A chitinase gene of Serratia marcescens ATCC 27117 was cloned and expressed in Escherichiu di. A genomic library of S, marcescens was constructed with pUC 19 and screened using the swollen chitin agar plate for chitinolytic clones. A positive clone showing chitinclearance contains a recombinant pCHI 89, composed of 8.9 Kb chromosomal DNA fragment and pUC 19. Plasmid pCHI 89 produced 58 KD chitinase in E. coli, which was coincided with one of five extracellular chitinases produced by S. nzarccscens. Restriction endonuclease cleavage sites of the 8.9 Kb insert DNA fragment were mapped. E. coli JM109 harboring pCHI 89 inhibits the growth of a plant pathogenic fungus, Fusarium oxysporum.

The ascomycetous fungus Taphrina wiesneri, the pathogen of cherry witches' broom, is highly pathogenic to Prunus yedoensis, the most widely planted cherry trees in Korea as park and roadside trees. A collection of 13 strains of the pathogen in Korea and Japan was characterized by 18S rDNA gene sequence and restriction fragment length polymorphism (RFLP) analysis. In cluster analysis based on 18S rDNA gene sequence the strains were divided into 2 clusters. In RFLP analysis of the rDNA-IGS region using HhaI, the strains were separated into four patterns, B, C, D and G, of which pattern G was new.

Three double-stranded RNAs (dsRNAs), approximately 1.85, 1.65 and 1.27 kb in size, were detected in an isolate of Cytospora sacchari from Iran. Partial nucleotide sequence revealed a 1,284 bp segment containing one ORF that potentially encodes a 405 aa protein. This protein contains conserved motifs related to RNA dependent RNA polymerases (RdRp) that showed similarity to RdRps of partitiviruses. The results indicate that these dsRNAs represent a novel Partitivirus that we tentatively designate Cytospora sacchari partitivirus (CsPV). Treatment of the fungal strain by cyclohexamide and also hyphal tip culture had no effect on removing the putative virus. Phylogenetic analysis of putative RdRp of CsPV and other partitiviruses places CsPV as a member of the genus Partitivirus in the family Partitiviridae, and clustering with Aspergillus ochraceous virus 1.
A soybean-sprout rot epidemic occurred in a mass production soybean sprout factory in 2000 and 2001 in Korea, which caused up to 20% production loss. Among the causal pathogenic bacteria and fungi, Pythium deliense was found to be the dominant pathogen of severe root and hypocotyls rot, particularly in recirculating water system. An average of 90% of the isolated fungi from the rotted sprout on potato dextrose agar were Pythium sp. The fungal density of Pythium in the sampled water was monitored in the recycled water system for 1 year using a selective medium (com meal agar with Pimaricin, 10 mg; Rifampicin, 10 mg; and Ampicillin, 100 mg per 1 liter). The drained water from the soybean-sprout cultivation always had a certain amount of fungus in it. The removal of Pythium from the recycling water system must be thorough, safe, and environment friendly. However, the pathogen in the water was easily found even after ozone and chlorine treatments, which were devised on the recycling system for the removal of microorganisms. 5-$\mu\textrm{m}$ pore size filter was applied and was able to successfully control the disease. As the sprout industry increasingly shifts into mass production, the demand for water will increase continuously. Recycling water for sprout production is eco-friendly. However, a process must be devised to be able to first decompose organic matters before Pythium zoospores are filtered.
A fungal disease of the cultivated mushroom, Ganoderma lucidum, caused by Xylogone sphaerospora was epidemic throughout all cultivation areas in Korea which caused a lot of yield losses in the mushroom production. For controlling the disease, the screening of effective fungicides against the pathogenic fungus were conducted. Thirty seven commercially available fungicides were tested for their inhibitory activities on potato dextrose agar media supplemented with these fungicides at various concentrations. Twenty one fungicides significantly inhibited mycelial growth of the pathogen, Xylogone sphaerospora, but 16 fungicides had no inhibitory effect. Among these 21 fungicides, 17 fungicides also inhibited mycelial growth of Ganoderma lucidum as well, but imazalil, procymidone, triforine, and vinclozolin had no inhibitory effects. However, vinclozolin showed no inhibitory effect on mycelial growth of the mushroom even at the concentration of 50 $\mu\textrm{g}$/ml vinclozolin solution for 2 hours, and then the pathogen was inoculated. After two month-cultivation of the mushroom, over 90% of logs treated with vinclozolin without pathogen inoculation produced fruiting bodies. However, fruiting bodies were not produced form the logs inoculated with the pathogen, but not treated with vinclozolin. Fifty seven percent of logs. which were pre-treated with vinclozolin and then inoculated with the pathogen produced fruiting bodies. Based on the results, vinclozolin is effective for the control of yellow disease of the Ganoderma lucidum caused by Xylogone sphaerospora.
Recently, Platypus koryoensis has been reported as a major pest of oak trees in Korea which can introduce the pathogenic fungus(Raffaelea sp.) by making galleries into the heartwood of the tree. As the beetle has developed its effective drilling mouthpart enough to make tunnels, we have analyzed the fine structural aspects of the mouthpart using the field emission scanning electron microscopy(FESEM) to reveal its mechano-dynamic characteristics. The mouthparts of this ambrosia beetle which consist of a labrum, a pair of mandibles, a pair of maxillae and the labium exhibit typical morphology of mycophagous coleopteran beetles and have those characteristics of chewing mouthparts that can excavate galleries in the hardwood. Both of maxillary and labial palpi have the functions of direct the food to the mouth and hold it while the mandibles chew the food. The distal ends of these palpi are flattened and have shovel-like setae. Females have larger maxillary palpi and a larger gular region than males in general.

