Bacteria in the viable but nonculturable (VBNC) state fail to produce colonies on routine bacteriological media, but are still alive in the state of very low metabolic activity. The aim of the present study was to induce the VBNC state of the Edwardsiella tarda using sea water microcosm under starvation conditions at $10^{\circ}C$ and to investigate resuscitation of the VBNC cells in temperatures changed from 10 to $25^{\circ}C$, with and without additives. E. tarda entered into the VBNC state within about 42-84 days of incubation in the microcosm. Throughout this period, the total cell counts as determined using acridine orange direct counting remained near the original inoculum level of ${\sim}10^8cells/ml$. The live cell counts measured with direct viable counting, on the other hands, declined to ${\sim}10^4cells/ml$. When the VBNC cells were incubated with addition of yeast extract, fish muscle extract or serum at $25^{\circ}C$, the ratios of resuscitated samples were 37%, 23%, and 37%, respectively. The characteristics of resuscitated E. tarda were consistent with those of the original E. tarda. When the resuscitated E. tarda were intraperitoneally injected into olive flounders, all fishes died within 5 days, indicating that the VBNC E. tarda might retain its pathogenic potential. Therefore, E. tarda under starvation conditions in the winter enter into the VBNC state and the VBNC E. tarda cells resuscitated at summer and autumn seawater temperature are considered to be pathogen continuously to olive flounder on the southern coast of Korea.
Olive flounder (Paralichthys olivaceus) is an important aquaculture fish species in Jeju Island, South Korea. Due to the intensification of flounder fish farming, huge amounts of chemical antibiotics are used against several fish diseases. This has many harmful side effects on fish, as well as human consumers. Hence, an alternative to chemical antibiotic agents is needed for disease control. In this study, three strains of rhizobacteria (BRH433-2, TRH415-2, and THJ609-3) were isolated from the rhizosphere of plants. Assays of their antibacterial activity against fish pathogens, such as S. iniae, S. parauberis, V. anguillarum, and E. tarda, were performed with untreated broth culture (without cell separation), supernatant, and precipitated pellets separated by centrifugation. Among these, the cell suspension prepared from the precipitated pellet showed significant antimicrobial activity when compared with that of the untreated broth culture and centrifugal supernatant. These results indicate that the three isolated rhizobacterial strains exhibit antibacterial activity. Analysis of the 16S rDNA sequences of the BRH-433-2, THJ609-3, and TRH415-2 strains showed the highest similarity to Burkholderia gladioli (99.5%), Pseudomonas baetica (97.7%), and P. koreensis and P. baetica (98.4%), respectively. We suggest that the strains hold promise in disease management of fish.
Early development growth and sexual phenomena of the hybrid between male spotted flounder (Verasper variegatus) and female olive flounder (Paralichthys olivaceus) were investigated. The hybrid produced was newly called the "Bumnupchi" in Korean name, "Mottled flounder" in English name. Fertilization rate, hatching rate and early survival rate of the mottled flounder were 56.9~72.3%, 86.7~92.5% and 89.1~96.0%, respectively. At 50th day and 240th day after hatching, they were 2.1$\pm0.2cm\;and\;22.9\pm2.1cm\;in\;total\;length, \;0.10\pm0.002g\;and\;179.0\pm38.5g$ in body weight, respectively. Eye position of the mottled flounder was mixed with both dextral (77.7%) and sinistral type (22.3%). Gonads were differentiated into ovarian type and testis type at 80th day after hatching, and its ratio was 1:1 (P>0.05). Most Gonads seemed to be sterile without having germ cell, whereas only 3.6% of investigated indiciduals turned out to have oocytes in the gonad. When the induction of sex reversal were attempted by daily oral administration of $17\beta$-methyltestosterone (0.5mg/kg fish) andestradiol-17$17\beta$(0.5mg/kg fish)from 40th to 100th day after hatching, both testis and ovary type were induced 81.9% and 87.7% of each hormone-treated fish, respectively.fish, respectively.

