아미그달린 저감화 매실청의 독성동태학적 및 경구독성 연구 (Toxicokinetics and oral toxicity of Maesil-cheongs with reduced amygdalin levels)

  • 본 연구에서는 플라즈마 및 UVC 처리에 의해 매실청에 존재하는 아미그달린 저감화 공정을 개발하여 아미그달린 저감화 매실청에 대한 독성을 독성동태학적 및 14일 반복 경구투여에 의해 평가하였다. 독성동태학적 연구 결과, 플라즈마 및 UVC 처리에 의한 아미그달린 저감화 매실청 투여에 의해 아미그달린의 경구 흡수량이 약 3.7, 3.4배 감소하여 저감화 효과를 확인할 수 있었다. 아미그달린 저감화 매실청의 14일 반복 경구독성시험 결과를 종합하면, 몸무게 변화, 식이 및 물 섭취량, 절대장기 및 상대 장기 무게, 혈액학적 및 혈청학적 분석, 조직 병리학적 분석에서 유의미한 독성현상이 관찰되지 않았다. 따라서 본 연구에서 개발 플라즈마 및 UVC 처리에 의한 아미그달린 저감화 매실청의 독성영향은 없는 것으로 보이며, 이러한 공정은 매실청뿐만 아니라 아미그달린 함유 다양한 제품의 저감화 공정에 활용될 수 있을 것이다.

A 4-week Repeated dose Oral Toxicity Study of Mecasin in Sprague-Dawley Rats to Determine the Appropriate Doses for a 13-week, Repeated Toxicity Test

  • Objectives: In this study, we investigated the 4-week repeated-dose oral toxicity of gami-jakyak gamcho buja decoction (Mecasin) to develop safe treatments. Methods: In order to investigate the 4-week oral toxicity of Mecasin, we administered Mecasin orally to rats. Sprague-Dawley (SD) rats were divided into four groups of five male and five female animals per group: group 1 being the control group and groups 2, 3, and 4 being the experimental groups. Doses of Mecasin of 500, 1,000, and 2,000 mg/kg of body weight were administered to the experimental groups, and a dose of normal saline solution of 10 mL/kg was administered to the control group. We examined the survival rate, weight, clinical signs, and gross findings for four weeks. This study was conducted under the approval of the Institutional Animal Ethics Committee. Results: No deaths occurred in any of the four groups. No significant changes in weights or food consumption between the control group and the experimental groups were observed. Serum biochemistry revealed that some groups showed significant decrease in inorganic phosphorus (IP) (P < 0.05). During necropsy on the rats, one abnormal macroscopic feature, a slight loss of fur, was observed in the mid dosage (1,000 mg/kg) male group. No abnormalities were observed in any other rats. In histopathological findings, the tubular basophilia and cast of the kidney and extramedullary hematopoiesis of the spleen were found. However, those changes were minimal and had occurred naturally or sporadically. No other organ abnormalities were observed. Conclusion: During this 4-week, repeated, oral toxicity test of Mecasin in SD rats, no toxicity changes due to Mecasin were observed in any of the male or the female rats in the high dosage group. Thus, we suggest that the doses in a 13-week, repeated test should be 0, 500, 1,000, and 2,000 mg/kg respectively.

Subacute Oral Toxicity Assesment of Ethanol Extract of Mariposa christia vespertilionis Leaves in Male Sprague Dawley Rats

  • The term Butterfly tea refers to decoction of Mariposa christia vespertilionis leaves which is widely consumed by cancer patients throughout Malaysia and has gained a huge popularity among Malaysians, not only cancer patients but also researchers to discover the real potential of this plant. Herein, the study is aimed at evaluating the possible toxicity in 28-day subacute oral toxicity of ethanolic extract M. christia vespertilionis in male Sprague Dawley rats. The 28-day subacute toxicity study was conducted to detect the no-observed adverse effect level (NOAEL). In this study, a total of 30 rats were divided into the control, 5% DMSO (vehicle), low dose (75 mg/kg), medium dose (125 mg/kg) and high dose (250 mg/kg) groups. The extract was administered daily from day 1 until day 28. At the end of the study, the animals were humanely sacrificed and assessed for the effect extract of Mariposa christia vespertilionis leaves on body weight and relative organ weights and haematological, biochemical and histopathological parameters. The haematological and serum biochemical parameters for the assessment of kidney and liver injuries were carried out. Results of haematological and serum biochemistry results showed no changes in the control and treated groups. In the histopathology, evaluation of kidney tissues in all treated groups showed no significant (p > 0.05) lesions. In contrast to kidney, liver tissues showed significant differences (p < 0.05) in lesions observed in low dose (430 mg), medium dose (700 mg) and high dose (1480 mg) groups with very mild, mild and mild to moderate lesion of hepatic necrosis, in the respective groups, and very mild hepatic degeneration and hepatitis were scored in all three groups.

