흰쥐의 스트레스성 위궤양에 대한 Alove vera의 항궤양작용 (The Antiulcer Effects of Alove vera on the Stomach ulcer Induced by Stress in rats)

  • This was performed to investigate the antiulcer effects of Aloe vera on the stomach ulcer induced by restraint and water-immersion stress in rats. For this experiment, 60 male Sprague-Dawley strain were used. The experimental groups were divided into five: a control(CA) and 4 aloe treatment groups. Each dose of aloe was 50 mg/kg BW(AA), 100mg/kg BW(AB), 200 mg/kg BW(AC), and 400 mg/kg BW(AD). The rats were allocated to each group by 12 and observed for 4 weeks. The results are as following. 1. The stomach surface pH in each group showed no significant difference, byt the values of aloe treatment groups were higher than the value of the control group. 2. The gastric wall mucus was significantly increased in all aloe treatment groups(p<0.05) compared with the control group. Especially in AC and AD group the differences were higher(p<0.01). 3. At shear rate rate 11.25, 45.0, 90.0, 225.0 sec-1, whole blood viscosity and plasma viscosity were measured. Most of the values of aloe treatment groups were significantly low compared with those of the control group(P<0.05). 4. The ulcer index of aloe treatment groups were significantly low campared with control group(p<0.05). Especially in AC and AD group the differences were more significant(p<0.01). 5. Less severe ulcers were observed in AA and AB group than in the control group. Tissues of AC and AD group had only slight ulcers and necrosis of tissue was not observed in these groups. Especially in AD group, there was more mucus than other groups and it seemed that alove vera stimulated the epithelial regeneration. From the results of this study, it can be concluded that the oral administration of Alove vera results in protection of stomach ulcer by stimulating the secretion of gastric mucus and the circulation of blood.

Establishment of Highly Tumorigenic Human Gastric Carcinoma Cell Lines from Xenograft Tumors in Mice

  • Patient's primary tumor-derived tumor cell lines likely represent ideal tools for human tumor biology in vitro and in vivo. Here, we describe eight human gastric carcinoma cell lines derived from established tumors in vivo upon subcutaneous transplantation of primary gastric carcinoma specimens in BALB/c nude mice. These xenografted gastric tumor cell lines (GTX) displayed close similarity with primary gastric tumor tissues in their in vivo growth pattern and genomic alterations. GTX-085 cells were resistant to cisplatin, while GTX-087 was the most sensitive cell line. GTX-085 was the only cell line showing a metastatic potential. Epithelial cell adhesion molecule (EPCAM) expression was especially strong in all tissue samples, as well as in cell cultures. GTX-139, the largest tumor graft obtained after injection, displayed distinct expression of CD44v6, fibroblast growth factor receptor 2 (FGFR2), and prominin 1 (PROM1, also known as CD133). In summary, we established eight xenograft gastric cancer cell lines from gastric cancer patient tissues, with their histological and molecular features consistent with those of the primary tumors. The established GTX cell lines will enable future studies of their responses to various treatments for gastric cancer.

5-Aminolevulinic Acid Fluorescence in Detection of Peritoneal Metastases

  • Background: The value of 5-aminolevulinic acid (ALA) in fluorescence detection of peritoneal metastases and the underlying mechanisms were evaluated in patients with peritoneal surface malignancies. Materials and Methods: Oral 5-ALA was administered at a concentration of 20 mg/kg body weight with 50 ml of water 2 hours prior to surgery (n=115). The diagnostic value of 5-ALA based fluorescence production was evaluated following white light inspection during prior to cytoreductive surgery and hyperthermic intraperitoneal chemotherapy. Then, peptide transporter PEPT1 (ALA influx transporter) and ATP-binding cassette transporter ABCG2 (porphyrin efflux transporter) gene expression was determined with quantitative real time (qRT)-PCR and pathological diagnoses confirmed for all tissue samples. Results: The 5-ALA based photodynamic detection rate was 17% for appendiceal mucinous neoplasms, 54% for colorectal cancers, 33% for gastric cancers, 67% for diffuse malign peritoneal mesotheliomas, and 89% for epithelial ovarian cancer of peritoneal metastases. 5-ALA was detected in all cases of peritoneal metastases originating from cholangiocarcinomas whereas it was not able to detect any in granulosa cell and gastrointestinal stromal tumor cases. Furthermore, PEPT1 was overexpressed whereas ABCG2 expression was downregulated in tumors detected with fluorescence. Conclusions: 5-ALA provided 100% specificity and high sensitivity to detect peritoneal metastases in subgroups of patients with peritoneal surface mailgnancies. ALA influx transporter PEPT1 and porphyrin efflux transporter ABCG2 genes are important in tumor specific 5-ALA induced fluorescence in vivo. Further studies should clarify diagnostic utility of 5-ALA in peritoneal surface malignancies.

