Study on the Cultural Conditions of Starch Utilizing Yeast Sporobolomyces holsaticus (전분이용성효모, Sporobolomyces holsaticus FRI Y-5의 배양조건에 관한 연구)

  • It was investigated under several cultural conditions to produce biomass directly from starch by an strictly aerobic and amylolytic yeast, Sporobolomyces holsaticus FRI Y-5. Its optimal temperature and initial pH of medium for growth were $23^{\circ}C$ and 6.9, respectively. Activation energy, Ea, for growth was calculated to be 17.33 Kcal/mole from the Arrhenius relationship. When each of 13 nitrogen sources was added to the basal medium, $(NH_2)_2CO$ had the best effect, on which concentration of cell after 3 day incubation was 10.6 g/l and cell yield was 0.451. The yeast growth was affected by $MgSO_4,\;K_2SO_4\;and\;ZnSO_4$ as a mineral source and was best on the medium containing all of them. The addition of yeast extract (5g/l) could enhance the production of biomass and cell yield to 77% and 32%, respectively.

Effect of GB 34-GB 39 Electro-acupuncture on Regional Cerebral Blood Flow in Stroke Patients and Normal Volunteers Evaluated by $^{99m}Tc-ECD$ SPECT (양릉천-현종 전침치료가 뇌경색환자 및 정상인의 뇌혈류에 미치는 영향 - SPECT와 SPM을 이용한 연구 -)

  • Objectives: Acupuncture has been applied in Asia for thousands of years, especially to rehabilitation after stroke. It has been reported that acupuncture increased cerebral blood supply and stimulated the functional activity of brain nerve cells shown by using brain imaging techniques. This study was to evaluate the effect of GB 34-GB 39 electro-acupuncture (EA) on regional cerebral blood flow (rCBF) in stroke patients and normal volunteers using single photon emission computed tomography (SPECT). Methods: The study procedure was divided into two parts: patients and volunteers studies. For the patients study, ten ischemic stroke patients (3 males, 7 females, mean age $68.5{\pm}8.9$ years old) were selected. Baseline brain SPECT was done with triple head gamma camera (MultiSPECT3, Siemens, USA) after intravenous administration of 1,110 MBq of $^{99m}Tc-ECD$. Fifteen-minute EA at GB 34 and GB 39 were applied on the affected limb. The same dose of $^{99m}Tc-ECD$ was injected during the EA, and the second set of SPECT images wasobtained. Using the computer software (ICON 7.1, Siemens, USA), 3 SPECT slices (upper, middle, lower) surrounding the brain lesion were selected and each slice was divided into 10-16 brain regions. Asymmetry indexes (AI) were analyzed in each brain region. We regarded over 10% changes of AI between before and after EA as significance. For the volunteers study, 10 healthy human volunteers (5 males, 5 females, mean age $28.1{\pm}6$ years old) were selected. In the resting state, $^{99m}Tc-ECD$ brain SPECT scans were performed. On the 7th day after the resting examination, 15 minute EA was applied at GB 34 and GB 39 on the right side of the subjects. Immediately after EA, the second SPECT images were obtained inthe same manner as the resting state. Significant increases and decreases of rCBF after EA were estimated by comparing their SPECT images with those of the resting state using paired t statistics at every voxel, which were analyzed by statistical parametric mapping with a threshold of p = 0.01, uncorrected (extent threshold: k=100 voxels). Results: In stroke patients, six of the eight (75%) had significantly increased perfusion in post-acupuncture scans compared to their baseline state. In normal volunteers, GB 34-GB GB EA increased rCBF in both hemispheres including right ventral posterior cingulate (Brodmann area (BA) 23), left superior temporal, anterior transverse temporal (BA 22, 41), left parastriate, peristriate (BA 18, 19), right occipitotemporal, angular (BA 37, 39), left rostral postcentral, caudal postcentral and preparietal (BA 2, 3, 5). However GB 34-GB 39 EA decreased rCBF in the right hemisphere including triangular and middle frontal lobes. Conclusions: The results demonstrated that OB 34-GB 39 EA increased cerebral perfusion in ischemic stroke patients and increased rCBF grossly in temporal lobes of normal volunteers. It is also suggested that there may be a correlation between the GB meridian and the territory of the middle cerebral artery.

