Protease-Activated Receptor 2 Is Involved in Th2 Responses against Trichinella spiralis Infection

  • In order to get a better understanding of the role of protease-activated receptor 2 (PAR2) in type 2 helper T (Th2) cell responses against Trichinella spiralis infection, we analyzed Th2 responses in T. spiralis-infected PAR2 knockout (KO) mice. The levels of the Th2 cell-secreted cytokines, IL-4, IL-5, and IL-13 were markedly reduced in the PAR2 KO mice as compared to the wild type mice following infection with T. spiralis. The serum levels of parasite-specific IgE increased significantly in the wild type mice as the result of T. spiralis infection, but this level was not significantly increased in PAR2 KO mice. The expression level of thymic stromal lymphopoietin, IL-25, and eotaxin gene (the genes were recently known as Th2 response initiators) of mouse intestinal epithelial cells were increased as the result of treatment with T. spiralis excretory-secretory proteins. However, the expression of these chemokine genes was inhibited by protease inhibitor treatments. In conclusion, PAR2 might involve in Th2 responses against T. spiralis infection.

  • Partition chromatography의 방법(方法)으로는 우선 cellulose powder colume을 시도해 보았으나 이는 얻어진 fraction의 용매제거의 난점(難點)이 있으므로 결국(結局) paper chromatography 방법(方法)만을 사용(使用)하였다. 연구결과(硏究結果)로는 우선 갈색색소의 fraction에 있어서 발효초기(醱酵初期)에 약간의 활성(活性)을 관찰(觀察)할수 있었으며 glycine이 함유된 fraction 에서는 발효초기에 벌써 상당한 활성을 보였으며 이는 또한 발효의 진행(進行)에 따라 점차 증가되었다. 그리고 N-glycoside의 fraction에서는 발효초기에 미약(微弱)한 활성(活性)을 관찰할 수 있었다. 그러나 paper chromatography의 방법(方法)은 본실험(本實驗)에 적합하지 않다고 생각되며 그 이유(理由)는 일회(一回)에 fractionation 할수있는 시료(試料)의 양(量)이 극(極)히 제한되어 있다는 점(點)과 또 fractionation 도중에 타물질(他物質)의 오염으로 인(因)해서 시료(試料)의 정확(正確)한 발효 활성(活性)을 측정(測定)할수 없다는 것이다.

  • 본(本) 실험(實驗)에 사용(使用)된 Gel로는 Sephadex G-200, G-100과 G-75로써 fractionation은 대체(大體)로 호조건에서 이루어 졌다. G-200과 G-100의 경우에 있어서는 갈색색소의 분자크기가 서로 다른 2개(個)의 Peak를 얻을 수 있었고 Colume에 투입된 시료(試料)는 완전(完全)히 회수(回收)되었다. G-75의 경우에는 갈색색소와 분자 크기가 작은 다른 물질(物質)과를 분리(分離)할 수 있었으나 약간의 시료(試料)는 Gel에 흡착되어 완전(完全)히 회수(回收)할 수 없었다. 발효실험의 결과를 종합해 보면 우선 갈색색소를 함유하늘 모든 fraction은 발효초기에 활성(活性)을 보이며 분자크기가 작은 물질의 구획(區劃)에서는 강하고도 점증되는 활성(活性)을 관찰할 수 있었다. Sephadex Gel에 의(依)한 fractionation은 첫째로 작업(作業)도중에 시료(試料)의 변질현상(變質現象)이 없다는 것과 둘째로 갈색색소를 그들의 분자크기에 따라 분획(分劃)할 수 있다는 이점(利點)들이 있다.

