A Method for Selecting Software Reliability Growth Models Using Partial Data (부분 데이터를 이용한 신뢰도 성장 모델 선택 방법)

  • Software Reliability Growth Models (SRGMs) are useful for determining the software release date or additional testing efforts by using software failure data. It is not appropriate for a SRGM to apply to all software. And besides a large number of SRGMs have already been proposed to estimate software reliability measures. Therefore selection of an optimal SRGM for use in a particular case has been an important issue. The existing methods for selecting a SRGM use the entire collected failure data. However, initial failure data may not affect the future failure occurrence and, in some cases, it results in the distorted result when evaluating the future failure. In this paper, we suggest a method for selecting a SRGM based on the evaluation goodness-of-fit using partial data. Our approach uses partial data except for inordinately unstable failure data in the entire failure data. We will find a portion of data used to select a SRGM through the comparison between the entire failure data and the partial failure data excluded the initial failure data with respect to the predictive ability of future failures. To justify our approach this paper shows that the predictive ability of future failures using partial data is more accurate than using the entire failure data with the real collected failure data.

Forging Die Design for Vent Forming of Square Cup Battery Case (사각 컵 배터리 케이스 바닥 벤트 성형을 위한 단조 금형 설계)

  • The demand for electric motor fuel cells has surged in the automotive industry, leading to a recent increase in the demand for square aluminum cans used as fuel cell battery casings. The air vent located on the bottom of the rectangular battery casing prevents large explosions by intermittent pressure release prior to the accumulation of abnormally high pressures. Conventionally, the square cup battery casing is produced via six-step deep drawing, with the outer shape of the vent being manufactured by welding to the square battery casing. On the other hand, this study directly incorporated the air vent outlet into the bottom surface of the rectangular casing. The product of a coupled finite element analysis technique applying the thickness and contour generated from the square cup multi-step deep drawing formation analysis was used as the forging input shape. The results yielded increased prediction accuracy and the advanced prediction of defects, such as swelling and fracture. Based on the results of the initial analyses, two of the generated forging shapes were determined to be suitable, with the optimal forging shape being determined by molding analysis. The results presented here were validated by mold fabrication and a subsequent comparison of the actual and analytical results.

  • The composting is a biological process that converts organic matter into useful resources such as fertilizers. It is a continuous transition of microbial communities to adapt changes in organic matter and environmental conditions (carbonation rate, temperature, humidity, oxygen supply, pH, etc.). Most of the composting plants are located in the proximity of the residential areas. It is a general scenario where government authorities receive complaints from the local residents due to release of odor from the composting, and has become a social problem in Korea. Identification of dominant microorganisms, understanding change in microbial communities and augmentation of specific microorganism for composting is vital to enhance the efficiency of composting, quality of the compost produced, and reduction of odor. In this paper, we suggest the optimum operation conditions and methods for compost depot to reduce odor generation. The selection of the appropriate microorganisms and their rapid increase in population are effective to promote composting. The optimal growth conditions of bacteria such as aeration (oxygen), temperature, and humidity were standardized to maximize composting through microbial degradation. The use of porous minerals and moisture control has significantly improved odor removal. Recent technologies to reduce odor from the composting environment and improved composting processes are also presented.

Hypoallergenic and Physicochemical Properties of the A2 β-Casein Fractionof Goat Milk

  • Goat milk has a protein composition similar to that of breast milk and contains abundant nutrients, but its use in functional foods is rather limited in comparison to milk from other sources. The aim of this study was to prepare a goat A2 ${\beta}$-casein fraction with improved digestibility and hypoallergenic properties. We investigated the optimal conditions for the separation of A2 ${\beta}$-casein fraction from goat milk by pH adjustment to pH 4.4 and treating the casein suspension with calcium chloride (0.05 M for 1 h at $25^{\circ}C$). Selective reduction of ${\beta}$- lactoglobulin and ${\alpha}_s$-casein was confirmed using sodium dodecyl sulphate-polyacrylamide gel electrophoresis and reverse-phase high-performance liquid chromatography. The hypoallergenic property of A2 ${\beta}$-casein fraction was examined by measuring the release of histamine and tumor necrosis factor alpha from HMC-1 human mast cells exposed to different proteins, including A2 ${\beta}$-casein fraction. There was no significant difference in levels of both indicators between A2 ${\beta}$-casein treatment and the control (no protein treatment). The A2 ${\beta}$-casein fraction is abundant in essential amino acids, especially, branched-chain amino acids (leucine, valine, and isoleucine). The physicochemical properties of A2 ${\beta}$-casein fraction, including protein solubility and viscosity, are similar to those of bovine whole casein which is widely used as a protein source in various foods. Therefore, the goat A2 ${\beta}$-casein fraction may be useful as a food material with good digestibility and hypoallergenic properties for infants, the elderly, and people with metabolic disorders.