  • Objectives: So far it has been reported that acupuncture increases cerebral blood supply and stimulates the functional activity of brain nerve cells. Previous studies have demonstrated that frequently used electro-acupuncture (EA) therapies for stroke increased regional cerebral blood flow (rCBF) in normal volunteers. Though ST 36-ST 41 EA is another prevailing therapy for stroke, there had been no report about its effect on rCBF. This study was to evaluate the effect of ST 36-ST 41 EA on rCBF in normal volunteers using single photon emission computed tomography (SPECT) and statistical parametric mapping (SPM). Methods: In the resting state, $^{99m}Tc$-ECD brain SPECT scans were performed on 10 normal volunteers (5 males, 5 female, mean age $23.6{\pm}0.5$ years). On the other study day, 7 days after the resting examination, 15 minutesEA were applied at ST 36 and ST 41 on the right side of the subjects. Immediately after ST36-ST41 EA, the second SPECT images were obtained in the same manner as the resting state. Significant increases and decreases of rCBF after EA were estimated by comparing their SPECT images with those of the resting state using paired t statistics at every voxel, which were analyzed by SPM with a threshold of p = 0.01, uncorrected (extent threshold: k=100 voxels). Results: EA applied at the right ST36-ST41 significantly increased rCBF in the right inferior parietal lobule (Brodmann area [BA] 40), right retrosubicular area (BA 48), left inferior parietal lobule (BA 40), left middle temporal gyrus (BA 21), left fusiform gyrus (BA 37), left inferior parietal lobule (BA 39), left inferior temporal gyrus (BA 20), and left somatosensory association cortex (BA 7). However, right ST36-ST41 EA significantly decreased rCBF in the right parahippocampal gyrus (BA 35), right cerebellum, left frontopolar area (BA 10), left orbitofrontal area (BA 11), left dorsolateral prefrontal cortex (BA 9), and left dorsal anterior cingulate cortex (BA 32). Conclusions: These results demonstrate that rightST36-ST41 EA increased rCBF prominently in both inferior parietal lobule (BA 40) and right retrosubicular area (BA 48), which suggest that there be correlation between specific EA and corresponding rCBF.

Biomechanical Effects of Facet Capsule Injuries in Posterior Lumbar Fusion Operations (후방경유 요추 융합수술시 척추후관절낭 손상의 운동역학적 영향)

  • Objectives : Although posterior lumbar fusion operations had been reported to accelerate spinal degeneration, there have been only a few studies for their biomechanical effects. We have studied the change of motion range at the vertebral joint one level above the fusion(UVJ) in pedicle screw fixation group(PSF)(n=13) where facet capsule was destroyed and in posterior lumbar interbody fusion group(PLIF)(n=8) where it was spared. Patients and Methods : The patients were divided into early(3 to 6 months) and late(over 12 months) according to postoperative follow-up period. The flexion, extension and flexion-extension angles(FA, EA, FEA) were measured at the UVJ with pre-operative, early and late post-operative films. Results : Mean age and male to female ratio were $52.7{\pm}9.3$ and 1 : 3.2. Mean follow-up periods were $144.1{\pm}30.0$ and $528.8{\pm}160.3$ days in early and late groups, respectively. The FEA and FA in the late PSF($11.8{\pm}3.1$, $8.5{\pm}2.9$) were significantly greater than pre-operative angles($7.8{\pm}3.9$, $5.1{\pm}3.7$)(p<0.01, p<0.05). All angles in the PLIF showed no significant changes with time. The FEA and FA in the late PSF($11.8{\pm}3.1$, $8.5{\pm}2.9$) were significantly greater than those of the late PLIF($7.6{\pm}2.3$, $3.4{\pm}2.0$)(p<0.01, p<0.001). All angles at early follow-up period were similar between PSF and PLIF. The EA showed no significant change in relation with follow-up period or fusion method. Conclusion : As a result, the facet capsule injury in pedicle screw fixation seems to be related with increased flexion angle or degeneration of the adjacent joint above the fused vertebra in the late phase.