Oral administration of fermented wild ginseng ameliorates DSS-induced acute colitis by inhibiting NF-κB signaling and protects intestinal epithelial barrier

  • Ginseng has been widely used for therapeutic and preventive purposes for thousands of years. However, orally administered ginseng has very low bioavailability and absorption in the intestine. Therefore, fermented ginseng was developed to enhance the beneficial effects of ginseng in the intestine. In this study, we investigated the molecular mechanisms underlying the anti-inflammatory activity of fermented wild ginseng (FWG). We found that FWG significantly alleviated the severity of colitis in a dextran sodium sulfate (DSS)-induced colitis mouse model, and decreased expression level of pro-inflammatory cytokines in colonic tissue. Moreover, we observed that FWG suppressed the infiltration of macrophages in DSS-induced colitis. FWG also attenuated the transcriptional activity of nuclear factor-κB (NF-κB) by reducing the translocation of NF-κB into the nucleus. Our data indicate that FWG contains anti-inflammatory activity via NF-κB inactivation and could be useful for treating colitis. [BMB Reports 2015; 48(7): 419-425]

Subchronic Inhalation Toxicity of Trichloroacetonitrile on the Sprague Dawley Rats

  • Trichloroacetonitrile is used as an intermediate in insecticides, pesticides, and dyes. In Korea alone, over 10 tons are used annually. Its oral and dermal toxicity is classified as category 3 according to the globally harmonized system of classification and labelling of chemicals, and it is designated a toxic substance by the Ministry of Environment in Korea. There are no available inhalation toxicity data on trichloroacetonitrile. Thus, the present study performed inhalation tests to provide data for hazard and risk assessments. Sprague-Dawley rats were exposed to trichloroacetonitrile at concentrations of 4, 16, or 64 ppm for 6 hour per day 5 days per week for 13 weeks in a repeated study. As a result, salivation, shortness of breath, and wheezing were observed, and their body weights decreased significantly (p < 0.05) in the 16 and 64 ppm groups. All the rats in 64 ppm group were dead or moribund within 4 weeks of the exposure. Some significant changes were observed in blood hematology and serum biochemistry (e.g., prothrombin time, ratio of albumin and globulin, blood urea nitrogen, and triglycerides), but the values were within normal physiological ranges. The major target organs of trichloroacetonitrile were the nasal cavity, trachea, and lungs. The rats exposed to 16 ppm showed moderate histopathological changes in the transitional epithelium and olfactory epithelium of the nasal cavity. Nasal-associated lymphoid tissue (NALT) and respiratory epithelium were also changed. Respiratory lesions were common in the dead rats that had been exposed to the 64 ppm concentration. The dead animals also showed loss of cilia in the trachea, pneumonitis in the lung, and epithelial hyperplasia in the bronchi and bronchioles. In conclusion, the no-observed-adverse-effect level (NOAEL) was estimated to be 4 ppm. The main target organs of trichloroacetonitrile were the nasal cavity, trachea, and lungs.

해조가 2,4,6-trinitrobenzene-sulfonic acid로 유발된 염증성 장질환 동물모델에 미치는 영향 (Effects of Sargassumpallidum on 2,4,6-Trinitrobenzene Sulfonic Acid-Induced Colitis in Mice)

