바이오에탄올 생산에 적합한 백합나무(Liriodendron tulipifera)의 oxalic acid 전처리 효과 탐색 (Effect of Oxalic Acid Pretreatment on Yellow Poplar (Liriodendron tulipifera) for Ethanol Production)

  • 본 연구에서는 국내 조림 수종인 백합나무를 바이오에탄올 생산용 자원으로 이용하고자 oxalic acid 전처리 방법을 도입하여 가능성을 타진하였다. $160^{\circ}C$에서 0.037 g/g oxalic acid로 20분 전처리하였을 때 $40.22g/{\ell}$의 발효가능한 당을 생산하였으며, 처리된 고형 바이오매스를 이용하여 동시당화발효를 수행한 결과 72시간 후 $8.6g/{\ell}$의 에탄올을 생산하였다. 같은 조건에서 반응시간을 증가시켜 40분 처리하였을 때 $32.66g/{\ell}$의 발효가능한 당을 생산하였고 동시당화발효로 72시간 후 $9.5g/{\ell}$의 에탄올을 생산하였다. 가수분해산물을 분석한 결과, 같은 조건에서 반응시간이 증가함에 따라 acetic acid, 5-HMF, furfural, total phenols와 같은 발효저해물질이 증가하였다. 이와 같은 발효저해물질은 반응시간보다는 초기 oxalic acid 첨가량에 영향을 받았다. Acetic acid 생산량은 저농도(0.013 g/g)의 oxalic acid를 사용하였을 때 $3.39{\sim}5.78g/{\ell}$로 나타났으며 xylose 분해산물인 furfural은 glucose의 분해산물인 5-HMF보다 2~3배 많게 가수분해산물에 존재하였다. 리그닌 분해산물로 예측되는 total phenols는 모든 조건에서 $5g/{\ell}$ 이상이 검출되었다. 가수분해산물과 동시당화발효로부터 에탄올 생산량을 분석한 결과 0.037 g/g oxalic acid로 20분 전처리한 가수분해산물과 고형 바이오매스로부터 가장 높은 에탄올 생산을 예측할 수 있다.

  • The characteristic of calcined BaTiO3 powder and sintered PTC ceramics was investigated varing with calcination temperature and time of BaTiO(C2O4)2.4H2O synthesized from BaCl2.2H2O, TiCl4, oxalic acid and ethanol by oxalic acid method. When the particle size was less than 0.1 ${\mu}{\textrm}{m}$ by controlling calcination temperature and time, the resistance at room temperature was measured very high (above M{{{{ OMEGA }}). However, when the calcined particle sizes ranged from 0.2 to 0.3 ${\mu}{\textrm}{m}$, the resistance was 100 {{{{ OMEGA }} (After sintering, the grain size was 10~30 ${\mu}{\textrm}{m}$ homogeneously with the addition of dopant in sintering, the resistivity, resistance, $\alpha$ value and jumped to were 110{{{{ OMEGA }}.cm, 24$^{\circ}C$/% and 106{{{{ OMEGA }}, respectively.

Fluoride and Calcium in Tea Leaves

  • The determination of fluoride and the nutritious calcium in infusion of teas are explained. Tea leaves were pulverized and were immersed in boiling water. The solution was filtered and fluoride, calcium and oxalic acid were determined by the ion chromatography. The quantities of fluoride, calcium and oxalate ions extracted from 100 g of tea leaves were calculated. Tea leaves were also immersed in 0.5 M hydrochloric acid and extracted oxalate and calcium ions were analyzed. The free oxalic acid and calcium were extracted in boiling water and the total ones were extracted in hydrochloric acid. The quantity of calcium oxalate was calculated from the total and the free oxalic acids. The free calcium was estimated to be nutritious.