Therapeutic Effects of Achyrantes Radix and Electroacupuncture of Zusanli acupoint(ST36) on Type II Collagen-Induced Arthritis (흰쥐의 제 2형 콜라겐 유도 관절염에 대한 우슬과 족삼리 전침 자극의 치료 효과)

  • Purpose: We investigated the effects of the combined therapy in rats with rheumatoid arthritis induced by type II collagen for 28 days, which consisted of the oral administration of the AR and EA applied to zusanli acupoint(ST36). Methods: Normal group was oral administered with 0.9% NaCl $0.5\;m{\ell}/day$ to normal rats. Control group was oral administered with 0.9% NaCl $0.5\;m{\ell}/day$ to arthritic rats. Group I was oral administered with AR 500 mg/kg $0.5\;m{\ell}/day$ to arthritic rats. Group II was given 2 Hz EA of ST36 in the test group for 30 min/day to arthritic rats. Group III was oral administered with AR 500 mg/kg $0.5\;m{\ell}/day$ and 2 Hz EA of ST36 in the test group for 30 min/day to arthritic rats. We Observed effect of the histopathological changes by H&E stain of liver, kidney, knee joint and ELSIA of cytokines($TNF-{\alpha}$). Results: 1. The vacuolization of liver tissue was decreased in group I, II, III comparing with control group. 2. The glomerular sclerosis of kidney tissue was decreased in group I, II, III comparing with control group. 3. The erosion of arthritic site of knee joint tissue was decreased group I, II, III comparing with control group. In particular group III was the most effective comparing with group I, II on the histopathological view. 4. In the ELSIA test of $TNF-{\alpha}$ concentration, Control group significantly increased in the concentration more than group I, II, III. The rate of increase in concentration slowed down in group III more than group I, II(p<0.05). Conclusion: It is concluded that 500 mg/kg of AR extracts and EA have clear therapeutic effect on the rheumatoid arthritis.

Needle Characteristics and Genetic Variation of Pinus pumila Natural Population in Mt. Seorak (설악산 눈잣나무 천연집단의 침엽특성과 유전변이)

  • This study was conducted to investigate the morphological and anatomical characteristics of needle and the genetic diversity of Pinus pumila Regel which is a unique and the southern peripheral population in South Korea. ANOVA test showed that there were significant differences among individuals within population in all 8 needle characteristics. Average characteristics of 66 individuals were 53.59 mm in needle length, 0.78 mm in needle width, 68.98 in needle index, 0.65 mm in needle thickness, 4.56 ea. in maximum stomata row, 3.80 ea. in minimum stomata row, 8.36 ea. in total stomata row and 1.71 ea. in resin canals, respectively. Resin canal per needle of this species ranged from one to three, depending on external type. Especially, arrangement types were 69.47% in two resin canals and 30.45% in a single resin canal. A total of 78 bands was generated from 9 selected I-SSR primers. The estimates of genetic variation were 61.5% in proportion of polymorphic bands (P), 1.698 in effective number of alleles ($A_e$), 0.388 in expected heterozygosity ($H_e$) and 0.567 in Shannon's information index (S.I.), respectively.

Purification and Characterization of a Recombinant Pea Chloroplastic Fructose-1, 6-bisphosphatase

  • A cDNA fragment encoding the chloroplastic fructose-1, 6-bisphosphatase (FBPase) was cloned via PCR from the cDNA library of pea leaves. The cloned cDNA, about 1.05 kbp without signal sequence, was introduced into a pET-28a vector for expression in E. coli strain BL21(DE3)pLysS. The recombinant FBPase was purified through $Ni^+-NTA$ affinity chromatography and characterized. Molecular mass of the monomer was about 42,000. Enzymatic activity of the purified enzyme as the native pea chloroplastic FBPase was the highest at alkaline pH (pH 9.0). The recombinant enzyme was activated by a reducing agent DTT and was insensitive to AMP. The activation energy (Ea) and Arrehenius frequency factor were 42.67 kcal/mol and $2.65{\times}10^{14}/s$, respectively, slightly higher than those of the native enzyme. $K_M$ and $V_{max}$ were $99.98{\mu}M$ and $52.9{\mu}M/min$, respectively, which were comparable with the native enzyme.

