This study was carried out to investigate the potential use of Zanthoxylum schinifolium and Zanthoxylum piperitum A.P. DC. as a source of antimicrobial agents against food borne pathogens. Essential oils of Z. schinifolium and Z. piperitum A.P. DC. were collected by steam distillation and analyzed by GC-MS. The antimicrobial activity of the essential oils was examined using the agar diffusion and micro-dilution assays. The effectiveness of Z. schinifolium essential oil was greater against Bacillus cereus, Staphylococcus aureus, and Vibrio parahaemolyticus than other pathogens, and the minimal inhibitory concentration (MIC) values were 1.25, 2.5, and 1.25, 2.5, and $1.25\;{\mu}g/mL$, respectively. Z. piperitum A.P. DC. essential oil was the most effective against all pathogens tested except for Escherichia coli O157:H7, and the MIC values against B. cereus, Salmonella choleraesuis, and V. parahaemolyticus were 1.25, 2.5, and $1.25\;{\mu}g/mL$, respectively. Limonene, the major component of Z. piperitum A.P. DC. essential oils, had the highest inhibitory activity toward V. parahaemolyticus with a MIC value of $0.15\;{\mu}g/mL$. Meanwhile, citronellal and geranyl acetate, major components of both essential oils, displayed antibacterial activity against only B. cereus with MIC values of 1.25 and $5\;{\mu}g/mL$, respectively. Therefore, these essential oils could be useful as antimicrobial agents against foodborne pathogens.
Objectives: The pathogenic Vibrios genus denotes halophilic bacteria that are distributed in aquatic environments, including both sea and freshwater. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are the most important species since they can be potent human pathogens and leading causes of septicemia, wound infections, and seafood borne gastroenteritis. The recent emergence of a potential pandemic clone, V. cholera serotype O1 and the cholera outbreak in South Korea in 2016 indicates the importance of consistent surveillance of pathogenic Vibrio genus within coastal areas. Methods: The present study was undertaken to determine where and how vibrios live in the aquatic environment adjacent to coastal areas of South Korea. For this survey, a total of 838 samples were obtained at 35 different sites in South Korean coastal areas during the period from January 2016 to December 2016. Pathogenic vibrios was determined using the real-time PCR method, and its clones were isolated using three selective plating media. We also monitored changes in seawater and atmospheric temperature, salinity, turbidity, and hydrogen ion concentration at the collection points. Results: The total isolation rates of V. vulnificus, V. cholera (non-pathogenic, non-O1, non-O139 serogroups), and V. parahaemolyticus from seawater specimens in 2016 were 14.2, 13.48, and 67.06%, respectively. Conclusions: The isolation rates of pathogenic vibrios genus showed a positive correlation with temperature of seawater and atmosphere but were negatively correlated with salinity and turbidity.

Objectives: To establish the expiration date of herbal decoction pouch, the change in antimicrobial activities was measured according to storage temperature and period. Methods: Five herbal formulas, Gyeji-tang, Jakyakgamcho-tang, Saengmaek-san, Paljung-san, and Dangguisu-san, with antimicrobial activity were selected from 51 herbal formulas commonly used in traditional Korean medical institutions. Five herbal formula decoction pouches were prepared and stored at room temperature (RT) or 4℃ from 0 to 12 weeks to evaluate the antimicrobial activity against Bacillus cereus, Listeria monocytogenes, or Vibrio parahaemolyticus. The antimicrobial activities were evaluated by the disk diffusion method (disk size: 10 mm). Results: The decoction pouches of Gyeji-tang and Jakyakgamcho-tang maintained antimicrobial activity against V. parahaemolyticus from week 0 (14 mm) to week 12 (14 mm) at both temperatures. Saengmaek-san pouch had antimicrobial activities against V. parahaemolyticus and B. cereus from week 0 (19 mm and 20 mm) to week 12 (both 18 mm), respectively. At RT and 4℃, the antimicrobial activity of Paljung-san pouch was maintained week 0 (17 mm) to week 12 (both 13 mm) against B. cereus. Against L. monocytogenes, the antimicrobial activity of Dangguisu-san pouch showed 27 mm at week 0 and 23 mm (RT) and 24 mm (4℃) at week 12, respectively. Conclusion: According to increase of the storage duration, the antimicrobial activities of 5 herbal formula decoction pouches decreased slightly but remained up to 12 weeks. These results can be used as important data to establish the expiration date of herbal formula decoction pouches.
