pH- and Temperature-Sensitive Bifunctional Hydrogels of N-Isopropylacrylamide and Sulfadimethoxine Monomer

  • pH- and temperature-sensitive bifunctional hydrogels composed of N-isopropylacrylamide (NiPAAm) and a sulfadimethoxine monomer (SDM) derived from sulfadimethoxine were prepared. These hydrogels exhibit simultaneous pH- and temperature-induced volume-phase transitions. The pH-induced volume-phase transition behavior is produced by the ionization/deionization of SDM and is very sharp. In the high pH region, the ionization of SDM induces swelling of the hydrogels. In the low pH region, the deionization of SDM induces deswelling of the hydrogels. The temperature-induced volume-phase transition behavior of the bifunctional hydrogels exhibits negative thermosensitivity because of the NiPAAm component. The hydrogels swell even at low pH as the temperature decreases. The hydrogels swell at low temperature and high pH, and deswell at high temperature and low pH. The volume of the hydrogels dependl on the balance of the swelling and deswelling produced by the two competing stimuli, pH and temperature.

Characterization of a pH/Temperature-Sensitive Hydrogel Synthesized at Different pH and Temperature Conditions (pH/온도-동시 민감성 Hydrogel의 합성조건에 따른 특성 연구)

  • A hydrogel, poly(N-isopropylacrylamide-co-N, N-dimethylaminopropylmethacrylamide), sensitive to both pH and temperature, was synthesized and characterized at $^13∼23{\circ}C$ and pH of 10.3∼12.3. The gel was more transparent and mechanically stronger at lower preparation temperature and pH. Large pores observed in scanning electron microscope seem to be responsible for the lower biomolecular separation efficiency. The lower critical solution temperature (LCST) decreased at a higher polymerization temperature. At $25^{\circ}C$, which is lower than the LCST, the gel was swollen regardless of the solution pH. At $40^{\circ}C$, however, the gel was swollen at neutral and acidic pHs even though the temperature was higher than the LCST. The gel collapse pH, defined as the point at which the gel made its largest volume decrease per unit pH increment, increased as the gel preparation temperature increased.

  • A hydrogel, poly(N-isopropylacrylamide-co-N,N'-dimethylaminopropyl methacrylamide), sensitive to both pH and temperature was synthesized and characterized for its welling behavior, lower critical solution temperature (LCST), and its appearance. The hydrogel with 5 mol% of N,N'-diemthylaminopropyl methacrylamide (DMAPMAAm) increased its volume phenomenonally in a lower pH range (ph 1~8) even at temperature ($37^{\circ}C$ and $40^{\circ}C$) higher than LCST. This behavior was unique compared to the temperature -sensitive hydrogel which did not exhibit any swelling in the same pH range. The hydrogel with 20 mol% of DMAPMAAm was swollen significantly at a higher pH of 12. With pH decrease from 12 to 2 water content in the gel increased from 38.8 wt% and 60.6 wt%, and 90.8 wt% for 5 mol% and 20 mol% gel, respectively. The transition pH that pH effect overwhelmed temperature effect occurred at a lower pH for a higher temperature ($40^{\circ}C$) and a lower mol% (5 mol%) of DMAPMAAm. Transparency and LCST of the gel increased with higher DMAPMAAm mol%.

Synthesis and pH-Dependent Micellization of a Novel Block Copolymer Containing s- Triazine Linkage

  • Novel pH-sensitive moieties containing an s-triazine ring were synthesized with sulfonamide and secondary amino groups. The synthesized pH-sensitive moieties were used for the synthesis of a pH-sensitive amphiphilic ABA triblock copolymer. The pH-sensitive triblock copolymer was composed of diblock copolymers, methoxy poly(ethylene glycol)-poly ($\varepsilon$-caprolactone-co-D,L-lactide) (MPEG-PCLA), and pH-sensitive moiety. These copolymers could be dissolved molecularly in both acidic and basic aqueous media at room temperature due to secondary amino and sulfonamide groups. The synthesized s-triazine rings containing pH-sensitive compounds were characterized by ${^1}H-NMR,\;{^13}C-NMR$, and LC/MSD spectral data. The synthesized diblock and triblock copolymers were also characterized by ${^1}H-NMR$ and GPC analyses. The critical micelle concentrations at various pH conditions were determined by fluorescence technique using pyrene as a probe. Furthermore, the micellization and demicellization study of the triblock copolymer was done with pH-sensitive groups. The sensitivity towards pH change was further established by acid-base titration.

Synthesis and Characterization of Temperature and pH Sensitive Graft Copolymers Based on Pluronic (Pluronic을 기초로 한 온도와 pH에 민감한 그래프트 공중합체의 합성과 특성)

  • Temperature and pH sensitive graft copolymers [Pluronic-$g$-poly(NIPAAm-$co$-MMA), Polymer A] and [Pluronic-$g$-poly( NIPAAm-$co$-MAA), Polymer C] were synthesized by macro radical graft polymerization with $N$-isopropylacrylamide (NIPAAM)/$N,N$-diethylaminoethylmethacrylate (DEAEMA) and $N$-isopropylacrylamide (NIPAAm)/methacrylic acid (MAA) based on Pluronic, respectively. The chemical structure and molecular weight of the graft copolymers was characterized by $^1H$ NMR and gel permeation chromatography. The aqueous solution properties of graft copolymers were measured using a UV-visible spectrophotometer, contact angle and dynamic light scattering equipment with different temperature and pH conditions. The obtained graft copolymers showed a very sensitive phase transition in response to temperature and pH in aqueous media which suggested that the amine group of DEAEMA segment and carboxylic group of MAA had a great influence on the lower critical solution temperatures (LCST) in Polymer A and C, respectively. The graft copolymers can be utilized for drug delivery system and molecular switching applications where responses to temperature and pH changes are relevant.