  • 본 연구에서는 집먼지 진드기 추출물은 호중구에 단독으로 작용하는 것보다, 림프구와 호중구의 공동배양에서 호중구의 세포고사를 더 억제시켰다. 집먼지 진드기는 알레르기 질환의 림프구에서 IL-6, IL-8, MCP-1, GM-CSF의 분비를 증가시켰다. 집먼지 진드기에 의해 증가된 사이토카인은 protein kinase C ${\delta}$의 억제제인 rottlerin과 p38 MAPK의 억제제인 SB202190에 의해서 감소하였다. 집먼지 진드기에 의해 활성화된 p38 MAPK은 protease-activated receptor (PAR2)의 억제제, rottlerin, SB202190에 의해서 억제되었다. Serine protease 억제제인 aprotinin과 cysteine protease 억제제인 E64은 림프구의 사이토카인의 증가와 관련이 없었다. 또한 집먼지 진드기에 의해 증가된 사이토카인의 변화는 천식과 알레르기 비염 환자에서 차이가 없었다. 림프구에서 집먼지진드기에 의해서 분비되는 분자들은 호중구의 유주운동을 억제시켰다. 본 연구를 통하여 집먼지진드기에 의해 유발되는 알레르기 질환의 병인기전을 규명하는데 유용한 결과가 될 것이다.

Molecular Cloning of the DNA Gyrase Genes from Methylovorus Sp. Strain SS1 and the Mechanism of Intrinsic Quinolone Resistance in Methylotrophic Bacteria

  • The genes encoding the DNA gyrase A (GyrA) and B subunits (GyrB) of Methylovorus sp. strain SS1 were cloned and sequenced. gyrA and gyrB coded for proteins of 846 and 799 amino acids with calculated molecular weights of 94,328 and 88,714, respectively, and complemented Escherichia coli gyrA and gyrB temperature sensitive (ts) mutants. To analyze the role of type II topoisomerases in the intrinsic quinolone resistance of methylotrophic bacteria, the sequences of the quinolone resistance-determining regions (QRDRs) in the A subunit of DNA gyrase and the C subunit (ParC) of topoisomerase IV (Topo IV) of Methylovorus sp. strain SS1, Methylobacterium extorquens AM1 NCIB 9133, Methylobacillus sp, strain SK1 DSM 8269, and Methylophilus methylotrophus NCIB 10515 were determined. The deduced amino acid sequences of the QRDRs of the ParCs in the four methylotrophic bacteria were identical to that of E. coli ParC. The sequences of the QRDR in GyrA were also identical to those in E. coli GyrA except for the amino acids at positions 83, 87, or 95. The $Ser^{83}$ to Thr substitution in Methylovorus sp. strain SS1, and the $Ser^{83}$ to Leu and $Asp^{87}$ to Asn substitutions in the three other methylotrophs, agreed well with the minimal inhibitory concentrations of quinolones in the four bacteria, suggesting that these residues play a role in the intrinsic susceptibility of methylotrophic bacteria to quinolones.

Transformation of an Alkalin Protease Overproducer, Vibrio metschnikovii Strain RH530, and Improvement of Plasmid Stability by the par Locus

  • Vibrio metschnikovii strain RH530 is a non-pathogenic, industrially-important alkaline protease producer which has been isolated from wastewater. In this paper, we report on the transformation of this strain by using the method of electroporation. A field strength of $7.5\;kVcm^{-1}$ and $25\;{\mu}F$, and using a 0.2-cm cuvette, appeared to be the optimal conditions for electroporation of the cells with the recombinant pSBCm plasmid carrying the vapK alkaline protease gene and the ColE1 replicon. Cells were subjected to osmotic shock in order to remove extracelluar DNase, and adding 200 mM of sucrose to electroporation buffer cells showed an increased transformation efficiency. Maximum efficiency of transformation was obtained at an early exponential growth phase. Using all of the conditions mentioned above, we routinely obtained a transformation efficiency of more than $10^4{({\mu}g\;plasmid\;DNA)}^{-1}$. The stability of the plasmid pSBCm in V. metschnikovii RH530 was 25% after 18h of growth (27 generations) in the medium without antibiotic selection. The insertion of the par locus to the pSBCm increased the stability of the plasmid up to 42% without selective pressure. The increase in plasmid stability was accompanied by the increase in the productivity of alkaline protease in the recombinant V. metschnikovii strain RH530. Determining optimal conditions for the transformation of the industrially-important, nonpathogenic Vibrio strain, and the improvement of plasmid stability by introducing the par locus into the high copy number plasmid vector, will allow the development of procedures involved in the genetic manipulation of this strain, particularly for its use in the production of industrial enzymes such as alkaline protease.

