EFFECT OF CONCENTRATION OF PHOSPHORIC ACID ON MICROLEAKAGE TO TOOTH STRUCTURE (인산농도에 따른 치질의 미세누출 효과)

  • The purpose of this study was to evaluate the micro leakage of composite resin when various phosphoric acid agents were used to etch the enamel and dentin. In this study, class V cavities were prepared on the buccal surfaces of fourty extracted human molar teeth, and they were randomly assigned into 4 groups with 10 teeth. The cavities of each groups were etched with 10%, 32%, 35% and 37% phosphoric acids for 15 seconds, washed and dried and the cavities were restored with composite resin after application of the adhesive. The specimens were immersed in 2% methylene blue solution for 3 days. And then, the specimens were sectioned buccoligually. Degree of dye penetration at tooth-restoration interfaces was examined by Inverted Metallurgical Microscope at the occlusal and gingival margins. The result were as follows : 1. The degree of microleakage at occlusal and gingival margin in all group was statistically difference among 10% and 35% (P<0.01), 10% and 37% (P<0.01), 32% and 35% (P<0.05) and 32% and 37% (P<0.05) acid concentrations, but was not statistically difference between 10% and 32%, 35% and 37% acid concentrations (P>0.05). The degree of microleakage was showed to dimish with increase of acid concentration. 2. The degree of microleakage at the occlusal margin was statistically significant difference among 10% and 32%,10% and 35%, 10% and 37% acid concentrations(P<0.05). The degree of microleakage among 32% and 35%, 32% and 37% and 32% and 37% acid concentrations was not statistically significant difference. 3. In comparison of microleakage at the gingival margin, the degree of microleakage above 32% acid concentration was not showed less than 10% acid concentration (P>0.05). In comparison of 32%, 35%, 37% acid concentrations, the degree of microleakage at 35% and 37% acid concentrations was showed less than 32% acid concentration(P<0.05).

General Population's View on Euthanasia (안락사에 대한 일반인들의 인식도)

  • Purpose : Amont the various issues concerning bio-ethics, the concern on euthanasia has increased along with the development of medical technology. Thus, the general public tends to have more liberal opinion. They have detail research data and real practices in US, Europe and Australia, but we lack such studies in our country. This study was undertaken to address the need of studies on the recognition of euthanasia among the public because the existing studies have been focused on the medical staff. Methods : Survey 413 people the age of 17 or more, from May to July 2000. Testify the data on the variation of demography and the recognition of euthanasia by using SAS 6.12, the statistic program. Results : 304 people (73.6%) think that euthanasia should be legislated, 156 people (37.8%) permit euthanasia to the rage of voluntary one, and 234 people (56.6%) permit passive euthanasia. When the subject of voluntary euthanasia was himself, more people whose age is 35 or more (P=0.001) responded that they will undertake euthanasia. And, on issues related to the passive euthanasia, one's educational background (P=0.046) and economic power (P=0.040) arrangement showed significant differences. When the subject of voluntary euthanasia is other people, more people whose age is 35 or more than 35 (P=0.001), whose sex is male (P=0.001), and married people (P=0.002) were for allowing the matter. For the subject of passive euthanasia, survey participant's occupation (P=0.016) created meaningful difference. More people whose age is 35 or more than 35 responded that they want voluntary euthanasia for themselves (P=0.001), and in the case when euthanasia is legislated, marital status (P=0.002) also shows meaningful difference. Passive euthanasia is permitted by the more people whose age is less than 35 for respondents other people (P=0.001), marital status show meaningful difference in case for respondent himself. In the case of legal euthanasia is more people whose age is 35 or more than 35 (P=0.001), sex is male (P=0.004) and more married people (P=0.001) responded that they want voluntary euthanasia for other people. And, age (P=0.002), sex (P=0.001), education (P=0.025) and economic power (P=0.001) show meaningful difference for case the subject of passive euthanasia. Conclusion : Most of general public responded that the legislation on euthanasia is required; and, age, education and economic power seem to have an influence on their decisions on euthanasia. Not only such a study of demographic and sociological correlation; but, various basic data on the legislation of euthanasia are needed.

Korean Red Ginseng protects dopaminergic neurons by suppressing the cleavage of p35 to p25 in a Parkinson's disease mouse model

  • Background: Ginseng is known to have antiapoptotic, anti-inflammatory, and antioxidant effects. The present study investigated a possible role of Korean Red Ginseng (KRG) in suppressing dopaminergic neuronal cell death and the cleavage of p35 to p25 in the substantia nigra (SN) and striatum (ST) using a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced Parkinson's disease mouse model. Methods: Ten-week-old male C57BL/6 mice were injected intraperitoneally with 30 mg/kg of MPTP at 24-h intervals for 5 d, and then administered KRG (1 mg/kg, 10 mg/kg, or 100 mg/kg) once a day for 12 consecutive days from the first injection. Pole tests were performed to assess the motor function of the mice, dopaminergic neuronal survival in the SN and ST was evaluated using tyrosine hydroxylase-immunohistochemistry, and the expressions of cyclin-dependent kinase 5 (Cdk5), p35, and p25 in the SN and ST were measured using Western blotting. Results: MPTP administration caused behavioral impairment, dopaminergic neuronal death, increased Cdk5 and p25 expression, and decreased p35 expression in the nigrostriatal system of mice, whereas KRG dose-dependently alleviated these MPTP-induced changes. Conclusion: These results indicate that KRG can inhibit MPTP-induced dopaminergic neuronal death and suppress the cleavage of p35 to p25 in the SN and the ST, suggesting a possible role for KRG in the treatment of Parkinson's disease.