  • Poly (ethylene glycol)(PEG) - Poly (${\varepsilon}-caprolactone(CL)$) - Poly (D,L lactide(LA) (PCLA-PEG-PCLA) was synthesized by ring-opening polymerization to form temperature sensitive hydrogel triblock copolymer. The triblock copolymer was acrylated by acryloyl chloride. ${\beta}-amino$ ester was used as a pH sensitive moiety, in this study ${\beta}$- amino ester obtained from 1,4-butandiol diacrylate, and 4, 4' trimethylene dipiperidine, it have pKb around 6.6. pH/temperature sensitive penta-block copolymer (PAE-PCL-PEG-PCL-PAE) was synthesized by addition polymerization from acrylated triblock copolymer, 1,4-butandiol diacrylate, and 4, 4' trimethylene dipiperidine. Their physicochemical properties of triblock and penta-block copolymers were characterized by $^1H-NMR$ spectroscopy and gel permeation spectroscopy. Sol-gel phase transition behavior of PAE-PCL-PEG-PCL-PAE block copolymers were investigated by remains stable method. Aqueous media of the penta-block copolymer (at 20 wt%) changed from a sol phase at pH 6.4 and $10^{\circ}C$ to a gel phase at pH 7.4 and $37^{\circ}C$. The sol-gel transition properties of these block copolymers are influenced by the hydrophobic/hydrophilic balance of the copolymers, block length, hydrophobicity, stereo-regularity of the hydrophobic of the block copolymer, and the ionization of the pH function groups in the copolymer depended on the changing of environmental pH, respectively. The degradation and the stabilization at pH 7.4 and $37^{\circ}C$, and the stabilization at pH 6.4 and $10^{\circ}C,\;5^{\circ}C,\;0^{\circ}C$, of the gel were determined. The results of toxicity experiment show that the penta block copolymer can be used for injection drug delivery system. The sol?gel transition of this block copolymer also study by in vitro test ($200{\mu}l$ aqueous solution at 20wt% polymer was injected to mouse). Insulin loading and releasing by in vitro test was investigated, the results showed that insulin can loading easily into polymer matrix and release time is around 14-16days. The PAE-PCL-PEG-PCL-PAE can be used as biomaterial for drug, protein, gene loading and delivery.

  • A new and useful poly(ethylene oxide)-based pH-sensitive block copolymer is introduced. Poly(ethylene oxide-b-N-phenylmaleimide) was first synthesized by anionic polymerization of N-phenylmaleimide (N-PMI) using mixed alkali metal polymeric alkoxide by sequential monomer addition method in the mixture of benzene/THF/DMSO (10/5/3, v/v/v) at room temperature. Reductive deimidation of the resulting block copolymer was performed using hydrazine monohydrate leading to the formation of the corresponding pH-sensitive poly(ethylene oxide-b-maleic acid).

  • A series of pH and temperature-responsive (N,N-diethylacrylamide-co-methylacrylic acid) copolymers were synthesized by radical copolymerization and characterized by elemental analysis, Fourier-transform infrared (FT-IR), nuclear magnetic resonance (NMR) $^1H$, $^{13}C$ and LLS. The effects of salt and pH on the phase transition behaviors of the copolymers were investigated by uv. With increasing NaCl concentration, significant salt effects on their phase transition behaviors were observed. UV spectroscopic studies showed that the phase transition became faster with increasing NaCl concentration. In addition, the phase transition behaviors of copolymers were sensitive to pH. The pH and temperature sensitivity of these copolymers would make an interesting drug delivery system.

  • Properties of purified isocitrate lyase from Saccharomycopsis lipolytica ATCC 44601 and MX9-11RX8 temperature-sensitive mutant were investigated. Purified isocitrate lyase from temperature-sensitive mutant was indistinguishable from the wild type enzyme with respect to the isoelectric pH(5.3), the thermostability and Km value for threo-Ds-Isocitrate(about 0.2 mM). When isocitrate lyase induced by acetate minimal medium at 33$^{\circ}C$, MX9-11RX8 mutant did not express enzyme activity but did synthesize polypeptide chain whose electrophoretic mobilities were equal to those of the purified enzymes.

  • pH/Thermal sensitive copolymers with the various acidic comonomer compositions composed of N-isopropylacrylamide (NIPAAm) with acrylic acid (AAc), 2-acrylamido glycolic acid (AAmGAc), and 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS) were synthesized by free radial polymerization. In this study, to characterize the effect of different acidic comonomer composition and pH on the lower critical solution temperature (LCST) behaviors of their copolymers. phase transition experiments were performed with a thermo-optical analyzer (TOA). The phase transition temperature (T$^{p}$ ) of aqueous poly(NIPAAm-co-AAc) solution was lowered with increasing the ionization of the acid group in AAc, that is, the ionized state induced the electrostatic repulsion of ionized groups. In contrast, when AAmGAc was introduced into PNIPAAm, T$^{p}$ was little changed at pH 1-3, whereas climbed up significantly from pH 1 to pH 3. In the range of pH 6-10, Tp was lower than that of pH 3-5. This result was considered to be \"Ionic Screen Effect\" and this effect had been also observed in the case of poly(NIPAAm-co-AMPS).-co-AMPS).
