 충전기 전과정평가)

  • Environmental impact of a mobile phone charger containing recycled plastic was quantified using LCA and the environmental benefits from the use of recycled and virgin plastic were compared. The assessment considers potential environmental impacts across the whole life cycle of the charger including; pre-manufacturing; manufacturing; distribution; product use; and end-of-life stages and quantified six environmental impact categories; Abiotic depletion; Acidification; Eutrophication; Global warming; Ozone layer depletion; and Photochemical oxidants creation. The study showed that the environmental impacts of the use stage accounted for 94.4% and 70% in the resource depletion and global warming impact categories, respectively, and the environmental impacts of the pre - manufacturing stage accounted for more than 98% in the other impact categories. The main cause of the environmental impacts in the use stage was electricity consumed by the charger. The main cause in the pre-manufacturing stage was PBA (Printed Board Assembly) and external case manufacturing. In order to quantify the environmental benefits of recycled PC (Polycarbonate) in the exterior case, the environmental impacts of 1 kg production of recycled PC and virgin PC were evaluated. The environmental impact on the abiotic depletion of the recycled PC is estimated to be 30% compared to the virgin PC, and the impacts on the other impact categories of the recycled PC were less than 5% of the virgin plastic. Sensitivity analysis was performed for 12 items including site data and assumptions made. The sensitivity of each item was less than 10%. The results of this study confirm that designing compact and light PBA, improving charging efficiency, and use of recycled plastic are important design factors to reduce the environmental impact of a charger.
