Current Status of Plasmodiophora brassicae Researches in Korea

  • Clubroot disease is caused by the soil-born obligate plant pathogen Plasmodiophora brassicae. This pathogen can infect all cruciferous vegetables and oil crops, including Brassica rapa, B. oleracea, B. napus, and other Brassica species. Clubroot disease is now considered to be a major problem in Chinese cabbage production in China, Korea, and Japan. We collected several hundreds of P. brassicae infected galls from Korea, and isolated the single spore from the collection. For establishment of novel isolation, and mass-propagation methods for singe spore isolates of P. brassicae pathogen, we developed new filtration method using both cellulose nitrate filter and syringe filter. Accurate detection of P. brassicae pathogen in the field was done by using real-time PCR in the potential infested soil. When we tested the different pathogenicity on commercial Chinese cabbage varieties, P. brassicae from collected galls showed various morphological patterns about clubroot symptom on roots. To date, 8 CR loci have been identified in the B. rapa genome using the quantitative trait loci (QTL) mapping approach, with different resistant sources and isolates. We are trying to develop the molecular marker systems for detect all 8 CR resistant genes. Especially for the study on the interaction between pathogens and CR loci which are not well understood until now, genome wide association studies are doing using the sequenced inbred lines of Chinese cabbage to detect the novel CR genes.

황토의 물리적 특성 및 수용액중의 중금속 이온의 흡착 특성에 관한 연구 (A Study on Physical Properties and Adsorption Characteristics of Heavy Metal Ions of Loess)

  • Removal of Cu(II), Cr(III) and Pb(II) ions from aqueous solutions using the adsorption process on the loesses has been investigated. Variations of contact time, pH, adsorption isotherms and selectivity of coexisting ions were experimental parameters. pH of KJ and YIK samples diluted to 1% solution, was rearly the same with each value of pH 5.58 and 5.49, and both samples showed weak acidic properties. From chemical analysis, both samples contain remarkably different amounts of ${SiO}_{2}$, ${Al}_{2}O_{3}$ and ${Fe}_{2}O_{3}$. From XRD measurement, quartz was mainly observed in both samples. Kaolinite was also observed, also in both samples, but Feldspar was only observed in KJ sample. Adsorption of metal ions on the loesses were reached at equilibrium by shaking for about 30min. The adsorption of Cr(III) ion was higher than that of Cu(II) oand Pb(II) ions. The order of amount adsorbed among the investigated ions was Cr(III)>Pb(II)>Cu(II). In acidic solution, the adsorptivity of loesses was increased as pH increased. The adsorption of Cr(III) ion on the loesses were fitted to the Freundlich isotherms. Freundlich constants(1/n) of KJ and YIK loesses were 0.54 and 0.55, respectively.

서울에 유통 중인 해산 어류의 부위별 중금속 분석 (Contents of heavy metals in marine fishes, sold in Seoul)

  • 서울시내에 유통 중인 대형어류 17종 89건에 대하여 원자흡광광도계(AAS) 및 수은분석기 등을 이용하여 납, 카드뮴, 수은, 크롬, 구리, 비소 함량을 근육, 간 및 아가미 등의 부위별로 분석하였다. 그 결과를 요약하면 다음과 같다($Mean{\pm}SD$, mg/kg). 어종별 평균 중금속 함량은 Hg $0.08{\pm}0.01mg/kg$, Pb $0.17{\pm}0.32mg/kg$, Cd $0.34{\pm}0.07mg/kg$, Cr $0.05{\pm}0.05mg/kg$, Cu $1.14{\pm}0.13mg/kg$, As $0.24{\pm}0.22mg/kg$ 이었다. 부위별 유해 중금속의 전체적인 측정치를 살펴보면 중금속과 어종에 상관없이 간의 측정치가 가장 높게 나타났다. 나라별 중금속을 비교한 결과 Hg, Pb, Cu는 뉴질랜드산, Cd, As는 노르웨이산, Cr은 국내산 어류의 평균함유량이 다른 나라 어류보다 다소 높은 측정치를 보였으나, 자연함량수준으로 오염의 가능성은 없다고 생각된다. 기준초과로 검출된 어종은 일본에서 수입된 황돔으로 두 건이었다. 어류의 간과 근육 부위 중금속 함량 간의 상관관계는 Hg (r=0.989, p<0.01), Pb (r=0.978, p<0.01), Cd (r=0.991, p<0.01), Cu (r=0.998, p<0.01), As (r=0.198, p<0.05)에서 상관관계를 나타냈으며, Cr (r=0.082)은 유의적 상관관계가 인정되지 않았다.