랫드에서 l-muscone의 급성독성 및 아급성독성시험 연구 (Acute and Subacute Toxicity Studies of l-Muscone in Rats)

  • l-Muscone is synthesized for use as substitutive material of musk which is the active ingredient of woohwangchungsimwon. The objective of this investigation was to evaluate the acute and subacute toxicity of l-muscone in rats. In oral acute toxicity test, SPF Sprague-Dawley male and female rats were gayaged with l-muscone of two doses(0, 5.0 g/kg). No dead animal and abnormal autopsy findings were found in control and treated group. Body weights were slightly decreased in both sexes of rats treated with 5.0 g/kg. Therefore, oral $LD_{50}$ of l-muscone was consider to be higher than 5.0 g/kg in male and female rats. In intraperitoneal acute toxicity test, rats were injected intraperitoneally with dosages of 0, 1,000, 1,316, 1,732, 2,279 and 3.000 mg/kg. Decreased body weights and motor activities were observed at high dose group. Intraperitoneal $LD_{50}$ of l-muscone were 1,920 mg/kg in male and female rats. In the subacute study, l-muscone was administrated orally to both sexes of rats for 4 weeks as several doses(0, 10, 100 and 1,000 mg/kg). There were neither dead animals nor significant changes of body weights during the experimental period. In addition, no differences were found between control and treated groups in clinical signs, urinalysis, hematology, serum biochemical analysist and other findings. Above data suggest that no observed adverse effect level of l-muscone in rats might be over 1,000 mg/kg/day in this study.

백서에서 식품 복합물 TB001의 단회 경구투여 독성 시험 연구 (Single Oral Dose Toxicity of TB001 in Rats)

  • Objectives: This study was performed to investigate single oral dose toxicity of TB001, the extract mixture of Purple loosestrife and Aceriphyllum rossii. Methods: The mortality, general symptom, change of weight: and necropcy findings was investigated for 14 days after a dose(2000 mg/kg B.W.) of TB001 was given, using SD rats of both male and female according to "The guideline of toxicity test for medicine and others". Results: The death of rats and abnormal finding was not observed. There was no the significant difference of weight between control group and TB001 group. Conclusions: The abnormalities at the necropsy finding of all survived rats was not detected. The study suggested that there is no toxicity in single dose(2000 mg/kg B.W.) of TB001.

Acute Toxicity Study of Modified Je-Ho-Tang in ICR Mice

  • Previous studies have shown that modified Je-Ho-Tang (MJHT) has anti-platelet effects. Je-Ho-Tang (JHT), a Korean court beverage, is a traditional Korean herbal medicine that has been used for the treatment of a disease attended by great thirst, and for prevention of illness in hot summers. We made MJHT from JHT by excluding honey. The present study was performed to determine the acute oral toxicity of crude extract of MJHT in male and female ICR mice. We investigated the in vivo single dose acute toxicity of MJHT hot-water extraction. This test was orally administered once by gavage to 20 mice of each sex received doses of 0 (control group), 1250, 2500 and 5000 mg/kg body weight. Mortalities, clinical findings, autopsy findings and body weight changes were monitored daily for 14 days following the administration. We observed survival rates, general toxicities, changes of body weight, and autopsy. No significant lethality was observed after single oral administration of MJHT at the different dosages. Autopsies on the animals revealed no gross abnormalities. Therefore, the LD50 value of MJHT for ICR mice was estimated more than 5000 mg/kg by the oral route. These results suggest that no toxic dose level of MJHT in mice is considered to be more than 5000 mg/kg. Consequently, it was concluded that MJHT have no effect on acute toxicity and side effect in ICR mice.