Two 15-week feeding trials were conducted to investigate how the rate and frequency of feeding affected the winter growth of olive flounder ($13^{\circ}C$). In the first experiment, triplicate groups of fish (initial mean weight $117{\pm}6.0$ g) were fed extruded pellets twice a day at feeding rates of 0.1, 0.25, 0.4, 0.55, and 0.57% (satiation) body weight per day (BW/d). The weight gain, specific growth rate, and feed efficiency increased significantly (P<0.05) with the feeding rates from 0.1 to 0.55% BW/d, but no significant differences in these parameters were found for fish fed diets of 0.55% BW/d and satiation. The moisture and ash contents of whole body of fish tended to decrease as the feeding rate increased, but the opposite trend was found for crude lipid content. In the second experiment, triplicate groups of fish (initial mean weight $117{\pm}6.3$ g) were fed extruded pellets to apparent satiation at the three different feeding frequencies: one meal 2 days, one meal a day and two meals a day. The weight gain and specific growth rate of fish fed one meal 2 days were significantly (P<0.05) lower than those of fish fed one or two meals a day, whereas no significant differences in the weight gain and specific growth rate were found between fish fed one and those fed two meals a day. Feed efficiency and condition factor were not significantly affected by feeding frequency. Based on these results, a feeding rate of 0.3% BW/d is recommended as maintenance feeding level, and the optimum feeding frequency is one meal a day with satiation feeding for the growth of olive flounder (117-147 g) during winter ($13^{\circ}C$).
This study analyzed the taste components of dorsal muscle and fin muscle collected from olive flounder cultured with extruded pellet (EP) compared with those of olive flounder cultured with raw fish moist pellet (MP) as a control. The olive flounder in this study were cultured for 10 months with either the formulated extrude pellets (FEP), commercial extruded pellets (CEP) or the MP feed, and the average weight was 1.15 kg. The major nucleotides and related compounds in the dorsal muscles of the fish fed with MP, CEP and FEP diets were AMP and IMP. The fin muscle of the olive flounder contained the largest amounts of IMP($5.91{\mu}mol/g$). The K value of the dorsal muscle in olive flounder fed with MP, CEP and FEP showed 9.63%, 9.83% and 5.84%, respectively. The free amino acids in the dorsal muscle of olive flounder showed significantly differences in the composition depending on the feed, but the distribution of free amino acids showed similar patterns. Among free amino acids, taurine showed the highest content in all the experimental groups, and the asparagine and citrulline contents were significantly higher in the groups fed with CEP and FEP than in the group fed with MP. The significant differences were observed in the contents of specific free amino acids in dorsal muscle and fin flesh depending on the feed for cultured olive flounder.
We evaluated the efficacy of $\beta$-hemolytic Streptococcus(S.) iniae vaccine on cultured olive flounder. Three hundred flounders(weight $50{\pm}5$ g) were obtained from two farm at Wando and Taean in the southern and western coast of Korea at May and June 2007, respectively. Twenty of flounders moved in 0.5 tons aquaria in land-marine tank system of National Veterinary Research and Quarantine Service. Seawater was transported from the sea of Inchon in western Korea, and water temperature maintained to $22^{\circ}C$ and $25^{\circ}C$ during the vaccination and challenge test, respectively. We used the formalin-inactivated $\beta$-hemolytic S. iniae vaccine produced by domestic manufacturers. The vaccine was intraperitoneally administered to fish. The vaccinated and control group were challenged with intraperitoneal injection by virulent S. iniae SI-36 isolates with $5.0{\times}10^8$ CFU/fish at 3 weeks after vaccination. We evaluated the vaccine efficacy by calculating numbers of dead fish, and observing of clinical signs, exterior and gross lesions, and examining bacteria isolation and identification. Thirty-four(25.2%) of 135 control and vaccinated group fish were dead with serious anemia, abdominal extension, and hernia of intestine during 3 weeks post vaccination. We isolated Neoheterobothrium hirame from the buccal cavity and Edwardsiella tarda from kidney of dead and diseased fish. When infected fish with these agents were challenged with S. iniae SI-36 isolates, the cumulative mortality of control and vaccinated group were 86.7, and 46.7%, respectively. However, significant differences(p<0.05) were observed on cumulative mortality between control(20.0%) and vaccinated group(95.0%) at second trials with 40 healthy, and relative percent survival(RPS) was 78.0%. We confirmed that the efficacy of $\beta$-hemolytic S. iniae vaccine on olive flounder were impacted by health condition such as bacterial and parasitic diseases.