알루미늄 합금의 연속식 양극산화법으로 형성시킨 이중 산화막층의 특성 (Properties of double-layered anodizing films on Al alloys formed by two consecutive anodizings)

  • In this study, double-layered anodizing films were formed on Al 5052 and Al 6061 alloys consecutively first in sulfuric acid and then in oxalic acid, and hardness, withstand voltage, surface roughness and acid resistance of the anodizing films were compared with single-layered anodizing films in sulfuric acid and oxalic acid electrolytes. Hardness of the double-layered anodizing film decreased with increasing ratio of inner layer to outer layer for both Al 5052 and Al 6061 alloys, suggesting that outer anodizing film formed in sulfuric acid electrolyte is damaged during the second anodizing in oxalic acid electrolyte. Withstand voltage of the double-layered anodizing films increased with increasing the thickness ratio of inner layer to outer layer. Surface roughness of the double-layered anodizing films were comparable with that of single-layered anodizing film formed in sulfuric acid but higher than that of single layer anodizing film formed in oxalic acid electrolyte. In acid resistance test, all of the double-layered and single-layered anodizing films showed good acid resistance more than 3 h without any visible gas evolution, which is attributable to sealing of pores. Based on the experimental results obtained in this work, it is possible to design a double-layered anodizing film with cost-effectiveness and improved physical and electrical properties by combining two consecutive anodizing processes of sulfuric acid anodizing and oxalic acid anodizing methods.

Pathogenicity Determinants of Sclerotinia sclerotiorum and Their Association to Its Aggressiveness on Brassica juncea

  • White rot or stem rot caused by Sclerotinia sclerotiorum is one of the most destructive fungal diseases that have become a serious threat to the successful cultivation of oilseed Brassicas. The study was designed with an aim to investigate the association between the pathogenic aggressiveness and pathogenicity determinants of this pathogen specifically in Brassica for the first time. For this, a total of 58 isolates of S. sclerotiorum from different geographical regions were collected and purified. These isolates were inoculated on a Brassica juncea cv. RL-1359 and they exhibited high level of variation in their disease progression. The isolates were grouped and then 24 isolates were selected for the biochemical analysis of pathogenicity determinants. The isolates varied significantly with respect to their total organic acids, oxalic acid production and pectin methyl esterase and polygalacturonase activity. The oxalic acid production corresponded to the disease progression of the isolates; the isolates with higher oxalic acid production were the more aggressive ones and vice-versa. This is, in our knowledge, the first study to establish a correlation between oxalic acid production and pathogenic aggressiveness of S. sclerotiorum on B. juncea. However, the pectinases' enzyme activity did not follow the trend as of disease progression. These suggest an indispensable role of oxalic acid in pathogenicity of the fungus and the potential to be used as biochemical marker for preliminary assessment of pathogenic aggressiveness of various isolates before incorporating them in a breeding program.

화학세척제를 이용한 도자기의 철산화물 제거 특성 연구 (Chemical Cleaning of Iron Stains on Ceramics)

  • 금속산화물에 오염된 도자기 유물을 보존처리 하기 위해서는 경우에 따라 화학적 세척과정이 필요하다. 본 연구에서는 화학적 세척과정을 침적법과 습포법으로 구분하여, 침적법에서는 옥살산과 구연산의 농도에 따른 철산화물 제거특성을 확인하고자 하였다. 나아가 앞선 침적법의 결과를 토대로 spot 테스트를 거친 후, 태안 마도에서 출토된 도자기 유물에 습포법을 적용하여 철산화물을 제거하였다. 옥살산에 60시간 침적시 도자기 빙렬 내의 철산화물은 제거되었지만, 육안관찰에서 침적 3시간 이후부터 철산화물이 관찰되지 않기 때문에 보존처리현장에서는 유물의 안정성을 고려하여 0.25M 이하의 옥살산에 1~3시간동안 침적하는 것이 적당할 것으로 판단된다. 구연산에서는 60시간 침적시 철산화물은 제거되지 않았다. 이는 옥살산과의 분자량 차이 및 산도(acidity) 등에 의한 것으로, 빙렬 내에 침투한 철산화물을 제거할 때에는 구연산보다 옥살산이 좀 더 효과적이었다. 옥살산을 용제로 제작한 습포팩(벤토나이트, 세피올라이트, 활성탄소섬유, 셀라이트)을 오염된 백자태토에 10시간동안 도포 처리하여 철산화물은 제거되었지만, 유물적용성은 벤토나이트와 세피올라이트가 뛰어났다. 그래서 앞선 침적법의 결과를 토대로, 0.25M의 옥살산과 세피올라이트를 습포 팩으로 제조하여 태안 마도 출토 청자 및 백자에 적용하였다. 습포팩 1회 처리로 유물표면의 철산화물은 대부분 제거되었으나 오염물의 형태에 따라 적용횟수는 달라질 수 있을 것으로 판단되며, 세피올라이트에 의한 2차 오염을 방지하기 위한 세척도 함께 병행되어야 할 것이다.