Ni-Cr-Mo-V강의 전기화학적 방법에 의한 내식성 평가 (Evaluation of corrosion resistance by electrochemical method for Ni-Cr-Mo-V steel)

  • When the structures are exposed to their own an application for a long period, a number of variables such as strength properties and corrosion resistance, so on are expected to change. In the present investigation the corrosion behavior and resistance for the original and degraded materials of Ni-Cr-Mo-V steel were evaluated under the conditions of pH 3, 6, 9 and 12 in a distilled water environment. The electrochemical polarization technique was employed in this investigation. Based upon the experimental results obtained, the following conclusions were drawn. A severe and uniform corrosion was observed for both original and degraded materials under the condition of pH 3. At pH 6 and pH 9, these materials showed the degradation by a pitting corrosion. The materials under pH 12 environment were degraded by a uniform corrosion. The corrosion rate per year were the highest in the pH 3 environment, followed by pH 12, pH 6 and pH 9 environment in order. The corrosion resistance was decreased from the original material, slow cooled material (10.deg. C/hr) and step cooled material in order.

공간적 가중을 이용한 강화 유무에 따른 3주간 수축-이완 중재가 넙다리뒤근 단축 대상자의 유연성과 균형능력에 미치는 효과 (Effects of Three-Week Contract-Relax Interventions with and without Reinforcement Using Temporal Summation for Flexibility and Balance Ability in Young People with Hamstring Shortening)

  • Purpose: The purpose of this study was to examine the effect of three-week contract-and-relax (CR) interventions with and without reinforcement using temporal summation for flexibility and dynamic balance ability in young people with hamstring shortening. Methods: This study was conducted on 20 female college students with hamstring shortening. The participants were divided equally into two groups using stratified randomization: the CR group (CRG) and the CR with reinforcement group (CRRG). All interventions were applied three times a week for three weeks. The passive straight leg raise (PSLR) test and functional reaching test (FRT) were conducted on each participant before and after the three-week intervention. Results: In both groups, PSLR and FRT improved significantly after the three-week intervention compared to before intervention (p < 0.01). The amount of change in PSLR after the three-week intervention was significantly higher in CRRG than in CRG (p < 0.05). Conclusion: Three-week CR interventions with and without reinforcement were effective in improving PSLR and FRT. To improve hamstring shortening, CR intervention with reinforcement may be more useful than CR intervention without reinforcement.

MMPH-0 (Enterobacter aerogenes)에 의한 6가 크롬 오염 지하수의 생지화학적 정화 (Biogeochemical Remediation of Cr(VI)-Contaminated Groundwater using MMPH-0 (Enterobacter aerogenes))

  • 오염환경에 서식하는 토착미생물은 환경정화에 중요한 역할을 담당하며 이 연구는 6가 크롬 오염 지하수에서 분리한 미생물을 이용해 반응성, 이동성, 발암성 높은 6가 크롬을 당대사 조효소인 3가 크롬으로 환원/침전시켜 경제적, 친환경적, 생지화학적 정화의 효율성을 알아보았다. 미생물 농화배양과 조성분석, 호기와 혐기환경의 6가 크롬 환원과 내성, 전자공여체별 6가 크롬 환원, 지화학적 변화, 미생물 외형과 Cr((III) 침전물의 광물특성을 연구한 결과, 분리한 MMPH-0(Enterobacter aerogenes)는 혐기/호기환경에서 6가 크롬 내성과 환원능(유기산 주입 1주 후 70%, 주입 안한 경우 4주 후 10 ~ 20%)이 있고, Eh는 미생물의 유기산 산화로 생성된 전자에 의해 산화에서 환원환경, pH는 중성에서 약산성으로 변화되어 $Cr(OH)_3$/Cr(III)침전물이 형성되었다. SEM/TEM-EDS 결과 $2{\sim}5{\mu}m$ 막대형 미생물과 세포 밖 Cr(III) 침전물은 지화학적 환경변화와 유기산 산화에 따른 전자공여에 의한 환원의 근거가 된다. 지화학적 촉매제 토착미생물의 활성화로 산화환원에 민감한 중금속 오염 지하수 정화에 효율적 기술 응용이 기대된다.

  • We examined the removal of hazardous heavy metals (Cd, Cr, and Pb) from laver Pyropia sp. using citric, hydrochloric, and nitric acids. Under the same conditions, the quality of the laver samples was also evaluated using the variation in absorbance and major mineral levels. The heavy metals that accumulated in raw laver samples after 3 days in seawater included Pb (117.79 μg/g), Cr (33.53 μg/g), and Cd (10.54 μg/g) in descending order. The rate of heavy metal removal from laver was higher at lower pH for all acids used. However, its color changed unsatisfactorily at pH 2.0. After 10 min in seawater at pH 2.5, the heavy metals in laver were eliminated in the order Cd (68.7–81.6%), Pb (57.7–67.0%), and Cr (31.9–49.4%) using the three acids. The differences in heavy metal removal among acid types were not significant. The laver quality was not affected after 20 min at the pH range of 2.5–4.0. The maximum removal of heavy metals was from laver soaked for 10 min in seawater at pH 2.5 using the organic acid, citric acid.

