Evaluation of DNA Markers for Fruit-related Traits and Genetic Relationships Based on Simple Sequence Repeat in Watermelon Accessions

  • Modern watermelon cultivars (Citrullus lanatus [Thunb.] Matsum.& Nakai var. lanatus) have fruits with diverse phenotypes, including fruit shape, rind patterns, and flesh color. Molecular markers enable efficient selection of plants harboring desirable phenotypes. In the present study, publicly available DNA markers tightly linked to fruit shape, rind stripe pattern, and flesh color were evaluated using 85 watermelon accessions with diverse fruit phenotypes. For fruit shape, the dCAPS SUN - Cla011257 marker revealed an 81% of marker - trait match for accessions with elongated or round fruits. For rind stripe pattern, the SCAR wsb6-11marker was effective for selecting Jubilee-type rind pattern from other rind patterns. For flesh color, the Clcyb.600 and Lcyb markers derived from a mutation in the Lycopene ${\beta}$ - cyclase (Lcyb) gene, were effective at selecting red or yellow flesh. Forty-eight accessions possessing diverse fruit - related traits were selected as a reference array and their genetic relationships assessed using 16 SSR markers. At a coefficient of 0.11, the 48 accessions grouped into two major clades: Clade I and Clade II. Clade I subdivided further into subclades I - 1 and I - 2 at a coefficient of 0.39. All accessions with colored flesh were classified into Clade I, whereas those with white - flesh were classified into Clade II. Differences in fruit traits between subclades I - 1 and I - 2 were observed for rind pattern and fruit color; a majority of the accessions with Crimson-type striped or non-striped rind were grouped together in subclade I - 1, while most accessions in subclade I - 2 had a Jubilee - type rind stripe pattern. These results imply that reference array watermelon accessions possess distinguishable genetic structure based on rind stripe pattern. However, no significant grouping pattern was observed based on other fruit-related traits.

Study on the Electrical Characterization of Inverted Staggered Pentacene Thin Film Transistor using Hydrogen Plasma Treatment (수소 플라즈마 처리를 이용한 역스테거드형 펜타센 트랜지스터의 전기적 특성 향상에 대한 연구)

  • In order to reach the high electrical quality of organic thin film transistors (OTFTs) such as high mobility and on-off current ratio, it is strongly desirable to study the enhancement of electrical properties in OTFTs. Here, we report the novel method of hydrogen plasma treatment to improve electrical properties in inverted staggered OTFTs based on pentacene as active layer. To certify the effect of this method, we compared the electrical properties of normal device as a reference with those of device using the novel method. In result, the normal device as a reference making no use of this method exhibited a field effect mobility of 0.055 $\textrm{cm}^2$/Vs, on/off current ratio of 10$^3$, threshold voltage of -4.5 V, and subthreshold slope of 7.6 V/dec. While the device using the novel method exhibited a field effect mobility of 0.174 $\textrm{cm}^2$/Vs, on/off current ratio of 10$\^$6/, threshold voltage of -0.5 V, and subthreshold slope of 1.49 V/dec. According to these results, we have found the electrical performances in inverted staggered pentacene TFT owing to this method are remarkably enhanced. So, this method plays a key role in highly improving the electric performance of OTFTs. Moreover, this method is the first time yet reported for any OTFTs.

The Effect of Arm Swing on Gait in Healthy Adults (팔 흔들기가 정상인의 보행에 미치는 영향)

  • This study was to performed to get the reference data of the kinetic parameters for normal subjects according to the arm swing type. Forty-five normal subjects($22.62{\pm}2.69years$) pariticpated in this study and preformed 4 sequence according to the arm swing type as follows; first procedure-normal arm swing, second procedure-one arm swing, thrid procedure-no arm swing, fourth procedure-fitness arm swing. There were significant differences according to the arm swing type in the kinetic parameters such as walking speed, hip power, ground reaction force of vertical in terminal stance phase(p<.05). These finding can be utilized (a) as a reference for kinetic data of gait analysis in normal subjects, and (b) as an aide in evaluating and treating patients who have problems relating to gait.