Study on the Selection of Solvent for Purificatino of p-Dioxanone by Crystallization Method (결정화에 의한 파라디옥산온의 정제를 위한 용매선정에 관한 연구)

  • In order to obtain a highly purified p-dioxanone (PDX) as monomer of biodegradable polymer, suitable solvent must be selected. The selection was based on the solubility of impurities, and partial layer melt crystallization were carried out under the presence of solvent. The solubility of PDX in various solvents such as ethyl acetate, tetrahydrofuran, acetone, alcohols (methanol, ethanol, 1-propanol, 1-butanol and 1-pentanol) were measured over the temperature range of $-10{\sim}15^{\circ}C$. As solubility parameters, the mixing and dissolution enthalpy between the PDX and the solvents was studied based on empirical equations and the regular solution theory. The solubility and the temperature dependency of the solubility with the solvents of acetone, ethylacetate, and tetrahydrofuran of PDX were shown to have relatively high values compared to the alcohol type solvents. Also, in same alcohols, the smaller molecules and higher polarity gave higher solvency. In partial layer melt crystallization, small amount of ethylacetate selectively dissolved impurities and gave highly purified p-dioxanone, over 99.9% purity.

The Variation of Cone, Seed and Germination Characteristics of Picea jezoensis (Siebold & Zuccarini) Carriere Populations in Korea (한국산(韓國産) 가문비나무 자생집단(自生集團)의 구과(毬果), 종자(種子) 및 발아특성(發芽特性) 변이(變異))

  • This study was conducted to investigate the variation of cone, seed and germination characteristics among populations and among individuals within populations of Picea jezoensis(Siebold & Zuccarini) Carriere distributed in Korea. Cone collected from 25 trees in two natural populations(Mt. Jiri and Mt. Dokyu) and their thirteen cone and seed characteristics as well as three germination behaviors were analyzed. Statistical analysis showed significant differences among populations and among individuals within populations in 9 traits except for seed breadth, seed weight, index of seed wing and mean germination time. Generally, morphological variation of cone and seed wings from Mt. Jiri population has smaller and longer than those of Mt. Dokyu population. Percentage of sound seeds and Tetrazolium test from Mt. Jiri population showed 1.79 and 1.87 higher values than Mt. Dokyu population, respectively. The maximum germination percentage was obtained at the optimum temperature of $20^{\circ}C$ and in this case, mean germination time and germination rate showed 7.5 days and 2.9 ea./day, respectively. In seed germination behaviors, percentage(40.7%) and rate(0.90 ea./day) of germination in Mt. Jiri population were more higher and faster than those of Mt. Dokyu population(17.7%, 0.37 ea./day). According to correlation analysis, P. jezoensis populations with small restricted distribution may have been reduced because seed qualities were correlated with increased levels of inbreeding and disproportion flowering.

Modification of Functional Properties of Soy Protein Isolate by Proteolytic Enzymes (단백분해효소에 의한 대두단백의 기능적 특성변화)

  • The effects of enzymatic modification with pepsin and actinidin was studied on molecular weight distributions and functional properties of hydrolysates from soy protein isolate (SPI) differing in degree of hydrolysis. The hydrolyzed SPI by pepsin showed 41.5% degree of hydrolysis after 5 min, and maximum hydrolysis was obtained after 2 hours. Actinidin hydrolyzed SPI 26.71% degree after 1 hour. On SDS-PAGE, native SPI showed 9 distinguishable bands on SDS-PAGE gel. Pepsin treated SPI showed one broad band in the lower part of gel. This band was shifted further to the bottom of the gel and became faint as hydrolysis time increased. While actinidin treated SPI showed different SDS-PAGE pattern from pepsin. However PAGE patterns were similar with pepsin and actinidin treated groups. With pepsin treatment, solubility of SPI distinctively increased around isoelectric point(pI). Emulsifying activity (EA) and emulsifying stability (ES) showed marked increase over pH range of $3.0{\sim}8.0$. 5 min modified group had most excellent foam expansion (FE). Foam stability (FS) was increased as pepsin treatment time increased at pI. With actinidin treatment, solubility was increased. 60 min modified SPI had the most effective EA at pH 4.5. However ES was not effected by actinidin treatment. 5 min modified group was most effect in FE. FS was higher at alkaline pH.