백강잠 추출물의 C57BL/6 마우스를 이용한 2주경구투여 독성시험 (Effects of a Two-Week-Oral-Toxicity Study of Bombyx batryticatus L. Extracts on C57BL/6 Mice)

  • Objectives : Bombyx batryticatus L. is the dried larval form of the silkworm (Bombyx mori L.) infected by Beauveria bassiania (Bals.) Vuill. It is used as a food and medicinal resource to treat asthma, headaches, epilepsy, and convulsions in traditional Korean and Chinese medicines. However, the research of the toxicity about B. batryticatus is not enough yet. Here, we investigate the effects of potential subacute toxicity following the repeated oral administration of B. batryticatus water extract to C57BL/6 mice, at various doses of 0, 50, 150, and 450 mg/kg/day during a two-week period. Methods : The following parameters were examined during the study period: body weight, gross findings, clinical signs, organ weight, hematology, serum biochemistry, histopathology, and mortality. At the end of the treatment period, all the mice were euthanized. Results : No changes were observed in the body weights, gross findings, clinical signs, organ weights, and mortality after two weeks of administration of the B. batryticatus extract. In addition, compared with the normal control group, no noticeable treatment-related changes were observed in the hematological, serum biochemical, and histopathological parameters in the treated group following treatment with doses of up to 450 mg/kg/day. Conclusion : Based on these findings, we conclude that the treatment of mice with the water extract of B. batryticatus did not cause considerable C57BL/6 toxicity, and therefore, it could be considered safe for further pharmacological studies.

Large-scale purification and single-dose oral-toxicity study of human thioredoxin and epidermal growth factor introduced into two different genetically modified soybean varieties

  • Thioredoxin (TRX) protein is an antioxidant responsible for reducing other proteins by exchanging cysteine thiol-disulfide and is also known for its anti-allergic and anti-aging properties. On the other hand, epidermal growth factor (EGF) is an important material used in the cosmetics industry and an essential protein necessary for dermal wound healing facilitated by the proliferation and migration of keratinocytes. EGF also assists in the formation of granulation tissues and stimulates the motility of fibroblasts. Hence, genetically modified soybeans were developed to overexpress these industrially important proteins for mass production. A single-dose oral-toxicity-based study was conducted to evaluate the potential toxic effects of TRX and EGF proteins, as safety assessments are necessary for the commercial use of seed-specific protein-expressing transgenic soybeans. To achieve this rationale, TRX and EGF proteins were mass purified from recombinant E. coli. The single-dose oral-toxicity tests of the TRX and EGF proteins were carried out in six-week old male and female Institute of Cancer Research (ICR) mice. The initial evaluation of the single-dose TRF and EGF treatments was based on monitoring the toxicity signatures and mortality rates among the mice, and the resultant mortality rates did not show any specific clinical symptoms related to the proteins. Furthermore, no significant differences were observed in the weights between the treatment and control groups of male and female ICR mice. After 14 days of treatment, no differences were observed in the autopsy reports between the various treatment and control groups. These results suggest that the minimum lethal dose of TRX and EGF proteins is higher than the allowed 2,000 mg·kg-1 limit.

Safety evaluation of cricket(Gryllus bimaculatus) extract in Sprague-Dawley rats

  • Recently, research investment in the improvement of food safety as a food source and specializing of nutritional source of edible insects is being actively conducted. Cricket especially has been attracting considerable interest in entomophagy; however, research on the safety assessment of cricket is limited. This study investigated the effects of cricket ethanol extract when orally administrated in Sprague-Dawley rats. Here, we performed a 4 wk repeated oral dose toxicity test in Sprague-Dawley rats following the Organization for Economic Cooperation and Development test guidelines 407 under Good Laboratory Practice regulation. Rats were randomly allocated 4 groups: vehicle control, 250, 500, 1,000 mg/kg test groups and administrated based on body weight for 28 d. The animals were observed for mortalities and clinical signs, body weight changes, food and water consumption. At the end of treatment period, blood and urine were collected and analyzed. Subsequently, the animals were sacrificed and subjected to gross pathological examination and organ weight measurement. The organs were preserved for histopathological examination. The results showed that there were no systemic toxicological effects related with the cricket ethanol extract in the 4 wk oral repeated dose toxicity study. It is considered that NOAEL of cricket ethanol extract is greater than 1,000 mg/kg/d and there was no target organ detected.

Acute Toxicity Study of Recombinant Human Interferon ${\alpha}A$ (LBD-007) in ICR Mice

  • The acute toxicity of a recombinant human interferon $\alpha$A (code name: LBD-007) was evaluated in both sexes of ICR mice, 5 weeks old, by the oral, subcutaneous and intravenous routes of administration. Based on the results, LBD-007 was not considered to induce any toxic effect on the mice in mortalities, clinical findigs, body weights and gross findings. It is suggested that LD$_{50}$ values in mice would be $>48{\times}10^8$ IU/kg in the oral, subcutaneous or intravenous routes.s.