  • Comment faciliter l'apprentissage du français pour rendre notre enseignement plus efficace? Beaucoup de recherches ont ${\acute{e}}t{\acute{e}}$ $men{\acute{e}}es$ sur cette question; en $r{\grave{e}}gle$ $g{\acute{e}}n{\acute{e}}rale$ l'enseignement de la langue d'une part et l'expression ${\acute{e}}crite$ et la $r{\acute{e}}flexion$ d'autre part ${\acute{e}}taient$ $men{\acute{e}}es$ $ind{\acute{e}}pendamment$. Notre intention dans ce $pr{\acute{e}}sent$ travail a ${\acute{e}}t{\acute{e}}$ d'introduire et $d^{\prime}e xp{\acute{e}}rimenter$ pendant $l^{\prime}ann{\acute{e}}e$ scolaire des cours associant le $d{\acute{e}}bat$ $d^{\prime}id{\acute{e}}e$ et la $r{\acute{e}}flexion$ $th{\acute{e}}matique$ avec l'apprentissage linguistique(grammaire, vocabulaire${\dots}$), afin de susciter $l^{\prime}i nt{\acute{e}}r{\hat{e}}t$ des apprenants, leur $curiosit{\acute{e}}$ et leur $vivacit{\acute{e}}$, au $del{\grave{a}}$ des $d{\acute{e}}fis$ $pos{\acute{e}}s$ par la langue française. Nous avons en particulier $utilis{\acute{e}}$ une $publicit{\acute{e}}$ comme support d'apprentissage des bases linguistiques et comme moteur $d^{\prime}{\acute{e}}veil$ ${\grave{a}}$ la civilisation française pour mobiliser leur attention et stimuler leur $activit{\acute{e}}$ cognitive par $l^{\prime}interdisciplinarit{\acute{e}}$. Nous avons donc $expos{\acute{e}}$ ici les exemples d'argumentation autour les deux $s{\acute{e}}quences$ publicitaires conduites avec les ${\acute{e}}tudiants$ et les arborescences $d{\acute{e}}gag{\acute{e}}es$ de ces analyses. Presque concomittamment ${\grave{a}}$ "voir" et "${\acute{e}}couter$", l'information est $trait{\acute{e}}e$ par $l^{\prime}activit{\acute{e}}$ "$d{\acute{e}}coder$". Nous approfondissons ensemble cette $interpr{\acute{e}}tation$ $s{\acute{e}}miotique$, et l'analyse de la façon dont la $publicit{\acute{e}}$ capte l'attention du spectateur nous $am{\grave{e}}ne$ ${\grave{a}}$ $d{\acute{e}}gager$ des faits de $soci{\acute{e}}t{\acute{e}}$ et ${\grave{a}}$ y $r{\acute{e}}fl{\acute{e}}chir$; comment, avec l'exemple de la $publicit{\acute{e}}$ jouant la musique $d{\acute{e}}licieuse$ de la gourmandise, nous entrevoyons la $r{\acute{e}}alit{\acute{e}}$ obscure de $l^{\prime}ob{\acute{e}}sit{\acute{e}}$ infantile. Ces approches interdisciplinaires ${\grave{a}}$ partir du multi-$m{\acute{e}}dia$ $r{\acute{e}}pondent$ ${\grave{a}}$ la $n{\acute{e}}cessit{\acute{e}}$ de distance critique que requiert la $soci{\acute{e}}t{\acute{e}}$ contemporaine. $L^{\prime}{\acute{e}}tude$ d'une $s{\acute{e}}quence$ publicitaire ne permet certes pas $d^{\prime}appr{\acute{e}}hender$ la $soci{\acute{e}}t{\acute{e}}$ française dans sa $globalit{\acute{e}}$, mais en $consid{\acute{e}}rant$ que notre travail consiste tout autant ${\grave{a}}$ stimuler la $facult{\acute{e}}$ critique $qu^{\prime}{\grave{a}}$ favoriser l'apprentissage linguistitique, notre $strat{\acute{e}}gie$ nous permet de $r{\acute{e}}aliser$ plusieurs objectifs autour d'un $m{\hat{e}}me$ axe d'enseignement.