  • 본 연구는 Cr 투여가 태권도 선수들에게 운동 수행능력, 혈장요소 및 무산소성 파워에 어떠한 영향을 미치는지를 규명하기 위하여 수행되었다. 이를 위하여 남자 고등학교 태권도 선수를 대상으로 위약집단, Cr 투여 집단으로 구분하여 Cr을 투여한 후 Cr 투여는 태권도 선수들에게 있어서 효과적인 영양 보조물 방법이고, 단기간의 Cr loading으로도 운동수행력의 향상에 긍정적인 영향을 미칠 수 있음을 확인할 수 있었다.

Effect of Chromium Dietary Supplementation on the Immune Response and Some Blood Biochemical Parameters of Transport-stressed Lambs

  • Forty-eight Naemi lambs (avg. BW 31.7 kg) were transported by truck for a distance of 1,450 km from Al-Jouf to Riyadh, Saudi Arabia. On arrival day, the lambs were randomly allocated to four groups receiving diets supplemented with 0.0, 0.3, 0.6 and 0.9 ppm organic chromium (Cr). Each group consisted of four separately housed replicates of three lambs each. The animals were fed ad libitum on a grower diet for 84 days. Blood samples were obtained shortly before transportation, upon arrival and at weekly intervals thereafter from all lambs for analysis of plasma and serum. Plasma glucose and serum cortisol, total protein, albumin, urea-N and total cholesterol concentrations were determined. A cursory clinical examination of the lambs, along with rectal temperature, was undertaken at different intervals during the experiment. The lambs were inoculated each with 2 ml i.v. chicken red blood cells (CRBC) on days 0, 21, and 42. Serum total, IgG and IgM antibody titers were determined at weekly intervals post-immunization. An in vivo intradermal hypersensitivity test was carried out on 6 lambs from each group on days 10 and 70. Transportation of the lambs resulted in a significant (p<0.001) elevation of serum cortisol, total protein and albumin levels, as well as increased plasma glucose concentration, with corresponding decrease in total cholesterol, while blood urea-N remained largely unchanged. These constituents returned to normal levels during subsequent weeks, with no significant differences in their concentrations being observed between the Cr-supplemented groups and controls. Rise in rectal temperature after transportation was reduced to a greater extent (p<0.05) in Cr-supplemented versus control lambs. Total, IgG and IgM antibody titers against CRBC rose significantly (p<0.05) during immunizations in all groups, with significantly and linearly higher (p<0.05) total and IgG titers in Cr-supplemented versus control lambs. By contrast, no significant effect due to Cr supplementation was recorded in IgG titers, which increased equally in Cr-fed and control groups. Skin thickness in response to intradermal inoculation of phytohaemagglutinin (PHA) was also significantly (p<0.01) increased as a result of Cr supplementation. These results indicate that dietary Cr supplementation might be useful during stress especially for enhancing immune responses in transport-stressed lambs.

Effects of Various Sources and Levels of Chromium on Performance of Broilers

  • Three hundred and twenty four one day old mixed sex broiler chicks were assigned at random into 9 treatment groups. The experimental design was a 3${\times}$3 factorial arrangement. During the starter period (week 1-3), chicks were fed ad libitum. A cornsoybean meal based diet contained 23% crude protein, 3,200 kcal/kg metabolizable energy (NRC, 1994), and supplemented with organic or inorganic forms of chromium. Two organic chromium products, chromium yeast (Cr-Yeast from Alltech Biotechnology Corporation Limited) and chromium picolinate (Cr-Pic) were supplemented at the rate of 200, 400 and 800 ppb. One inorganic product, chromium chloride, was supplemented at the rate of 200, 400 and 800 ppb. During the finishing period (week 4-7), the corn-soybean meal based diet contained 20% crude protein, 3,200 kcal/kg metabolizable energy (NRC, 1994), and the same levels of chromium as in the starter period were added. No significant difference was observed among treatment groups in average daily gain, feed intake, body weight gain, feed conversion ratio and mortality. The carcass percentage of broilers receiving 200 and 400 ppb organic chromium (Cr-Yeast or Cr-Pic) was significantly increased (p<0.01). In addition, the supplementation of organic chromium reduced (p<0.05) breast meat fat content but increased breast meat protein content. The addition of chromium in the diet had no effect on boneless breast, skinless boneless breast, boneless leg, skinless boneless leg but reduced percentage of sirloin muscle. Total cholesterol and triglycerides were reduced by organic Cr supplementation. Supplementation with 200 and 400 ppb of both Cr-Yeast and Cr-Pic showed the lowest total cholesterol. The effects of type of Cr on HDL and LDL were variable, however, LDL increased with increasing level of Cr supplementation. This trial indicates that organic chromium tended to improve growth performances and carcass composition, reduced total cholesterol and triglycerides. The optimum level of organic chromium supplementation was at 200 ppb.