  • Objectives : The aim of the current study was to investigate the effects of Sargassum (Sargassum pallidum (TURN.) C. AG.; SP) on the experimental colitis induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS) in mice. Methods : ICR mice were divided into 7 groups (NOR, CON, $SS50\times5$, $SP20\times3$, $SP50\times3$, $SP20\times5$, $SP50\times5$). TNBS processing was intrarectally applied to all experimental groups on the 3rd experiment day, except the normal group (NOR). For investigating the prophylactic effect, SP at doses of 20 mg/kg ($SP20\times5$) and 50 mg/kg ($SP50\times5$) were orally administered for 5 days. The SP at doses of 20 mg/kg ($SP20\times3$) and 50 mg/kg ($SP50\times3$) were orally administered for 3 days after the colitis induction in order to check the effect of treatment. As a positive control group, sulfasalazine 50 mg/kg ($SS50\times5$) was administrated. Macroscopic findings of epithelial tissue on mice were measured by colon length and macroscopic score. Histologic findings were also checked by crypt cell, epithelial cell, inflammatory cell and edema of submucosa. We measured the ability of SP to inhibit lipid peroxidation and myeloperoxidase activity. We also measured levels of the inflammatory markers, interleukin (IL)-$1\beta$ and cyclooxygenase-2 (COX-2), its transcription factor activation, phospho-NF-${\kappa}B$ (pp65), in the colon by enzyme-linked immunosorbent assay and immunoblot analysis. We measured activation of fecal bacterial enzyme, $\beta$-glucuronidase and degradation activation of fecal glycosaminoglycan (GAG), and hyaluronic acid. Results : Oral administration of SP on mice inhibited TNBS-induced colon shortening and myeloperoxidase activity in the colon of mice as well as IL-$1\beta$ and COX-2 expression. SP also inhibited TNBS-induced lipid peroxidation and pp65 activation in the colon of mice. SP inhibited $\beta$-glucuronidase activation and fecal hyaluronic acid degradation activation as well. Conclusions : SP could be a possible herbal candidate and preventive prebiotic agent for treating inflammatory bowel disease (IBD). Further experiments to differentiate effects of SP on IBD, such as other solutions and extracting times, might be promising.

Bombina orientalis 피부손상 후 과립선 재생에 관한 미세구조적 연구 (Fine Structural Approach of Granular Gland Regeneration after Skin Injury in Bombina orientalis)

  • 두꺼비 등 피부손상 후 과립선의 재생과정을 주사전자현미경과 투과전자현미경으로 관찰하였다. 절개에 의하여 피부손상을 가한 후 실험을 위하여 특수히 제작된 cage 내에서 최대 20일간 사육하였다. 투과전자현미경 관찰에서 손상 4일 후 미성숙 형태의 신생 과립선이 관찰되었으며, 상피세포는 신생 과립선의 첨단부로 이동하여 있었다. 상피세포의 표면은 편평하였으며 desmosome 에 의해 서로 연결되어 있지 않았다. 미토콘드리아를 많이 함유한 세포돌기 (MRC)들이 선의 내강을 형성하고 있었고, 이들 돌기에서는 hemidesmosome이 관찰되었다. 신생선의 기저강은 MRC, 과립형성전세포 및 과립형성 세포 등으로 이루어져 있었다. 특히, 손상 후 10일에 xanthophore가 상피세포의 기저부로 이동하여 있음이 관찰되었다. 이들 세포는 다수의 크기가 큰 pterinosome 과 중등도의 전자밀도를 가진 carotenoid vesicle을 포함하고 있었다. 손상 후 13일에, xanthophore는 많은 carotinoid vesicle과 lammelated pterinosomes을 포함하고 있었다. Iridophore는 손상 16일에 분화중인 xanthophore 주변에서 관찰되었다. 이러한 소견은 손상으로부터의 회복 과정에 선조세포(glandular precursor cell)로부터 과립선이 재생되며, 선세포의 팽대는 이들 신생 세포의 성숙 및 증식에 의한 것을 의미한다.

Er;YSGG 레이저를 이용한 치은 색소침착 제거 증례보고 (Gingival pigmentation treatment using Er;YSGG laser)

  • 치은의 매력 필수 요소로는 잇몸의 색상, 형태 그리고 치아와 잇몸 경계부의 형태와 위치 등에 따라 아름다운 모습인지 아닌지에 따라 결정된다. 치은의 일반적인 아름다움과 균형감 그리고 건강하다는 기준을 받아 들이는 것은 모두 다르지만 치은의 색상 만큼은 분홍빛의 건강한 치은 색상이 검거나 갈색의 치은 보다는 좀 더 심미적이라고 생각하는 기준은 모두 비슷하다고 할 수 있다. 비교적 간단한 소수술로 검거나 갈색인 잇몸의 색을 아름다운 핑크색으로 바꾸는 치은착색 제거술을 진행한다면 좀 더 자신 있는 미소를 가질 수 있도록 도움을 줄 수 있다. 건강한 잇몸의 색은 연분홍색에서 진한 푸르스름한 보라색까지 다양하다. 이러한 정상적인 한계 사이에는 주로 혈관 공급, 상피 두께, 각질화 정도, 상피 내 색소의 존재에 따라 많은 색 변화가 관찰된다. 멜라닌, 카로틴, 감소된 헤모글로불린, 옥시헤모글로불린은 구강 점막의 정상적인 색상에 기여하는 주요 색소들이다. 잇몸의 건강과 형태는 매력적인 미소의 필수 요소다. 빈번하게 발생되는 착색증은 상피의 기저층과 초기저층에 있고 멜라노솜의 형태로 저장되는 멜라닌 침전물이 과도하게 발생하기 때문이다. 이런 착색을 제거하는 술식은 매우 다양하지만 본 증례에서는 Er;YSGG 레이저를 이용한 치은 색소침착을 제거하였으며 다소 임상적인 경험을 얻어 이에 공유 하는 바이다.

