This paper describes a new type of replaceable fuse for moment resisting frames. Column-tree connections with beam splice connections are frequently preferred in the moment resisting frames since they eliminate field welding and provide good quality. In the column-tree connections, a part of the beam is welded to the column in the shop and the rest of the beam is bolted with the splice connection in the field. In this study, a replaceable reduced beam section (R-RBS) connection is proposed in order to eliminate welding process and facilitate assembly at the site. In the proposed R-RBS connection, one end is connected by a beam splice connection to the beam and the other end is connected by a bolted end-plate connection to the column. More importantly is that the proposed R-RBS connection allows the replacement of the damaged R-RBS easily right after an earthquake. Pursuant to this goal, experimental and numerical studies have been undertaken to investigate the performance of the R-RBS connection. An experimental study on the RBS connection was used to substantiate the numerical model using ABAQUS, a commercially available finite element software. Additionally, five different finite element models were developed to conduct a parametric study. The results of the analysis were compared in terms of the moment and energy absorption capacities, PEEQ, rupture and tri-axiality indexes. The design process as well as the optimum dimensions of the R-RBS connections are presented. It was also demonstrated that the proposed R-RBS connection satisfies AISC criteria based on the nonlinear finite element analysis results.
In a nuclear reactor, gamma radiation is the dominant energy deposition in non-fuel regions. Heat is generated upon gamma deposition and consequently affects the mechanical and thermal structure of the material. Therefore, the safety of samples should be carefully considered so that their integrity and quality can be retained. To evaluate relevant parameters, an in-core gamma thermometer (GT) was used to measure gamma heating (GH) throughout the operation of the McMaster nuclear reactor (MNR) at four irradiation sites. Additionally, a Monte Carlo reactor physics code (Serpent-2) was utilized to model the MNR with the GT located in the same irradiation sites used in the measurement to verify its predictions against measured GH. This research aids in the development of modeling, calculation, and prediction of the GH utilizing Serpent-2 as well as implementing a new GH measurement at the MNR core. After all uncertainties were quantified for both approaches, comparable GH profiles were observed between the measurements and calculations. In addition, the GH values found in the four sites represent a strong level of radiation based on the distance of the sample from the core. In this study, the maximum and minimum GH values were found at 0.32 ± 0.05 W/g and 0.15 ± 0.02 W/g, respectively, corresponding to 320 Sv/s and 150 Sv/s. These values are crucial to be considered whenever sample is planned to be irradiated inside the MNR core.
Interferon regulatory factor 3 (IRF3) integrates both immunological and non-immunological inputs to control cell survival and death. Small GTPases are versatile functional switches that lie on the very upstream in signal transduction pathways, of which duration of activation is very transient. The large number of homologous proteins and the requirement for site-directed mutagenesis have hindered attempts to investigate the link between small GTPases and IRF3. Here, we constructed a constitutively active mutant expression library for small GTPase expression using Gibson assembly cloning. Small-scale screening identified multiple GTPases capable of promoting IRF3 phosphorylation. Intriguingly, 27 of 152 GTPases, including ARF1, RHEB, RHEBL1, and RAN, were found to increase IRF3 phosphorylation. Unbiased screening enabled us to investigate the sequence-activity relationship between the GTPases and IRF3. We found that the regulation of IRF3 by small GTPases was dependent on TBK1. Our work reveals the significant contribution of GTPases in IRF3 signaling and the potential role of IRF3 in GTPase function, providing a novel therapeutic approach against diseases with GTPase overexpression or active mutations, such as cancer.
Concrete-filled steel tubes (CFSTs) nowadays are widely used as the main parts of momentous structures, and its connection has gained increasing attention as the complexity in configuration and load transfer mechanism. This paper proposes a novel CFST pier-to-footing incorporating tube-confined RC encasement. Such an innovative approach offers several benefits, including expedited on-site assembly, effective confinement, and collision resistance and corrosion resistance. The seismic behavior of such CFST pier-to-footing connection was studied by testing eight specimens under quasi-static cyclic lateral load. In the experimental research, the influences on the seismic behavior and the order of plastic hinge formation were discussed in detail by changing the footing height, axial compression ratio, number and length of anchored bars, and type of confining tube. All the specimens showed sufficient ductility and energy dissipation, without significant strength degradation. There is no obvious failure in the confined footing, while local buckling can be found in the critical section of the pier. It suggests that the footing provides satisfactory strength protection for the connection.