A Determination of Bias between Calculational Methods for the Criticality Safety Analysis of Spent Fuel Storage Pool with Burnup Credit (연소를 고려한 사용후핵연료저장조 핵임계 안전성분석에서 계산체제간의 편차결정)

  • A test is made for a method to determine reliable bias in the criticality safety analysis of spent fuel storage pool with turnup credit between the reference and rack criticality calculation methods. The spent fuel pool of Kori Unit 1 is conceptually redesigned to the most compact rack with turnup credit, and its multiplication factors are calculated depending on fuel enrichment and burnup, by the Monte Carlo code KENO-IV as a reference and by a two-dimensional collision probability code FATAC as a practical method. Then, the computed values with the help of the above two computer codes are compared to evaluate the bias and its trend in terms of multiplication factor on fuel enrichment and turnup. The result indicates that the bias can be determined with reliability basis but without any disadvantage in criticality safety margin compared with the conventional method.

Objective evaluation of the color of tongue substance using L*a*b* color coordinates

  • The purpose of this study was to analyze whether quantitative evaluation of the color of the tongue substance using $L^*a^*b^*$ color coordinates system could minimize the problems arising from the different illuminating conditions or not. In controlled 4 different illuminating conditions (by natural light, flashlight, f-number, shutter speed),12 healthy subjects were photographed of their tongue substance through a digital camera (C-2100uz, Olympus Co.), both on the top surface and on the bottom surface of the tongue substance by two examiners, twice at 3 day intervals. Clinician evaluation was also performed grading the redness of the tongue substance in the form of 5-points scale by 6 clinicians. As a result, there was no significant difference in color differences between the color of the tongue substance and the reference red card in the 4 different illuminating conditions. Intra-rater reliability was satisfied and even though limitedly, inter-rater reliability was satisfied. Color differences were significantly correlated with the results by the clinicians, although they were applicable limitedly to specific illuminating conditions. Our results indicate that the application of the color differences in tongue diagnosis could not only evaluate the color information quantitatively, but also minimize the problems arising from the different illuminating conditions and that there was the significant difference in the visual evaluation of the red color of the tongue substance, both between the clinicians and between the illuminating conditions.

Studies on the Phosphate Metabolism in Chlorella, with Special Reference to Polyphosphate (Chlorella의 인산대사에 관한 연구)

  • Yung Nok Lee (Dept. of Biology, Korea University) : Studies on the phosphate metabolism in Chlorella, with special reference to polyphosphate. Kor. J. Microbiol., Vol.2, No.1, p1-11 (1964). 1. Uniformly $^{32}P$-labeled Chlorella cells which were irradiated with Cobalt-60 gamma-rays of about 70, 000 $\gamma$ dose, were further grown in a standard "cold" medium ("hot".rarw."cold"), and some portions of the algae were taken out at the begining of, and at intervals during the culture, and subjected to analyze the contents of $^{32}P$- and total P in various fractions of the cell materials. Results obtained were compared with those of nonirradiated normal cells. 2. Amounts of phosphate in various fractions of the nonirradiated normal Chlorella cells were measured using uniformly $^{32}P$--labeled cells. Analysis of the $^{32}P$--labeled algal cells showed that the highest value in P-content was the fraction of RNA followed by those of lipid, polyphosphate "C" polyphosphate "B", DNA, nucleotidic labile phosphate compounds, polyphosphate "A" and protein. It was observed that content of total polyphosphates in a single Chlorella cell was almost equal to RNA-P content in the cell, and the amount of RNA-P was almost equal to ten times of DNA-P content. 3. When the $^{32}P$--labeled algae which were irradiated with gamma-rays were grown in a normal "cold" medium, phosphate contents in the fraction of DNA, nucleotidic labile phosphate compounds and protein decreased markedly, while the contents of phosphate in the fractions of polyphosphate "C" and potyphosphate "B" increased in comparison with those of unirradiated normal cells. So, it was considered that the pretreatment of above mentioned dose of gamma-ray inhibited DNA and protein synthesis from polyphosphate in Chlorella cells. 4. Proceeding the culture of $^{32}P$--labeled Chlorella in a "cold" standard medium, whose synthetic activity of DNA and protein from polyphosphate was disturded by gamma-ray irradiation, the amounts of $^{32}P$-in the fraction of polyphosphate "C" increased, in contrast with those of polyphosphate "B" fraction. A LATERAL CEPHALOMETRIC STUDY OF THE SIZE OF TONGUE AND INTERMAXILLARY SPACE IN KOREAN (한국인 설과 악간극의 크기에 관한 방사선학적 연구)