마른김의 유기산조성(有機酸組成)과 저장중의 변화 (Changes in Organic Acid Composition of Dried Lavers during Storage)

  • 마른김의 유기산조성(有機酸組成)과 이것의 저장중(中)의 변화(變化)를 GLC 및 GC-MS 를 사용(使用)하여 조사검토(調査檢討)하였다. 1. 마른김에 함유(含有)된 유기산(有機酸)으로서 동정(同定) 및 정량(定量)을 한 것은 pyruvic, lactic, oxalic, malonic, fumaric, succinic, malic 및 citric acid외 8종(種)이며, 양적(量的)으로 주요산(主要酸)은 oxalic, succinic, malic 및 citric acid였다. 2. oxalic, malic 및 succinic acid는 저습도하(低濕度下)에서는 큰 변화(變化)를 보이지 않으나, 고습도하(高濕度下)에서는 oxalic 및 malic acid는 급속(急速)히 감소(減少)하고, 반대(反對)로 succinic acid는 현저(顯著)히 증가(增加)하는 대조적(對照的)인 변화(變化)를 보였다. citric acid만은 저습도(低濕度)에서나 고습도(高濕度)에서나 다같이 저장기간(期間)에 따라 급속(急速)히 감소(減少)하였다. 3. 말동정성분중(末同定成分中), 그 mass spectrum으로 보아 환상구조(環狀構造)의 산(酸)이라고 추정(推定)되는 2성분(成分)은 양적(量的)으로 극히 많았으며 저장중(中)의 변화(變化)는 저습도하(低濕度下)에서는 서서(徐徐)히 감소(減少)하나, 고습도하(高濕度下)에서는 신속(迅速)히 감소(減少)하여 45일간(日間)의 저장으로 거의 소실(消失)하였다. 4. 휘발산량(揮發酸量)의 변화(變化)는 저장기간(期間)에 따라 증가(增加)하였으며, 저습도(低濕度)에서보다 고습도(高濕度)에서의 증가경향(增加傾向)이 높았다.

Changes of Organic Acids, Polyphenols, Pigments and Fiber Concentration with a Different Stalk Position and Grade of Korean Flue-cured Leaf Tobacco

  • This study was carried out to analyze the organic acids, polyphenols, pigments and fiber materials concentration with a different stalk position and grade of korean leaf tobaccos. Eight kinds of flue-cured leaf tobaccos which were different stalk position and grade were used for this study. Three kinds of major organic acids(citric, malic and oxalic), 2 kinds of polyphenols(chlorogenic acid and rutin), 3 kinds of pigments($\beta$-carotene, chlorophyll-a and chlorophyll-b), and 2 kinds of fiber components(pectin and lignin) were analyzed. All of these chemical components were changed with a different stalk position. When the citric acid, malic acid, $\beta-carotene$, chlorophyll-a, and lignin concentration were low in the middle stalk position and high in both bottom and upper position, oxalic acid and chlorogenic acid show the highest concentration in the middle stalk position. All of these chemical components also changed with a different grade of leaf tobaccos. As the citric acid, malic acid, $\beta-carotene$, chlorophyll-b, and lignin concentration decreased as the grade ascended, the oxalic acid and chlorogenic acid concentration increased as the grade ascended. This results assumed that the quality of korean leaf tobacco was directly proportional to oxalic acid and chlorogenic acid concentration but it was inversely proportional to citric acid, malic acid, $\beta-carotene$, chlorophyll-b and lignin concentration.