In order to study the growth and survival of enteric pathogens causing water-borne infections in sewage, the filter-sterilized and autoclaved sewages of Dae Gu City were inoculated with Salmonella typhimuriuim, Shigella flexneri 2a, Sh. sonnei I, Vibrio eltor and V. parahaemolyticus, as test series and Escherichia coli as control. After varying periods of incubation up to 15 days at $4^{\circ}$, $15^{\circ}$, $25^{\circ}$ and $37^{\circ}C$, viable cells in the inoculated sewages were counted by colony count technique. Distilled water and 0.9% saline were subjected to inoculation of the organisms was observed in the filter-sterilized and autoclaved sewages at $4^{\circ}$ and the sewages became sterile within a few days. At $15^{\circ}$, no growth and rapid inactivation of the organisms in the filter-sterilized sewage and slight or no growth in the autoclaved sewage was noted. Some viable cells were found in the autoclaved sewage after 15 days. A considerable growth was observed in the filter-sterilized and autoclayed sewages, at $25^{\circ}$ and $37^{\circ}$, and large numbers of viable cells were found even after 15days of incubation. In general, the autoclaved sewage supproted the growth more noticeably than the filter-sterilized, except for V.parahaemolyticus which grew well in filter-sterilized sewage. No marked difference was noted between incubations at $25^{\circ}$ and $37^{\circ}$, but V. parahaemolyticus showed a slightly more active growth at $25^{\circ}$ than at $37^{\circ}$. Distilled water inactivated the organisms within a few days, but saline supported the growth at $25^{\circ}$ and $37^{\circ}$. Marked differences were noted in the survival test of sewages pathogens of different origins.
This research has been performed by our own investigation, also cooperated with Health and Sanification Division and each of district offices in Busan metropolitan city. After choosing, we collected water samples five times for microbiological examination. As a result of investigating in 160 water samples from urban areas, we could detect 88 cases of Vibrio spp. Furthermore, there were four cases exceeding the acceptable limit of aquarium water (100,00/mL) and another four cases exceeding the limit of Coliform group (1,000 below/100 mL). As a result of investigating that we performed for 271 cases of water samples from coastal areas from April to November, we could detect 130 cases of vibrio species and 10 cases of Coliform group. After performing 17 kinds of antibiotic susceptibility test for 41 cases of isolated Vibrio parahaemolyticus, 27 cases showed tolerance to Amplicllin (AM), all of 31 cases showed intermediate resistance only to Cefazolin (CF) but had sensitivity to the rest of them. As a result of performing antibiotic susceptibility test, serum test and PFGE gene analysis on each 10 pair of Vibrio parahaemolyticus detected concurrently from intake-pipe water and, aquarium water, we couldn't get data showing that they are clearly same species in three kinds of test. In addition, UV sterilization, Ozonization and so on. Based on our research, intake pipe didn't have a problem with microbiological contamination so we are sure that the germ came from supplied fish had caused that kind of contamination. For effective management, UV sterilization or Ozonization which can be handled consistently should be adopted in aquarium.
In order to develop a natural food preservative, freeze dried plantain (Plantago asiatica L.) was extracted with several solvents, and the antimicrobial activity was investigated. The methanol extract exhibited antimicrobial activites against five strains of bacteria such as Bacillus subtilis, Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, and Vibrio parahaemolyticus. The methanol extract at the concentration of 1.0 mg/ml completely inhibited the growth of B. subtilis and V. parahaemolyticus. Antimicrobial activity of the ethylacetate fraction from the methanol extract was the strongest compared with those of other solvent fractions such as n-hexane, chloroform, n-butanol and water. The ethylacetate fraction showed the inhibitory effect at the concentration of 0.5 mg/disc on the growth of B. subtilis and V. parahaemolyticus.