Development of a Plant Transformation Vector, pKCHI (고등식물 형질전환용 유전자 운반체 pKCHI의 개발)

  • We have developed a plasmid vector, pKCH1, for the purpose of higher plant transformation. It contains the promoter region of cauliflower mosaic virus 35S transcript (P35s) and the terminator region of nopaline synthase gene (Tnos) with unique cloning sites, Bam HI and Xba I, between them. After inserting a foreing gene at the cloning sites, P35s-foreign gene-Tnos cassette can be recovered by using a restriction enzyme Hind III.

Preformulation Study of Prokidin : Chemical Stability

  • The effects of pH and temperature on the degradation of prokidn in various buffered aqueous solutions(pH 1.32~9.66) and temperatures (35, 45 and 6$0^{\circ}C$ were investigated. The effect of ionic strength on the degradation of prokidin was also measured by varying ionic strength (0.0466~1.5) at pH 1.35 and 45$^{\circ}C$ The effect of metal ions on the degradation of prokidin at pH 7.35 and 3.98 was observed. The degradation of prokidin followed the pseudo- first- order kinetics. The degradation rate of prokidin showed pH-dependent and temperature-dependent patterns. Prokidin was very stable at the pH below 3.95, where half-lives at 35, 45 and 6$0^{\circ}C$were 294, 206 and 107 day, respectively. However, it degraded very rapidly at pH above 6.49; the half-lives at 35, 45 and 6$0^{\circ}C$were 60, 25 and 13 day, respectively. As ionic strength increased, the degradation rate of prokidin increased. Some metal ions increased the degradation rate in the rank order of Mn > Fe > Cu >Fe On the other hand. other metal ions such as Bi, Ba. Zn, Ni, Co did not show unfavorable effect.

Dietary protein requirement of Pacific white shrimp Litopenaeus vannamei in three different growth stages

  • A study of three feeding trials was conducted to investigate the dietary protein requirements of Pacific white shrimp (Litopenaeus vannamei) at three different growth stages. Six experimental diets were formulated to include increasing protein levels of 25, 30, 35, 40, 45, and 50% (designated as P25, P30, P35, P40, P45, and P50, respectively) for three feeding trials. The three feeding trials were conducted in different-sized shrimp at 0.65 g (trial 1), 4.80 g (trial 2), and 10.5 g (trial 3). Triplicate groups of shrimp were fed one of the experimental diets for 36, 42, and 48 days in trials 1, 2, and 3, respectively. In trial 1, the growth performance was not affected by the dietary protein levels. However, protein efficiency ratio was significantly higher in P30 diet compared to P40, P45, and P50 diets. In trial 2, growth rate was significantly higher in P35 diet than in P25 diet. In trial 3, the lowest growth performance was obtained in P25 diet which significantly differed from that of other experimental diets. Broken line analysis of growth data indicates that the optimal dietary level of crude protein is 34.5, 35.6, and 32.2% for small-, medium-, and large-sized (juvenile, sub-adult, and adult stages) Pacific white shrimp, respectively.

Establishment of Quantitative Analysis Method for Genetically Modified Maize Using a Reference Plasmid and Novel Primers

  • For the quantitative analysis of genetically modified (GM) maize in processed foods, primer sets and probes based on the 35S promoter (p35S), nopaline synthase terminator (tNOS), p35S-hsp70 intron, and zSSIIb gene encoding starch synthase II for intrinsic control were designed. Polymerase chain reaction (PCR) products (80~101 bp) were specifically amplified and the primer sets targeting the smaller regions (80 or 81 bp) were more sensitive than those targeting the larger regions (94 or 101 bp). Particularly, the primer set 35F1-R1 for p35S targeting 81 bp of sequence was even more sensitive than that targeting 101 bp of sequence by a 3-log scale. The target DNA fragments were also specifically amplified from all GM labeled food samples except for one item we tested when 35F1-R1 primer set was applied. A reference plasmid pGMmaize (3 kb) including the smaller PCR products for p35S, tNOS, p35S-hsp70 intron, and the zSSIIb gene was constructed for real-time PCR (RT-PCR). The linearity of standard curves was confirmed by using diluents ranging from $2{\times}10^1{\sim}10^5$ copies of pGMmaize and the $R^2$ values ranged from 0.999~1.000. In the RT-PCR, the detection limit using the novel primer/probe sets was 5 pg of genomic DNA from MON810 line indicating that the primer sets targeting the smaller regions (80 or 81 bp) could be used for highly sensitive detection of foreign DNA fragments from GM maize in processed foods.