  • To obtain basic information for the genetic analysis and breeding of Flammulina velutipes, some factors affecting the release of protoplasts from the fungus were studied. Potato Dextrose peptone Agar medium was suitable for the growth of the mycelium and the protoplast formation of F. velutipes. The culture age for the high yields of protoplast was 5 days on PDPA. Few protoplasts were formed from the mycelium cultured on Mushroom minimum Media. The highest yield of protoplasts was obtained in enzyme solution containing Novozyme 234 plus cellulase CP at 10 mg $ml^{-1}$ concentration, while a half amount of protoplasts was obtained in enzyme solution containing Novozyme 234 only. The optimal reaction time of the mycelium in the Iytic enzyme mixtures was 3 hours. The best osmotic stabilizer for the protoplast formation of the mycelium was 0.6M sucrose without buffer at pH 6.2.

Characteristics and Drug Release Profiles of Multilamellar Vesicle(MLV) and Microemulsified Liposome(MEL) Entrapped 5-Fluorouracil and Its derivatives (5-Fluorouracil과 그 유도체를 봉입한 Multilamellar Vesicle(MLV)과 Microemulsified Liposome(MEL)의 특성 및 약물방출 거동)

  • Although liposome has many advantages as a pharmaceutical dosage form, its application in the industrial field has been limited because of some problems such as preparation method, reproducibility, scale-up, stability and sterilization etc. Liposomes prepared by microemulsification method had defined size, narrow size distribution, reproducibility and high entrapment efficiency. For enhancing the stability, the dry form of liposome was recommended. These types of liposome are proliposome and freeze-dried liposome. The liposome must have some properties for preparing of freeze-dried liposome; small size $(50{\sim}200\;nm)$, narrow size distribution and cryoprotectant. In this experiment, the liposomes containing 5-Fluorouracil(5-FU) and its prodrug(pentyl-5-FU-1-acetate; PFA, hexyl-5-FU-1-acetate; HFA) were made with soybean phosphatidylcholine, cholesterol, stearylamine(SA) and dicetyl phosphate(DCP) employing hydration method or microemulsification method using $Microfluidizer^{TM}$. Both or liposome types were MLV and MEL. After preparation, freeze drying and rehydration were performed. In the process of freezing, trehalose(Tr) was added as a cryoprotectant. Their evaluation methods were as follows; entrapment efficiency, mean particle size and size distribution, dissolution test, retain of entrapment efficiency and turbidity after freeze-drying. The results are summarized as belows. The entrapment efficiency of 5-FU was dependent on total lipid concentration and cholesterol content but that of PFA and HFA was decreased when cholesterol was added. When DCP and SA were added, entrapment efficiency was decreased. As the partition coefficient of drug was increased, entrapment efficiency was increased. Under the same condition, entrapment efficiency of MEL is similar to that of MLV. The mean particle size and size distribution of MEL were smaller than those of MLV. Dissolution rates of drug from both liposome types were comparatively similar. Dissolution rates of drugs with serum and liver homogenate were faster than without these material. After preparation of liposome, free drug was removed efficiency by Dowex 50W-X4. When liposome was freeze-dried and then rehydrated in the presence of Tr, characteristics of liposome were maintained well in MEL than MLV. Tr Was used successfully as a cryoprotectant in the process of freeze drying and the optimal ratio of Tr:Lipid was 4:1(g/g).

Optimal Culture Conditions and XAD Resin on Tropane Alkaloid production in Scopolia parviflora Hairy Root Cultures (미치광이풀 모상근의 배양조건 구명 및 XAD Resin 처리에 의한 Tropane Alkaloid 생산)

  • The optimum culture conditions for tropane alkaloid production in hairy root cultures of Korea native Scopolia paviflora Nak. were investigated. Hairy root was induced from the rhizome of the mother plant on B5 medium containing 1.0 mg/L IBA. Among the culture media examined, 1/2 B5 medium was the best for tropane alkaloid production, whereas the growth of hairy root increased in SH medium. The best result on the growth of hairy root was obtained in 1.0 mg/L NAA, and tropane alkaloid production was obtained in plant growth regulator-free medium. Of the carbone sources tested, 3% sucrose promoted the growth of hairy root, whereas 5% sucrose increased tropane alkaloid production. Optimum inoculum densities for root growth and tropane alkaloid production were 0.5 g and 1 g, respectively. The addition of XAD resins (1 % w/v) to hairy root cultures led to increases in tropans alkaloid production, and the release of alkaloid into the medium and its adsorption by the resin accounted for about 50 to 80% of total production. It is concluded that optimized culture conditions and the addition of XAD resins could be used in the development of a bioprocess for tropane alkaloid production in hairy root cultures of S. paviflora Nak.