Since molecular structure of hnRNP is not available in foreseeable future, it is best to construct a working model for hnRNP structure. A geometric problem, assembly of $700{\pm}20$ nucleotides with 48 proteins, is visualized by a frame work in which all the proteins participate in primary binding, followed by secondary, tertiary and quaternary binding with neighboring proteins without additional import. Thus, 40S hnRNP contains crown-like secondary structure (48 stemloops) and appearance of 6 petal (octamers) rose-like architectures. The proteins are wrapped by RNA. Co-transcriptional folding for RNP fibril of FMR1 gene can produce 2,571 stem-loops with frequency of 1 stem-loop/15.3 nucleotides and 53 40S hnRNP beaded structure. By spliceosome driven reactions, there occurs removal of 16 separate lariated RNPs, joining 17 separate beaded exonic structures and anchoring EJC on each exon junction. Skipping exon 12 has 5'GU, 3'AG and very compact folding pattern with frequency of 1 stem-loop per 12 nucleotides in short exon length (63 nucleotides). 5' end of exon 12 contains SS (Splicing Silencer) element of UAGGU. In exons 10, 15 and 17 where both regular and alternative splice sites exist, SS (hnRNP A1 binding site) is observed at the regular splicing site. End products are mature FMR-1 mRNP, 4 species of Pri-microRNAs derived from introns 7,9,15 and 3'UTR of exon17, respectively. There may also be some other regulatory RNAs containing ALU/Line elements as well.
Minimum Letter Flips(MLF) and Weighted Minimum Letter Flips(WMLF) can perform the haplotype reconstruction more accurately from SNP fragments when they have many errors and gaps by introducing the related genotype information. And it is known that WMLF is more accurate in haplotype reconstruction than those based on the MLF. In the paper, we analyze two models under the conditions that the different rates of homozygous site in the genotype information and the different confidence levels according to the sequencing quality. We compare the performance of the two models using neural network and genetic algorithm. If the rate of homozygous site is high and sequencing quality is good, the results of experiments indicate that WMLF/GI has higher accuracy of haplotype reconstruction than that of the MCIH especially when the error rate and gap rate of SNP fragments are high.
Pipe pile walls are commonly used as retaining structures for excavation projects, particularly in densely populated coastal cities such as Hong Kong. Pipe pile walls are preferred in reclaimed land due to their cost-effectiveness and convenience for installation. However, the pre-bored piling techniques used to install pipe piles can cause significant ground disturbance, posing risks to nearby sensitive structures. This study reports a well-documented case history in a reclamation site, and it was found that pipe piling could induce ground settlement of up to 100 mm. Statutory design submissions in Hong Kong typically specify a ground settlement alarm level of 10 mm, which is significantly lower than the actual settlement observed in this study. In addition, lateral soil movement of approximately 70 mm was detected in the marine deposit. The lateral soil displacement in the marine deposit was found to be up to 3.4 and 3.1 times that of sand fill and CDG, respectively, mainly due to the relatively low stiffness of the marine deposit. Based on the monitoring data and site-investigation data, a 3D numerical analysis was established to back-analyze soil movements due to the installation of the pipe pile wall. The comparison between measured and computed results indicates that the equivalent ground loss ratio is 20%, 40%, and 20% for the fill, marine deposit and CDG, respectively. The maximum ground settlement increases with an increase in the ground loss ratio of the marine deposit, whereas the associated influence radius remains stationary at 1.2 times the pipe pile wall depth (H). The maximum ground settlement increases rapidly when the thickness of marine deposit is less than 0.32H, particularly for the ground loss ratio of larger than 40%. This study provides new insights into the pipe piling construction in reclamation sites.
Irradiation of gold nanorods (GNRs) with laser light corresponding to the longitudinal surface plasmon oscillation results in rapid conversion of electromagnetic energy into heat, a phenomenon commonly known as the photothermal effect of GNRs. Herein, we propose a facile strategy for increasing the photothermal conversion efficiency of GNRs by integration to form graphene oxide (GO) nanocomposites. Moreover, conjugation of iron oxide (IO) with the GO-GNR nanohybrid allowed magnetic enrichment at a specific target site and the separated GO-IO-GNR assembly was rapidly heated by laser irradiation. The present GO-IO-GNR nanocomposites hold great promise for application in various biomedical fields, including surface enhanced Raman spectroscopy imaging, photoacoustic tomography imaging, magnetic resonance imaging, and photothermal cancer therapy.
Since the year 2008, the first demonstration project of the superconducting cable for the KEPCO grid operation has been in progress. To apply the HTS (High Temperature Superconducting) cable system into the commercial network, a new test complex was built in the 154kV Icheon substation and the on site assembly of 22.9kV, 50MVA HTS cable with the cryogenic refrigeration system was finished. A full scale of HTS cable fabricated with 2nd generation wire is to be in service in KEPCO network. In this paper, a series of processes of the installation and operation test are introduced.