Evaluation of Mulberry Germplasm (Morns spp.) for Leaf Yield and Quality through Bioassay

  • Twenty - four elite mulberry germplasm each of indigenous and exotics were studied for their leaf yield and compared with commercial check ($V_1$ and Kosen). Accession MI-0416 and ME-0169 out yielded the check accession in leaf yield/plant. The other few mulberry germplasm were also performed at par with the checks. For quality test and bioassay were conducted with the leaves of selected mulberry germplasm. Among the selected twelve mulberry accessions used for bioassay, MI-0376 and ME-033 performed better than check ($V_1$, Kosen). Other mulberry accessions i.e., MI-0310 and MI-0437 are on par with the check as far as the bioassay is concerned. MI-0376 and ME-0033 out yielded in rearing parameters and qualified for 11 and 10 rearing and related traits. Other mulberry accessions i.e., MI-0310 and MI-0437 were also qualified for eight rearing traits along with check ($V_1$). The mulberry accessions tested after selection from the preliminary characterization seems to be better and equally good in rearing and leaf yield compared to check ($V_1$, Kosen), which provides scope for selection and further evaluation. The selected mulberry accession may be included in crop improvement programme.

윤영선 희곡 연구: 죽음과 애도의 글쓰기 (Une étude sur les textes théâtrals de Young-Sun Yun: L'écriture théâtrale sur la mort et le deuil)

