Subacute Oral Toxicity of Taeumjowi-tang in SD Rats (태음조위탕(太陰調胃湯)의 경구 투여에 의한 SD계 흰쥐의 아급성 독성시험 연구)

  • Taeumjowi-tang is originated in Donguisusebowon edited by Lee Je-Ma. The objective of this study was to investigate the subacute toxicity of Taeumjowi-tang in rats. Several doses(0, 125, 250, 500, 1,000, and 2,000 mg/kg) of Taeumjowi-tang were administered to rats for 4 weeks. The mortality, clinical signs, body weights and gross findings were examined for experimental period. No dead animals were found during the experimental period. In addition, any differences were not found between control and treated groups in clinical signs, hematology, serum biochemistry, and other findings. In conclusion, the no observed adverse effect level(NOAEL) for Taeumjowi-tang was over 2,000 mg/kg/day in rats.

Acute Toxicity Study on Coptidis Rhizoma in Mice (황련의 급성독성에 관한 연구)

  • In order to evaluate acute toxicity of Coptidis rhizoma, 6 week- and 13 week-old male ICR mice received Coptidis rhizoma extract (600~4,800 mg/kg body weight) orally, and toxicological responses were observed for consecutive 7 days. In the mice received relatively high concentration of Coptidis rhizoma($\geq$1,200mg/kg), death occurred within 3 hrs after oral administration, and its ratio in 13 week-old mice was conspicuously higher than that in 6 week-old mice. $LD_{50}$ of Coptidis rhizoma were estimated to bi 2,575 mg/kg and 1,490 mg/kg body weight in 6 week and 13 week-old mice, respectively. Coptidis rhizoma-treated animals manifested a variety of abnormal clinical findings such as ptosis, crouching, lethargy, convulsion, bizarre behavior and truning sideway. These abnormalities also ranked highly in the 13 week-old mice compared to those in the 6 week-old mice. In addition to abnormal behaviors, Coptidis rhizoma($\geq$1,200 mg/Kg) significantly elevated the urinary contents of bilirubin, urobilirubin, protein and glucose, and values in 13 week-old mice was higher than those in 6 week-old animals. No toxicological response was observed at concentration less than 600 mg/kg. Our results clearly demonstrate that susceptibility of mice to Coptidis rhizoma may be related with age, indicating that younger age mice is more resistant to the Coptidis rhizoma than the older, and toxicological mechanism of Coptidis rhizoma may be closely associated with its pharmacological mechanism.

A 4-week Repeated Oral Dose Toxicity Study of CJ-10882 in Dogs

  • The present study was conducted to investigate the potential subacute toxicity of CJ-10882 by a 4-week repeated oral dose in dogs. The test article was administered once dally by gavage to dogs at dose levels of 0, 2, 10, and 50 mg/kg/day for 4 weeks. During the test period, clinical signs, mortality, body weights, food consumption, ophthalmoscopy, urinalysis, hematology, serum biochemistry, gross finding, organ weight, and histopathology were evaluated. Several clinical sign were observed in treated dogs at 50 mg/kg, including salivation and vomiting. Increase in the serum level of ALT and albumin observed in the female 50 mg/kg group was considered as a toxic effect related to the test article since the histopathological change in Liver was accompanied. There were no treatment-related effects on mortality, food and water consumption, ophthalmoscopy, urinalysis, hematology, serum biochemistry, necropsy findings and organ weights in any treatment group. Based on these results, target organ was not observed and the no-observed-adverse-effect level (NOAEL) was 10 mg/kg/day and the absolute toxic dose was 50 mg/kg for both male and female dogs.

Chronic Treatment of Ethanol Inhibits Proliferation of Normal Fibroblasts, but Not Oncogenic ras-Transformed Cells

  • The adverse effects of ethanol on cell proliferation have been described for a variety of tissues and cells. In the present study, we investigated whether chronic ethanol intoxication impairs the cell proliferation and DNA synthesis induced by oncogenic $H-ras^{V12}$ - and $v-K-ras^{V12}$-transformed cells. Ethanol treatment inhibited the cell proliferation and the DNA synthesis of control parental fibroblasts in a time- and dose-dependent manner. In contrast, ethanol did not suppress the proliferation of either oncogenic $H-ras^{V12}$ - or $v-K-ras^{V12}$ -transformed fibroblasts. Microinjection of oncogenic $H-Ras^{V12}$ protein induces DNA synthesis and ethanol treatment did not interfere with the DNA synthesis. The antiproliferative toxicity of ethanol was rescued by antioxidants, such as N-acetylcysteine and 4-methlpyrazole. These results indicate that the antiproliferative action site of ethanol toxicity lies upstream or is independent of Ras and ethanol exerts its toxicity through a free radical formation.