Optimum Concentration of Alizarin Complexone (ALC) for Otolith Marking of Pacific Cod Gadus macrocephalus (대구 Gadus macrocephalus 이석 표지를 위한 Alizarin Complexone의 최적 농도)

  • Optimum concentration of alizarine complexone (ALC) for staining of otoliths for Pacific cod was determined by examination of stained quality, growth and survival rates. Small juveniles ($34.1{\pm}1.8mm$ TL, $mean{\pm}SD$) were immersed in four different concentrations (20, 40, 60, 80 mg/L) and large ones ($73.8{\pm}1.2mm$ TL, $mean{\pm}SD$) in five different concentrations (1, 5, 10, 20, 40 mg/L) of ALC for 24 hrs. The fish were then reared and observed for 14 days. The stained quality of otoliths was evaluated by dividing them into four grades by the presence of fluorescent stains on the otoliths under G-excited UV light. The small juveniles stained with the different ALC concentrations had 100% staining success, and their growth rate and body weight did not differ significantly between treatments. Optimal ALC concentration for the smaller juveniles was 20 mg/L in the present experiment. On the other hand, distinct fluorescent rings were confirmed on otoliths of larger juveniles in the 10 mg/L treatment with high survival rate. These results suggest that ALC staining at concentrations of 10 mg/L with larger juveniles will be helpful for release-recapture experiments for stock enhancement of Pacific cod.

Effects of rhBMP-2 with various carriers on bone regeneration in rat calvarial defect (백서 두개골 결손에서 rhBMP-2와 다양한 carrier의 골재생 유도효과)

  • Purpose: Bone morphogenetic protein (BMP) is a potent differentiating agent for cells of the osteoblastic lineage. It has been used in the oral cavity under a variety of indications and with different carriers. However, the optimal carrier for each indication is not known. This study evaluated the bone regenerative effect of rhBMP-2 delivered with different carrier systems. Materials and Methods: 8 mm critical-sized rat calvarial defects were used in 60 male Sprague-Dawley rats. The animals were divided into 6 groups containing 10 animals each. Two groups were controls that had no treatment and absorbable collagen membrane only. 4 groups were experimentals that contained rhBMP-2 only and applied with absorbable collagen sponge($Collatape^{(R)}$), $MBCP^{(R)}$, Bio-$Oss^{(R)}$ each. The histological and histometric parameters were used to evaluate the defects after 2- or 8-week healing period. The shape and total augmented area were stable in all groups over the healing time. Results: New bone formation was significantly greater in the rhBMP-2 with carrier group than control group. rhBMP-2/ACS was the highest in bone density but gained less new bone area than rhBMP-2/$MBCP^{(R)}$ and rhBMP-2/Bio-$Oss^{(R)}$. The bone density after 8 weeks was greater than that after 2 weeks in all groups. However, rhBMP-2 alone failed to show the statistically significant difference in new bone area and bone density compared to control group. Also $MBCP^{(R)}$ and Bio-$Oss^{(R)}$ particles remained after 8 weeks healing period. Conclusion: These results suggest that rhBMP-2 with carrier system is an excellent inductive agent for bone formation and we can use it as the predictable bone tissue engieering technique. Future study will likely focus on the kinetics of BMP release and development of carriers that is ideal for it.

Acid Tolerance of Lactobacillus brevis Isolated from Kimchi (김치에서 분리한 Lactobacillus brevis의 내산성)

  • We isolated wild lactic acid bacteria from kimchi and identified as Lactobacillus brevis by using API 50 CHL Kit, some morphological and physiological tests. In order to evaluate the acid tolerance of Lactobacillus brevis, its survivals rate, glycolysis assay, membrane permeability, and pH profiles of $H^+-ATPase$ were also determined. When Lactobacillus brevis were incubated in Lactobacilli MRS broth adjusted to various levels of pH for 2 hours, the decreases in its population at pH 4.0 and 3.0 were about 2.61 log cycles/mL and 5.89 log cycles/mL, respectively, but there was no decrease at pH 6.0 and 5.0. Glycolysis by Lactobacillus brevis had optimal pH about 6.5 and glucose degradation was reduced by 50% at a pH of 5.2. $Mg^{++}$ release from Lactobacillus brevis cells in medium with pHs of 4.0 and 3.0 was 24.3 and 71.2% of totals, respectively. The $H^+-ATPase$ of Lactobacillus brevis showed a maximal activity between pH values of approximately 6.5 to 7.0.
