Effects of Non-protein Energy Intake on Whole Body Protein Synthesis, Nitrogen Retention and Glucose Turnover in Goats

  • The responses of whole body protein and glucose kinetics and of nitrogen (N) metabolism to non-protein energy intake (NPEI) were determined using an isotope dilution approach and measurement of N balance in three adult male goats. The diets containing 1.0, 1.5 and 2.0 times ME maintenance requirement, with fixed intake of CP (1.5 times maintenance) and percentage of hay (33%), were fed twice daily for each 21 d experimental period. After an adaptation period of 11 d, N balance was determined over 3 d. On day 17, whole body protein synthesis (WBPS) and glucose irreversible loss rate (ILR) were determined during the absorptive state by a primed-continuous infusion of [$^2H_5$]phenylalanine, [$^2H_2$]tyrosine, [$^2H_4$]tyrosine and [$^{13}C_6$]glucose, with simultaneous measurements of plasma concentrations of metabolites and insulin. Ruminal characteristics were also measured at 6 h after feeding over 3 d. Nitrogen retention tended to increase (p<0.10) with increasing NPEI, although digestible N decreased linearly (p<0.05). Increasing NPEI decreased (p<0.01) ammonia N concentration, but increased acetate (p<0.05) and propionate (p<0.05) concentrations in the rumen. Despite decreased plasma urea N concentration (p<0.01), increased plasma tyrosine concentration (p<0.05), and trends toward increased plasma total amino N (p<0.10) and phenylalanine concentrations (p<0.10) were found in response to increasing NPEI. Increasing NPEI increased ILR of both glucose (p<0.01) and phenylalanine (p<0.05), but did not affect ($p{\geq}0.10$) that of tyrosine. Whole body protein synthesis increased (p<0.05) in response to increasing NPEI, resulting from increased utilization rate for protein synthesis (p<0.05) and unchanged hydroxylation rate of phenylalanine ($p{\geq}0.10$). These results suggest that increasing NPEI may enhance WBPS and glucose turnover at the absorptive state and improve the efficiency of digestible N retention in goats, with possibly decreased ammonia and increased amino acid absorption. In addition, simultaneous increases in WBPS and glucose ILR suggest stimulatory effect of glucose availability on WBPS, especially when sufficient amino acid is supplied.

  • Processing conditions for dried condiments with oyster, pen shell and cockle shell were investigated. The enzymatic hydrolysis for 3 hours was more profitable than hydrothermal extraction to develop flavoring matters from oyster, pen shell and cockle shell. As a result of omission tests, nucleotides were predominated in the taste compounds of shellfish hydrolysates rather than free amino acids, and the contribution of nucleotides and free amino acids to the taste of shellfish hydrolysates was remarkable. The major flavoring components of shellfish hydrolysates were free amino acids and oligopeptides below 500 dalton. When shellfish hydrolysates were separated with membrane (molecular weight cutoff 500 dalton) for recovering flayer, recovering yields of amino type nitrogen were $92.1\~92.8\%$. Moisture contents of dried shellfish condiments prepared with pretense hydrolyzed oyster, pen shell and cockle shell were $3.5\%,\;3.8\%$ and $3.7\%$, respectively. Contents of total nitrogen were $69.4\%,\;78.8\%$ and $74.2\%$, and those of amino nitrogen were $45.5\%,\;48.9\%$ and $45.4\%$, respectively. Drying yield, solubility and absorption rates at Aw 0.88 were $11.7\%,\;78.4\%$ and $6.8\%$ in oyster, $8.2\%,\;73.6\%$ and $6.1\%$ in pen shell, $9.8\%,\;76.9\%$ and $6.6\%$ in cockle shell, respectively.

  • Oxides of nitrogen (NO$_x$) in exhaust gases was determined by absorbing the gas in alkaline peroxide solution containing 0.03${\%}$ H2O2 and 0.1N NaOH. About 100 ppm of NO$_x$ was rapidly oxidized to NO$_2$ or N$_2$O$_5$ by H$_2$O$_2$ and required a contact time of 2 minutes with the absorbing solution for complete absorption. With vigorous shaking including air or oxygen gas, high concentration of NO$_x$ (>200 ppm) can be absorbed within 30 minutes. The remaining H$_2$O$_2$ affect the absorbance of color solution strongly. However, the excess H$_2$O$_2$ was completely decomposed by zinc powder 0.5g and the sample solution should be adjusted to the pH range 6.1∼6.6 before the reduction so that conversion of nitrate to nitrite ion is possible. The absorbed NO$_x$ is determined colorimetrically by the diazotization-coupling method with sulfonilamide and NEDA as the coupling agent. The sensitivity of the new method was 4.48 ${\times}$ 10$^4$ as molar absorptivity which was high sensitive compared with that obtained for the usual zinc reduction NEDA method with O$_3$. This method was far more rapid, brief and accurate than previously published O$_3$-NEDA method in Korean industrial standard.