9세 소아에서 나타난 비구개낭의 치험례 (CASE REPORT : A NASOPALATINE DUCT CYST IN 9-YEAR-OLD CHILD)

  • 비구개낭은 절치관낭이라고도 불리우며 가장 흔한 비치성낭이다. 발육성으로 비구개관내에서 태생 상피잔사가 증식하여 낭변성을 일으킴으로써 발생한다. 비구개낭은 악골낭의 약 1.7-11.9%를 차지하며 남자에게서 악 3배 정도 호발한다. 연령 분포는 매우 광범위하나 대개 40대에서 60대 사이에서 발견되며 10세 이전에 발견되는 경우는 매우 드물다. 비구개낭을 위한 적절한 치료는 적출이며 비구개신경에 손상을 가하지 않도록 해야 한다. 낭이 커서 치아의 생활력에 영향을 주거나 비구강누공이나 구강 상악동 누공을 형성 할 우려가 있을 경우에는 조대술로 치료할 수 있다. 본 증례의 환아는 구개부의 무통성 종창을 주소로 내원하여 임상 및 방사선 검사를 실시한 결과 소아에서 매우 드물게 나타나는 비구개낭으로 방사선사진에 $32{\times}25.5mm$크기를 나타내었으며, 매우 팽창되어 순측 피질골판을 관통하고 상순소대 하방과 전정부위까지 종창되어 있었다. 병소와 인접한 상악 4전치 치근은 심하게 이개되어 있었다. 낭의 크기가 매우 커서 조대술을 시행하였으며 obturator를 장착하고 있으며 향후 계속 관찰이 요한다.

치주인대섬유모세포의 분화과정에서 아미노산 수송계 L의 발현 (Expression of amino acid transport system L in the differentiation of periodontal ligament fibroblast cells)

  • The periodontium is a topographically complex organ consisting of epithelial tissue, soft and mineralized tissues. Structures comprising the periodontium include the gingiva, periodontal ligament (PDL) , cementum and the alveolar bone. The molecular mechanism of differentiation in PDL fibroblast cells remain unclear. Amino acid transporters play an important role in supplying nutrition to normal and cancer cells and for cell proliferation. Amino acid transport system L is a major nutrient transport system responsible for the Na+-independent transport of neutral amino acids including several essential amino acids. The system L is divided into two major subgroups, the L-type amino acid transporter 1 (LAT1) and the L-type amino acid transporter 2 (LAT2). In this study, the expression pattern of amino acid transport system L was, therefore, investigated in the differentiation of PDL fibroblast cells. To determine the expression level of amino acid transport system L participating in intracellular transport of amino acids in the differentiation of PDL fibroblast cells, it was examined by RT-PCR, observation of cell morphology, Alizaline red-S staining and uptake analysis after inducing experimental differentiation in PDL fibroblast cells isolated from mouse molar teeth. The results are as follows. 1. The LAT1 mRNA was expressed in the early stage of PDL fibroblast cell differentiation. This expression level was gradually reduced by differentiation- inducing time and it was not observed after the late stage. 2. The expression level of LAT2 mRNA was increased in time-dependent manner during differentiation induction of PDL fibroblast cells. 3. There was no changes in. the expression level of 4F2hc mRNA, the cofactor of LAT1 and LAT2, during differentiation of PDL fibroblast cells. 4. The expression level of ALP mRNA was gradually increased and the expression level of Col I mRNA was decreased during differentiation of PDL fibroblast cells. 5. The L-leucine transport was reduced by time from the early stage to the late stage in PDL fibroblast cell differentiation. As the results, it is considered that among neutral ammo acid transport system L in differentiation of PDL fibroblast cells, the LATl has a key role in cell proliferation in the early stage of cell differentiation and the LAT2 has an important role in the late stage of cell differentiation for providing cells with neutral amino acids including several essential amino acids.
