Thermodynamics of the Micellization of Cetylpyridinium Bromide in Some Aqueous Alcohol Solutions (몇 가지 알코올-수용액에서 Cetylpyridinium Bromide의 미셀화의 열역학적 성질)

  • The effects of added alcohols on the critical micelle concentration(CMC) of cetylpyridinium bromide(CPB) were investigated by the UV-Vis spectrophotometer at the temperature range of 8∼45$^{\circ}C$. The CMC of CPB was increased with the addition of methanol in the whole temperature region studied, while decreased with the addition of ethanol and propanol. The increase of CMC with the addition of methanol may be attributable to the increasing solvent power of the methanol-water mixture, because methanol was scarcely solubilized into the palisade layer of the micelle of CPB. The decrease of CMC with the inclusion of ethanol and propanol may be derived from the solubilization of alcohols into the micelles. On the other hand, the CMC was decreased with the temperature rise in the low-temperature region below about 25$^{\circ}C$, and the CMC was increased in the high-temperature region above that. The thermodynamic parameters (${\Delta}G_M^{\circ},\;{\Delta}H_M^{\circ},\;and\;{\Delta}S_M^{\circ}$) of the micellization of CPB were obtained in some aqueous alcohol solutions. In the whole temperature region (8∼45$^{\circ}C$), the values of ${\Delta}G_M^{\circ}$ were negative, while those of ${\Delta}S_M^{\circ}$ were positive. And in the temperature region below about 25$^{\circ}C$ the ${\Delta}H_M^{\circ}$ values were positive, while in the temperature region above that the values were negative.

A Study on Structure and Differentiation of Seed Coat of Panax ginseng C.A. Meyer (인삼(Panax ginseng C.A. Meyer) 종피의 구조 및 분화에 관한 연구)

  • Structure and differentiation mechanism of the seed coat of Panax ginseng are studied with light and electron microscopes to clarify the developmental processes of seed coat and the structural changes during the differentiation of the seed. The seed coat of ginseng is differentiated from the inner cell layers of ovary wall, which can be compared with the seed coat differentiated from integument(s) in other plants. The single integument is differentiated into endothelium, which is degenerated to one layer of 4${\mu}{\textrm}{m}$ in thickness, composed of remants of cell wall components in fully ripened seed. The ripened seed coat is composed of three layers; fringe layer, inner layer and palisade layer, and all of the them are crossed at right angles with one another. This may be the cause of protection of the kernel from other mechanical injuries. The thickness of fully ripened seed coat is about 300~600 ${\mu}{\textrm}{m}$, and arrangements of sclereids are irregular. However, the raphe region of seed coat is thin about 200 ${\mu}{\textrm}{m}$ in thickness and sclereids in that region are arranged regularly. This is the important cause for the cleavage of the seed coat during post-maturation process. The vascular bundles on the raphe are still remaining after sarcocarps are removed, and one of the branches of vascular bundles entered into the seed coat through the hilum and extended to chalazal region. During post-maturation process, the supply of water being necessary for growth of embryo may be accompolished by the vascular bundles entered into the seed coat through the opened hilum.

Solubilization of 4-Alkylbenzoic Acid Isomers by the Aqueous Solution of Tetradecyltrimethylammonium Bromide (테트라데실트리메틸암모늄 브로마이드의 수용액에서 4-알킬벤조산 이성질체들의 가용화에 대한 연구)

  • The interaction of 4-alkylbenzoic acid isomers with the micellar system of TTAB (tetradecyltrimethylammonium bromide) was studied by the UV/Vis spectrophotometric method. The solubilization constants ($K_s$) of 4-alkylbenzoic acid isomers into the TTAB micellar system and the critical micelle concentration (CMC) of TTAB have been measured with the change of temperature. Various thermodynamic parameters have been calculated and analyzed from those measurements. The results show that the values of ${\Delta}G^o{_s}$ for the solubilization of all the isomers are negative and the values of ${\Delta}H^o{_s}$ and ${\Delta}S^o{_s}$ are all positive within the measured temperature range. The effects of additives (n-butanol and NaCl) on the solubilization of 4-alkylbenzoic acid isomers have been also measured. There was a great change on the values of $K_s$ and CMC simultaneously with these additives. From these changes we can postulate that the solubilization sites of 4-alkylbenzoic acids are the core or deep palisade region of the TTAB micelle.