Effect of Soil Salinity on Nitrogen Mineralization of Livestock Manure Compost in Salt-Affected Coastal Soils

  • We conducted a short-term incubation experiment in order to understand the effect of the salinity of reclaimed coastal soils on nitrogen mineralization of livestock manure compost (LMC). Two soils with the same soil texture but different EC levels were collected from the same field. These samples were treated with 0%, 1%, 2%, and 3% of LMC by weight basis and incubated at $25^{\circ}C$ to observe changes in inorganic N contents, pH, and dehydrogenase activity with respect to time. As a result, regardless of the soil EC level, as the LMC increased, the total content of the inorganic N ($NH_4{^+}+NO_3{^-}$) increased. Difference in the soil EC level did not affect N mineralization of LMC greatly. The soil EC had negligible effect on the dehydrgenase activity as with the case of inorganic nitrogen. The $NH_4{^+}$ contents remained very low throughout the experimental period starting from the first week of incubation. We believe this is due to the high pH level (pH 7.9 and pH 8.3) of the original soils leading to ammonia volatilization. On the other hand the $NO_3{^-}$ content maintained high level as the LMC treatment level increased and reached maximum at the third week. The pH of the soil during incubation period decreased as the $NO_3{^-}$ contents increased and increased slightly after three weeks. The rise of pH level is believed to be from the $NO_3{^-}$ absorption for immobilization by microbes. In conclusion, the high soil $EC_{1:5}$ level of $12dS\;m^{-1}$ conducted in this experiment did not affect the growth in terms of soil microbes involved in N mineralization of LMC.

Isolation of Alfalfa Nodule Bacteria ana Assessement of their Nitrogen Fixing Capacities (알팔파근류균(根瘤菌)의 분리(分離) 및 질소고정능력(窒素固定能力)의 비교(比較))

  • A series of experiments was planned for practical application of rhizobia in grass lands in Korea. This is the report for the studies mainly on the isolation and characterization of alfalfa nodule bacteria, and on the assessment of their nodulation abilities and nitrogen fixation capacities. 1. Total number of 47 strains was isolated from nodules which were taken from alfalfa grown in Daekwanyong, Cheju and other places. 2. Morphological and cultural characteristics of the strains were studied, and attempts. were also made to investigate their antigenic properties and to demonstrate lysogenic strains. The results were; i) the isolates varied in their cultural characteristics on yeast mannitol broth and agar, and in degree of congo red absorption; ii) similarities in their antigenic prorerties were found between the strains: SU 47/M-11, M-13/M-15, and M-3/M-5; iii) no lysogeny was found in the strains. 3. Plant infection test by test tube method in light room were carried out to elucidate the ability of the strains to nodulate Luna alfalfa and of the capacity of such nodules to fix atmospheric nitrogen. The isolates were grouped info non-invasive, ineffective, or effective to the legumes. Those strains which produced effective nodules, supporting similar/higher level of growth as nitrate control were: M-8, 9, 14, 20, 21, 25 and 34.

Isolation and Characterization of Indole-3-acetic acid- and 1-aminocylopropane-1-carboxylyic Acid Deaminase-producing Bacteria Related to Environmental Stress (환경스트레스와 관련된 indole-3-acetic acid 및 1-aminocylopropane-1-carboxylyic acid deaminase 활성을 갖는 박테리아의 분리와 특성 연구)

  • In this study, strains isolated from soil samples collected from Busan, Changwon, and Jeju Island were examined to verify their abilities of phosphate solubilization and nitrogen fixation, production of indole-3-acetic acid (IAA), siderophore, and 1-aminocylopropane-1-carboxylyic acid (ACC) deaminase in order to select strains that promote plant growth and play a role in biocontrol of pests or pathogens. According to the results of this study, most of the isolated strains were found to have ability of phosphate solubilization, nitrogen fixation, IAA production, siderophore production, and production of ACC deaminase. These isolated strains might help plant growth by directly improving absorption of nutrients essential for phosphate solubilization and nitrogen fixation. In addition, they can promote plant growth and control resistance to plant diseases through extracellular enzyme activity and antifungal activity. In addition, most of the selected strains were found to survive in various environmental conditions such as temperature, salinity, and pH. Therefore, Pseudomonas plecoglossicida ANG14, Pseudarthrobacter equi ANG28, Beijerinckia fluminensis ANG34, and Acinetobacter calcoaceticus ANG35 were finally selected through a comparative advantage analysis to suggest their potential as novel biological agents. Further studies are necessary in order to prove their efficacy as novel biological agents through formulation and optimization of effective microorganisms, their preservation period, and crop cultivation tests.