Single Oval Dose Toxicity Study of the Extract of Aralia elata in Mice (마우스에서 두릅 추출물의 단회경구투여 독성시험)

  • This study was conducted to investigate the potential acute toxicity of Aralia elata by a single oral dose in ICR mice. Thirty mice of each sex were randomly assigned to three groups of 10 mice each. The test articles were administered once by the gavage to mice at dose levels of 0, 2,500 and 5,000 mg/kg body weight. The mortality and changes on body weight and clinical signs of gross observation were monitored for 14 days after dosing. At the end of 14-day observation period, all animals were sacrificed and complete gross postmortem examinations were performed. There were no dead animal and adverse effects on clinical signs, the body weight and the gross finding. As the results, we could not find any toxic effect at the dose levels of 2,500 or 5,000 mg/kg in mice and the minimal lethal dose was considered to be over 5,000 mg/kg body weight in mice.

Single Oral Toxicity Study on the Polysaccharide Fraction of Pueraria lobata in Rats (갈근 다당체 분획의 단회경구투여 독성에 관한 연구)

  • Acute toxicity on the polysaccharide fraction of Pueraria lobata was examined using male and female Sprague-Dawley rats. The polysaccharide fraction of Pueraria lobata was orally administered at a dose of 5 mg/kg, 50 mg/kg, 500 mg/kg, 2,000 mg/kg and 5,000 mg/kg and observed for two weeks. No mortality and abnormal clinical signs were observed at the doses used. There were not any significant differences in parameters of blood biochemical values and urinalysis by the treatment of test material. All rats were appeared to be healthy and normal throughout the observation period. Also there was no difference in net body weight gain and gross pathological findings among the groups rats treated with different doses of the polysaccharide fraction with Pueraria lobata.

Acute Oral Toxicity of A Novel Combined Antibiotic(Cefatrizine / Clavulanic Acid) in Rats

  • The acute toxicity study of combined antibiotic (Cefatrizine / Clavulanic Acid), a formulation consisting of cafatrizine and clavulanic acid in a ratio of 2 : 1, was evaluated in rats. The antibiotic was orally administered with single dose in dose levels up to 5 g/kg (0, 1.25, 2.5, 5 g/kg). Treatment-related effects were limited to soft stool excretion and caecal dilatation, but histologically no morphological changes could be detected in caecum. In hematology, serum-chemistry parameters and histopathology, no drug-related changes were found. The results of the present study indicate that cefatrizine / clavulanic acid has a low toxic potential and the oral $LD_{50}$values exceed 5 g / kg in rats

  • Objectives: The study is aimed at evaluating the possible toxicity in 90-day repeated oral administration of modified Samjung-hwan (mSJH) in Sprague-Dawley (SD) rats. This study was conducted to detect the no-observed adverse effect level (NOAEL). Methods: Modified SJH extract was administered orally in male and female SD rats at dose of 0, 1,000, 2,000, 4,000 mg/kg. Each group consisted of 10 rats of each gender. The modified SJH extract was given once a day for 90 days. We monitored the changes of mortalities, clinical signs, body weight changes, food consumption, ophthalmologic findings, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histological markers of all animals treated with modified SJH extract during the study period. Results: There were no toxicologically significant changes in mortalities, clinical signs, body weight gains, food consumption, ophthalmologic findings, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histological markers in any of rats tested. Conclusions: The NOAEL of the modified SJH extract in male rats and no observed effect level (NOEL) in female rats are considered 4,000 mg/kg.