Solubilization of Cresol Isomers by the Cationic Surfactant of TTTAB in Aqueous Solution of n-Butanol and NaCl (n-부탄올 및 NaCl 수용액에서 양이온 계면활성제인 TTAB에 의한 크레졸 이성질체들의 가용화에 대한 연구)

  • We tried to investigate the effects of substituent position, temperature, and additives such as NaCl and n-butanol on the solubilizations of cresol isomers by tetradecyltrimethylammonium bromide (TTAB), using the UV-Vis spectrophotometric method. The measured solubilization constants (Ks) values for each cresol isomer increased in the order o-cresolo and ∆Ho values for the solubilizations of cresols were all negative values but the ∆So values were all positive values within the measured ranges. The values of ∆Go increased also with increasing the concentration of n-butanol but decreased with increasing the concentration of NaCl. From these facts, we could conclude that both the enthalpy and entropy changes contribute together for the solubilizations of cresols isomers by cationic surfactant of TTAB and they are solubilized in the polar palisade region or at the surface of micelle.

The Phenotype of the Soybean Disease-Lesion Mimic (dlm) Mutant is Light-Dependent and Associated with Chloroplast Function

  • The dlm (disease lesion mimic) mutant of soybean (Glycine max L. Merr) shows the similar lesion of a soybean disease caused by a fungus, Corynespora cassilcola. The lesion was examined at cellular and molecular level. Trypan blue staining result indicated that cell death was detectable in the entire region of leaves excluding veins when the lesions had already been developed. We found that the mesophyll cells of palisade layer in the dim mutant appeared to be wider apart from each other. The chloroplasts of the dim mutant cells contained bigger starch granules than those in normal plants. We also found that the lesion development of dlm plant was light-dependent and the starch degradation during the dark period of diurnal cycle was impaired in the mutant. Three soybean pathogenesis-related genes, PR-1a, PR-4, and PR-10, were examined for their expression patterns during the development of disease lesion mimic. The expression of all three genes was up-regulated to some extent upon the appearance of the disease lesion mimic. Although the exact function of DLM protein remains elusive, our data would provide some insight into mechanism underling the cell death associated with the dim mutation.

Thermodynamic Study on the Solubilization of p-Halogenated Phenol Derivatives in TTAB Solution (TTAB 수용액에서 p-할로겐화 페놀유도체들의 가용화에 대한 열역학적 연구)

  • The interaction of p-halogenated phenol derivatives with the micellar system of tetradecyltrimethylammonium bromide (TTAB) was studied by the UV/Vis spectrophotometric method. Effects of substituents, additives, and temperatures on the solubilization of phenol derivatives have been measured. The results show that all the values of ${\Delta}G^o$ and ${\Delta}H^o$ were negative and the values of ${\Delta}S^o$ were positive for all phenol derivatives within the measured temperature range. The calculated thermodynamic parameters depended on the size, the electro-negativity, and the hydrophobic property of halogen substituents. The addition of n-butanol results in the decrement in tthe Ks values and the addition of NaCl caused the increment in the Ks values for all the phenol derivatives. From these changes we can postulate that the solubilization sites of the phenol derivatives in the micelle depend severely on properties of the halogen-substituent.

Structural Characteristics of Vascular Tissue in Carrot Seedlings with Anomalous Cotyledon Developed from Somatic Embryos (당근의 체세포배로부터 발생한 이상자엽 유식물의 유관속 조직의 구조적 특성)

  • The somatic embryos of Daucus carota L. cv. Hongshim developed in MS basal medium far 4 weeks had varied number of cotyledons. Palisade and spongy parenchyma of cotyledon were not clearly discriminated in all seedlings developed from the embryos. No independent existence of collateral vascular bundle was observed in all seedlings with various types of cotyledon ; instead, vascular bundles were either interconnected or partially connected with one another. Most of the cotyledonary bases on hypocotyl showed short cylinder structure which encircle plumule. The vascular tissues of cotyledonary bases and nodes of seedlings with jar-shaped or 1 cotyledon were connected in ring forms, showing the pattern of ectophloic shiphonostele, and similar ring form structure was also found in the vascular arrangement of 5 cotyledon seedlings. The vascular bundles of seedlings with 2, 3 and 4 cotyledons in many cases had independently arranged within the cotyledonary bases and nodes, showing the pattern of eustele. In hypocotyl, tetrarch or hexarch xylems prevailed in seedlings with jar-shaped cotyledon or 1 and 5 cotyledon; tetrarch xylems prevailed in 2 cotyledon seedlings; and triarch xylems prevailed in 3 cotyledon seedlings. In most of seedlings, cortex vascular bundles were dispensed in the region from cotyledonary node to hypocotyl, but double vascular bundles were also observed occasionally. In roots, diarch xylems were observed in most of seedlings with 2 cotyledons, triarch xylems in half of seedlings with 3 cotyledons, and diarch xylems in most of the remaining seedlings with the occasional occurrences of tetrarch xylems.