A Review of Orchid Mycorrhizae in Korea

  • Orchids are evolutionally known to be the most advanced plants in the order Liliales, and comprise approximately 1,000 genera and 35,000 species world-wide. In Korea, more than 110 species of Orchidaceae have been reported to be cultivated or to be collected in the wild. Orchids aye mostly dependant on orchid mycorrhizae(OM) throughout or in part of their life cycle. The OM endomycorrhizae belonging to basidiomycetes or rarley ascomycetes are needed for orchid seed germination. Various fungi, including plant pathogenic, antagonistic and symbiotic fungi, were isolated from the roots of orchid native to Korea. The OM fungi collected from the roots of Cymbidium goeringii were three species of Rhizoctonia namely, R. repens (anamorph state of Tulsanella repens), R. endophytica (Ceratobasidium cornigerum), and an unidentified species (possibly an anamorph of T. calospora). These symbiotic fungi induced peloton in the cortical cells of orchid roots, and differed biologically and in 18s rDNA sequences from plant pathogenic Rhizoctonia species. Also, the mycorrhyzal fungi enhanced the orchid root absorption of nitrogen sources and minerals from the soil. The activity of mycorrhizal fungal hyphae in the roots caused prevention from pathogenic fungi. In nature, the peloton is observed in the cortical cells of Cymbidium goeriingii roots, indicating mycorrhizal colonization in the native orchid roots. On the other hand, pathogenic fungi such as Fusarium and/or Rhizoctonia species are mostly isolated from commercial orchid plants. These suggest that application of symbiotic mycorrhizal fungi should be needed for orchid cultivation in nurseries and at the time of transplanting.

Ab Initio and Experimental Studies on Dibenzothiazyl-Disulfide

  • Ab initio calculations of the structure, atomic charges and natural bond orbital (NBO) have been performed at HF/6-311G** and B3LYP/6-311G** levels for the title compound of dibenzothiazyl-disulfide. The calculated results show that the two nitrogen atoms have the biggest negative charges and they are the potential sites to react with the metallic ions, which make the title compound become a di-dentate ligand. Vibrational frequencies of the title compound have been obtained and compared with the experimental value and the comparison indicates that B3LYP/6-311G** level is better than HF/6-311G** level to predict the vibrational frequencies for the system studied here. For the title compound, electronic absorption spectra calculated by time?ependent density functional theory (TD-DFT) are more accurate than Hartree-Focksingle-excitation CI (CI-Singles) method. NBO analyses show that the electronic transitions are mainly derived from the contribution of bands $\pi\rightarrow\pi^{*}$. Thermodynamic calculated results show that the formation of the title compound from 2-mercaptobenzothiazole is a spontaneous process at room temperature with the change of free Gibbs being negative value.

Isolation and Characteristics of Photosynthetic Bacterium, Erythrobacter longus SY-46 which Produces Bacterial Carotenoids (Bacterial Carotenoids를 생산하는 광합성세균 Erythrobacter longus SY-46의 분리 및 특성)

  • The aerobic photosynthetic bacterium, which produces bacterial carotenoids was isolated and identified from coastal marine environments. This bacterium was identified by 16S rDNA sequencing and designated as Erythrobacter longus SY-46. E. longus SY-46 was Gram negative and rod shape, and the optimal culture conditions were $25^{\circ}C$, pH 7.0, and 3.0% NaCl concentration, respectively. The carbon and nitrogen sources required for the optimal growth were lactose and tryptone, respectively. Fatty acid compositions of E. longus SY-46 were $C_{18:1}$(78.32%), v-linolenic acid($C_{18:3n9.12.15c}:3.83%$), margaric acid($C_{17:0}$: 3.38%), palmitic acid($C_{16:0}$: 3.07%), and docosahexaenoic acid($C_{22:6n3}$: 2.21%). In addition, E. longus SY-46 showed the characteristic absorption peaks of bacterial carotenoids(in the region of 450 to 480 nm) and bacteriochlorophyll(770 to 772 nm). Major carotenoids of E. longus SY-46 were polyhydroxylated xanthophylls such as fucoxanthin and zeaxanthin.

Method for nutrient solution extraction from used diposed diapers (일회용 폐기저귀에서 양액 추출 방안)

  • Used disposable diapers have been considered for a long time as a type of waste difficult to recycle and valorize due to their composite nature including plastic, cellulose pulp, a super absorbent polymer and either urine, feces or both. Therefore, the fate of disposed diapers often is either incineration or landfill burial which both have various adverse environmental impacts. However, used disposable diapers contain nutrients: cellulose is an organic matter while urine and feces contain non negligible amounts of nitrogen, phosphorus and potassium which are primary nutrients included in most chemical fertilizers used in agriculture. In a scope of waste recycling and valorization, this study focuses on developing a method to achieve nutrient solution extraction from used disposable diapers. The experiment essentially consists in shredding the diapers and letting them macerate in solutions of sodium hydroxide with various concentrations to allow breaking down of the cellulose and super absorbent polymer and release of urine and feces before sterilizing the solutions in an autoclave to remove potential coliform bacteria. At the end of the experiment, a set of parameters is measured for the final solution to identify concentrations of nutrients as well as presence or absence of harmful substances. Results are discussed and directions for future studies are suggested, which include mechanization of the diapers shredding process or added aeration to enhance nitrification and absorption of extracted nutrients from plants.