  • Cette ${\acute{e}}tude$ est pour l'objectif de rechercher sur $l^{\prime}{\acute{e}}criture$ $th{\acute{e}}{\hat{a}}trale$ de la mort et le deuil dans les $th{\acute{e}}{\hat{a}}tres$ chez Young-sun Yun(1954-2007) ${\grave{a}}$ travers des textes dramatiques ${\acute{e}}crits$ par $lui-m{\hat{e}}me$, qui ${\acute{e}}tait$ aussi le metteur en $sc{\grave{e}}ne$ depuis 1994 dans le $th{\acute{e}}{\hat{a}}tre$ contemporain en $Cor{\acute{e}}e$. Cette $th{\grave{e}}se$ est $commenc{\acute{e}}$ ${\grave{a}}$ ${\acute{e}}crire$ pas pour apprendre ses textes mais pour comprendre la ralation entre les textes et les ${\exp}{\acute{e}}riences$ de l'auteur. Ici, pour cela, j''ai $analys{\acute{e}}$ ses 4 $pi{\grave{e}}ces$ $th{\acute{e}}{\hat{a}}trales$ produites ${\grave{a}}$ cette ${\grave{e}}poque$, dont (1994), (2000), $d{\acute{e}}biteur$, mon $p{\grave{e}}re$ est $progress{\acute{e}}$ de cancer>(1996), (2005), en referrant les rapports entre $l^{\prime}{\acute{e}}criture$ dramatique et de la mort et le deuil de $p{\grave{e}}re$. Cette ${\acute{e}}tude$ $pr{\acute{e}}suppose$ un certain situation psychologue de l'auteur sur la mort et le deuil de personnage '$p{\grave{e}}re$'. L'auteur dramatique et le metteur en $sc{\grave{e}}ne$ du $th{\acute{e}}{\hat{a}}tre$ Young-sun Yun propose certaines orientations de la mort de son $p{\grave{e}}re$ dans ses texts dramatiques: le sentiment de victime sous le pouvoir absolu de son $p{\grave{e}}re$. Chez lui, ${\acute{e}}crire$ le texte dramatique, c'est transmettre les amertumes d'esprit et une forme de $r{\acute{e}}flextion$ vis-${\grave{a}}$-vis de la relation avec son $p{\grave{e}}re$ surtout le $probl{\grave{e}}me$ $inachev{\acute{e}}$ de deuil. c'est-${\grave{a}}$-dire fournir des sentiments coupables et des $anxi{\acute{e}}t{\acute{e}}s$ qui $r{\acute{e}}sident$ au fond de son coeur, tout au long de sa vie. Pour cette objecif, il est ${\acute{e}}vident$ qu'il faudra toujours partir du texte et y revenir. J'ai $essay{\acute{e}}$ de ne pas $r{\acute{e}}duiser$ $l^{\prime}{\acute{e}}tude$ des textes au respect myope des paroles des personnage et rester la $r{\acute{e}}flextion$ pshchologique sur la mort et le processu de deuil. J'ai $essay{\acute{e}}$ d'appuyer mes remarques sur les sources $autoris{\acute{e}}es$: psychanalytiques en la $mati{\grave{e}}re$. C'est pourquoi j'ai cru devoir $pr{\acute{e}}senter$ cette recherche de l'analyse sous la forme d'une $synth{\grave{e}}se$ des principaux concepts de la $th{\acute{e}}orie$ de la pshchologie moderne qui permettent l'analyse des textes dramatiques.(Dany Nobus, David K. Switzer, Fritz Riemann, Giles Deleuze, Sigmund Freud, Julia Kristeva) Et auiss j'ai $essay{\acute{e}}$ de explorer l'imaginaire des paroles dans les textes en ses rapports avec l'auteur et les personnages et entre les personnages. Ainsi conçu, j'ai $analys{\acute{e}}$ les formes de paroles en ce qui concerne la mort de $p{\grave{e}}re$ et le deuil. Cette $mani{\grave{e}}re$ s'est $r{\acute{e}}v{\acute{e}}l{\acute{e}}e$ efficace pour montrer comment des traces de sentiment se transforment dans des textes. Dans les pluparts des personnages chez les textes de Young-sun Yun, on peut voir si $d{\acute{e}}finitivement$ leur cicatrices personnelles, $sensibilit{\acute{e}}s$ $m{\hat{e}}l{\acute{e}}s$, ce 'je-ne-pourrai-rien-contre-mon-$p{\grave{e}}re$' ou 'je-ne-sais-quoi-et-presque-$apr{\grave{e}}s$-la mort-de-mon pere'. Pour $d{\acute{e}}pouiller$ cet aspect, j'ai $montr{\acute{e}}$ qu'il n'y avait pas de $v{\acute{e}}rit{\acute{e}}s$ objectives, mais que la $subjectivit{\acute{e}}$ doit trouver le jeu triangulaire entre l'inconscient de l'auteur par les paroles des textes et par les rapports entre le symbolisant et le $symbolis{\acute{e}}$ dans les $repr{\acute{e}}sentation$ socio-culturelle en $Cor{\acute{e}}e$ d'aujourd'hui.

  • 우리나라 남해안, 특히 여수반도 주변의 도서들은 수많은 반도와 만, 섬 등의 자연경관으로 특별한 관심을 불러일으킨다. 이와 같은 자연경관을 지속가능한 생태관광자원으로 활용하기 위해서는 이를 잘 보존하는 노력 또한 중요하다. 현재 사도를 비롯한 여수시의 많은 도서들은 빼어난 자연경관에 비해 생태관광해설이 미비한 편이며, 자연경관 또한 염풍화와 침식으로 보존에 많은 어려움을 겪고 있다. 따라서 본 연구는 생태관광의 맥락 속에 여수시 사도일원의 자연경관에 대한 조사를 통해, 생태관광지 선정 기준에 의한 생태관광 탐방로를 개발하고 생태관광해설 프로그램을 제시함으로써 생태관광의 활성화와 지역발전에 기여하고자 한다. 사도일원은 사도의 사빈과 해식애, 중도의 공룡화 석지와 연흔 건열, 시루섬의 육계사주와 응회암층 노두 암맥, 장사도의 풍화혈과 암맥, 그리고 나끝~추도 구간의 해할 현상 등의 생태관광자원을 가지고 있으며, 전반적으로 중생대 퇴적층 및 화석과 관련된 지형들이 발달하였다. 탐방경로는 사도 선착장을 기점으로 북쪽에서 남쪽으로 구성된다.