The aim of this study was to investigate the effects of environmental factors such as temperature, salinity, turbidity and pH on the growth of pathogenic Vibrios. In this study, we was obtained the samples from 2 different sites of the Incheon coastal area between January 2012 and December 2012. The water temperature in August and September was the high. the Incheon port changes the width of a small, wherease in the case of Hanjin harbor of changes of larger width. Salinity and turbidity showed significant differences, whereas temperature and hydrogen ion concentration was not significant. Pathogenic vibrios was determined using the real-time PCR method. Pathogenic vibrios in the Incheon port and Hanjin harbor were detected in 11 samples (91.67%) and 9 samples (75.0%) of Vibrio cholerae, 7 samples (58.3%) and 6 samples (50.0%) of V. vulnificus, 10 samples (83.3%) and 12 samples (100.0%) of V. parahaemolyticus, respectively. Pathogenic Vibrio bacteria were the highest at $26.8^{\circ}C$ of seawater in August. Quantitative results were the following: 102 $cell/m{\ell}$ in Vibrio cholerae, 7.876 $cell/m{\ell}$ in V. vulnificus, and 503.4 $cell/m{\ell}$ in V. parahaemolyticus, respectively. The enumeration of pathogenic vibrios showed a positive correlation with temperature and pH, but was negatively correlated with salinity and turbidity.
Vibrio parahaemolyticus is a halophilic bacterium associated with seafood gastroenteritis. An unusual strain of Kanagawa-positive urease producing Vibrio parahaemolyticus O1:K1 was isolated from the environment and identified . A polymerase chain reaction assay revealed that this strain harbored both the tdh and the genes. The urease from this strain was studied. Maximum urease production was induced in LB medium containing 0.2% urea, 0.5% glucose, 2% NaCl and pH 5.5 with 6h of culti-vation at 37$\^{C}$ under aeration. Purification of urease was achieved by the process of whole cell lysate, 65% ammonium sulphate precipitation, DEAE-cellulose ion exchange column chromatography, Sepharose CL-6B gel filtration and oxirane activated Sepharose 6B-urea affinity chromatography with 203 fold purification and 2.2% yield. Analysis of the purified enzyme by SDS-PAGE demonstrated the presence of the subunits with a molecular weight of 85kDa, 59kDa, 41kDa and the molecular weight for the native enzyme by nondenaturing PAGE and gel filtration chromatography was 255kDa. The purified urease was stable at pH 7.5 and the opeimal pH in HEPES buffer was 8.0 The enzyme was stable at 60$\^{C}$ for 2 h with a residual activity of 32% . The addition of 10$\mu$M if NiCl$_2$maintained stability for 30 min. The Km value of the purified enzyme was 35.6 mM in urea substrate. The TD$\_$50/(median toxic dose) of the purified urease was 2.5$\mu\textrm{g}$/ml on human leukemia cells.

The antimicrobial resistance patterns to the 10 antimicrobial agents of potential pathogenic vibrios isolated from seawater and a variety of farmed fishes, including oliver flounder (Payalichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagyus major) and sea bass (Lateolabyax japonicus), were investigated from May to October, 2004. A total of 314 strains of the genus vibrios were isolated from 126 collected samples, and the number of isolated strains of Vibrio payahaemolyticus and Vibrio alginolyticus were 194 and 120, respectively. Apparently $98.5\%$ of V. parahaemolyticus and $100\%$ of V. alginolyticus isolates demonstrated antimicrobial resistance against at least one antimicrobial agent. The resistance of V. parahaernolyticus isolates to ampicillin ($97.9\%$) was highest, followed by oxolinic acid ($26.8\%$), amikacin ($19.1\%$) and tetracycline and amoxicillin/clavulanic acid ($6.7\%$). V. alginolyticus isolates were resistant to ampicillin ($100\%$), solfamethoxazoleit, imethopenem ($25\%$), amikacln ($21.7\%$), amoxicillin/clavulanic acid ($15.8\%$), ciprofloxacin ($13.3\%$), and tetracycline and doxycycline ($11.7\%$). The rate of multiple antimicrobial resistance to at least four antimicrobials was higher in the V. alginolyticus isolates ($20.8\%$) than in the V. parahaemolyticus ($6.7\%$).