Biochemical Characteristics of the Granulosis Viruses DNA of Common Cabbage Worm, Pieris rapae and Pieris brassicae (배추흰나비 과립병바이러스 DNA의 생화학적 특성)

  • This study was carried out to acquire some basic biochemical informations on the granulosis virus (GV) DNA of Pieris rapae and Pieris brassicae. The thermal denaturation temperature (Tm) and G+C content of the DNA of the viruses were $83.7^{\circ}C$ and 35.5% for P. rapae GV, $84.0^{\circ}C$ and 35.9% for P. brassicae GV, respectively. There were some differences in the DNA fragmentation patterns of the two GV's produced by digestion with restriction endonucleases such as EcoR I , BamH I and Hind m . The homololgy between the two DNAs was caculated to be 97.0%. The size of the genome was estimated to be 103 kbp for P. rapae GV and 108 kbp for P. brassicae GV.

A Study on the Marginal Fit of Dental Prosthesis According to the W/P Ratio of Investment (매몰재의 혼수비가 치관 보철물 변연의 적합성에 미치는 영향에 관한 연구)

  • This test is verifying for influence of W/P ratio of investment upon dental prosthesis Firstly we made 40 MOD die and divide 4 groups, each group take 0.2 different w/p ratio grade such as 0.35, 0.37, 0.39 and 0.41. The method of data analysis applied were by ANOVA, Tukey test and Mann-Whiney and Kruskal Wallis test. The finding of this study were as follows : 1. The 0.37 w/p ratio group showed most accurte margin due to $15.8{\mu}m$ gap between margin and die which keep up with manufacture's instruction and 0.35 w/p ratio group was followed as second accuracy group o 새 $48.3{\mu}m$ gap between margin and die, 0.39 w/p ratio group showed $101{\mu}m$ gap and 0.41 w/p ratio group showed $129.8{\mu}m$ gap. 2. As to the relationship of the margin accuracy between 4 different grade of w/p ratio groups, each group was statistically significant(P<0.01). Also the all groups were statistically significant except between 0.35 and 0.37 groups between 0.39 and 0.41 groups, and between 0.35 and 0.39 groups. 3. A consequence of Mann-Whitney & Kruskal-Wallis test for marginal accuracy between 4 different grade of w/p ratio was statistically significant as same as above ANOVA test result. Also Tukey test for verifying similarity from each group showed same as above.

Antioxidant Enzymes of Strains Panax ginseng C.A. Mey. and Panax quinquefolius L.

  • The strains of Panax ginseng C.A. Mey., P. quinquefolius L. and selected strains P. ginseng-B, P.ginseng-A, P. quinquefolius-C were investigated. Activities of SOD, catalase and peroxydase were determined by methods of Fridovich et al. (1979), Komov et al.(1975), Bovaird et al.(1982) respectively. Activities of SOD, catalase, peroxydase were investigated every day 5 in cycle of cultivation. For P. ginseng it was the 35 days, P. quinquefolius the 70 days, P. quinquefolius-C 90 days. P. ginseng-B 90 days, P. ginseng-A 60 days. The P. quinquefolius, P. quinquefolius-C, P. ginseng-B had clear differentiation and developed tracheid elements, which are absent in strain of P. ginseng. The peaks of protein content for P. ginseng (4.5 units/g) and for P. quinquefolius (3.5 units/g) were on day 10 and remained unchanged till the last cultivation. The strain P. ginseng-A had two peaks of protein content (2.5 mg/g) on day 15 and on day 30. For P. ginseng-B strain these peaks were on day 5 and day 40 (3.5 mg/g). Peroxydase activity peak (60 units/g) in P. ginseng strain was on day 10. This activity in P. ginseng-B had two peaks on day 15 and day 35 and reached 95 units/g , increasing to 150 units/g to day 80. In strain of P. ginseng-A was only one maximum of this activity -130 units/g on day 45. In P. quinquefolius peroxydase activity was 103 units/g on day 40, increasing to 135 units/g to day 90. For P. quinquefolius-C this activity peak was 136 units/g on day 60. Peroxydase activities for the upper and lower layers of biomass was different and varied considerably from 28-35 units/g in lower to 270-290 units/g for upper layer. The SOD activity had two peaks in P. ginseng strain the 80 units/g and the 70 units/g on day 20 and day 35 respectively. Activity of SOD in P. quinquefolius strain reached 53 units/g on day 40 and increased up to 83 units/g to day 60.The similar increase of SOD activity was marked for P. ginseng-B to 85 units/g on day 90. In P. ginseng strain the 6 molecular isoforms SOD was defined. One of them with RfO,6 was determined in all days of cycle, three other (Rf-0.43; 0.54;0.80) only on day 10 and day 20. The isoform of SOD with Rf-0,29 was detected only on day 10 and with Rf-0,35 only on day 35. The catalase activity decreased in all strains to the last days of cultivation. The changes of SOD, catalase and peroxydase activities reflect the differences between the strains of Panax ginseng and Panax quinquefolius and their selected forms. The correlation between maximum life span of strains and activities of their antioxydant enzymes were detected.