Simultaneous determination and acute toxicity study of Fructus mume extracts in ICR mice (오매(烏梅)의 다성분동시분석 및 마우스를 이용한 경구 단회투여 급성독성시험)

  • Objectives : Previous studies have shown that Fructus mume (FM) has anti-platelet effects. The present study was performed to determine the acute oral toxicity and quality control of a crude extract of FM in ICR mice. Methods : We investigated the in vivo single dose acute toxicity of FM 95% ethanol extract. This test was orally administered once by gavage to 20 male and 20 female mice at dose levels of 0 (control group), 1250, 2500 and 5000mg/kg body weight, respectively. Mortalities, clinical findings, autopsy findings and body weight changes were monitored daily for the 14 days following the administration. HPLC analysis was performed for the simultaneous determination of ursolic acid and p-hydroxycinnamic acid in FM. Reverse-phase chromatography using a C18 column and photodiode array detection at 211 nm was used for quantification of the two maker components. The mobile phase for gradient elution consists of water and acetonitrile. Results : We observed survival rates, general toxicity, change of body weight, and autopsy. The mice did not die after single oral administration of maximum dose of FM. Autopsy of animal revealed no abnormal gross finding. Therefore, $LD_{50}$ value of FM for ICR mice was more than 5000mg/kg on oral route. The HPLC analysis showed that ursolic acid and p-hydroxycinnamic acid amounts to 9.75- and 0.12% in the extract with the retention times of 47.99- and 15.38 minutes, respectively. Conclusions : These results suggest that no toxic dose level of FM in mice is considered to be more than 5000mg/kg. Consequently, it was concluded that FM have no effect on acute toxicity and side effect in ICR mice. For the quality control of FM extract, simultaneous determination of ursolic acid and p-hydroxycinnamic acid was established.

Analysis of Aconitine Contents in Aconiti Radix Lateralis Preparata and Sambutang-P that Contains Aconiti Radix Lateralis Preparata and Single Oral Toxicity Test (포제부자(炮製附子) 및 포제부자(炮製附子) 함유 삼부탕(蔘附湯)의 aconitine 함량 분석과 단회투여 독성시험)

  • Objectives : The contents of aconitine in aconiti radix lateralis preparata, purified hot water extract of Aconiti Radix lateralis preparata, and purified hot water extract of Sambutang-P that contains Aconiti Radix lateralis preparata was analyzed to compare toxicity. Toxicity of Sambutang-P that contains Aconiti Radix lateralis preparata was assessed with a single oral toxicity test on 6-week-old male and female Sprague-Dawley rats. Methods : 1. The contents of aconitine in Aconiti Radix lateralis preparata, purified hot water extract of Aconiti Radix lateralis preparata, and purified hot water extract of Sambutang-P that contains Aconiti Radix lateralis preparata was analyzed using the purity test according to the "Korean Herbal Pharmacopoeia". 2. 2,000mg/kg was injected for the single oral toxicity test of purified hot water extract of Sambutang-P that contains Aconiti Radix lateralis preparata, and the test was done for a test group (injection) and a control group, each with 5 male and 5 female rats. For 14 days after injection, rats were observed for general symptoms and changes in weight. Afterwards, blood biochemical test, autopsy, and histophathological exam of the liver was conducted. Results : 1. The contents of aconitine was 0.0785% for Aconiti Radix lateralis preparata, 0.1510% for purified hot water extract of Aconiti Radix lateralis preparata, and 0.1248% for purified hot water extract of Sambutang-P that contains Aconiti Radix lateralis preparata. 2. There was no death of either male or female rats in both the control group and the test group (injection of 2,000mg/kg). 3. No unusual symptom was observed in both the control group and the test group (injection of 2,000mg/kg). 4. No significant change in weight was observed for both male and female rats in the test group (2,000mg/kg). 5. The histopathological exam of ALT, AST, ALP, GGT and LDH showed no significant changes for both male and female rats in the test group (2,000mg/kg). 6. According to the autopsy results, no visible abnormality of organs or tissues was found in both the control group and the test group (2,000mg/kg). 7. According to the histopathological exam of the liver, the effect of the injected material was not observed for either male or female rats in the test group (2,000mg/kg). Conclusions : The contents of aconitine in Aconiti Radix lateralis preparata was lower for decoction of Sambutang-P with ginseng radix alba than for decoction of only Aconiti Radix lateralis preparata. This suggests that ginseng radix alba can dilute toxicity of Aconiti Radix lateralis preparata. As for a single oral toxicity test of Sambutang-P that contains Aconiti Radix lateralis preparata, no abnormal reaction was observed even when the injection amount far exceeded a toxic dose or a lethal dose. Thus, it is deemed that using Sambutang-P at a clinically prescribed dose would not lead to hepatoxicity.