A large patch disease caused by Rhizoctonia solani AG2-2(IV) is a serious problem in turfgrass sites including golf courses and sports fields in Korea. The objectives of this study were to isolate some antagonistic microorganisms and to explain some involving mechanisms. Initially single colonies which were formed from the filtrates of various soil samples were obtained from LB culture and then co-cultured with R.solani AG2-2(IV) on PDA plate to explore some antagonistic microbes against for large patch fungus, Rhizoctonia solani AG2-2(IV). Out of total 82 antagonistic isolates which commonly had inhibition effect on Rhizoctonia solani AG2-2(IV) mycelial growth, one candidate (YPIN22) showed the most antifungal effect, which was confirmed by the longest distance from the edge of bacterial colony to the mycelial edge of the Rhizoctonia solani AG2-2(IV) in the dual culture. A succeeding investigation was to test any potential effect of the isolate on growth inhibition of 5 other turfgrass pathogens including R. solani solani AG2-2(IIIB), P. ultimum, C. caudatum, C. lunata, and F.oxysporum. Preliminary result indicated that the new isolate YPIN22 was also found to have antagonistic potential on the growth inhibition of those turfgrass pathogenic fungi, which was explained by inhibition zones ranging from 8 to 22mm. A further explanation of some characteristics of the isolate YPIN22 will be discussed in detail.
Okra (Abelmoschus esculentus (L.) Moench) has gained more popularity as an economically significant plant for its nutritional and medicinal value, especially in China. During 2014-2016, the root disease of okra was discovered in four okra commercial fields surveyed in China. A fungul was isolated from the infected tissues, and was identified by Verticillium dahliae based on morphological characteristics. Pathogenicity test demonstrated that the fungus was pathogenic on okra, and fulfilled Koch's postulates. The analysis of three sequences revealed 99-100% identity with the reported V. dahliae strain in GenBank. Neighbor-joining analysis of the gene sequences revealed that the representative isolates were clustered with V. dahliae. To the best of our knowledge, this is the first report of Verticillium wilt of okra in China.
Heterotrimeric G proteins (G proteins) are conserved in all eukaryotes and are crucial components sensing and relaying external cues into the cells to elicit appropriate physiological and biochemical responses. Basic units of the heterotrimeric G protein signaling system include a G protein-coupled receptor (GPCR), a G protein composed of ${\alpha},\;{\beta},\;and\;{\gamma}$ subunits, and variety of effectors. Sequential sensitization and activation of these G protein elements translates external signals into gene expression changes, resulting in appropriate cellular behaviors. Regulators of G protein signaling (RGSs) constitute a crucial element of appropriate control of the intensity and duration of G protein signaling. For the past decade, G protein signaling and its regulation have been intensively studied in a number of model and/or pathogenic fungi and outcomes of the studies provided better understanding on the upstream regulation of vegetative growth, mating, development, virulence/pathogenicity establishment, and biosynthesis of secondary metabolites in fungi. This review focuses on the characteristics of the basic upstream G protein components and RGS proteins, and their roles controlling various aspects of biological processes in the model filamentous ascomycete fungus Aspergillus nidulans. In particular, their functions in controlling hyphal proliferation, asexual spore formation, sexual fruiting, and the mycotoxin sterigmatocystin production are discussed.
In the present study, we determined the antifungal effect of obacunone isolated from Phellodendri Cortex against Candida ablicans, a pathogenic fungus. The antifungal effect was analyzed by an in-vitro susceptibility test and in a murine model of disseminated candidiasis. Possible mechanism of the antifungal activity was also examined. Analyses of data resulting from the susceptibility test revealed that the compound inhibited C. albicans growth. At 25 ${\mu}g$ obacunone/ml, there was app. 45% reduction of CFUs (colony forming units) as compared to obacunone-untreated C. albicans yeast cells (P<0.01). In the murine model of disseminated candidiasis due to C. albicans, obacunone enhanced resistance of mice against disseminated candidiasis. During an entire period of 30-day observation, control animals all died within 14 days, whereas 60% of obacunone-treated mice survived (P<0.05). In addition, obacunone inhibited the hyphal production, a major virulence factor of C. albicans, from the blastoconidial form. Thus, obacunone appears to have antifungal activity for C. albicans infection. This may possibly be mediated by the blockage of hyphal production.