When fed on one of the six isonitrogenous (45%) and isolipic (8%) feed containing 5 or 10% Undaria, 2% wasabi leaf, 2% wasabi stem and 0.5% herb (Obosan) for a period of 8 weeks, 95-98% juvenile flounder survived. Growth, feed efficiency and condition factor of the flounder fed on diet containing 0.5% herb were significantly (p<0.05) higher than those fed on diet supplemented with 10% Undaria. There was no significant (p>0.05) differences in moisture, crude protein, lipid and ash of the flounder receiving the different diets. The flounder fed on diet supplemented with 10% Undaria had the highest moisture but the lowest lipid in liver. Hematological parameters such as red blood cell, hematocrit and hemoglobin and serum constituents such as glucose, total cholesterol and glutamic-oxaloacetic transaminase of the flounder fed on the diets varied but no specific trend became apparent. Lysozyme activity in the serum of the flounder fed on diet supplemented with 5% Undaria and the herb, as well as nitroblue tetrazolium (NBT) reduction of macrophage in the head kidney of the flounder fed on diet containing the herb and 2% wasabi stem were significantly (p<0.05) higher than those receiving control diet. Briefly, the herb supplementation promoted growth and that of Undaria and wasabi stem enhanced non-specific immune response.

A 10-week feeding trial was conducted to investigate the effects of feeding rate on growth and body composition of adult flounder during the summer season. Three replicated groups of the fish (initial mean weight of $535{\pm}4.1\;g$) were hand-fed a extruded diet twice a day at the different feeding rates of satiation (100%), 90%, 80% and 70% of satiation for 10 weeks. Mean Water temperature was $19.2{\pm}2.8^{\circ}C$ during the feeding trial. Weight gain and condition factor increased significantly with increase in daily feeding rate (P<0.05), but there was no significant difference in weight gain of fish fed the diet between 100% and 90% of satiation. Survival and feed efficiency were not significantly different among all groups. Proximate composition of dorsal muscle was not significantly affected by feeding rate. Based on the results of this study, optimum feeding rate for growth and condition factor of adult flounder (535-928 g) seemed to be 90% of satiation.

Difference of carotenoid pigments in integuments of the wild and cultured flounder, Paralichthys olivaceus and yellowtail, Seriola quinqueradiata were studied. Total carotenoid contents in integuments of the wild and cultured flounder were 1.38mg% and 1.l6mg%, respectively. The main carotenoids in integuments of the wild flonder were zeaxanthin (19.22%), $\beta$-carotene type triol (17.80%), tunaxanthin C (17.77%), lutein (16.44%) and tunaxanthin B (13.70%). In addition, tunaxanthin A (5.42%), $\alpha$-cryptoxanthin (4.80%), astaxanthin (0.69%) and $\beta$-cryptoxanthin (0.24%) were also contained in small amounts. But in the cultured flounder, lutein (38.21%) and zeaxanthin (29.69%) were contained as main carotenoids. In addition, $\beta$-carotene type triol (7.80%), tunaxanthin C (7.05%), $\alpha$-cryptoxanthin (4.34%), tunaxanthin B (4.21%), as-taxanthin (2.40%) and $\beta$-cryptoxanthin (1.30%) were present in small amounts. Consequently, the wild flounder contained higher amounts of tunaxanthin and trios but contained lower amounts of lutein and zeaxanthin than the cultured flonder. The contents of carotenoids from integuments of wild and cultured yellow-tail were 1.08mg% and 0.09mg%. Wild and cultured yellowtail have similar carotenoid patterns, consisting of tunaxanthin C (44.11%, 43.37%), tunaxanthin B (33.56%, 29.23%) and tunaxanthin A (18.22%, 21.68%), respectively.

Currently the fish aquaculture industry of Korea is focused on the mass culture of flatfish (Paralichthys olivaceus) and and rockfish (Sebastes schlegeli) with completely controlled culture techniques. Recently, there has been considerable interest in new species development, such as the starry flounder (Platichthys stellatus). The value of starry flounder (Platichthys stellatus) as a raw fish increases with time because it is tasty, light, and bouncy. In this paper, the growth characteristics dependent on the LED wavelengths and the optimal growth conditions of the starry flounder were studied. In these experiments 4 different kinds of LED lighting, configurations were designed and prepared using red, green, blue and white, respectively. The fish aquaculture experiments were conducted over 10 weeks in four fish tanks, each installed with a different color of LED lighting. 10 starry flounders of 13 ~ 17g were placed into each tank. The effects of each color of light on the growth rate of the starry flounders were then examined. As a result, the starry flounders under the green LED lighting showed the highest growth rate, followed by the white, red, and blue LED lighting. Based on these results, a green light provides a suitable breeding environment for the starry flounder.