  • A study was performed to investigate the size of tongue area and intermaxillary space area, and compare the sexual differences between normal Korean children and adults by introducing planimetric and linear analysis of the lateral cephalograms. The cephalograms were composed of 41 child male aged 10.8, 40 child female aged 10.5, 38 adult male aged 21.3 and 40 adult female aged 20.8 respectively. In order to study and measure the intermaxillary space area, the followings were selected, as reference items: occlusal plane, anterior intermaxillary space height, posterior intermaxillary space height, length of intermaxillary space. Among those reference items anterior intermaxillary space height and posterior intermaxillary space height were perpendicular to the maxillary plane. An. index, (equation omitted) While the tongue area was plotted by outline of tongue shadow, above a line extending from the vallecula to the most anterior point on the hyoid body, and above a line from the most anterior point of the hyoid body to the menton. The obtained results were as follows: 1. In general the measurements of male were larger than those of female in intermaxillary space area in childhood and adulthood group. but intermaxillary space area of childhood group showed no significant sexual difference, and that of adulthood group showed significant sexual difference when evaluated statistically. 2. In both groups the measurements of male were larger than those of female in tongue area and there are also statistical significance of sexual differences in both age groups. 3. Considerable growh changes between the childhood and adulthood groups were revealed in intermaxillary space area and tongue area, and the tongue had tendency to become relatively smaller when compared with the intermaxillary space in both sex.

Normal Walking Versus Toe-walking in Healthy Subjects: An Electromyographic Analysis (정상 보행과 발가락 보행의 하지 근육 근 활성도 비교)

  • This study was designed to identify the effects of walking conditions (normal walking vs. toe-walking) on electromyographic (EMG) activity of gastrocnemius, tibialis anterior, and soleus muscle. Seven healthy adult males participated in this study. The exclusion criteria were orthopedic or neurologic disease, congenital anomaly or acquired deformity, or pain in low back or lower extremities. The maximal voluntary isometric contraction for each muscle was used for the reference contraction, and EMG activity of each muscle during normal walking and toe-walking was expressed as a percentage of reference contraction. The gait cycle was determined with two foot switches, and gait was normalized as 100% gait cycle for each condition. The maximal values of EMG activity in terminal stance (30~50% of gait cycle) of each condition were compared for data analysis. No significant differences were found in EMG activity of the tibialis anterior and soleus (p>.05), whereas significant decrement was found in EMG activity of gastrocnemius during toe-walking compared to normal walking (p<.05). There is a limitation to generalize the results of this study, because small number of subjects participated for this study and only EMG was used for data collection. The treatment methods should be developed to improve gait efficiency by substituting the weakened muscles secondary to upper motor neuron, or by strengthening the distal muscles in lower extremity.

A Analysis of the Relevance between Home Economics and Other Subjects in the Middle School (중학교 가정교과와 다른교과의 교육내용 관련성 분석)

  • The purpose of this study is two-fold. One is to provide the teachers of Home Economics with the basic reference data of the relevance between Home Economics and other subjects taught in the middle schools for the purpose of bringing out more efficient classroom teaching. The other is to help the teachers, the textbook writers, and the curriculum reformers improve the curricula of the middle school on the basis of this research. The conclusions drawn from this study are as follows: 1) The units of Human Development and Family Relationship should be taught with reference to Korean Language, Ethics, and Music. 2) The unit of Adolescence Development is dealt with in Physical Education and in Ethics as well, which seems to indicate that there was no interdisciplinary consideration and cooperation before/when the curricula were made. 3) The unit of Nutrition which appears in the first grade in Home Economics has a close relationship with the unit of Morphology and Physiology of Animals which appears in the second grade in Natural Science. It is thought that the interdisciplinary themes ought to be organized cooperatively among subjects for efficiency of study. 4) Since the unit of Clothing is not dealt with in the other subjects, ti can be developed in Home Economics on its own. 5) As the unit of Housing is closely related to Technology, it should be taught on the basis of Technology. 6) The textbooks of all subjects should be written based on the interdisciplinary research and cooperation. They should be organized in order not to overlap or repeat the same topic in the same level if it is not necessary.

  • The electro-optic properties of polymer-dispersed liquid crystal cells containing BDVE(Butanediol vinyl ether) in PN393 base pre-polymer were examined. The higher the contents of BDVE, the smaller becomes the droplet size. However, the droplet size was saturated around $3{\mu}m$ even at 40 wt% of BDVE. Both of contrast ratio and response time of PDLC cell fabricated with a new formula were found to be superior to the reference cell with PN393 by the factor of 4.9 and 0.15, respectively. However, the new formula made the operating voltage go higher compared to the reference cell of PN393 formula. Except for contrast ratio, response time as well as operating voltage were found to be highly stabilized by adding BDVE in PN393 base pre-polymer over the temperature range of $0{\sim}60^{\circ}C$ studied.
